A new stackable modular system was developed for continuousin vivoproduction of phytoseiid mites. The system consists of cage units that are filled with lima beans,  Phaseolus lunatus, or red beans,P. vulgaris, leaves infested with high levels of the two-spotted spider mites,Tetranychus urticae. The cage units connect with each other through a connection cup, which also serves for monitoring and collection. Predatory mites migrate upwards to new cage units as prey is depleted. The system was evaluated for production ofPhytoseiulus persimilis. During a 6-month experimental period,20,894.9±10,482.5(mean ± standard deviation) predators were produced per week. The production consisted of4.1±4.6% nymphs and95.9±4.6% adults. A mean of554.5±59.8predatory mites were collected per harvested cage and the mean interval length between harvests was6.57±6.76days. The potential for commercial and experimental applications is discussed.

Participatory genetic improvement is a component of the management of genetic diversity, aiming at the systematic inclusion of farmers' knowledge, skills, experiences, practices and preferences. This modality of improvement is based on the knowledge of conventional genetics, physiology and economics, combined with those of anthropology, sociology, knowledge of the producers and with the principles of product development. The development of participatory strategies in the improvement of different crop species in the farmers field involves some fundamental aspects such as: rescue of different species and of different varieties within each species, valorization of local varieties, development of new varieties, valorization of cultural aspects and nutrition, agroecosystem management, agro-ecological crop systems, adaptation to the local environment with increased production from participatory improvement. Initially a search was made for communities in the forest area of ​​Pernambuco, Brazil, which preserve, store and use native varieties. After the identification of the localities contemplated. The seeds were obtained in the municipality of Lagoa de Itaenga. The proposal was submitted to farmers living in the Marrecos community. At the same time, the importance of the conservation of local cultivars, the maintenance of the genetic identity of these genotypes, the importance of the isolation of the cultures, mainly the Creole ones and the practice of the participatory improvement were also treated. In addition, seed samples were collected and stored by the farmers, for later implantation of the germplasm bank (in vivo) at the Federal Institute of Pernambuco. At the time of collection, the forms of storage adopted by farmers were observed, being of great relevance for the guarantee of the genetic reserves of the crops. The experiments were conducted in a phyto-bacteriology laboratory and in a greenhouse of the Department of Agriculture of the Federal Institute of Pernambuco (IFPE) in Vitória de Santo Antão-PE. The plants were cultivated in the field in an organic cultivation system. The results showed that there is genetic divergence, based on agro-morphological markers, among fava varieties studied as a function of germplasm origin. Thus, samples from the Germplasm Collection of IFPE-Camps Vitoria correspond to different materials. The existence of genetic divergence among the varieties indicates the possibility of selection of superior genotypes. The evaluated characteristics present high potential to differentiate the genotypes and should serve as parameters for other studies.

High blood pressure is a crucial risk factor for many cardiovascular diseases, and a diet rich in whole-grain foods may modulate blood pressure. This study investigated the effects of dehulled adlay consumption on blood pressure in vivo. We initially fed spontaneous hypertensive rats diets without (SHR group) or with 12 or 24% dehulled adlay (SHR + LA and SHR + HA groups), and discovered that it could limit blood pressure increases over a 12-week experimental period. Although we found no significant changes in plasma, heart, and kidney angiotensin-converting enzyme activities, both adlay-consuming groups had lower endothelin-1 and creatinine concentrations than the SHR group; the SHR + HA group also had lower aspartate aminotransferase and uric acid levels than the SHR group did. We later recruited 23 participants with overweight and obesity, and they consumed 60 g of dehulled adlay daily for a six-week experimental period. At the end of the study, we observed a significant decrease in the group’s systolic blood pressure (SBP), and the change in SBP was even more evident in participants with high baseline SBP. In conclusion, our results suggested that daily intake of dehulled adlay had beneficial effects in blood-pressure management. Future studies may further clarify the possible underlying mechanisms for the consuming of dehulled adlay as a beneficial dietary approach for people at risk of hypertension.

AbstractOur study aimed to identify the novel acaricidal compound in Xenorhabdus szentirmaii and X. nematophila using the easyPACId approach (easy Promoter Activated Compound Identification). We determined the (1) effects of cell-free supernatant (CFS) obtained from mutant strains against T. urticae females, (2) CFS of the acaricidal bioactive strain of X. nematophila (pCEP_kan_XNC1_1711) against different biological stages of T. urticae, and females of predatory mites, Phytoseiulus persimilis and Neoseiulus californicus, (3) effects of the extracted acaricidal compound on different biological stages of T. urticae, and (4) cytotoxicity of the active substance. The results showed that xenocoumacin produced by X. nematophila was the bioactive acaricidal compound, whereas the acaricidal compound in X. szentirmaii was not determined. The CFS of X. nematophila (pCEP_kan_XNC1_1711) caused 100, 100, 97.3, and 98.1% mortality on larvae, protonymph, deutonymph and adult female of T. urticae at 7 dpa in petri dish experiments; and significantly reduced T. urticae population in pot experiments. However, the same CFS caused less than 36% mortality on the predatory mites at 7dpa. The mortality rates of extracted acaricidal compound (xenocoumacin) on the larva, protonymph, deutonymph and adult female of T. urticae were 100, 100, 97, 96% at 7 dpa. Cytotoxicity assay showed that IC50 value of xenocoumacin extract was 17.71 μg/ml after 48 h. The data of this study showed that xenocoumacin could potentially be used as bio-acaricide in the control of T. urticae; however, its efficacy in field experiments and its phytotoxicity need to be assessed in future.

SUMMARYThe voluntary food intake, in vivo digestibility and site of nutrient digestion was measured in 12 Polled Dorset ram lambs, from 6 to 24 weeks of age, offered clover hay. Lambs were weaned at 6 weeks of age. Six were fitted with a permanent T-shaped cannula in the abomasum and a temporary rumen catheter during the periods of digesta flow measurement.Surgical modification of lambs did not influence intake (g/kg live weight (W) per day), in vivo digestibility or marker retention time in the rumen. However, intact lambs grew significantly faster than cannulated lambs (139 v. 109g/day, P < 0·05) apparently because of an overall higher cumulative intake over the whole experimental period rather than any change in efficiency of use of nutrients or maintenance requirement.Relative to live weight, intake increased rapidly until c. 10 weeks of age and remained relatively constant at c. 37 g DM/kg W per day for the remainder of the experiment. Age had no effect on in vivo digestibility of organic matter (OM) or neutral detergent fibre (NDF) (average 0·69).The proportion of digestible organic matter apparently digested in the rumen (DOMADR) decreased from 0·63 to 0·47 between 8 and 24 weeks of age whilst intake between these measurement periods increased. Nonammonia N (NAN) flow past the abomasum was similar at all ages in relation to digestible organic matter intake (55·1 g NAN/kg DOMI) but was higher at 16 and 24 weeks of age than at 8 and 12 weeks of age, respectively, in relation to organic matter apparently digested in the rumen (OMADR) (116 and 85 g NAN/kg OMADR, respectively, P < 0·05).Fractional outflow rate of digesta markers from the rumen, when estimated from abomasal digesta, was significantly higher than when estimated from faeces (103Ru-p 0·121 v. 0·09; 51Cr-EDTA 0·169 v. 0·111/h; P > 0·05).It was concluded that cannulation did not affect intake and digestion characteristics in these young lambs. The changes in the site of nutrient digestion as the animals aged may have been due to agerelated changes in digestive function but appeared to be associated with the increase in intake that occurred between measurement periods of the cannulated lambs.

Lima beans are an important crop in Delaware and the Mid-Atlantic Region. In the summer of 2000, five commercial cultivars (3–28, 184–85, C-elite Sel, Butter Bean, and Jackson Wonder) of lima bean in Delaware, Maryland, and New Jersey were observed with white, appressed mycelia on infected pods that appeared distinctly different from signs of downy mildew infection caused by Phytophthora phaseoli. Isolations were made by placing diseased pods between layers of rye media (1). A fungus that produced white mycelia with sporangia was consistently isolated. All Phytophthora isolates from the infected pods were heterothallic, grew at 35°C, had as much as 100 μm long pedicles on varying shapes of caducous sporangia with tapering base and >2 papillae, and were identified as P. capsici (2). Initially, three surface-disinfected pods from cv. Early Thorogreen plants grown in the greenhouse were floated on 20 ml of sterile water in a petri dish, and each was inoculated with a disk of P. capsici. This was repeated for nine isolates obtained from lima bean. After incubation for 7 days at room temperature, all 27 pods were infected, and P. capsici was reisolated from all the pods. A pathogenicity test was performed on the same cultivars from which the original field isolates were collected. Three seedlings and two plants with mature pods were inoculated with a sporangial suspension of each of the nine isolates and placed in a dew chamber for 5 days at 20 to 25°C and 100% relative humidity. White mycelial growth was observed on seedlings and mature pods. One inoculated plant developed brown-to-black stem lesions with white mycelia. All pods on the mature plants showed appressed, white mycelia identical to that observed in the commercial lima bean fields. P. capsici was consistently reisolated from all inoculated plants. In 2000, most infected pods in infested fields were observed low in the plant canopy or touching the soil. However, in 2001, infected pods were mostly in the lower and mid-portion of the plants observed in baby lima bean fields in Kent County, DE. References: (1) C. E. Caten and J. L. Jinks. Can. J. Erucaria pinnata (Viv.) is a wild annual plant growing in North-Western Coastal Region in Egypt. This study reports for the first time the cytotoxic activity of different extracts of Erucaria pinnata plant against HEP-G2 cell line. The 70% methanolic extract (E1) recorded the best potent cytotoxic activity (IC50=13.6 µg/ml), so we analysis the flavonoids constituent of this extract using HPLC, which show that our extract is rich with important flavonoids compounds (rutin, quercetin, leutolin, etc.). We evaluated its antitumor activity against hepatocellular carcinoma (HCC) induced by diethylnitrosamine (DEN) (200mg/Kg. b.wt., i.p, single dose) after two weeks, animals received carbontetrachloride (CCl4) (3ml/Kg. b.wt., SC, once a week for 6 weeks) and the experiment continued for 44 weeks in rats. After the experimental period, the administration of DEN/CCl4 showed significant increase in the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total bilirubin, gamma-glutamyl transferase (γGT) and significant decrease in the levels of total proteins and albumin content in the serum with reduction in the liver antioxidants, including superoxide dismutase (SOD) and catalase (CAT). This was accompanied by increases in serum specific tumor markers (AFP). The 70% methanolic plant extract (E1) was orally administrated (400mg/kg/day respectively) for the whole study period, and it showed a significant improvement at the different biological liver functions, remodeled the antioxidant enzymes activity and down-regulated the serum AFP. All these findings were confirmed by histopathological studies of the liver samples obtained from all groups. In addition, we evaluated its antiangeogenic activity by docking study against VEGFR-2 tyrosine kinase after it showed an ability to inhibit the VEGFR-2 expression in vitro and inhibit the concentration of VEGF-A in vivo. The hepatoprotective effect of our extract was attributed to its antioxidant and antiangeogenic activity.
