Cisplatin (CP), an antitumor agent, has been shown to cause ovarian injury and dysfunction in both animal and human studies. The present study was conducted to investigate the protective effect of oxytocin (OT) on CP-induced ovarian toxicity in rats. Twenty-one adult female rats were included in the study. Fourteen rats were administered intraperitoneally CP (2 mg/kg/day) twice a week for 5 weeks. Control group (n=7) did not receive any treatment. Following treatment, CP-received rats were randomly divided into two groups and treated with either saline (1 mL/kg/day,n=7) or OT (160 μg/kg/day,n=7) for 5 weeks. Then, ovarian toxicity and effects of OT were evaluated by histomorphological and biochemical analysis. Our findings revealed a significant reduction in the number of follicles at each grade in saline-treated group. AMH level was significantly lower in saline group compared to control (P<0.0005). OT treatment significantly attenuated CP toxicity in ovaries and increased AMH levels compared to saline group (P<0.005). Also, administration of OT lessened lipid peroxidation and prevented glutathione depletion in CP-treated rats (P<0.05). These results indicated that OT could lessen the CP-induced ovarian damage and improve follicular reserve by preventing oxidative damage.

<p class="Abstract">The aim of this study was to evaluate the diuretic activity of <em>Eysenhardtia polystachya</em> bark aqueous extract at different doses in a rat model. Different doses of <em>E. polystachya</em> (125, 250, 500 and 750 mg/kg body weight), furosemide (4 mg/kg) and vehicle were administered per os to female rats (n=6 animals per group). After 6 hours in metabolic cages, the effect on urinary flow, glomerular filtration rate and electrolyte balance of sodium and potassium were assessed in all animals. <em>E. polystachya</em> at the doses of 500 and 750 mg/kg induced diuretic activity, since markedly increased (p&lt;0.05) the urinary flow rate, similar to that of furosemide treated group. Only the dose of 750 mg/kg produced an increment in urinary excretion of sodium but not of potassium compared with control group. These findings indicate that<em> E. polystachya</em> bark-induced diuretic activity, providing evidence for its folkloric use.</p><p> </p>

Abstract Background: Premature ovarian failure is among the most important side effects of chemotherapy during reproductive period. Preserving ovarian function is gradually gaining importance during oncologic treatment. The present study aims to investigate the potential of melatonin to protect from cisplatin-induced ovarian toxicity in rats. Twenty nine female rats were divided to three groups: Saline control group (Group 1), cisplatin group (Group 2), and cisplatin+melatonin group (Group 3). While the rats in Groups 2 and 3 were administered 5 mg/kg single dose of cisplatin via intra-peritoneal (IP) route, the rats in Group 3 were started on melatonin (20 mg/kg IP) before cisplatin administration and continued during 3 consecutive days. Ovaries were removed one week after cisplatin administration in all groups. Blood samples were obtained before the rats were decapited. Histological evaluation, follicle count, and classification were performed. TAp63 mRNA expression was evaluated using mRNA extraction and real-time polymerase chain reaction (PCR) method. Serum estradiol (E2) and anti-mullerian hormone (AMH) values were measured with enzyme immune-assay technology. Results: While primordial follicles were seen to decrease in Group 2 as compared to Group 1 (p:0.023), primordial follicle count was observed to be preserved significantly in melatonin group as compared to Group 2 (p:0.047). Moreover, cisplatin-induced histo-pathological morphology was preserved in favor of normal histology in melatonin group. A significant difference was not observed between groups with regard to mean serum AMH and E2 values (p:0.102 and p:0.411, respectively). While TAp63 gene expression significantly increased in Group 2 as compared to control group (p:0.001), we did not detect a statistically significant difference in cisplatin+melatonin group, although gene expression decreased (p:0.34). Conclusion: We conclude that concurrent administration of melatonin and cisplatin may protect from ovarian damage.

Abstract The biomechanical and biochemical properties of aortas from female rats treated with biosynthetic human GH (b-hGH) for 80 days were investigated. b-hGH was administered at a dose of 5 mg·kg−1·d−1. Treatment with b-hGH increased the body weight by 75% and the diameter of the aorta by 14% compared with the control group. The concentration of collagen and the relative amount of collagen type I were increased, and the concentration of elastin was decreased. Aortas from the b-hGH-treated group showed increased extensibility in the regions corresponding to physiological load values (i.e. 100-200 mmHg), and increased stiffness in regions with higher load values. The increased extensibility at low load values corresponds well with the loss of elastin, and the increased stiffness at higher load values with the increase of collagen and relative increase of collagen type I. These alterations induced by the growth hormone treatment might influence the elasticity and recoiling properties of the aorta.

Objectives: Oxaliplatin causes hematological toxicities in clinical setting whichlimits its efficacy. The aim of this study is to investigate the therapeutic effects of Andrographispaniculata against hematological toxicity caused by oxaliplatin. Study design: Experimentalanimal study. Period: Study takes 8 month from March 2015 to Oct 2015. Setting: Dow universityanimal house. Method: Wistar albino male rats, divided into 3 equals groups (n=6): GroupN* was a control group (0.9% normal saline), Group NP0 was Oxaliplatin treated group andGroup NP1 was prophylactically treated with Andrographis paniculata followed by Oxaliplatinin order to assess the protective effects of Andrographis paniculata against the hematologictoxicity caused by Oxaliplatin. Results: Prophylactic treatment with Andrographis paniculata(NP1) significantly increases the levels of platelets and neutrophile count compared with thestandard (NP0) (p<0.01) and increases the RBCs count and levels of hemoglobin comparedwith the standard (NP0). Conclusion: Prophylactic treatment with Andrographis paniculata(NP1) was effective in reducing risk of thrombocytopenia, anemia and neutropenia associatedwith Oxaliplatin.

Cadmium (Cd) present in the air, drinking water and food has the potential to affect the health of people, mainly those who live in highly industrialized regions. Cd affects placental function, may cross the placental barrier and directly modify fetal development. It is also excreted into milk. The body is particularly susceptible to Cd exposure during perinatal period. The effect on rat oral epithelium (floor of the mouth) after continuous exposure to drinking water containing low levels of Cd during lactation was studied. Female rats were supplied with ad libitum drinking water containing 300 mg/l of CdCl2 throughout the whole lactation period. Control animals received a similar volume of water without Cd. Lactating rats (21 day-old) were killed by lethal dose of anesthetic. The heads were retrieved, fixed in "alfac" solution (alcohol, acetic acid and formaldehyde) for 24 h, serially sectioned in frontal plane, at the level of the first molars. The 6 µm sections were then stained with hematoxylin and eosin. Nuclear epithelium parameters were estimated, as well as cytoplasm and cell volume, nucleus/cytoplasm ratio, numeric and surface densities, and epithelial thickness. Mean body weight was 34.86 g for the control group and 18.56 g for the Cd-treated group. Histologically, the floor of the mouth epithelium was thinner in the treated group, with smaller and more numerous cells. In this experiment, Cd induced epithelial hypotrophy, indicating a direct action in oral mucosa cells, besides retarded development of the pups.

ABSTRACT Background and objective Polycystic ovary syndrome (PCOS) can be induced in Wistar rats by over production of nitric oxide (NO). This study evaluated the efficacy of naloxone on the breeding characteristics of rats suffering from nitric oxide induced PCOS. Materials and methods Twenty-four female Wistar rats(200–250 gm) were kept as virgin under standard conditions. They were divided into four groups (n = 6). One group of the animals received L-arginine (50 mg/kg) intraperitoneally (i.p.) for 9 days/once a day. Another group was administered naloxone hydrochloride (0.4 mg/kg, i.p.) prior to injection of L-arginine. The third group was injected solely naloxone. Control group received saline solution (1 ml/kg, i.p.). After the treatments, all female rats were coupled with the intact males. They were then separated by observation of vaginal plaques; it was considered as day 0 of pregnancy. Eventually, they were operated on days 18 to 19 of the gestation to collect the animals’ ovaries. The samples were studied for pathological evidence. The fetal number and weight along with the fetal crown-rump length (CRL) were measured. Results The ovaries obtained from the L-arginine treated group had large cysts with thickened granulosa cell layer in contrast to those of the control or naloxone treated rats (p < 0.0001). The number of fetus though showed a decrease in the L-arginine treated rats (3 ± 1), but the fetal weight or fetal CRL did not change (p > 0.05). Conclusion This study may clearly illustrate the polycystic characteristics in the L-arginine treated group. It may particularly display the breeding efficacy of naloxone in rats with PCOS. The mortality of individuals suffering from depression has been increasing, noticeably of postmenopausal women; consequently, their care and treatment are significant to retain a high quality of life. The aim of this study was to examine the effect of Camellia sinensis (CS) on repeated stress-induced changes of the depression related function on the tail suspension test (TST), forced swimming test (FST) in ovariectomized female rats. After behavioral test, we evaluated the changes in the neurotransmitter by measuring the level of dopamine in the nucleus accumbens (NaC) and the serum levels of estrogen and oxytocin. We used 18F-2-fluoro-deoxy-D-glucose positron emission tomography (18F-FDG-PET) to examine the effects of CS on glucose metabolism in ovariectomized rats. Female rats were randomly segregated into three groups. Nor group was considered as nonoperated and nonstressed group, while the control was the ovariectomized and stressed group (OVX+ST), and CS was the ovariectomized, stressed and CS treated group. The rats were exposed to immobilization stress (IMO) for 14 d (2 h/d), and CS (300 mg/kg, i.p.) was treated 30 min before IMO stress. Significant reduction of immobility in the TST and FST was indicated in rats treatment with CS compared to the control group (OVX+ST). The levels of estrogen in the serum of the Nor and CS groups were significantly elevated compared to the OVX+ST group. Also, CS activated brain glucose metabolism in the cortex. The present findings suggested that CS had antidepressant effectiveness in a menopausal depression animal model. These findings suggest evidence that CS plays a crucial role in stressful situation, providing that CS might be a dependable antidepressant medicine to treat menopausal depression.

The experiment was conducted on “Cobb-500” broiler chicks to study the effects of Curcumazedoaria (shoti) on growth performances and haemato-biochemical parameters. A total of twentybroilers chicks (16 days old) were randomly divided into five equal groups (n=4). Three groups ofbroilers fed shoti powder at the rate of 10%; 20%; 30% (w/w) with other ingredients for 22 days.Broilers chicks fed on maize-soybean were considered as positive control while non-protein controlwas regarded as negative control. The results showed that body weight of broilers fed 10% and 20%shoti meal grew very close to those of control (maize-soybean) group. After having completed thefeeding trial, the birds were sacrificed to collect blood sample for and biochemical analysis .Weightsof different organs and length of intestine were also measured. Results showed that decreaseddressed weight, weight of skin and liver were found in 30% shoti treated group compared to proteincontrol group. Intestinal weight was highest in 30% shoti treated group but length was similar to theprotein control group. Total erythrocytes count, hemoglobin content and PCV value were founddecreased in shoti treated group compared to protein control group but ESR value was highest in10% shoti treated group. Total cholesterol was found to be decreased in the birds fed on shoticompared to protein control and significantly higher than non-protein control group. Triglyceride valuedecreased in 10% shoti treated group compared to protein and non-protein control group. IncreasedHDL value was found in 10% shoti treated group compared to control groups and 20 % and 30 %shoti treated groups. Result of this experiment showed that body weight of birds significantly (p<0.05)increased in all shoti treated groups. Further investigations are necessary by using differentpercentages of shoti and other shoti type feed to determine the effects of this feed supplement onanimal’s physiology. Res. Agric., Livest. The objective of present work is Development of Hepatotoxicity model in rats and to evaluate hepatoprotective activity of cell wall contents of probiotics. Animals were divided in four groups. The groups were normal saline group, diseased control group, standard drug treated group and 4th group was CCl4 +Cell wall contents of probiotics. In diseased control group chronic liver injury was induced by administration of Carbontetrachloride (CCl4) via intraperitoneal route (1 ml/kg) for seventy days. For standard drug treated group 1 ml Silymarin suspension (Orally) and CCl4 was given for seventy days. In fourth group cell wall contents (1 x 10 12 CFU/ml/animal) and CCl4 was given for seventy days. During disease induction and treatment period blood samples were collected and serum was separated which was used to analyse various parameters like Alanine aminotransferase (ALT), Aspartate aminotransferase, (AST), Alkaline phosphate (ALP), direct bilirubin, total protein and albumin levels to asses liver function. Along with these cholesterol, Glucose and Malondialdehyde were also measured. Liver fibrosis and cirrhosis was quantified by histopathological studies of small portion of the excised liver. Serum AST, ALT, ALP, and direct bilirubin were found to be significantly higher in CCl4 intoxicated rats. Total protein and albumin was decreased. Manondialdehyde was found to be significantly higher in CCl4 intoxicated rats which was main end product of Lipid Peroxidation. Whereas in cell wall contents probiotics and silymarin treated group improve the liver functions in CCl4 toxicated rats. We conclude that protein oxidation may play a role in the pathogenesis of CCl4 induced liver injury and that the accumulation of oxidised proteins may be an early indication of CCl4 induced liver damage. Silymarin and cell wall contents of probiotics were effective in liver injury by inhibiting protein oxidation.

Abstract Introduction: Parabens are used commonly as preservatives in a range of cosmetics applied to the under arm and breast area as well as popular preservatives because of their cost.Aim of the work: This study was done to evaluate the neprohepatic toxicity of parabens. Materials and methods: Thirty adult female rats were used and given paraben orally for six months at parabens at dose of 10 % of the LD50 equal to 4.6mg\kg.bw. Mushroom was given orally to at dose of 10 mg/kg/day for six months too. Results: Oral administration of BP induced biochemical and histopathological changes. Biochemical changes: BP toxicity manifested by changes in the liver and kidney function tests manifested by increase AST, ALT, Bilirubin, urea and createnine with decreases to plasma proteins in comparison to control group. Giving mushroom caused amelioration to the nephrohepatic toxicity by inducing recovery in liver and kidney functions in comparison to paraben treated group. For histopathological findings: BP induced vascular congestion in liver and kidney in association with necrotic changes in the hepatorenal epithelium which improved after mushroom treatment. Conclusion: BP induced hepatorenal toxicity which improved by mushroom treatment.
