The present study is to investigate which kinds of solvent extracts of Inulae Flos inhibit the chemokine productions in HaCaT cell and whether the inhibitory capacity of Inulae Flos is related with constitutional compounds. The 70% methanol extract showed comparatively higher inhibition of thymus and activation-regulated chemokine (TARC/CCL17) in HaCaT cells, therefore this extract was further partitioned with n-hexane, chloroform, ethyl acetate, butanol, and water. The ethyl acetate fraction inhibited TARC, macrophage-derived chemokine (MDC/CCL22), and regulated on activation of normal T-cell-expressed and -secreted (RANTES/CCL5) production in HaCaT cells better than the other fractions. The compounds of Inulae Flos, such as 1,5-dicaffeoylquinic acid and luteolin, inhibited TARC, MDC, and RANTES production in HaCaT cells. 1,5-Dicaffeoylquinic acid was contained at the highest concentrations both in the 70% methanol extract and ethyl acetate fraction and inhibited the secretion of chemokines dose-dependently more than the other compounds. Luteolin also represented dose-dependent inhibition on chemokine productions although it was contained at lower levels in 70% methanol extract and solvent fractions. These results suggest that the inhibitory effects of Inulae Flos on chemokine production in HaCaT cell could be related with constituent compounds contained, especially 1,5-dicaffeoylquinic acid and luteolin.

AbstractPeople of north-eastern states of India consume raw areca-nut (RAN) and lime which could lead to oral, esophageal and gastric cancers. However, the incidence of these cancers are significantly lesser in those who consume pieces of Potentilla fulgens root along with RAN. Since evaluation of anticancer role, if any, of P. fulgens on RAN-mediated genetic alterations in human is difficult because of other compounding factors, this study was undertaken in mice to focus on gastric carcinogenesis since ad libitum administration of RAN extract with lime in drinking water induced stomach cancer due to greater exposure of its lining. A total of 160 mice were used at different time points and either methanol extract of P. fulgens roots (PRE) or mixture of four compounds of ethyl-acetate fraction (EA-mixture) was mixed with mice feed. Histological studies revealed that RAN + lime induced cancer in all the mice and interestingly only 20% developed cancer when PRE/EA-mixture was provided along with RAN + lime. Higher frequency of precocious anaphase and over expression of p53 and Securin genes were significantly reduced by PRE/EA-mixture. Thus PRE/EA-mixture mitigates the RAN-induced tumor-initiating process in stomach by maintaining expression of tumor suppressor and check-point genes under control.

Bioassay-guided fractionation of the cytotoxic ethyl acetate fraction of the sequential methanol extract from the leaves and twigs of Dasymaschalon sootepense led to the isolation of a new 7-hydroxy aporphine alkaloid, 6a,7-dehydrodasymachaline (1) along with the five known compounds (-)-nordicentrine (2), dicentrinone (3), (-)-sinactine (4), aristolactam AII (5) and epiberberine (6). Their structures were elucidated by spectroscopic methods. This is the first report of alkaloids 1–2 and 5–6 from the genus Dasymaschalon. Compounds 1 and 5 showed cytotoxicity against a panel of cancer cell lines.

AbstractThe phytochemical and antimicrobial activity of the petroleum ether and crude methanol extracts, chloroform and ethyl acetate fractions of the leaves of Lannea kerstingii were investigated. Phytochemical screening revealed the presence of steroids and triterpenes in the petroleum ether extract, steroid, triterpene, flavonoids and tannins in both crude methanol extract and chloroform fraction while the ethyl acetate fraction contained only flavonoids and tannins. The extracts exhibited antimicrobial activities with zones of inhibition ranging from 17.00 to 21.03, 20.10 to 25.24, 25.32 to 34.02 and 22.28 to 27.20 mm for petroleum ether extract, methanol extract, chloroform and ethyl acetate fractions respectively. The minimum inhibitory concentration was between 5 and 10mg/ml, 5mg/ml for the petroleum ether and methanol extract respectively, and between 2.5 and 5 mg/ml, 5mg/ml for the acetate fractions. The minimum bactericidal concentration for all the extracts was 40mg/ml respectively except for chloroform fraction which ranged from 20 to 40mg/ml. The minimum fungicidal concentration for all the extracts was found to be 40mg/ml respectively. This result indicates the broad spectrum antimicrobial potential of L. Kerstingii and justifies the use of this plant in traditional medicine.

Urinary calculi are stones (urolithiasis) that can form anywhere in urinary tract outside of the kidneys and mostly composed of calcium oxalate and phosphate, additionally with elevated throughout the last two decades in the world. Chemical composition plays a major part in nephrolithiasis. Therefore, the high concentrations of lithogenic substances in urine enhance the crystallization method in urine tract system. The most kidney stones form from calcium oxalate, the present study was inspected the effect of the crude aqueous extract as well as the fractionated methanol extract (ethyl acetate, isopropanol, acetone and methanol residue) of paronychia argentea on the crystallization of calcium oxalate salts. The effect of aqueous extract and fractionated methanol extract on the size, number, type of calcium oxalate crystals. Paronychia argentea both the crude aqueous and the fractionated extract, especially ethyl acetate fraction have antiurolithic activity via reducing crystal size as well as activate the formation of calcium oxalate dihydrate (COD) crystals out from calcium oxalate monohydrate (COM) with increasing concentration of extract. The shifting of crystallization process to producing calcium oxalate dihydrate (COD) rather than oxalate monohydrate (COM) and the reducing the crystal size and calcium ion concentration, in addition to the diuretic action of extract plays an important role in controlling urolithiasis.

Background: Micro-organisms are responsible for the transmission of a large number of diseases. It is hard to comprehend the amount of diseases, deaths and economic losses caused by micro-organisms alone. Plants are good sources of eco-friendly and readily available antimicrobial agents. Purpose: The aim of this study was to evaluate the chemical constituents and antimicrobial characteristics of three fractions from ethnomedicinal Icacina trichantha. Oliv. (Icacinaceae).Methods: Methanol extract from Icacina trichantha. Oliv was obtained by maceration and fractionated successively using hexane, and ethyl acetate. The antimicrobial properties of Icacina trichantha. Oliv was assessed using agar cup diffusion method on MRSA, P. aeruginosa, S. typhi, C. krusei, S. dysenteriae, S. pyrogenes, E. coli, K. pneumoniae, C. albicans, and C. tropicalis. Phytochemical screening on fractions was also evaluated using standard methods.Results: Phytochemical screening on fractions revealed the presence of saponins, alkaloids, steroids, tannins, and glycosides. Agar diffusion assay on fractions showed growth inhibitory effect on all the organisms except P. aeruginosa, S. typhi, and C. tropicalis. The MIC revealed that n-hexane fraction was active against MRSA, S. pyrogenes, E. coli, K. pneumonia, C. albicans and C. krusei at 10 mg/mL while S. dysenteriae was active at 5 mg/mL. The ethyl acetate fraction was active against all the organisms at a concentration of 5 mg/mL except P.aeruginosa, S.typhi and C.tropicalis. Methanol fraction showed activity of 5 mg/mL against MRSA, S. pyrogenes, E. coli, S. dysenteriae, C. albicans and C. krusei except for K. pneumoniae with activity at 10 mg/mL. Minimum bactericidal concentration/fungicidal concentration MBC/MFC evaluated on the n-hexane fraction revealed that MRSA, S. pyrogenes, E. coli, K. pneumoniae, S. dysenteriae, C. albicans, and C. krusei were activeat 20 mg/mL, while the ethyl acetate fraction had MBC/MFC of 10 mg/mL against all the organisms except P. aeruginosa, S. typhi, C. tropicalis. Methanol extract had MBC/MFC of 10 mg/mL against MSRA, E.coli and S. dysenteriae whereas S. pyrogenes, K. pneumoniae, C. albicans and C. krusei had MBC/MFC at 20 mg/mL.Conclusion: Icacina trichantha. Oliv. contain constituents with concentration dependent antimicrobial properties based on type of organism. The plant could be useful in the prevention and treatment of multi-resistant disease causing microorganisms.

Date seeds are waste from palm fruit processing that has not been utilized optimally. Date seeds contain flavonoids which are known to act as antioxidants. The aim of this study was to determine the antioxidant activity and flavonoid of methanol extract which was carried out by soxhletation and its fractionation results. Date seeds are made into simplicia, soxhlet extraction with methanol, then the methanol extract is fractionated in stages by liquid-liquid partition with n-hexane, ethyl acetate, and n-butanol. Each fraction and extract was determined by its antioxidant activity with the DPPH method and its flavonoid was determined by spectrophotometry. Ethyl acetate fraction has the strongest antioxidant activity with the lowest IC50 value, that is equal to 5.74 ± 0.05μg/mL, followed by methanol extract of 9.55±0.53 μg/mL, n-butanol fraction 19.73±0,58 μg/mL, and n-hexane fraction of 289.59±10.52 μg/mL, while the IC50 value for vitamin C as a positive control was 4.29±0.74 μg/mL. The highest flavonoids were produced from ethyl acetate fractions which amounted to 1484.33 ± 161.47 mg Quercetin Equivalent (QE) / 100 g, followed respectively by methanol extract of 282.84±13.72 mg QE/100 g.

The antibacterial activity of methanol extract from the root bark of Akanda (Calotropis gigantea L.) and its petroleum ether, chloroform and ethyl acetate fractions were investigated. Both of methanol extract and its chloroform fraction showed activity against Sarcina lutea, Bacillus megaterium and Pseudomonas aeruginosa. Petroleum ether fraction showed activity against Bacillus subtilis and Shigella sonnei whereas ethyl acetate fraction showed activity against Pseudomonas aeruginosa and Escherichia coli at 20ìg/disc, 30ìg/disc and 40ìg/disc doses. Among the tested materials, methanol extract and its chloroform fraction showed comparatively better results. Minimum inhibitory concentration (MIC) for methanol extract and each fraction were also determined by serial dilution technique.
