Combined therapy using photothermal and photodynamic treatments together with chemotherapeutic agents is considered one of the most synergistic treatment protocols to ablate hypoxic tumors. Herein, we sought to fabricate an in situ-injectable PEG hydrogel system having such multifunctional effects. This PEG hydrogel was prepared with (i) nabTM-technique-based paclitaxel (PTX)-bound albumin nanoparticles with chlorin-e6 (Ce6)-conjugated bovine serum albumin (BSA-Ce6) and indocyanine green (ICG), named ICG/PTX/BSA-Ce6-NPs (~175 nm), and (ii) an albumin-stabilized perfluorocarbon (PFC) nano-emulsion (BSA-PFC-NEs; ~320 nm). This multifunctional PEG hydrogel induced moderate and severe hyperthermia (41−42 °C and >48 °C, respectively) at the target site under two different 808 nm laser irradiation protocols, and also induced efficient singlet oxygen (1O2) generation under 660 nm laser irradiation supplemented by oxygen produced by ultrasound-triggered PFC. Due to such multifunctionality, our PEG hydrogel formula displayed significantly enhanced killing of three-dimensional 4T1 cell spheroids and also suppressed the growth of xenografted 4T1 cell tumors in mice (tumor volume: 47.7 ± 11.6 and 63.4 ± 13.0 mm3 for photothermal and photodynamic treatment, respectively, vs. PBS group (805.9 ± 138.5 mm3), presumably based on sufficient generation of moderate heat as well as 1O2/O2 even under hypoxic conditions. Our PEG hydrogel formula also showed excellent hyperthermal efficacy (>50 °C), ablating the 4T1 tumors when the irradiation duration was extended and output intensity was increased. We expect that our multifunctional PEG hydrogel formula will become a prototype for ablation of otherwise poorly responsive hypoxic tumors.

This dissertation reports the synthesis of unique Si0₂ based nanostructures by exposing glass samples to MHz repletion rate femtosecond laser irradiation. A three-dimensional fibrous nanoparticle agglomerate network was observed on soda-lime glass (73% SiO + other compounds) when exposed to femtosecond laser irradiation at 8.4 MHz and 12.6 MHz repetition rate and 0.5 ms dwell time, in air. By irradiating silica glass (96% SiO₂+ trace elements) sample under ambient conditions with femtosecond pulses at 12.6 MHz and dwell time in excess of 3.0 ms; long continuous nanofibers of extremely high aspect ration (certain fibers up to 100000:1) were obtained. The mechanisms that promote such nanostructures with distinct morphologies have been explored. A deeper insight into the fundamentals of femtosecond laser interaction with dielectrics led to the understanding that variations in bandgap alters ablation dynamics and dictates the response of glass to femtosecond laser irradiation, ultimately resulting in the formation of structures with dissimilar morphology on silica and soda-lime glass.

Detailed three-dimensional numerical simulations have been carried out to find the velocity and temperature fields, in combination with shear and normal stresses, of the fluid flow inside a rectangular channel with large aspect-ratio. The channel under analysis is aimed to cool a thermochromic liquid crystal material (TLC) that is able to capture laser irradiation in the terahertz range. The TLC is manufactured on an extremely-thin substrate. The overall objective of the cooling system is to maintain a nearly-homogeneous temperature of the TLC-domain that is not exposed to the direct laser irradiation, while minimizing the deformation in the TLC caused by the fluid-solid interaction. The fluid flow, stress-strain and heat transfer simulations are carried out on the basis of three-dimensional Navier-Stokes and energy equations for an incompressible flow, coupled with the stress-strain equation for the TLC-layer, to determine values of velocity, pressure and temperature for the fluid inside the channel and the stresses and deformation of the TLC layer, under different operating conditions. These values are then used to find, from a specific set, the value of the channel gap that enables a nearly-uniform temperature distribution in the fluid and the least amount of deformation in the solid layer, within the expected operating conditions. Results from this analysis indicate that, for all the inlet velocities considered, there is a common value of the channel gap, that represents the optimum for the cooling system.
