Under present experiments, EPS and IPS production, monosaccharide composition, and antioxidant activities ofC. graciliswere studied for the first time under submerged culture conditions. Effect of different factors on polysaccharides production was studied by orthogonal experiments using one-factor-at-a-time method. Incubation of culture in the medium with capacity 200 mL (675.12 ± 5.01 and 385.20 ± 5.01 mg/L), rotation speed 150 rpm (324.62 ± 3.32 and 254.62 ± 4.62 mg/L), 6-day culture incubation time (445.24 ± 1.11, 216.60 ± 1.71 mg/L), pH 6.0 (374.81 ± 2.52 and 219.45 ± 2.59 mg/L), and temperature 23°C (405.24 ± 1.11 and 215.60 ± 1.71 mg/L) produced higher EPS and IPS, respectively. Maximum EPS and IPS production was observed in the medium supplemented with glucose as a carbon source (464.82 ± 2.12 and 264.42 ± 2.62 mg/L) and yeast extract as a nitrogen source (465.21 ± 3.11 and 245.17 ± 3.24 mg/L), respectively. Carbon to nitrogen ratio for maximum EPS and IPS production was observed as 10 : 1 (395.29 ± 2.15 and 235.62 ± 1.40 mg/L), respectively. Glucose was found to be the major monosaccharide (62.15 ± 7.33%). Both EPS and IPS ofC. gracilisshowed significant DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power, and iron chelating activity.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

Background: Cataract is a major cause of visual impairment and blindness around the world. This study evaluated the in vitro antioxidant and anti-cataract activities of Cnidoscolus aconitifolius leaves extract and fractions. Antioxidant activities were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azinobis (3-ethylbenzothiozoline-6-sulfonic acid) (ABTS), total reducing power, and hydrogen peroxide scavenging assays. Anti-cataract potential was evaluated in vitro using goat lenses divided into eight groups of different treatments and incubated in artificial aqueous humor at 37 °C for 72 hours. Glucose-induced opacity in the lenses was observed and biochemical indices quantified (catalase, Malondialdehyde (MDA) and total protein in the lens homogenate). Results: The crude extract and its fractions possess substantial antioxidant activities. The aqueous fraction exhibited the best DPPH radical scavenging activity (IC50 value 78.599 µg/ml); while the dichloromethane fraction exhibited the highest ABTS radical scavenging activity with IC50 66.68 µg/ml. The anti-cataract evaluation of crude and fractions at 250 μg/ml showed a significant increase (p<0.05) in the total protein and catalase activity compared to the cataract control group. The malonaldehyde level decreased significantly (p<0.05) in all the treated groups.

Pulverized rhizome of Curcuma peliolata on hydrodistillation, afforded light yellow essential oil in the yield of 0.13% v/w. The chemical constituent of the essential oil was determined by GC-MS. Nine compounds representing 95.34 % of the total oil were identified. The major compounds of the oil were 2-methyl-5-pentanol, 1H-pyrrol-1-amine,2-(4-methoxyphenyl)-n,n,5-trimethyl, and curcumol. The distillated aqueous was extracted with dicloromethane (DCM), ethyl acetate (EtOAc), and n-butanol. The DPPH radical scavenging and reducing power of the extracts were investigated. Curcumin and ascorbic acid were used as standard references. The DPPH radical scavenging activity was decrease in order: curcumin ascorbic acid EtOAc extract > DCM extract > n-butanol extract > aqueous extract. The EtOAc extract showed significantly highest inhibition of the radical scavenging activity with IC50 0.092 ± 0.001 mg/mL (curcumin and ascorbic acid (IC50 0.012 and 0.013 mg/mL, respectively (P<0.05)). While, the DCM extract showed highest reducing power with IC50 = 0.271 mg/mL which lower than that of standard curcumin (IC50 = 0.013 mg/mL) and ascorbic acid (IC50 = 0.002 mg/mL).Introduction

The study was designed to investigate the antioxidant activities of the methanolic extract of leaves of Begonia roxburghii by five different in vitro methods namely Folin-Ciocalteu, total flavonoid content, phosphomolybdenum, total reducing power determination, and DPPH scavenging activity. Phenol content (Gallic acid equivalent) was found as 180.40±0.03 mg/g of the dry sample by the Folin-Ciocalteu method. Flavonoid assay method revealed the content of flavonoids (Quercetin equivalent) as 60.43±0.27 mg/g of the dry sample. Total antioxidant capacity of the extract was observed 243.43±0.11 mg/g ( ascorbic acid equivalent) by phosphomolybdenum method. Free radical scavenging activity (measured by the DPPH test) of the methanolic extract was found 158.36±0.22 μg/mL whereas ascorbic acid, the standard, showed 19.53±0.04 μg/mL. The methanolic extract of Begonia roxburghii leaves exhibited positive and linear dose dependent reducing power activity and its activity was comparable to the standard ascorbic acid for a given dose. Phytochemical screening of the extract showed positive results for alkaloids, carbohydrates, flavonoids, glycosides, saponins, steroids, and tannins, which demonstrate definite pharmacological actions on human body. Callus cultures from stems, leaves and roots of colocynth were initiated on MS media supplemented with various combinations of 2,4 dichlorophenoxyacetic acid (2,4-D) with kinetin (KIN) and benzyladenine (BA) with α-naphthaleneacetic acid (NAA). The highest percentage of callus formation frequency (98.9%) was obtained from stem explants grown on MS media supplemented with (1.0 mg/L) 2,4-D + (1.0 mg/L) KIN. The total phenolics and flavonoid content of the colocynth callus cultures were measured. The results showed that the MS medium supplemented with 6.0 mg/L 2,4-D + 2.0 mg/L KIN (MD3) gave the highest content of total phenolics (19.2 mg/100g d.w.) in leaf-derived calli. The highest content of flavonoids (47.3 mg/100g d.w.) was obtained in stem derived calli grown on the same medium (MD3). Antioxidant activities of extracts were determined using different assays, including DPPH radical scavenging activity, hydrogen peroxide (H2O2) scavenging activity and ferric reducing power. Leaf-derived calli cultured on MS medium + 2.0 mg/L 2,4-D + 1.0 mg/L KIN (MD1) showed the highest DPPH radical scavenging activity (85.3%). The highest percentage of H2O2 scavenging activity (61.4%) was detected in leaf explant-derived calli growing on MD1. The leaf-derived calli growing on (MD3) gave the highest ferric reducing power (22.3 μg/g d.w.), compared to the activities of stems, leaves and roots of in vitro grown seedlings (3.28, 12.9 and 2.85 μg/g d.w.), which were used as controls. On the basis of the current findings, we conclude that MS media supplemented with different combinations of 2,4-D and KIN yields higher phenolics, flavonoids contents and antioxidant activities than MS media supplemented with BA and NAA.

Heterogeneous method for grafting of syringaldehyde (SD) onto chitosan by laccase oxidation, yielding a product with improved antioxidant activities. The results showed that about 1.1% amino groups participated in this reaction. The FTIR showed that the absorption of amino groups were decreased, the absorption of benzene ring structures were disappeared and a new band appeared at 1637 cm-1 corresponding to C=N vibrations that were due to the Schiff base reaction between the chitosan and syringaldehyde. Additionally, the chitosan had insignificant reducing power and DPPH free radical scavenging activity, SD-chitosan had reducing power of A700=0.684, DPPH free radical scavenging activity of 53.2%. SD-chitosan presented improved antioxidant properties.

AbstractIn the present study, the objective was to evaluate the effects of barley kernel size, grind level, and extract solvent on the antioxidant activities associated with total phenolic content and phenolic acid compositions. Three barley varieties (Kindred, Azure, and Tradition), were used and the results showed that with the exception of superoxide anion radical scavenging activity, thin kernel size fraction (2.0 mm) showed much higher antioxidant activities and total phenolic content than those in the 2.4 mm and 2.8 mm fractions when averaged across barley cultivar, grind level, and extract solvent. A similar trend was found for individual phenolic acid compositions among kernel sizes. Simple correlation analysis revealed that total phenolic content showed strong correlation with DPPH· scavenging activity, ABTS + scavenging activity, and reducing power. Generally, fine grind and 80% acetone extract showed averaged highest values in antioxidant activities, total phenolic content, and individual phenolic acid compositions. Stepwise linear regression showed that extract solvent was the most important factor for DPPH· scavenging activity, ABTS + scavenging activity, reducing power, and iron chelating activity. The sum of syringic acid and caffeic acid exhibited a dominant role in explaining the major variation in antioxidant activities except for superoxide anion radical scavenging activity.

Six purple corn grains were chosen to investigate their antioxidant activities and the relationships between anthocyanins and phenolics contents with antioxidant activities. Purple grains were screened for their antioxidant potentials using various in-vitro models such as superoxide anion radical scavenging activity and reducing power at various concentrations. The result showed that ZS39 and FS11 exhibited the highest anthocyanins and total phenolics contents. ZS39 and FS11 also exhibited the highest superoxide anion radical scavenging activity. There was no significant difference among the reducing power of six different purple corn grain extracts. It was found that there were positive correlations between anthocyanins and superoxide anion radical scavenging activity or reducing power, R2=0.9911, R2=0.9873, respectively. Whereas the correlations between total phenolics and superoxide anion radical scavenging activity or reducing power, R2=0.9863, R2=0.9826, respectively. It suggested that purple corn appeared to be a rich and interesting source of natural antioxidants.

Abstract Background Mushrooms have been recognized both as medicine and nutritional food in many countries for thousands of years, owing to the presence of significant amounts of carbohydrates, proteins, lipids, fibers, and antioxidants. This study aims at evaluating the nutritional contents and antioxidant potentials of seven types of mushrooms cultivated in Bangladesh. Methods Proximate composition analysis of the collected mushrooms was performed to determine moisture content, pH, lipid, crude fibre, total ash, protein, carbohydrate content, and calorific value. Antioxidant potential of collected mushrooms were evaluated by mean of eight different methods including total flavonoid content, phenolic content, tannin content, total antioxidant activity, hydroxyl radical scavenging activity, DPPH assay and reducing power capacity. Results The results demonstrated that investigated mushrooms were found rich in proteins (20–45 g/100 g), carbohydrate (11–61 g/100 g in dry sample) and fibre (5–40 g/100 g). The ash content was found 6–10 g/100 g and glucose content 54–160 mg/100 g. However, all the mushrooms showed a lower content of lipid (1–4%). Results also revealed that the total antioxidant capacity of the extracts were found in the concentration range of 0.08–0.21 mg/mL, whereas the hydroxyl radical and DPPH radical scavenging activity were 0.88–1.40 and 0.05–0.63 mg/mL. Conclusion The findings of the current investigation proved that the studied Bangladeshi mushrooms are good source of nutritional and antioxidant components. Therefore, this study can help spreading awareness among Bangladeshi people regarding consumption of mushrooms as functional foods.

In this study, antioxidant activities of H. fursei were investigated. Different model were used for assay. IC50 for DPPH radical-scavenging activity was 7.5±1.2 μg ml-1. The IC50 value for BHA was 92.9±4.5 µg ml-1. The extract showed very good reducing power that was comparable with vitamin C (p> 0.05) but showed weak nitric oxide scavenging activity and Fe2+ chelating ability. Extract showed good activity in scavenging of H2O2. Inhibition was 99.1% at 800 μg ml-1. The IC50 values for extract and BHA were 333.1±14.7 and 52.0±4.5 μg ml-1, respectively. Total phenol compounds, by the Folin Ciocalteu method, was 274±9.6 mg gallic acid equivalent/g of extract and the total flavonoid content, by AlCl3 method was 23.9 ± 1.4 mg quercetin equivalent/g of extract. Antioxidant activity may be attributed to the presence of phenols and flavonoids in the extract.
