Infections caused by parasites in humans represent one of the main public health concerns. Amoebiasis, a parasitic infection caused byEntamoeba histolytica(E. histolytica), is considered endemic in Mexico, whereArgemone mexicana(A. mexicana) has been used in traditional medicine to treat intestinal parasitic diseases. The objective of this work was to evaluate the potential biological activity ofA. mexicanaonE. histolytica. For this purpose, a methanolic extract was prepared fromA. mexicanaleaves, and a differential fractionation was carried out with solvents of different polarities. The inhibitory capacities of the extract and its fractions were evaluated in vitro using HM1-IMSS, a strain ofEntamoeba histolytica.A. mexicanaextract was found to have a growth-inhibiting activity forE. histolytica, showing IC50 = 78.39 μg/mL. The extract was characterized phytochemically, and the methanolic extract fractions were analyzed by liquid chromatography (HPLC) and mass spectrometry (MS). Berberine and jatrorrhizine were present in the active fractions, and these compounds may be responsible for the antiparasitic activity. The identification of amoebicidal activity ofA. mexicanaonE. histolyticagives support to the traditional use. Further studies with berberine and jatrorrhizine will be carried out to understand the mechanism involved.

Infections caused by <i>Trichomonas vaginalis</i> in humans are one of the main public health problems caused by sexually transmitted diseases. Objective of this study was to evaluate potential biological activity of the medicinal plant <i>Argemone mexicana</i> (Mexican poppy) on <i>T. vaginalis</i>. Methanolic extracts of the stems and leaves of <i>A. mexicana</i>, and different fractions were prepared with solvents of different polarities. The extracts and functional groups were detected containing sterols, triterpenes, quinones, flavonoids and, alkaloids. Extracts from both the stems and leaves of <i>A. mexicana</i> inhibited the growth of <i>T. vaginalis</i> with half-maximal inhibitory concentration value of 70.6 and 67.2 μg/ml, respectively. In the active fractions, the most abundant compounds were berberine and jatrorrhizine, with presumed antiparasitic activity.

Genetically uniform plant material, derived from Lychnis flos-cuculi propagated in vitro, was used for the isolation of 20-hydroxyecdysone and polypodine B and subjected to an evaluation of the antifungal and antiamoebic activity. The activity of 80% aqueous methanolic extracts, their fractions, and isolated ecdysteroids were studied against pathogenic Acanthamoeba castellani. Additionally, a Microtox® acute toxicity assay was performed. It was found that an 80% methanolic fraction of root extract exerts the most potent amoebicidal activity at IC50 of 0.06 mg/mL at the 3rd day of treatment. Both ecdysteroids show comparable activity at IC50 of 0.07 mg/mL. The acute toxicity of 80% fractions at similar concentrations is significantly higher than that of 40% fractions. Crude extracts exhibited moderate antifungal activity, with a minimum inhibitory concentration (MIC) within the range of 1.25–2.5 mg/mL. To the best of our knowledge, the present report is the first to show the biological activity of L. flos-cuculi in terms of the antifungal and antiamoebic activities and acute toxicity. It is also the first isolation of the main ecdysteroids from L. flos-cuculi micropropagated, ecdysteroid-rich plant material.

Giardia lamblia es el protozoario parásito causante de la giardiasis, la cual se caracteriza por molestia abdominal, pérdida de peso y desnutrición. La droga de elección para su tratamiento es el metronidazol sin embargo, presenta diversos efectos secundarios adversos en el paciente. Por otra parte, en la herbolaria se conoce a la semilla de Cucurbita pepo por sus propiedades desparasitante, principalmente sobre Entamoeba histolytica y Taenia solium.  Por lo anterior se planteó la hipótesis de que las semillas de C. pepo contienen principios activos capaces de inhibir el crecimiento de G. lamblia bajo condiciones axénicas in vitro. El objetivo de esta investigación fue evaluar la actividad biológica del extracto acuoso y metanólico de las semillas de  C. pepo sobre el crecimiento de G. lamblia.  Tanto el extracto metanólico como el acuoso se identificaron siete grupos funcionales y la actividad antioxidante no fue significativa. En el extracto metanólico se observó mayor efecto giardicida seguido del acuoso, esta diferencia puede deberse a las saponinas que se encuentran sólo en el extracto metanólico. Los resultados sugieren que las semillas contienen principios activos que pueden emplearse para la investigación de nuevos tratamientos para la  giardiasis.

Amebiasis caused by Entamoeba histolytica is nowadays a serious public health problem worldwide, especially in developing countries. Annually, up to 100,000 deaths occur across the world. Due to the resistance that pathogenic protozoa exhibit against commercial antiprotozoal drugs, a growing emphasis has been placed on plants used in traditional medicine to discover new antiparasitics. Previously, we reported the in vitro antiamoebic activity of a methanolic extract of Lippia graveolens Kunth (Mexican oregano). In this study, we outline the isolation and structure elucidation of antiamoebic compounds occurring in this plant. The subsequent work-up of this methanol extract by bioguided isolation using several chromatographic techniques yielded the flavonoids pinocembrin (1), sakuranetin (2), cirsimaritin (3), and naringenin (4). Structural elucidation of the isolated compounds was achieved by spectroscopic/spectrometric analyses and comparing literature data. These compounds revealed significant antiprotozoal activity against E. histolytica trophozoites using in vitro tests, showing a 50% inhibitory concentration (IC50) ranging from 28 to 154 µg/mL. Amebicide activity of sakuranetin and cirsimaritin is reported for the first time in this study. These research data may help to corroborate the use of this plant in traditional Mexican medicine for the treatment of dyspepsia.

 Objective: The objective of the present work is to study the in vitro anti-inflammatory and antioxidant activity of medicinal plants. The extent and correlation between anti-inflammatory and antioxidant activity have been studied.Method: Methanolic and aqueous extracts of five medicinal plants, namely, Ficus racemosa, Aloe vera, Cannabis sativa, Datura stramonium, and Calotropis gigantean have been taken for in vitro anti-inflammatory and total antioxidant activity.Result: The study showed that the inhibition of protein (albumin) denaturation was maximum in aqueous extract of A. vera with 97.55±1.45%. Proteinase inhibitory action of different plant extracts showed significant action and was found to be maximum in aqueous extract of D. stramonium with 87.89±2.58%. Heat-induced hemolysis showed that maximum inhibition was with aqueous extract of F. racemosa with 90.72±3.33%. When hypotonicity-induced hemolysis activity was done it was found maximum in methanolic extract of C. gigantea with 90.58±3.04%. Anti-lipoxygenase activity was found maximum in methanolic extract of F. racemosa with 94.05±4.24%. When total antioxidant activity was done, it was found highest in F. racemosa (4.38±0.546 mM equivalent of ascorbic acid/g tissue).Conclusion: An overall strong positive correlation between anti-inflammatory and antioxidant activity was observed, indicating that antioxidant activity of the plant species studied might be responsible for their anti-inflammatory property. Further work needs to be undertaken to fully elucidate the antioxidants responsible for anti-inflammatory action and to develop better herbal drug formulations.

Objective: Evaluation of beneficial effects of the seed of Linum usitatissimum in congestive heart failure.Methods: Methanolic extract of seeds of Linumusitatissimum (MELU) was prepared using soxhlet apparatus and oil of seed of Linumusitatissimum (OLU) was isolated using Clevenger apparatus. The positive inotropic action of methanolic extract of seeds of Linum usitatissimum was evaluated using Langendorff’s assembly (in vitro study). Beneficial effects of methanolic extract and oil of seeds of Linum usitatissimum were carried out by doxorubicin (1 mg/kg, i. p. within 3 w) to induce congestive heart failure (in vivo study). Parameters like electrocardiogram (ECG) recording and cytosolic Ca2+level and histopathology of the heart were carried out. In same study diuretic action was evaluated using Lipschitz model.Results: Methanolic extract of seeds of Linum usitatissimum showed significantly increased in positive inotropic effect (force of contraction 48.8±1.53 mm) as compared to control group (force of contraction 17.5±0.76 mm) on Langendorff”s study (in vitro study). In doxorubicin-induced congestive heart failure both MELU and OLU showed significant decreased QT (Note: In cardiology, the QT interval is a measure of the time between the start of the Q wave and the end of the T wave in the heart's electrical cycle. There is no full form for this medical word) interval. The histopathologic study indicated the least damage to the architecture of myocardial membrane. MELU and OLU increased urine output (5.66±0.16 ml and 6.58±0.15 ml respectively) significantly in Lipschitz model as compared to disease control group (4.58±0.15 ml).Conclusion: Present research work emphasizes that the seeds of Linum usitatissimum is beneficial in the management of congestive heart failure because of having positive inotropic effect, diuretic activity and control of rhythmicity of heart.

Based on the collected ethnobotanical data from the Traditional Medicine and Materia Medica Research Center (TMRC), Iran,Myrtus communisL. (myrtle) was selected for the assessment ofin vitroandin vivoantimalarial and cytotoxic activities. Methanolic extract of myrtle was prepared from the aerial parts and assessed for antiplasmodial activity, using the parasite lactate dehydrogenase (pLDH) assay against chloroquine-resistant (K1) and chloroquine-sensitive (3D7) strains ofPlasmodium falciparum. The 4-day suppressive test was employed to determine the parasitemia suppression of the myrtle extract againstP. berghei  in vivo. The IC50values of myrtle extract were 35.44 µg/ml against K1 and 0.87 µg/ml against 3D7. Myrtle extract showed a significant suppression of parasitaemia (84.8 ± 1.1% at 10 mg/kg/day) in mice infected withP. bergheiafter 4 days of treatment. Cytotoxic activity was carried out against mammalian cell lines using methyl thiazol tetrazolium (MTT) assay. No cytotoxic effect on mammalian cell lines up to 100 µg/mL was shown. The results support the traditional use of myrtle in malaria. Phytochemical investigation and understanding the mechanism of action would be in our upcoming project.

Aims: This study was carried out to investigate the antimicrobial activity of Chrysophyllum albidum leaves extract on selected Gastro-instestinal bacteria such as Salmonella typhimurium, Shigella dysentariae, Vibrio cholera, Escherichia coli and Clostridium perfringens. Methodology: The leaves were extracted using ethanol, methanol and distilled water; the concentration of the extracts employed were 100 mg/ml, 200 mg/ml, 400 mg/ml and 500 mg/ml respectively; however the   leaf extracts of Chrysophyllum albidum were screened for anti-microbial activity using the in vitro cup-plate method of agar diffusion technique with concentration of 10-5cells/ml of the selected bacteria. Simultaneously, 30 µg tetracycline and 30 µg metronidazole were used as positive control. Results: The result showed that the most active among them is Tetracycline; followed by ethanolic extract, aqueous extract, methanolic extract and metronidazole extract respectively on the tested bacteria. Conclusion: This research justifies the traditional use of the leaves of Chrysophyllum albidum for the therapeutic purposes; hence can be commercialized by pharmaceutical outfit; if not for anything but its availability and readily for human consumption.
