Temperature and ontogenetic effects on color change in the larval salamander species Ambystoma barbouri and Ambystoma texanum

Temperature has been shown to affect body color in several species of amphibians. The interaction between color and temperature may also change over larval ontogeny, perhaps because of age-related or seasonal changes in selection pressures on color. We quantified the effects of temperature on the color of the salamander sister species Ambystoma barbouri and Ambystoma texanum over larval ontogeny. We found that early-stage larvae responded to cold temperatures with a dark color relative to that of the warm temperature response. Both species then exhibited an ontogenetic shift in larval color, with larvae becoming lighter with age. Interestingly, older larvae showed decreased plasticity in color change to temperature when compared with younger stages. Older A. barbouri larvae showed no color response to the two temperature treatments, whereas older A. texanum larvae exhibited a reversal in the direction of color change, with cold temperatures inducing a lighter color relative to warm temperatures. We suggest that the overall pattern of color change (a plastic color response to temperature for young larvae, a progressive lightening of larvae over development, and an apparent loss of color plasticity to temperature over ontogeny) can be plausibly explained by seasonal changes in environmental factors (temperature, ultraviolet radiation) selecting for body color.

As age advances, breast cancer (BC) tends to change its biological characteristics. This study aimed to explore the natural progression of such changes. The study included 2383 women with clinically T0-2N0-1M0 BC, managed by primary surgery and optimal adjuvant therapy in a dedicated BC facility. Tissue micro-arrays were constructed from their surgical specimens and indirect immunohistochemistry was used for analysis of a large panel (n = 16) of relevant biomarkers. There were significant changes in the pattern of expression of biomarkers related to luminal (oestrogen receptor (ER), progesterone receptors (PgR), human epidermal growth factor receptor (HER-2), E-cadherin, MUC1, bcl2 CK7/8, CK18 and bcl2) and basal (CK5/6, CK14, p53 and Ki67) phenotypes, lymph node stage, histological grade and pathological size when decade-wise comparison was made (p < 0.05). The ages of 40 years and 70 years appeared to be the milestones marking a change of the pattern. There were significantly higher metastasis free and breast cancer specific survival rates among older women with ER positive tumours while there was no significant difference in the ER negative group according to age. Biological characteristics of BC show a pattern of change with advancing age, where 40 years and 70 years appear as important milestones. The pattern suggests <40 years as the phase with aggressive phenotypes, >70 years as the less aggressive phase and 40–70 years being the transitional phase.

For many years, the dogma has been that insulin resistance precedes the development of hyperinsulinemia. However, recent data suggest a reverse order and place hyperinsulinemia mechanistically upstream of insulin resistance. Genetic background, consumption of the “modern” Western diet and over-nutrition may increase insulin secretion, decrease insulin pulses and/or reduce hepatic insulin clearance, thereby causing hyperinsulinemia. Hyperinsulinemia disturbs the balance of the insulin–GH–IGF axis and shifts the insulin : GH ratio towards insulin and away from GH. This insulin–GH shift promotes energy storage and lipid synthesis and hinders lipid breakdown, resulting in obesity due to higher fat accumulation and lower energy expenditure. Hyperinsulinemia is an important etiological factor in the development of metabolic syndrome, type 2 diabetes, cardiovascular disease, cancer and premature mortality. It has been further hypothesized that nutritionally driven insulin exposure controls the rate of mammalian aging. Interventions that normalize/reduce plasma insulin concentrations might play a key role in the prevention and treatment of age-related decline, obesity, type 2 diabetes, cardiovascular disease and cancer. Caloric restriction, increasing hepatic insulin clearance and maximizing insulin sensitivity are at present the three main strategies available for managing hyperinsulinemia. This may slow down age-related physiological decline and prevent age-related diseases. Drugs that reduce insulin (hyper) secretion, normalize pulsatile insulin secretion and/or increase hepatic insulin clearance may also have the potential to prevent or delay the progression of hyperinsulinemia-mediated diseases. Future research should focus on new strategies to minimize hyperinsulinemia at an early stage, aiming at successfully preventing and treating hyperinsulinemia-mediated diseases.

Intravitreal injection of anti-vascular endothelial growth factor (anti-VEGF) is essential for the treatment of macular diseases such as wet age-related macular degeneration and macular edema. Although continued treatment is needed to maintain good vision, some patients cannot continue such injections for various reasons, including specific phobias. Here, we report a case of a patient with a specific phobia of intravitreal injections who could resume treatment after undergoing combined drug and cognitive-behavioral therapy (CBT). A 74-year-old Japanese man diagnosed with retinal angiomatous proliferation by fluorescein angiography and indocyanine green angiography was treated with intravitreal anti-VEGF injection. However, at 8 months after the first treatment, he became difficult to treat because of a phobia of injections. He was treated with photodynamic therapy, but his macular edema did not improve. After a psychiatric consultation, he was diagnosed with a specific phobia of intravitreal injections. Combined drug and CBT enabled him to resume receiving intravitreal injections. This case demonstrates that a specific phobia of intravitreal injections may benefit from combined drug and CBT. In this regard, some patients with high anxiety and fear of intravitreal injections should be referred to a psychiatrist at an early stage.

Denitrifying woodchip bioreactors (WBR), which aim to reduce nitrate (NO3−) pollution from agricultural drainage water, are less efficient when cold temperatures slow down the microbial transformation processes. Conducting bioaugmentation could potentially increase the NO3− removal efficiency during these specific periods. First, it is necessary to investigate denitrifying microbial populations in these facilities and understand their temperature responses. We hypothesized that seasonal changes and subsequent adaptations of microbial populations would allow for enrichment of cold-adapted denitrifying bacterial populations with potential use for bioaugmentation. Woodchip material was sampled from an operating WBR during spring, fall, and winter and used for enrichments of denitrifiers that were characterized by studies of metagenomics and temperature dependence of NO3− depletion. The successful enrichment of psychrotolerant denitrifiers was supported by the differences in temperature response, with the apparent domination of the phylum Proteobacteria and the genus Pseudomonas. The enrichments were found to have different microbiomes’ composition and they mainly differed with native woodchip microbiomes by a lower abundance of the genus Flavobacterium. Overall, the performance and composition of the enriched denitrifying population from the WBR microbiome indicated a potential for efficient NO3− removal at cold temperatures that could be stimulated by the addition of selected cold-adapted denitrifying bacteria.

AbstractEpoxy polymer with damage indicating ability was very usable for ships and bridges to detect the cracks at an early stage and to prevent corrosion. 2′, 7′-dichlorofluorescein (DCF), as a damage indicator, was used to report the mechanical damage of epoxy-amine polymer by a strong color change from a light yellow to bright red due to the molecular structure transition from the acid molecular form to the base ion form. The effect of water on damage indicator and damaged epoxy-amine polymer film was evaluated by an immersion test and the properties were characterized by ultraviolet-visible spectrophotometry (UV-Vis), scanning electronic microscopy (SEM), energy dispersive X-ray spectrometer (EDS), zeta potential and thermal gravimetric analysis (TGA). The results showed that DCF was an easy, stable and permanent indicator for epoxy-amine polymer and the water only had a slight influence on the indication stability of damaged epoxy polymer.

White matter hyperintensity (WMH) is common in healthy adults in their 60s and can be seen as early as in their 30s and 40s. Alterations in the brain structural and functional profiles in adults with WMH have been repeatedly studied but with a focus on late-stage WMH. To date, structural and functional MRI profiles during the very early stage of WMH remain largely unexplored. To address this, we investigated multimodal MRI (structural, diffusion, and resting-state functional MRI) profiles of community-dwelling asymptomatic adults with very early-stage WMH relative to age-, sex-, and education-matched non-WMH controls. The comparative results showed significant age-related and age-independent changes in structural MRI-based morphometric measures and resting-state fMRI-based measures in a set of specific gray matter (GM) regions but no global white matter changes. The observed structural and functional anomalies in specific GM regions in community-dwelling asymptomatic adults with very early-stage WMH provide novel data regarding very early-stage WMH and enhance understanding of the pathogenesis of WMH.

Purpose. Analysis of the AMD prevalence in the Republic of Bashkortostan, depending on the disease stage, gender and the nature of the respondent settlement. Methods. A population-based study of Republic of Bashkortostan residents “Ural Eye and Medical Study” was conducted on the basis of the State Budget Institution “Ufa Research Institute of Eye Diseases of the Academy of Sciences of the Republic of Bashkortostan”. There were 5899 participants in the study, 2581 (43.7 %) were men, 3318 (52.3 %) were women. There were 2501 urban residents (42.3 %), rural residents — 3398 (59.7 %). Study design: cross-sectional one-stage analytical research. Statistical data analysis was performed using the IBM SPSS statistic application package. Results. AMD was diagnosed in 550 individuals (11.5 %): early stage — in 392 (7.9 %), intermediate — in 119 (2.4 %), later — in 39 individuals (0.8 %). The number of men with AMD was 210 (10.8 %), women — 340 (11.4 %). Among men, an early stage was observed in 144 (7.4 %), intermediate — in 52 (2.7 %), later — in 14 individuals (0.7 %). Among women, early AMD was observed in 248 (8.3 %), intermediate — in 67 (2.2 %), later — in 25 individuals (0.8 %). The number of urban residents with AMD was 194 individuals (9.7 %), rural residents — 356 individuals (12.1 %). At the same time, among urban residents, an early stage was observed in 138 (6.9 %), intermediate — in 43 (2.2 %), and later stage — in 13 individuals (0.6 %). Among rural residents, the early stage of AMD is in 254 (8.6 %), intermediate — in 76 (2.6 %), later — in 26 individuals (0.9 %). Conclusion. The prevalence of AMD in the Republic of Bashkortostan according to the “Ural Eye and Medical Study” is 11.1 %, which exceeds the global figure. The lack of prevalence of the AMD among women (among women — 11.4 %, among men — 10.8 %) is a distinctive feature for this region. The prevalence of this pathology among rural residents (12.1 %), among urban (9.7 %). The influence of population characteristics for the prevalence of AMD in regions with a peculiar ethno-cultural composition, geographical features.

AbstractWe investigated differences between Alzheimer's disease (AD) and vascular dementia (VaD) from the appearance of the first cognitive symptoms, focusing on both time of onset and rate of accelerated decline for different cognitive functions before dementia diagnosis. Data from a longitudinal population-based study were used, including 914 participants (mean age = 82.0 years, SD = 5.0) tested with a cognitive battery (word recall and recognition, Block Design, category fluency, clock reading) on up to four occasions spanning 10 years. We fit a series of linear mixed effects models with a change point to the cognitive data, contrasting each dementia group to a control group. Significant age-related decline was observed for all five cognitive tasks. Relative to time of diagnosis, the preclinical AD persons deviated from the normal aging curve earlier (up to 9 years) compared to the preclinical VaD persons (up to 6 years). However, once the preclinical VaD persons started to decline, they deteriorated at a faster rate than the preclinical AD persons. The results have important implications for identifying the two dementia disorders at an early stage and for selecting cognitive tasks to evaluate treatment effects for persons at risk of developing AD and VaD.
