The novel highly sulfated (35%) fucoidan fractionCf2, which contained, along with fucose, galactose and traces of xylose and uronic acids was purified from the brown algaCoccophora langsdorfii. Its structural features were predominantly determined (in comparison with fragments of known structure) by a rapid mass spectrometric investigation of the low-molecular-weight fragments, obtained by “mild” (5 mg/mL) and “exhaustive” (maximal concentration) autohydrolysis. Tandem matrix-assisted laser desorption/ionization mass spectra (MALDI-TOF/TOFMS) of fucooligosaccharides with even degree of polymerization (DP), obtained by “mild” autohydrolysis, were the same as that observed for fucoidan fromFucus evanescens, which have a backbone of alternating (1 → 3)- and (1 → 4) linked sulfated at C-2 and sometimes at C-4 of 3-linkedα-L-Fucpresidues. Fragmentation patterns of oligosaccharides with odd DP indicated sulfation at C-2 and at C-4 of (1 → 3) linkedα-L-Fucpresidues on the reducing terminus. Minor sulfation at C-3 was also suggested. The “exhaustive” autohydrolysis allowed us to observe the “mixed” oligosaccharides, built up of fucose/xylose and fucose/galactose. Xylose residues were found to occupy both the reducing and nonreducing termini of FucXyl disaccharides. Nonreducing galactose residues as part of GalFuc disaccharides were found to be linked, possibly, by 2-type of linkage to fucose residues and were found to be sulfated, most likely, at position C-2.

We describe the gas chromatography/mass spectrometric (GC/MS) analysis of O-alkyl methylphosphinates (AMPs), which are included in Schedule 2B4 chemicals in the Chemical Weapons Convention (CWC). GC/MS analysis of a variety of AMPs and their deuterated analogs revealed that their fragmentations were determined by α-cleavages, McLafferty +1 and hydrogen rearrangement. Based on the obtained electron ionization mass spectra of AMPs, the fragmentation routes were rationalized, which were substantiated by the GC/MS analysis of deuterated analogs.

Peptide-based small molecule drug conjugates for targeted tumor therapy are currently in the focus of intensive research. Anthracyclines, like daunomycin, are commonly used anticancer drug molecules and are also often applied in peptide-drug conjugates. However, lability of the O-glycosidic bond during electrospray ionization mass spectrometric analysis hinders the analytical characterization of the constructs. “Overprotonation” can occur if daunomycin is linked to positively charged peptide carriers, like tuftsin derivatives. In these molecules, the high number of positive charges enhances the in-source fragmentation significantly, leading to complex mass spectra composed of mainly fragment ions. Therefore, we investigated different novel tuftsin-daunomycin conjugates to find an appropriate condition for mass spectrometric detection. Our results showed that shifting the charge states to lower charges helped to keep ions intact. In this way, a clear spectrum could be obtained containing intact protonated molecules only. Shifting of the protonation states to lower charges could be achieved with the use of appropriate neutral volatile buffers and with tuning the ion source parameters.

Abstract The diagnosis of bladder cancer (BC) is currently based on cystoscopy, which is invasive and expensive. Here, we described a non-invasive, low-cost BC diagnosis method based on a desorption, separation, and ionization mass spectrometry platform (DSI-MS) that adopts N, N- Dimethylethylenediamine (DMED) as a differential labeling reagent. The DSI-MS platform avoids the interferences from intra- and/or inter-samples, while the DMED increases detection sensitivity and distinguishes carboxyl, aldehyde, and ketone groups from untreated samples. Carbonyl metabolic fingerprints of urine from 28 BC patients and 38 controls were portrayed and significant differences of some potential biomarkers were observed. The mechanisms of the changes have been discussed. Logistic regression (LR) was applied to discriminate BC from controls and an accuracy of 87% was achieved. We believe this patient-friendly method provides a hopeful approach for BC rapid point-of-care diagnostic.
