Sex and species differences in plasma oxytocin using an enzyme immunoassay

The neuropeptide hormone oxytocin (OT) is released peripherally and centrally and has been implicated in both physiology and behavior, especially sociosexual behaviors. Knowledge of OT levels in blood or other sources would be useful but these are rarely reported. Radioimmunoassay following extraction is the most commonly used method for measuring OT but is not ideal for use in small mammals in which blood volumes and concentrations of OT are low. Here we report a chemical and biological validation for a commercially available enzyme immunoassay for OT in unextracted plasma. In addition, comparisons of OT were made across species to allow comparison of the monogamous prairie vole (Microtus ochrogaster (Wagner, 1842)) to the polygynous Sprague Dawley rat. These species were chosen because OT plays a role in the formation of social bonds and we predicted that the highly social prairie vole would have higher plasma OT than the less social rat. Results of this comparison confirmed our hypothesis. Further, OT was significantly higher in females than in males in both species. Our results indicate that this enzyme immunoassay can be used to assay plasma OT in rodents and that the predicted correlations exist between plasma OT and gender as well as species-typical social behavior.

The prairie vole (Microtus ochrogaster) is an important model for the study of social monogamy and dual parental care of offspring. Characterization of specific host species-microbe strain interactions is critical for understanding the effects of the microbiota on mood and behavior. The five metagenome-assembled genome sequences reported here represent an important step in defining the prairie vole microbiome.

The introduction first sets out some preliminary definitions of sex, sexuality, and gender. It then turns from the sexual part of Sexual Identities to the identity part. A great deal of confusion results from failing to distinguish between identity in the sense of a category with which one identifies (categorial identity) and identity in the sense of a set of patterns that characterize one’s cognition, emotion, and behavior (practical identity). The second section gives a brief summary of this difference. The third and fourth sections sketch the relation of the book to social constructionism and queer theory, on the one hand, and evolutionary-cognitive approaches to sex, sexuality, and gender, on the other. The fifth section outlines the value of literature in not only illustrating, but advancing a research program in sex, sexuality, and gender identity. Finally, the introduction provides an overview of the chapters in this volume.

Morality and gender are intersecting realms of human thought and behavior. Reasoning and action at their intersection (e.g., views of women’s rights legislation) carry important consequences for societies, communities, and individual lives. In this chapter, the authors argue that children’s developing views of morality and gender reciprocally shape one another in important and underexplored ways. The chapter begins with a brief history of psychological theory and research at the intersection of morality and gender and suggests reasons for the historical failure to view gender attitudes through moral lenses. The authors then describe reasons for expecting morality to play an important role in shaping children’s developing gender attitudes and, reciprocally, for gender attitudes to play an important role in shaping children’s developing moral values. The authors next illustrate the importance and relevance of these ideas by discussing two topics at the center of contentious debate in the United States concerning ethical policy and practice: treatment of gender nonconformity and gender-segregated schooling. The chapter concludes with suggestions for future research.

ABSTRACTObjectives: The purpose of this study was to evaluate self-reported oral health attitude and behavior among a group of Turkish dental students and to compare differences in oral health attitudes between years of study and gender.Methods: This study included 267 (153 female, 114 male) dental students. A modified English version of Hiroshima University Dental-Behavioral Inventory (HU-DBI) which consists of twenty-eight dichotomous responses (yes-no) was used.Results: Totally 141 preclinical (1, 2 and 3rd years of study) and 126 clinical students (4 and 5th years of study) who were mean age of 21.16 participated in the study. Statistically significant differences were found between years of study for brushing each of teeth carefully, cleaning the teeth well without using toothpaste, using a toothbrush which has hard bristles and for having had their dentist tell that they brush very well. There were statistically significant differences between females and males for using a toothbrush which has hard bristles and using tooth floss regularly. Statistically significant differences were found for brushing each of the teeth carefully and using mouth wash on regular basis between smokers and non-smokers.Conclusions: This study confirmed that oral and dental health behavior and attitudes and also their knowledge about oral and dental health care of dental students improved with increasing level of education while oral and dental health care of female students were better than males and oral and dental health care of non-smokers were better than smokers. Various molecular forms of CCK reduce food intake in rats. Although CCK-8 is the most studied form, we reported that CCK-58 is the only detectable endocrine peptide form in rats. We investigated the dark-phase rat chow intake pattern following injection of CCK-8 and CCK-58. Ad libitum-fed male Sprague-Dawley rats were intraperitoneally injected with CCK-8, CCK-58 (0.6, 1.8, and 5.2 nmol/kg), or vehicle. Food intake pattern was assessed during the dark phase using an automated weighing system that allowed continuous undisturbed monitoring of physiological eating behavior. Both CCK-8 and CCK-58 dose dependently reduced 1-h, dark-phase food intake, with an equimolar dose of 1.8 nmol being similarly effective (−49% and −44%). CCK-58 increased the latency to the first meal, whereas CCK-8 did not. The intermeal interval was reduced after CCK-8 (1.8 nmol/kg, −41%) but not after CCK-58. At this dose, CCK-8 increased the satiety ratio by 80% and CCK-58 by 160%, respectively, compared with vehicle. When behavior was assessed manually, CCK-8 reduced locomotor activity (−31%), whereas grooming behavior was increased (+59%). CCK-58 affected neither grooming nor locomotor activity. In conclusion, reduction of food intake by CCK-8 and CCK-58 is achieved by differential modulation of food intake microstructure and behavior. These data highlight the importance of studying the molecular forms of peptides that exist in vivo in tissue and circulation of the animal being studied.

Elections and pandemics highlight how much one’s safety depends on fellow community members, a realization that is especially threatening when this collective perceives political realities inconsistent with one’s own. Two longitudinal studies examined how people restored safety to social bonds when everyday experience suggested that fellow community members inhabited inconsistent realities. We operationalized consensus political realities through the negativity of daily nationwide social media posts mentioning President Trump (Studies 1 and 2), and the risks of depending on fellow community members through the pending transition to a divided Congress during the 2018 election season (Study 1), and escalating daily U.S. COVID-19 infections (Study 2). On days that revealed people could not count on fellow community members to perceive the same reality of President Trump’s stewardship they perceived, being at greater risk from the judgment and behavior of the collective community motivated people to find greater happiness in their family relationships.

Background. Recovery of motor function after stroke appears to be related to the integrity of axonal connections in the corticospinal tract (CST) and corpus callosum, which may both be affected after cortical stroke. Objective. In the present study, we aimed to elucidate the relationship of changes in measures of the CST and transcallosal tract integrity, with the interhemispheric functional connectivity and sensorimotor performance after experimental cortical stroke. Methods. We conducted in vivo diffusion magnetic resonance imaging (MRI), resting-state functional MRI, and behavior testing in twenty-five male Sprague Dawley rats recovering from unilateral photothrombotic stroke in the sensorimotor cortex. Twenty-three healthy rats served as controls. Results. A reduction in the number of reconstructed fibers, a lower fractional anisotropy, and higher radial diffusivity in the ipsilesional but intact CST, reflected remote white matter degeneration. In contrast, transcallosal tract integrity remained preserved. Functional connectivity between the ipsi- and contralesional forelimb regions of the primary somatosensory cortex significantly reduced at week 8 post-stroke. Comparably, usage of the stroke-affected forelimb was normal at week 28, following significant initial impairment between day 1 and week 8 post-stroke. Conclusions. Our study shows that post-stroke motor recovery is possible despite degeneration in the CST and may be supported by intact neuronal communication between hemispheres.
