We investigated the antiplasmodial properties of crude extracts fromCarica papayaleaves to trace the activity through bioassay-guided fractionation. The greatest antiplasmodial activity was observed in the ethyl acetate crude extract.C. papayashowed a high selectivity forP. falciparumagainst CHO cells with a selectivity index of 249.25 and 185.37 in the chloroquine-sensitive D10 and chloroquine-resistant DD2 strains, respectively.Carica papayaethyl acetate extract was subjected to bioassay-guided fractionation to ascertain the most active fraction, which was purified and identified using high-pressure liquid chromatography (HPLC) and GC-MS (Gas chromatography-Mass spectrometry) methods. Linoleic and linolenic acids identified from the ethyl acetate fraction showed IC50of 6.88 μg/ml and 3.58 μg/ml, respectively. The study demonstrated greater antiplasmodial activity of the crude ethyl acetate extract ofCarica papayaleaves with an IC50of2.96±0.14 μg/ml when compared to the activity of the fractions and isolated compounds.

Abstrak. Penelitian ini bertujuan untuk menguji aktivitas ekstrak etil asetat babadotan dari berbagai ketinggian tempat dan konsentrasi terhadap pertumbuhan gulma bayam duri. Penelitian ini dilaksanakan di Laboratorium Ilmu Gulma Program Studi Agroteknologi dan Kebun Percobaan Fakultas Pertanian Universitas Syiah Kuala Banda Aceh, serta Laboratorium Analisis Kimia Fakultas Matematika dan Ilmu Pengetahuan Alam (FMIPA), sejak September 2017– Januari 2018. Bahan yang digunakan adalah benih bayam duri, yang diperoleh dari Kecamatan Delima Kabupaten Pidie dan daun babadotan yang diperoleh dari 3 daerah dengan ketinggian yang berbeda yaitu Kecamatan Indrapuri Kabupaten Aceh Besar (19 mdpl), Kecamatan Lembah Seulawah Saree Kabupaten Aceh Besar (261 mdpl) dan Kecamatan Takengon Kabupaten Aceh Tengah (1259 mdpl) dengan konsentrasi 5, 10 dan 15% sebagai bioherbisida. Rancangan yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) pola faktorial 3  3 dengan 3 kali ulangan. Faktor pertama yaitu ketinggian tempat dengan 3 taraf dan faktor kedua konsentrasi dengan 3 taraf. Ekstrak yang dihasilkan pada ketinggian tempat yang berbeda dan konsentrasi ekstrak etil asetat berpengaruh nyata terhadap laju tinggi, jumlah daun, diameter batang, persentase pengendalian dan panjang akar. Terdapat interaksi antara ketinggian tempat dan konsentrasi terhadap persentase pengendalian pada 7 dan 14 HSA. Ekstrak yang dihasilkan pada ketinggian  9 mdpl dengan konsentrasi 10% dapat mengendalikan gulma bayam duri hingga 100% pada 7 HSA.The Test of Effectiveness Whiteweed Ethyl Acetate Extract (Ageratum conyzoides L.) from Various Altitudes And Concentration Against the Growth of Spiny Amaranth Weed (Amaranthus spinosus L.)Abstract. This study aims to examine the activity of ethyl acetate fraction of whiteweed extract from various altitudes and concentration againts the growth of spiny amaranth. This  research was conducted at Laboratory of Weed Science, Agrotechnology Department, and Experimental Garden, Agriculture Faculty, Syiah Kuala University Banda Aceh, and Laboratory of Chemical Analysis Mathematics Faculty and Natural Science, from September 2017 to January 2018. The materials used is spiny amaranth seed that obtained from Subdistrict of Delima, District of Pidie and babadotan leaves obtained from three regions with different altitude that is Subdistrict of Indrapuri, District of Aceh Besar (19 masl), Subdistrict of Subdistrict of, District of Aceh Besar (261 masl) and Subdistrict of Takengon District of Aceh Tengah (1259 masl) with each concentration 5, 10 and 15% as bioherbicide. This research using Randomized Completely Design with Factorial pattern which has 3 × 3 treatments and 3 replication. The first faktor is altitude and second is concentration of ethyl acetate extract. Ethyl acetate extracts that produced in different altitudes and concentrations were significantly influence the high rate of spiny amaranth, number of leaves, stem diameter, control percentage and root length. There is an interaction between altitude and concentration on the percentage of control at 7 and 14 HSA. Extracts produced at an altitude of 9 masl with a concentration of 10% can control spinach weeds up to 100% at 7 HSA.

Various species of Kalanchoe plants have been widely used as raw materials in traditional medicines. This study was the continuation of the researches on secondary metabolites from Indonesia Kalanchoe plants, focused on Kalanchoe blossfeldiana. Fresh K. blossfeldiana leaf was extracted with methanol at room temperature to obtain the concentrated extract. The concentrated methanol extract was dissolved in water and then partitioned successively with n-hexane and ethyl acetate. The methanol, n-hexane, ethyl acetate extracts were tested using antiplasmodial assay against Plasmodium falciparum 3D7. The IC50 of methanol, n-hexane, and ethyl acetate extract were 13.002; 2.807, and 11 nM, respectively. Ethyl acetate extract was separated by the combination of chromatography on silica and ODS. This process produced the yellow solid. The chemical structure of the compound was determined based on UV, IR, MS, 1H-NMR, and 13C-NMR analyses and the comparison of data obtained from the literature and identified as phenolic compound, namely 3,3',4',5,7-pentahydroxyflavone or quercetin (1), and displayed antiplasmodial activity with IC50 3.97x10-2 nM.

Background: In recent years, the study of the structure and biological activity of medicinal plants has a particularly important to search for diabetes medicine. Ruellia tuberosa is used to treat various diseases such as diabetes by inhibiting the activity of α-glucosidase. Objective: In this study, experiment was designed to isolated isolate and identified identify α-glucosidase inhibitory extracts and compounds from Ruellia tuberosa L. through bio-assay guided isolation. Method: Dry powder of Ruellia tuberosa L. was extracted with 70% ethanol, then liquid-liquid partition with n-hexane, ethyl acetate and butanol, respectively. The extracts were evaluated with α-glucosidase inhibition. The potential extracts were isolated and identified compounds. The effects of these compounds on the α-glucosidase inhibitory were evaluated. Results: The a-glucosidase inhibitory activities showed that the n-hexane, ethyl acetate and the butanol extract had the α-glucosidase inhibition with an IC50 of 46.2 0.9, 6.6 2.9 and 8.9  μg/mL, respectively. From the n-hexane and ethyl acetate extracts, the structures of four compounds were elucidated by NMR spectroscopic method, including lupeol (1), syringaresinol (2), apigenin (3), verbascoside (4). The a-glucosidase inhibitory activities showed that all isolated compounds were more active than the positive control - acarbose with an IC50 of 37.5  0.4; 19.5  0.2; 20.1  0.3; 69.3  0.2 µg/mL, respectively. Conclusion: The ethyl acetate extract showed strong activity about 19 times more than positive control - acarbose. The compound syringaresinol (2) was the most powerful α-glucosidase inhibitor. Therefore, Ruellia tuberosa L. contains many compounds that can inhibit α-glucosidase activity.

The objective of this study was to assess the ethyl acetate extracts of Gastrodia elata Blume (GEB) on vascular tone and the mechanisms involved. GEB was extracted with 95% EtOH followed by a further extraction with ethyl acetate. The effects of GEB and its ingredients on the isometric tensions of the aortic rings from rats were measured. The ethyl acetate extract of GEB induced a vasodilatory effect on rat aorta, which was partially dependent on endothelium. Four chemical compounds isolated from GEB were identified as 3,4-dihydroxybenzaldehyde (DB), 4-hydroxybenzaldehyde (HB), 4-methoxybenzyl alcohol (MA), and 4,4′-dihydroxydiphenyl methane (DM), respectively. All of these compounds induced vasodilatations, which were dependent on the endothelium to different degrees. After pretreatment with Nω-nitro-l-arginine methyl ester, indomethacin, or methylene blue, the vasodilatations induced by DB, HB, and MA were significantly decreased. In addition, the contractions of the rat aortic rings due to Ca2+ influx and intracellular Ca2+ release were also inhibited by DM. Furthermore, the administration of DB significantly enhanced the productions of nitric oxide (NO) and the activities of the endothelial NO synthase in aorta and in endothelial cells. Thus, GEB may play an important role in the amelioration of hypertension by modulating vascular tones.

Kasturi (Mangifera casturi) is a typical plant of South Kalimantan which has only been used for consumption because of its distinctive taste and aroma. Mangifera indica, which is in the same genus as Mangifera casturi, shows antioxidant, antibacterial, antifungal, and antidiarrheal activity, so it is suspected that this plant has the same activity. Crude methanol extract from the meat of Casturi mango was fractionated using n-hexane, ethyl acetate, and methanol-water. Testing for antibacterial activity and minimum inhibitory concentration (MIC) were carried out using the disk diffusion method (Kirby-Bauer). The positive control used was Chloramphenicol because it has an antibacterial range that is rather broad and the negative control used was 10% DMSO. The fraction that had the greatest antibacterial activity was the ethyl acetate fraction; with the formation of a clear zone of 7.98mm against Pseudomonas aeruginosa and 7.52mm against Bacillus cereus, with a minimum inhibitory concentration of 2mg/mL. The next test was a cell leakage analysis which revealed an increase in absorbance at MIC 1 and MIC 2. The Gas Chromatography – Mass Spectrometry (GC-MS) results from the ethyl acetate fraction revealed the presence of phenolic, flavonoid, and glycoide content, with the highest compound concentration of 5-Hydroxymethylfurfural. Mangifera indica yang masih satu genus dengan Mangifera casturi menunjukkan aktivitas antioksidan, antibakteri, antijamur, dan antidiare sehingga diduga tanaman ini memiliki aktivitas yang sama. Ekstrak kasar metanol daging buah mangga kasturi difraksinasi dengan menggunakan n-heksan, etil asetat, dan metanol-air. Pengujian aktivitas antibakteri dan kadar hambat minimum dilakukan dengan menggunakan metode difusi cakram (Kirby- Bauer). Kontrol positif yang digunakan adalah Chloramfenikol karena memiliki spektrum antibakteri yang cukup luas dan kontrol negatif yang digunakan adalah DMSO 10%. Fraksi yang memiliki aktivitas antibakteri terbesar pada penelitian ini adalah fraksi etil asetat dengan terbentuknya zona bening sebesar 7,98mm terhadap Pseudomonas aeruginosa dan 7,52mm terhadap Bacillus cereus dengan kadar hambat minimum 2mg/mL. Uji selanjutnya adalah uji kebocoran sel yang menyatakan adanya peningkatan absorbansi pada KHM 1 dan KHM 2. Hasil pemeriksaan Gas Chromatography – Mass Spectrometry (GCMS) dari fraksi etil asetat menyatakan adanya kandungan fenolik, flavonoid, dan glikosida dengan senyawa tertinggi yaitu 5-Hydroxymethylfurfural. Disease cancer caused by abnormal growth of tissue where there has been an error, fast and out of control. Judging from the fact of gender, more than 270,000 women die every year caused by cervical cancer. To inhibit the growth of cancer cells, a compound is needed that causes the cell cycle to stop so that the ability of cell proliferation decreases. Alkaloid compounds can inhibit proliferation through oxidative inhibition processes that can cause cancer. Mangrove plants have potential as anticancer, antimicrobial, and antioxidant. The content of chemical compounds found in mangroves are flavonoids, steroids, alkaloids, phenolites, saponins and tannins. These compounds show high antioxidant activity and are shown to have a real relationship with the properties of the material's bioactivity against cancer cells. One of the mangrove species is Rhizophora apiculata. The purpose of this study was to determine the IC50 value produced by Rhizophora apiculata mangrove leaf extract on HeLa cell viability and to see the effect of Rhizophora apiculata mangrove leaf extract dosage on HeLa cell viability. The method used in this research is the experimental method. The research parameters included yield, proximate test, phytochemical test, toxicity test, total phenol test, cytotoxicity test and LC-MS test. The experimental design used was a simple and complex completely randomized design (CRD) with the Tukey test.The results of this study showed that the highest yield was in the ethanol extract of 5.91%, while the n-hexane and ethyl acetate extracts respectively had yields of 1.18% and 1.31%. The results of the proximate test on the water content of leaves and powder were 64.53% and 13.86%, respectively, the results of the ash content in the leaves and powder of Rhizophora apiculata were 3.94% and 8.41%, respectively. while the water content in the extract obtained the highest yield in the ethanol extract of 21.42%, while the n-hexane extract and ethyl acetate extract were 11.08% and 15.42%, respectively. For phytochemical results, it was found that n-hexane extract only contained alkaloids, flavonoids and steroids. Ethyl acetate extract contains steroid compounds. Meanwhile, the ethanol extract contains the most bioactive compounds, namely saponins, flavonoids, tannins and triterpenoids. The toxicity test using the Brine Shrimp Lethality Test (BSLT) method resulted in the lowest IC50 of ethanol extract at 49.45 ppm while the n-hexane and ethyl acetate extracts were 251.63 ppm and 920.45 ppm respectively. In the total phenol test, the n-hexane extract was 66.79 mg GAE / 100 gr, 222.97 mg GAE / 100 gr ethyl acetate extract and 929.04 mg GAE / 100 gr ethanol extract. HeLa cell cytotoxicity testing using the MTT method (3- (4,5-dimethiltiazol-2-yl) -2,5-dipheniltetra zolium bromide) assay resulted in the highest cell viability value at a dose of 125 ppm of 46.97%. As for the doses of 250 ppm, 500 ppm 1000 ppm, and 2000 ppm resulted in a percentage of viability of 42.95% 37.70% 35.82% and 32.12%, respectively. The IC50 value of Rhizophora apiculata leaf extract was 64.42 ppm. This value indicates that the Rhizophora apiculata extract is toxic to HeLa cells.

AbstractCrinum asiaticum is an evergreen bulbous perennial shrub of Amaryllidaceae family with ethnomedicinal importance and our earlier study described a comparative account antimicrobial and antioxidant properties of the different solvent-mediated sequential extract fractions. The present study aimed to analyze their comparative account of induced phytotoxicity and chironomid toxicity. For phytotoxicity assessment, germination inhibition and seedling’s root and shoot growth retardation effects on Triticum aestivum and Cicer arietinum were analyzed and for lethal concentration determination, the freshwater bottom-dwelling first instars chironomid larvae were used. The crude aqueous, petroleum ether and ethyl acetate extracts showed significant toxic effects on both meristematic tissue and aquatic midges. The phytotoxic assays indicate that the ethyl acetate fraction contains the most potent growth inhibitors, followed by the crude aqueous and petroleum ether fractions. The last aqueous fraction was found to be the least toxic, the highest LC50 and LT50 values and ethyl acetate extract fraction having highest toxicity. Thus the present study supplements to our earlier report, that indicated the last aqueous extract fraction of C. asiaticum has potent antioxidant and antibacterial potentials as well as its prospective use in livestock maintenance, as it is least toxic and the ethyl acetate extract, the most toxic fraction identified here, is needed to explore for pharmaceutical importance.

This study was started by extraction of Eucalyptus deglupta Blume. Using organic solvent   (n-hexane, ethyl acetate, ethanol and water) followed by phytochemical screening and toxicity test using Brine Shrimp Lethality Test (BSLT) method. Isolation and identification of chemical compounds contained in the fraction were done by column chromatography and Gas Chromatography-Mass Spectrometry (GC-MS) analysis. Phytochemical screening revealed the presence of alkaloids, flavonoids, steroids and phenolics in the extract. Toxicity test results showed that the ethyl acetate extract was potentially active with LC50 value of  617.95 ppm. The extract was continued to isolation stage and gave fraction EKEA-3.1 with LC50 value of 2759.93 ppm. Identification of chemical compounds in EKEA-3.1 with KG-MS analysis showed that EKEA-3.1 was suspected to be Stigmastan-3,5-diene.

Objective: The objective of the present investigation was to perform the Gas Chromatography-mass spectrometry (GCMS) analysis of endophytic fungi Curvularia aeria MTCC-12847 isolated from Tribulus terrestris L. to find out the active compound present in the extract. Methods: The endophytic fungi were isolated from the plant Tribulus Terrestris L., leaf which was cultivated in optimized media for the production of secondary metabolites and was extracted using ethyl acetate. Ethyl acetate extract was used for the Gas Chromatography-mass spectrometry (GCMS) analysis. Results: GC-MS analysis of ethyl acetate extract of endophytic fungi revealed the presence of various secondary metabolites, the highest amount present was Palmitic acid (24.54%) and Lowest was Dimethyl 1-phenyl-7-methyl-1-hydroxy-1,4-dihydronaphthalene-2,3-dicarboxylate (5.76%). Conclusion: The endophytic fungal Curvularia aeria MTCC-12847 extract isolated from the Tribulus terrestris L. shows the presence of various bioactive compounds.

A total of 17 endophytic fungal isolates were obtained from the leaves of Mentha cordifolia Opiz (Lamiaceae). Seven isolates were identified to the level of genus by using taxonomically relevant morphological traits. Colletotrichum and Phomopsis species were dominant among these strains. All strains were separated from M. cordifolia leaf for the first time. The ethyl acetate extracts of all endophytic fungi were tested for antibacterial activity against Salmonella typhimurium TISTR1166 and Pseudomonas aeruginosa TISTR781. Most endophytes exhibited antibacterial activity. Ustilago sp. MFLUCC15-1024 presented the highest inhibition zone diameter with a MIC of 31.25 μg/mL against the tested pathogens. The chemical composition of the ethyl acetate extract of this strain was investigated using gas chromatography-mass spectrometry. Twenty-one components were identified. 2-Phenylethanol (38.7%), E-ligustilide (12.4%), α-eudesmol (10.2%), β-vetivone (4.6%), β-ylangene (3.7%) and verbanol (3.4%) were the major components of the extract. The strong antibacterial activity of Ustilago sp. MFLUCC15-1024 ethyl acetate extract may be attributed to the presence of a high concentration of bioactive compounds including phenyl ethyl alcohol, E-ligustilide and α-eudesmol. The results indicate that there is high diversity of endophytic fungi in M. cordifolia leaf, and that Ustilago sp. MFLUCC15-1024 strain could be an excellent resource of natural antibacterial compounds.
