Schisandrae Fructus (SF), which possesses five tastes: sweet (fruit skin), sour (pulp), bitter/pungent (seed core), and saltiness (all parts), can produce a wide spectrum of biological activities in the body. Here, we investigated the effects of the ethanolic extract of SF pulp, seed, or their combination (namely, EtSF-P, EtSF-S, or EtSF-P/S, resp.; collectively called EtSF) on the metabolism of lipids and glucose in normal diet- (ND-) and hypercholesterolemic diet- (HCLD-) fed mice. Supplementation with EtSF significantly reduced hepatic triglyceride and cholesterol levels by 18–47% in both ND- and HCLD-fed mice. EtSF supplementation reduced serum triglyceride levels (approximately 29%), whereas EtSF-P and EtSF-S/P elevated serum cholesterol (up to 26 and 44%, resp.) in HCLD-fed mice. Treatment with EtSF decreased hepatic glucose levels (by 9–44%) in both ND- and HCLD-fed mice. Supplementation with EtSF-S or EtSF-S/P (at 1 and 3%) increased biliary or fecal TC contents in HCLD-fed mice. However, supplementation with EtSF-S/P at 9% reduced biliary TC levels in HCLD-fed mice. EtSF-P or EtSF-S/P supplementation reduced serum alanine aminotransferase activity in HCLD-fed mice. The findings suggested that supplementation with EtSF lowered lipid and glucose accumulation in the liver and increased fecal cholesterol contents in mice. Dietary supplementation with EtSF-P or EtSF-S/P attenuated liver damage in HCLD-fed mice.

Schisandrae Fructus (SF), the fruit of Schisandra chinensis (Turcz.) Baillon, has been used for the treatment of liver injury and metabolism-related disorders in China. The objective of this study was to investigate the effects of supplementation with ethanol extract of SF seed (EtSF-S) on serum/hepatic lipid and glucose levels as well as fecal total cholesterol (TC) contents in mice fed a normal diet (ND) or high-fat/fructose diet (HFFD) containing 15% lard oil and 15% fructose. Female ICR mice (18–20 g in body weight) were fed with ND or HFFD for 3 months, and then EtSF-S was added to both chow diets at increasing concentrations of 1, 5, and 10% (w/w). Thirty days later, serum and hepatic lipids, including TC, triglyceride (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL), and glucose, were measured. Dietary supplementation with EtSF-S reduced hepatic TC (36 and 18%) and TG levels (38 and 28%) and increased serum HDL/LDL ratio (16 and 26%) in both ND- and HFFD-fed mice, respectively. Moreover, supplementation with EtSF-S elevated serum HDL (31%) in HFFD-fed mice and reduced serum LDL (27%) in ND-fed mice. EtSF-S treatment reduced fat mass (40%) in ND-fed mice and increased fecal TC contents (33%) in HFFD-fed mice. EtSF-S supplementation decreased hepatic glucose contents (29%) in both ND- and HFFD-fed mice. However, diet supplemented with EtSF-S elevated serum TG levels (up to 123%) and hepatic size (28%), but more importantly, suppressed the body weight gain (approximately 130%) in mice fed with HFFD. These findings suggested that dietary supplementation with EtSF-S as natural herbal function food may be a useful strategy for the treatment of patients with fatty liver disease or overweight without a high intake of sugar and fat.

World Health Organization (WHO) estimated that 80% of the population of developing countries use traditional medicines, mostly natural plant products, for their primary health care needs. In the past few decades, the medicinal value of plants has been assumed more important dimension owing largely to the discovery that extracts from plants contain not only primary metabolites but also a diverse array of secondary metabolites with antioxidant potential. Medicinal plants are potential sources of natural compounds with biological activities and therefore attract the attention of researchers worldwide. Antioxidants are vital substances which possess ability to protect the body from damage due to free radical-induced oxidative stress. The purpose of current study was to determine the antioxidant activities and bioactive components of Foeniculum vulgare (fennel) (Samonsabar) seeds by using UV Visible Spectrophotometer (UV-Vis) and Gas Chromatography-Mass Spectrometry (GC-MS). Aqueous extract of fennel seeds showed more antioxidant activity (IC50: 0.28 ug/ml) than ethanolic extract (IC50: 0.83 ug/ml) and comparable to standard antioxidant, ascorbic acid (IC50: 0.59 ug/ml). GC-MS analysis was fruitful in identification of compounds based on peak area, retention time, molecular formula, molecular weight, MS Fragmentions and pharmacological actions. Ten bioactive phytochemical compounds from aqueous extracts and 11 from ethanolic extract of fennel seeds were identified. These findings indicated that fennel seeds are potential to provide preventive properties against oxidative damage. These results will give scientific information for quality control of indigenous drug to herbal medicine users and local practitioners using fennel for different types of ailments

Acacia senegalL. (Fabaceae) seeds are essential ingredient of “Pachkutta,” a specific Rajasthani traditional food. The present study explored antiatherosclerotic and cardioprotective potential ofAcacia senegalseed extract, if any, in hypercholesterolemic diet-induced atherosclerosis in rabbits. Atherosclerosis in rabbits was induced by feeding normal diet supplemented with oral administration of cholesterol (500 mg/kg body weight/day mixed with coconut oil) for 15 days. Circulating total cholesterol (TC), HDL-cholesterol (HDL-C), LDL-cholesterol (LDL-C), triglycerides, and VLDL-cholesterol (VLDL-C) levels; atherogenic index (AI); cardiac lipid peroxidation (LPO); planimetric studies of aortal wall; and histopathological studies of heart, aorta, kidney, and liver were performed. Apart from reduced atherosclerotic plaques in aorta (6.34±0.72) and increased lumen volume (51.65±3.66), administration with ethanolic extract ofAcacia senegalseeds (500 mg/kg/day, p.o.) for 45 days to atherosclerotic rabbits significantly lowered serum TC, LDL-C, triglyceride, and VLDL-C levels and atherogenic index as compared to control. Atherogenic diet-induced cardiac LPO and histopathological abnormalities in aorta wall, heart, kidney, and liver were reverted to normalcy byAcacia senegalseed extract administration. The findings of the present study reveal thatAcacia senegalseed extract ameliorated diet-induced atherosclerosis and could be considered as lead in the development of novel therapeutics.

Nelumbinis Semen (NS, the seeds of Nelumbo nucifera) extract is a traditional Korean medicine with anti-oxidant activity. The present study examined the anti-obesity and antidiabetic effects of NS powder in high-fat diet (HFD)-induced obese C57BL/6 mice. Mice (n = 8/group) were fed a normal diet (CON), HFD, HFD containing 5% NS powder (HFD-NS5%), or HFD containing 10% NS powder (HFD-NS10%) for 12 weeks. Food intake was relatively higher in groups HFD-NS5% and HFD-NS10%, while the food efficiency ratio was highest in group HFD (p < 0.05). HFD-NS5% reduced the body weight (−39.1%) and fat weight (−26.6%), including epididymal fat and perirenal fat, and lowered the serum triglyceride levels (−20.6%) compared with HFD. Groups HFD-NS5% and HFD-NS10% showed hepatoprotective properties, reducing the serum ALT levels (p < 0.05) and fat globules (size and number) in the liver compared with group HFD. HFD-NS5% and HFD-NS10% regulated the blood glucose, improved the glucose intolerance, and showed a 12.5% and 15.0% reduction in the area under the curve (AUC) of intraperitoneal glucose tolerance test (IPGTT), and a 26.8% and 47.3% improvement in homeostatic model assessment insulin resistance (HOMA-IR), respectively, compared with HFD (p < 0.05). Regarding the expressions of genes related to anti-obesity and antidiabetes, there was a 1.7- and 1.3-fold increase in PPAR-α protein expression, 1.4- and 1.6-fold increase in PPAR-γ protein expression, and 0.7- and 0.6-fold decrease in TNF-α protein expression, respectively, following HFD-NS5% and HFD-NS10% treatments, compared with HFD, and GLUT4 protein expression increased relative to CON (p < 0.05). These results comprehensively provide the fundamental data for NS powder’s functional and health-promoting benefits associated with anti-obesity and antidiabetes.

Abstract Aim and Objective The present study aims to investigate whether the antihyperglycemic effect of Murraya koenigii is mediated by antioxidant properties and insulin mimetic effect. Methods Thirty Spraque-Dawley rats were induced hyperglycemia by streptozotocin and nicotinamide (STZ-NA). The STZ-NA diabetic rats were treated with an ethanolic extract of Murraya koenigii 200 mg/kg b.w and 400 mg/kg b.w. One group was treated with glibenclamide (1 mg/kg b.w). After the administration of Murraya koenigii extract and glibenclamide for four weeks, the rats were sacrificed. Blood and organ samples were collected under a fasting condition. The body weight and blood glucose levels were measured. Hepatic enzymes were determined using a commercial kit, protein levels were estimated by Bradford’s method, and plasma insulin was assayed by an ELISA kit. Malondialdehyde (MDA) and reduced glutathione (GSH) levels were estimated by the TBA-Wills method and Ellman’s method, respectively. Results Ethanolic extract of Murraya koenigii showed a significant reduction in blood glucose level at both doses, 200 and 400 mg/kg b.w. In addition, Murraya koenigii exhibited a profound antioxidant effect with decreased MDA level and increased GSH level, particularly at the 200 mg/kg b.w. and significantly decreased the HOMA-IR index. Conclusions The present study reveals that Murraya koenigii possesses antidiabetic activity and antioxidant effects on STZ-NA induced diabetes mellitus.

In the Indian ayurvedic system of medicine, Rumex nepalensis Spreng. (Polygonaceae) commonly known as Nepal Dock has wide-spectrum therapeutic potencies and is extensively used for centuries in traditional medicine systems. They act as a possible food supplement and are largely used in pharmaceutical industry. Extracts and metabolites from this plant exhibits pharmacological activities including anti-inflammatory, antioxidant, antibacterial, antifungal, antiviral, insecticidal, purgative, analesic, antipyretic, anti-algal, central nervous system depressant, genotoxic, wound healing and skeletal muscle relaxant activity. Due to its remarkable biological activities, it has the potential to act as a rich source of drug against life threatening diseases. The aim of the present study is to examine Rumex nepalensis roots for phytochemical profile. Qualitative analysis of various phytochemical constituents and quantitative analysis of total phenolics and flavonoids were determined by the well-known test protocol available in the literature. Quantitative analysis of phenolic and flavonoids was carried out by Folins Ciocalteau reagent method and aluminium chloride method respectively. Phytochemical analysis revealed the presence of phenols, flavonoids, tannins, saponins, alkaloids, fixed oil and fats. The total phenolics content of roots ethanolic extract was (1.658 mg/100mg), followed by flavonoids (1.048mg/100mg). The present study concluded that the crude extract of Rumex nepalensis is a potential source of various activates and this justifies its use in folkloric medicine. Eriobotrya japonica locally called as loquat in Kashmir has been studied in various parts of the world but little work has been reported on Kashmiri loquat. The chemical nature of fruits and vegetables offers a great diversity of biological properties and plays an important role in the field of pharmacology. There is a quest for newer drugs with few adverse effects and this poses a challenge for the development of new drugs. The study was undertaken to study the activities of ethanolic extract of Eriobotrya japonica fruits in streptozotocin induced diabetic rats. The phytochemical screening of the plant was also done. The animals were divided into five groups. Normal Control group received only the vehicle. Toxic group included those animals in which diabetes was induced by streptozotocin. The 3rd group was those animals which received streptozotocin and standard antidiabetic drug-glibenclamide. 4th group included those diabetic animals which received 50 mg/kg b.w dose of fruits of Eriobotrya japonica. 5th group animals included those diabetic animals which received 100 mg/kg b.w of the plant extract. The biochemical parameters that were evaluated were blood glucose levels and lipid profile tests. The body weight was also checked. Histopathology of pancreas was also done. The results showed significant decrease in blood glucose levels, lipid profile tests in animals treated with different doses of the plant extracts. Histopathology of pancreas also showed positive results.

Background: To study the hepatoprotective effect of Ethanolic extract of Stem Bark of Berberis lycium Royale in isoniazid (INH) induced hepatotoxicity in mice model.Methods: The study design was lab based randomized controlled in-vivo study in mice conducted from 9th April 2014 till 9th May 2014 at animal house of National Institute of Health, Islamabad. Group A was on normal diet and water and hepatotoxicity was produced by giving isoniazid (50mg/kg BW) in mice of Group B. Group C and D were given isoniazid (INH) plus low dose and high dose of Ethanolic extract of stem bark of Berberis Lycium Royle respectively.Results: INH induced hepatotoxicity was depicted by elevated serum LFT’s, hepatocytic ballooning, severe steatosis and inflammation. Mice getting concurrent treatment of INH, low and high dose of Ethanolic extract of Berberis Lycium Royle showed decreased serum levels of biomarkers and their liver sections manifested improved histological picture but more significant reduction in toxic effects were observed in animals receiving high dose.Conclusions: High dose of Ethanolic extract of stem bark of Berberis lycium Royale showed more marked hepatoprotective activity as compare to low doses. The hepatotoxicity of INH can be reduced by concurrent use of INH with ethanolic extracts of Berberis Lycium Royle.

Abstract Inbred Carworth Farms Nelson (CFN) congenitally hyperlipidemic rats had significantly shorter coagulation and prothrombin times and higher levels of coagulation factors, II, V, VII, VIII, and X than did controls. Conversely, congenitally hypolipidemic rats of the same strain had significantly longer coagulation and prothrombin times and lower levels of factors II, V, VII, X and XII and of blood platelets than did controls. A loop-shaped polyethylene cannula was inserted into the aorta to assess the potential for thrombosis. The hyperlipidemic group obstructed this significantly faster and the hypolipidemic group slower than did the controls. Normal CFN rats made hypertensive by unilateral renal artery clip developed hypertension together with significantly elevated serum cholesterol and factor VII and X levels. Rhesus monkeys with diet-induced hyperlipidemia showed shorter prothrombin times and higher factor X levels than did controls on normal diet. By selective breeding, two groups of squirrel monkeys were obtained. Both groups had similar serum cholesterol levels on a normal diet but one group (hyperresponders) showed higher serum cholesterol levels on a cholesterol-containing diet than did the other (hyporesponder) group. Both groups showed significantly elevated levels of factors II, V, VII, IX and X on a cholesterol-containing diet. There was good correlation between the levels of many coagulation factors and serum cholesterol in both rats and monkeys. If thrombosis is important in the genesis of atherosclerosis, these findings could indicate that elevation of plasma lipids may play a role, via the coagulation pathway, in the production of human vascular disease.
