Metacestodes of moose, Alces alces, of the Chapleau Crown Game Preserve, Ontario

Lungs, liver, spleen, heart, and kidneys of moose, Alces alces, collected from 1963 to 1965 in the Chapleau Crown Game Preserve of northeastern Ontario were examined for parasites. Thirty-eight of 51 moose (75%) were infected with metacestodes of Taenia hydatigena Pallas, 1766; 40 of 54 (74%) with Taenia krabbei Moniez, 1879; and 36 of 54 (67%) with Echinococcus granulosus (Batsch, 1786). Twenty-two of 51 (43%) moose harboured all three species. Each species was more prevalent in older moose than in young moose and intensity of infections increased with the age of moose. Occurrence of small hydatid cysts decreased and large cysts increased with increasing age of moose. Of 1154 hydatid cysts, 95.3% were in lung tissue, 3.6% in liver, 0.9% in spleen, and 1 cyst (0.1%) was recovered from each of heart and kidney. Degenerate cysticerci of T. hydatigena and T. krabbei were observed in all age groups of moose. There was no apparent relationship between intensity of infection with metacestodes and physical condition of moose.

AbstractHydatid cysts were recovered from 35.2% (233/661) of camels (Camelus dromedarius) slaughtered in five different regions of Iran. The degree of prevalence between males (34.4%) and females (36.6%) was not statistically significant. The highest rate of infection (59.3%) was found in the Isfahan region (in the central part of Iran) while the lowest (25.7%) was found in Kerman province. The organ distribution of cysts was 49.4% in lungs alone, 30.0% in both liver and lungs, 14.6% in liver only and 6.0% in other organs. Therefore, the lungs were the predominant sites of the hydatid cyst. The range in the number of cysts was 1–48 in infected animals. The majority of the camels had 1–5 cysts, with 21.9%, 11.6% and 5.6% of infected camels having 6–10, 11–20 and 21 or more cysts respectively. There was a direct relationship between the rate and intensity of infection and host age. The fertility rate of lung cysts (69.7%) was higher than that of liver cysts (58.7%) and other organs (50.0%) whilst the viability rate of protoscoleces of liver fertile cysts (80.3%) was significantly higher than that of lung cysts (55.8%) and other organs (57.1%). The role of camels in the epidemiology of Echinococcus granulosus in Iran is discussed.

Abstract Background The species complex of Echinococcus granulosus sensu lato (s.l.) causes cystic echinococcosis distributed worldwide. There is no genotype information from hydatid cysts in the intermediate hosts in Central Iran. Therefore, in this study, we analyzed the hydatid cysts in livestock slaughtered in an abattoir in this region. Six hundred fifty-seven hydatid cysts were isolated from 97 animals, including sheep, cattle, camels, and goats slaughtered in Yazd abattoir from September 2018 to January 2020. The demographic data was collected as well as cyst location, fertility, and viability. Out of 657 samples, 164 samples were genotyped. Then, phylogenetic analysis was performed using MEGAX. Statistical analyses were done using SPSS version 16.0 by chi-square with a significant difference of less than 0.05. Results Out of 164 samples, the G1-G3 complex genotype had the most frequency in samples, with 135 cases recognized. The G6/G7 was observed in 19 isolates and G5 was reported in nine samples. One sample was detected as Taenia hydatigena. Conclusions This study showed that G1-G3 and G6/G7 genotypes were presented in all animals, but G5 was reported only in cattle, goats, and camels. It is the first molecular identification of cystic echinococcosis in Central Iran. Hence, reporting G5 in livestock in this area should be considered due to transmission to humans.

Livers of 164 white-tailed deer (Odocoileus virginianus) from the Peterborough Crown Game Preserve in central Ontario were examined for Fascioloides magna and Taenia hydatigena. Fascioloides magna was recovered from 2.6% of fawns and 68.3% of all older deer, and mean intensity of infection increased with age. Both prevalence and intensity of unencapsulated flukes were similar among all age-groups of deer 1 year of age and older. While the majority (96%) of encapsulated flukes were within capsules containing more than one fluke, aggregation of flukes is probably not required for either encapsulation or maturation. Infection with F. magna had no detectable effect on the condition of yearling or adult male deer, where condition was measured by depth of fat on the rump and sternum. Cysticerci of T. hydatigena were most prevalent among yearlings (70%) as compared with all other age-groups (29%). Mean intensity of infection with T. hydatigena tended to increase with age.

Echinococcus granulosus sensu lato (s.l.) is the causative agent of cystic echinococcosis in animals and humans. Different E. granulosuss.l. genotypes exhibit great diversity in their life cycle, host selectivity and pathogenicity. For this reason, the study of genetic variation within Echinococcus species is of importance for their epidemiological implication. We employed two SNP genotyping technologies to distinguish G1 and G3 E. granulosus sensu stricto (s.s.). genotypes. The genotypes of DNA samples (n = 28) extracted from hydatid cysts of different animal species were identified by amplification and sequencing of a fragment of the mitochondrial nad5 gene. Two SYBR green and three TaqMan real time PCR assays were developed for targeting of three nad5 informative positions (SNP758, 1123, and 1380) known to be able to discriminate G1 from G3. Genotyping by SYBR Green PCR based on cycle threshold (Ct) with melting temperature (Tm) analysis and performed on SNP1123 and SNP1380 failed to identify one DNA sample. TaqMan assays for SNP758, 1123 and 1380 effectively confirmed genotype identification obtained by Sanger sequencing. Our results demonstrated that the combination of the three Taqman assays developed in this study represents a valuable and cost effective tool alternative to DNA sequencing for E. granulosus s.s. genotyping.

The Italian wolf population in human-modified landscapes has increased greatly in the last few decades. Anthropisation increases the risk of transmission of many zoonotic infections and in this context, control of taeniid cestode species needs to be addressed from a One Health perspective. Predator-prey interactions are at the root of taeniid cestode transmission, and the wolf plays a key role in the maintenance and transmission of taeniids. To date, all available data on the taeniids of wolves in Italy refer to populations living in a wild habitat. Between 2018 and 2019, we investigated taeniids in a wolf pack living in a highly anthropic hilly agro-ecosystem. Thirty-eight faecal samples were collected and analysed, 4 of which were also genetically characterised for individual wolves and belonged to three different animals. Samples collected were analysed microscopically and by molecular analysis in order to identify the taeniid species. Taeniid eggs were detected in 34.2% (13/38) of samples. Within samples positive to taeniid eggs only Echinococcus granulosus s.s. and Taenia hydatigena were identified in 26.3% and 10.5% of the samples, respectively. On microscopic examination, Capillaria spp., Ancylostomatidae and Toxocara canis eggs, Crenosoma vulpis larvae, and coccidian oocysts were also found. The combination of low biodiversity of taeniid species with a high occurrence of E. granulosus s.s. recorded in this study could be the consequence of a deeper link occurring between wolves and livestock in human-modified landscapes than in wild settings.

Fifty-eight percent of woodland caribou (Rangifer tarandus caribou) of the George River herd, Labrador, were infected with up to 110 giant American liver flukes (Fascioloides magna). The prevalence and intensity of infection did not differ between 2 sampling years or between sexes. Prevalence tended to increase with age while intensity did not differ among age groups. Liver weight increased with intensity of infection but condition, as measured by the mean depth of back fat, did not differ between infected and uninfected animals. Caribou must be considered a suitable host of F. magna since the growth and maturation of the parasite and lesions produced in Rangifer resemble those in deer and wapiti.
