Corneal anatomy of marine mammals

The corneal anatomy of fin whales (Balaenoptera physalus), minke whales (Balaenoptera acutorostrata), harp seals (Phoca groenlandica), ringed seals (Phoca hispida), and bearded seals (Eriganthus barbatus) was examined to determine if marine mammals have evolved specialized corneas for life in a marine habitat. One to seven eyes of each species were analyzed: paraffin sections stained with haematoxylin and eosin for light microscopy; and ultrathin sections for transmission electron microscopy. All corneas contain the five typical mammalian layers: epithelium, Bowman's layer, stroma, Descemet's membrane, and endothelium. The corneas of these marine mammals are thicker than human corneas because of a thicker stromal layer. The other layers are thinner than those found in humans, except for the epithelial layer in the bearded seal and the cetaceans where it may provide extra protection for the eye during feeding behaviour. The epithelial cells in all corneas studied have an abundance of tonofilaments, which may strengthen the cells and distribute force across the corneal surface. No special organization of collagen fibrils was found in the stroma that would offer protection from ultraviolet radiation or glare for pinnipeds when on ice. The thickness of the sclera in the cetaceans may serve to hold the inner globe of the eye in an elliptical shape, while the thinning of the sclera in the equatorial region in pinnipeds may flatten the eye in air to reduce aerial myopia.

This paper documents net entrapments of baleen whales in the inshore waters of Newfoundland and Labrador. The species involved are principally minke whales (Balaenoptera acutorostrata) and humpback whales (Megaptera novaeangliae). Finback whales (Balaenoptera physalus) are also caught in nets but far less frequently. The timing of these entrapments corresponds with the spawning season for capelin (Mallotus villosus) on the beaches around the province. The magnitude and implications of this problem, both to the whales and the fisherman, are assessed. Many marine mammals rely on sound for foraging, maintaining group cohesion, navigation, finding mates, and avoiding predators. These behaviors are potentially disrupted by anthropogenic noise. Behavioral responses to sonar have been observed in a number of baleen whale species but relatively little is known about the responses of minke whales (Balaenoptera acutorostrata). Previous analyses demonstrated a spatial redistribution of localizations derived from passive acoustic detections in response to sonar activity, but the lack of a mechanism for associating localizations prevented discriminating between movement and cessation of calling as possible explanations for this redistribution. Here we extend previous analyses by including an association mechanism, allowing us to differentiate between movement responses and calling responses, and to provide direct evidence of horizontal avoidance responses by individual minke whales to sonar during U.S. Navy training activities. We fitted hidden Markov models to 627 tracks that were reconstructed from 3 years of minke whale (B. acutorostrata) vocalizations recorded before, during, and after naval training events at the U.S. Navy’s Pacific Missile Range Facility, Kauai, Hawaii. The fitted models were used to identify different movement behaviors and to investigate the effect of sonar activity on these behaviors. Movement was faster and more directed during sonar exposure than in baseline phases. The mean direction of movement differed during sonar exposure, and was consistent with movement away from sonar-producing ships. Animals were also more likely to cease calling during sonar. There was substantial individual variation in response. Our findings add large-sample support to previous demonstrations of horizontal avoidance responses by individual minke whales to sonar in controlled exposure experiments, and demonstrate the complex nature of behavioral responses to sonar activity: some, but not all, whales exhibited behavioral changes, which took the form of horizontal avoidance or ceasing to call.

Experienced observers from the community of Rigolet, Newfoundland and Labrador, recorded marine mammal observations at Rigolet on the coast of Labrador from July to September 2012. During this study, a total of nine species of marine mammals were documented. A daily average of 2.6 (range 1–5) marine mammal species were observed, with Harp Seals (Pagophilus groenlandicus) and Common Minke Whales (Balaenoptera acutorostrata) being the most common. The area is thought to be regionally important for Ringed Seal (Pusa hispida) whelping; however, Ringed Seals were not observed in large numbers. Beluga Whales (Delphinapterus leucas) were also reported anecdotally, but were less common than the other species, with the possible exception of the Harbour Porpoise (Phocoena phocoena). Labrador is experiencing significant industrial growth related to hydroelectric development and offshore oil and gas exploration. These data should aid environmental assessment and environmental effects monitoring and assist researchers in understanding regional marine mammal ecology, including species diversity, seasonal occurrence, and relative abundance.

An overview of the estimates of consumption by predators on the main fish stocks in the Barents Sea is given. The main predators are cod (Gadus morhua), harp seal (Phoca groenlandica) and minke whale (Balaenoptera acutorostrata). The results indicate that cod is the most important predator, consuming about as much food annually as harp seals and minke whales combined. The consumption estimates, together with data on the amount of fish removed by commercial fisheries, are compared to estimates of the abundance and removal through natural mortality of the various species of fish prey. The consistency between these estimates is discussed. The natural mortality values for cod and haddock used in assessments are found to be reasonably consistent with the consumption estimates. The consumption of capelin is found to be higher than what is available for predation in years of low capelin abundance, while in years of high herring abundance the consumption of herring does not explain all the mortality. The way in which the consumption estimates are and can be utilised in theassessment and management of fish stocks in the Barents Sea using multispecies models and approaches is described.

Seven cases of lymphocytic hypophysitis in women have been reported previously in association with various degrees of hypopituitarism. We report two pregnant patients who presented with mass lesions of the sella turcica, clinically mimicking pituitary adenoma. However, pathologic examination revealed extensive infiltration of the anterior pituitary by lymphocytes and plasma cells with destruction of the gland. To our knowledge, the ultrastructural features of lymphocytic hypophysitis have not been studied so far.For transmission electron microscopy, tissue from surgical specimens was fixed in glutaraldehyde, postfixed in OsO4, dehydrated and embedded in epoxy-resin. Ultrathin sections were stained with uranyl acetate and lead citrate and examined with a Philips 300 electron microscope.Electron microscopy revealed adenohypophysial cells of all types exhibiting varying degrees of injury. In the areas of most dense inflammatory cell infiltration pituitary cells contained large lysosomal bodies fusing with secretory granules (Fig. 1), as well as increased numbers of swollen mitochondria, indicating oncocytic transformation (Fig. 2).

Mast cell and basophil granules of the vertebrate contain heparin or related sulfated proteoglycans. Histamine is also present in mammalian mast cells and basophils. However, no histamine is detected in mast cell granules of the amphibian or fish, while it is shown in those of reptiles and birds A quantitative x-ray microanalysis of mast cell granules of fresh frozen dried ultrathin sections of the tongue of Wistar rats and tree frogs disclosed high concentrations of sulfur in rat mast cell granules and those of sulfur and magnesium in the tree frog granules. Their concentrations in tree frog mast cell granules were closely correlated (r=0.94).Fresh frozen dried ultrathin sections and fresh air-dried prints of the tree frog tongue and spleen and young red-eared turtle (ca. 6 g) spleen and heart blood were examined by a quantitative energy-dispersive x-ray microanalysis (X-650, Kevex-7000) for the element constituents of the granules of mast cells and basophils. The specimens were observed by transmission electron microscopy (TEM) (80-200 kV) and followed by scanning transmission electron microscopy (STEM) under an analytical electron microscope (X-650) at an acceleration voltage of 40 kV and a specimen current of 0.2 nA. A spot analysis was performed in a STEM mode for 100 s at a specimen current of 2 nA on the mast cell and basophil granules and other areas of the cells. Histamine was examined by the o-phthalaldehyde method.

In secretory cells a dense and complex network of actin filaments is seen in the subplasmalemmal space attached to the cell membrane. During exocytosis this network is undergoing a rearrangement facilitating access of granules to plasma membrane in order that fusion of the membranes can take place. A filamentous network related to secretory granules has been reported, but its structural organization and composition have not been examined, although this network may be important for exocytosis.Samples of peritoneal mast cells were frozen at -70°C and thawed at 4°C in order to rupture the cells in such a gentle way that the granule membrane is still intact. Unruptured and ruptured cells were fixed in 2% paraformaldehyde and 0.075% glutaraldehyde, dehydrated in ethanol. For TEM (transmission electron microscopy) cells were embedded in Lowicryl K4M at -35°C and for SEM (scanning electron microscopy) they were placed on copper blocks, critical point dried and coated. For immunoelectron microscopy ultrathin sections were incubated with monoclonal anti-actin and colloidal gold labelled IgM. Ruptured cells were also placed on cover glasses, prefixed, and incubated with anti-actin and colloidal gold labelled IgM.

Mammals are known to utilize wax esters with an efficiency of less than 50%. The purpose of the present study was to examine whether or not minke whales (Balaenoptera acutorostrata), which at times may eat considerable amounts of wax-ester-rich krill, represent an exception to this general pattern. Samples of fresh undigested forestomach, as well as colon, contents were obtained from minke whales (n5) that had been feeding on krill (Thysanoessa inermis) for some time. The samples were analysed for dry mass, energy density, lipid content and the major lipid classes, including wax esters. The concentrations of wax esters were compared with previous estimates of dry-matter disappearance of the same type of prey using anin vitrotechnique, to calculate the dry-matter digestibility of wax esters (DMDwax). Wax esters contributed 21% of the energy and 47% of total lipids in the krill diet. The energy density of gut contents decreased by 50% after their passage from forestomach to the end of the colon. The DMDwaxwas 94·1 (SD 2·8)% (n5). This high DMDwaxand the occurrence of fatty alcohols, one of the products of wax-ester hydrolysis, in faeces show that minke whales are very efficient digesters of wax esters and absorb most of the energy-rich products of this process.
