Functional Connectivity Between Auditory Areas Field L and CLM and Song System Nucleus HVC in Anesthetized Zebra Finches

A key discovery that has emerged from studies of the vocal system in songbirds is that neurons in these regions respond preferentially to playback of the bird's own song (BOS). This BOS selectivity is not a general property of neurons in primary and secondary auditory forebrain regions, field L and caudolateral mesopallium (CLM). Moreover, anatomical studies have been unable to conclusively define a direct projection from field L and/or CLM to HVC, a central structure for integrating sensory and motor information in the vocal system. To examine the communication between these regions, we used simultaneous dual-electrode recording in anesthetized male zebra finches and cross-correlation analysis to estimate the functional connectivity between auditory areas, field L and CLM, and HVC. We found that ≥18% of neurons in field L and 33% of neurons in CLM are functionally connected to HVC, most with auditory forebrain leading-HVC latencies ranging from 0.5 to 15 ms. These results indicate that field L and CLM communicate extensively with HVC through both direct and indirect anatomical connections. To further explore the role of the auditory forebrain cells that are functionally connected with HVC, we assessed their responsiveness and selectivity for a variety of natural and synthetic auditory stimuli. We found that field L and CLM neurons that are functionally connected to HVC exhibit generic auditory forebrain properties including the lack of BOS selectivity. This finding puts further constraints on the neural architecture and the nature of the nonlinearity that leads to BOS-selective auditory responses in the vocal control nuclei.

AbstractMuch as humans acquire speech in early childhood, the zebra finch (Taeniopygia guttata) songbird learns to sing from an adult “tutor” during the first three months of life. Within a well-defined critical period (CP), juvenile zebra finches memorize a tutor song that will guide subsequent motor patterning. This sensory learning process is mediated by tutor experience-dependent neuroplasticity within the auditory forebrain. Here, we used longitudinal resting-state fMRI analyses to investigate whether tutor experience also modifies patterns of functional connectivity (FC) within the juvenile zebra finch brain. Eighteen male zebra finches (only males sing) were scanned before, during, and at the end of the CP, as well as at the young adult stage. Prior to the onset of the CP, birds were separated into rearing conditions: Normal (aviary-housed; N=5), Tutored (one adult male tutor and one adult female; N=7), and Isolate (two adult females, isolated from male song; N=6). Brain-wide voxel-wise analyses identified a single cluster overlapping the left caudomedial nidopallium (NCM) of the auditory forebrain that showed developmentally decreasing FC strength in Isolates but stable or increasing FC in Normal and Tutored birds. Additionally, FC between left NCM and left dorsal cerebellum showed a parallel developmental difference. Developmental changes in left NCM FC strength statistically mediated condition-related differences in song stereotypy. These results extend previous reports of tutor experience-dependent plasticity in NCM at epigenetic, genomic, molecular, and cellular levels to the whole-brain functional network level by demonstrating that tutor experience also influences the development of NCM FC. Moreover, these results link NCM FC to the emergence of song stereotypy.

AbstractIntegration of information across the senses is critical for perception and is a common property of neurons in the cerebral cortex, where it is thought to arise primarily from corticocortical connections. Much less is known about the role of subcortical circuits in shaping the multisensory properties of cortical neurons. We show that stimulation of the whiskers causes widespread suppression of sound-evoked activity in mouse primary auditory cortex (A1). This suppression depends on the primary somatosensory cortex (S1), and is implemented through a descending circuit that links S1, via the auditory midbrain, with thalamic neurons that project to A1. Furthermore, a direct pathway from S1 has a facilitatory effect on auditory responses in higher-order thalamic nuclei that project to other brain areas. Crossmodal corticofugal projections to the auditory midbrain and thalamus therefore play a pivotal role in integrating multisensory signals and in enabling communication between different sensory cortical areas.

AbstractThe present study is interested in the role of the corpus callosum in the development of the language network. We, therefore, investigated language abilities and the language network using task-based fMRI in three cases of complete agenesis of the corpus callosum (ACC), three cases of partial ACC and six controls. Although the children with complete ACC revealed impaired functions in specific language domains, no child with partial ACC showed a test score below average. As a group, ACC children performed significantly worse than healthy controls in verbal fluency and naming. Furthermore, whole-brain ROI-to-ROI connectivity analyses revealed reduced intrahemispheric and right intrahemispheric functional connectivity in ACC patients as compared to controls. In addition, stronger functional connectivity between left and right temporal areas was associated with better language abilities in the ACC group. In healthy controls, no association between language abilities and connectivity was found. Our results show that ACC is associated not only with less interhemispheric, but also with less right intrahemispheric language network connectivity in line with reduced verbal abilities. The present study, thus, supports the excitatory role of the corpus callosum in functional language network connectivity and language abilities.
