Coumarin (2H-1-benzopyran-2-one) is a plant-derived natural product known for its pharmacological properties such as anti-inflammatory, anticoagulant, antibacterial, antifungal, antiviral, anticancer, antihypertensive, antitubercular, anticonvulsant, antiadipogenic, antihyperglycemic, antioxidant, and neuroprotective properties. Dietary exposure to benzopyrones is significant as these compounds are found in vegetables, fruits, seeds, nuts, coffee, tea, and wine. In view of the established low toxicity, relative cheapness, presence in the diet, and occurrence in various herbal remedies of coumarins, it appears prudent to evaluate their properties and applications further.

Aim of the review paper is to give an enlightenment on potential phyto-constituents of Gentianaceae.  Bitter principles (secoiridoid glycosides) and xanthones are widely distributed among plants belonging to the family Gentianaceae. Phytochemicals like amarogentin, bellidifolin, gentianine and swerchirin have been reported to possess significant anti-inflammatory, analgesic, anti-asthmatic, anticonvulsant, antihistaminic, antimalarial, antiamoebic, cytoprotective, diuretic, hepatoprotective and hypoglycemic properties. Topoisomerase inhibition is significant pharmacological activity reported from amarogentin. Further investigations exploring possible use of these phytochemicals as therapeutic agents are warranted. The articles reviews certain pharmacological investigations carried out on phytochemicals reported from Gentianaceae

Diversity-Oriented Synthesis (DOS) represents a strategy to obtain molecule libraries with diverse structural features starting from one common compound in limited steps of synthesis. During the last two decades, DOS has become an unmissable strategy in organic synthesis and is fully integrated in various drug discovery processes. On the other hand, natural products with multiple relevant pharmacological properties have been extensively investigated as scaffolds for ligand-based drug design. In this article, we report the amino dimethoxyacetophenones that can be easily synthesized and scaled up from the commercially available 3,5-dimethoxyaniline as valuable starting blocks for the DOS of natural product analogs. More focus is placed on the synthesis of analogs of flavones, coumarins, azocanes, chalcones, and aurones, which are frequently studied as lead compounds in drug discovery.

Coumarone is a chemical compound found in many plants. Coumarone having diverse pharmacological properties popularly known as an antimicrobial, Analgesic anti-inflammatory, Anticancer, Antiviral, anticoagulant, antihypertensive, anticonvulsant, antihyperglycemic, antioxidant, activities. Coumarone was also observed in all green color veggies, and also in fruits and their seeds, dark coffee, tea leafs, further it is used for herbal remedies due to having less toxicity, very cheaper. The most useful method for the synthesis of Coumarone is from phenol and ethyl acetate and also by using catalyst. In this paper we tried to update the observations of authors towards the biological and medicinal significance of novel the natural and synthetic coumarone derivatives.

The review of the 2016–2017 marine pharmacology literature was prepared in a manner similar as the 10 prior reviews of this series. Preclinical marine pharmacology research during 2016–2017 assessed 313 marine compounds with novel pharmacology reported by a growing number of investigators from 54 countries. The peer-reviewed literature reported antibacterial, antifungal, antiprotozoal, antituberculosis, and antiviral activities for 123 marine natural products, 111 marine compounds with antidiabetic and anti-inflammatory activities as well as affecting the immune and nervous system, while in contrast 79 marine compounds displayed miscellaneous mechanisms of action which upon further investigation may contribute to several pharmacological classes. Therefore, in 2016–2017, the preclinical marine natural product pharmacology pipeline generated both novel pharmacology as well as potentially new lead compounds for the growing clinical marine pharmaceutical pipeline, and thus sustained with its contributions the global research for novel and effective therapeutic strategies for multiple disease categories.

The lipoxazolidinone family of marine natural products, with an unusual 4-oxazolidinone heterocycle at their core, represents a new scaffold for antimicrobial discovery; however, questions regarding their mechanism of action and high lipophilicity have likely slowed follow-up studies. Herein, we report the first synthesis of lipoxazolidinone A, 15 structural analogs to explore its active pharmacophore, and initial resistance and mechanism of action studies. These results suggest that 4-oxazolidinones are valuable scaffolds for antimicrobial development and reveal simplified lead compounds for further optimization.

Azetidin-2-ones (β-lactams) and its derivatives are an important group of heterocyclic compounds that exhibit a wide range of pharmacological properties such as antibacterial, anticancer, anti-diabetic, anti-inflammatory and anticonvulsant. Efforts have been made over the years to develop novel congeners with superior biological activities and minimal potential for undesirable side effects. The present review aimed to highlight some recent discoveries (2013-2019) on the development of novel azetidin-2-one-based compounds as potential anticancer agents.
