Marking of the largest (F1) and the second largest (F2) follicles of the ovaries of the sheep with India ink has allowed us to observe the fate of these follicles after different treatments.All the F1 and F2 follicles of the controls marked on the 10th day of the cycle became atretic, but when the marking was done on the 14th day, only the F1 (of the marked follicles) ovulated. Destruction of the F1 with or without removal of the corpora lutea allowed some of the F2 to ovulate, whereas only removal of the corpora lutea decreased the number of F1 ovulating. It thus appears that the fate of the F1 is related to the time of the cycle and that the F1 could have an inhibitory effect on the ovulation of the F2. Removal of the corpora lutea also induced the development of several cystic follicles and decreased the total ovulation rate.

Twelve hours after standing estrus was first detected, 30 ewes had their four largest follicles identified and marked with India ink. Seven days later, ovulation rates and origin of the corpora lutea (CL) from previously marked follicles were determined. Feeding a high energy ration from day 10 to estrus did not change the ovulation rate as compared to feeding a low energy ration. Its only effect was to increase the diameter of the third and fourth largest follicles. The diameter of the largest follicle was identical in single and multiple ovulators. However, in multiple ovulators, the diameter of the second largest follicle was larger than in single ovulators. The largest follicle identified at estrus developed into a CL that was 15% heavier in single than in multiple ovulators. In multiple ovulators, the second largest follicle developed into a CL of weight similar to that from the largest follicle.

The morphology and the responsiveness of ovarian follicles to an ovulating stimulus imposed in the presence or absence of previously induced corpora lutea (CL) were studied in anestrous DLS ewes (Dorset × Leicester × Suffolk). Induction of CL formation was done with 500 IU of PMSG given intravenously. In CL-bearing ewes, the diameter of the largest follicle was significantly larger 12 and 16 days after PMSG injection, while that of the second largest follicle was increased only on day 12. When ovulation was induced with 500 IU PMSG on day 16, the presence of a previously induced CL did not modify the ovulation rate. The two largest follicles identified with India ink markings 4 days prior to PMSG injection did not respond by ovulation and most of these follicles had become completely atretic by day 21 of the experiment. When the two largest follicles, however, were identified and marked on the day of PMSG injection, 65% of them responded to the stimulus. This indicated that turnover of the large follicles was occurring during the anestrous season as is the case during the normal breeding season.

The attainment of puberty in September-born early-maturing ewe lambs was studied at Zaragoza (latitude 41° 40' N). Thirty twin Salz females were allocated to two groups receiving two nutrition levels after 3 months of age: high (500 g/day lucerne hay and 500 g/day concentrate) (H) and low (500 g/ day lucerne hay) (L). Oestrus was detected daily by aproned rams. Corpora lutea were counted after oestrus and plasma progesterone levels monitored each week.In the first breeding season (January to February) the percentage of females showing sexual activity (silent emulation or oestrus and ovulation) was higher in the H compared with the L group (67 and 20%; P < 0/05). Nonpubertal oestrus before the main breeding season was detected in 67% of animals. In the main breeding season and for H and L groups respectively, percentage of females showing silent ovulation before puberty was 67 and 33% and mean age at puberty extended to 319 (s.e. 4-8) and 314 (s.e. 3·7) days. Ovulation rate at puberty was 1·73 (s.e. 0·13) and 1·33 (s.e. 0·15) respectively (P < 0·05).

ABSTRACTOver 2 years, 104 Greyface ewes were mated at a synchronized oestrus either in early October or early November. From late September until slaughter at return to service or between 4 and 7 weeks after mating, ewes were fed to maintain their body condition score as previously adjusted by differential group feeding on grass. Ovulation rate and potential lambing rate were derived from counts of corpora lutea and viable embryos at slaughter. Both rates were higher in the early-mated ewes in comparable body condition. Stress factors were implicated as contributing to the decline in ovulation rate with time, while ewes which failed to hold to first mating were largely responsible for the decline in potential lambing rate.

ABSTRACTIn two experiments over 2 years, 57 North Country Cheviot and 82 South Country Cheviot hill ewes were differentially group-fed indoors over a 2-month period to achieve either good or moderate body con- dition. Over 5 weeks prior to mating, ewes in good condition were brought down in condition by restricted feeding and ewes in mod- erate condition were raised in condition by a high level of feeding. The ewes were thus in moderately-good condition at mating. After mating, ewes were maintained in this condition until killed either on return to service or at 29 ± 8 days for counts of corpora lutea and viable embryos.Ovulation rate in each breed was positively related to the level of pre-mating food intake at the condition level studied. Embryo mortality, as ova loss, was not influenced overall by the level of pre-mating food intake but loss of multiple-shed ova was greater than that of single-shed ova in ewes which had been on restricted feeding before mating. Although a greater proportion of ewes in the North Country Cheviot breed were not pregnant at slaughter, this could not be identified as a breed difference since the breeds were studied in different years.

The dose-response relationship for the inhibitory effect of indomethacin on implantation and continuance of pregnancy was examined in four groups of rabbits administered with indomethacin (2.5, 5.0, 7.5 and 10.0 mg/kg) during the implantation period and compared with a control group. Implanted fetuses and corpora lutea were counted by laparotomy, and the number of offspring born was noted. The inhibitory effect of indomethacin on implantation was found to be dose–dependent, and the birth rate decreased in the indomethacin groups compared with the control group. As a result, even where implantation had been achieved, death of the implanted fetuses occurred at a high rate in rabbits administered with indomethacin during the implantation period.

The transforming growth factor β (TGFB) superfamily proteins bone morphogenetic protein 15 (BMP15) and growth differentiation factor 9 (GDF9), are essential for mammalian fertility. Recent in vitro evidence suggests that the proregions of mouse BMP15 and GDF9 interact with their mature proteins after secretion. In this study, we have actively immunized mice against these proregions to test the potential in vivo roles on fertility. Mice were immunized with either N- or C-terminus proregion peptides of BMP15 or GDF9, or a full-length GDF9 proregion protein, each conjugated to keyhole limpet hemocyanin (KLH). For each immunization group, ovaries were collected from ten mice for histology after immunization, while a further 20 mice were allowed to breed and litter sizes were counted. To link the ovulation and fertility data of these two experimental end points, mice were joined during the time period identified by histology as being the ovulatory period resulting in to the corpora lutea (CL) counted. Antibody titers in sera increased throughout the study period, with no cross-reactivity observed between BMP15 and GDF9 sera and antigens. Compared with KLH controls, mice immunized with the N-terminus BMP15 proregion peptide had ovaries with fewer CL (P<0.05) and produced smaller litters (P<0.05). In contrast, mice immunized with the full-length GDF9 proregion not only had more CL (P<0.01) but also had significantly smaller litter sizes (P<0.01). None of the treatments affected the number of antral follicles per ovary. These findings are consistent with the hypothesis that the proregions of BMP15 and GDF9, after secretion by the oocyte, have physiologically important roles in regulating ovulation rate and litter size in mice.

The efficacy of melatonin implants inserted around the spring equinox to improve fertility and ovulation rate or litter size in Mediterranean ewes has been previously reported (Chemineau et al., 1996; Forcada et al., 2002a.), indicating the ability of the hormone to regulate the hypothalamic activity (Viguié et al., 1995). Moreover, a direct effect of melatonin on corpora lutea and embryonic development has been also reported (Wallace et al., 1988; Abecia et al., 2002). The use of prolific Rasa Aragonesa (RA) ewes (a Mediterranean breed) before culling as embryo donors has been previously tested in the breeding season (Forcada et al., 2002b.). The aim of this experiment was to improve embryo production during the seasonal anoestrus period in selected superovulated RA ewes at the end of their reproductive lives through the use of melatonin.

<p>The aim of this study was to estimate the limiting litter size components in rabbit females from a Synthetic line (n=32) and a Local population (n=34). Ovulation rate, number of implanted and live embryos were counted by laparoscopy at 12 d after mating. Prolificacy (total newborn, number born alive and mortality) and embryonic, foetal and prenatal survival at day of birth of the 3<sup>rd</sup> gestation were measured. The analysed traits were body weight of the female at mating, ovulation rate, implanted, live and resorbed embryos, embryonic, foetal and prenatal survival, as well as total newborn, number born alive and mortality at birth. Synthetic line females had a higher ovulation rate compared to the Local population (11.03±0.23 vs. 8.41±0.23 corpora lutea; <em>P</em>&lt;0.0001). Synthetic line displayed a higher number of implanted embryos (10.00±0.25 vs. 7.85±0.25 embryos; <em>P</em>&lt;0.0001). No difference was found between groups for number of resorbed embryos. Similar embryonic, foetal and prenatal survival rates were reported between the Synthetic line and the Local population. Additionally, total newborn was higher in the Synthetic line than in the Local population (+1.46 kits; <em>P</em>&lt;0.05). A principal components analysis was performed. The first four principal components (PC) explained more than 90% of the total variation in both lines. Total newborn, number born alive and live embryos were the main variables defining the 1<sup>st</sup> PC. Resorbed embryos and foetal survival were located in the 2<sup>nd</sup> PC. Ovulation rate and embryonic survival were the predominant variables defining the 3<sup>rd</sup> PC. The body weight of females was located in the 4<sup>th</sup> PC. The phenotypic correlation between total newborn and its components were high and positive in both lines, except for ovulation rate and total newborn, where it was moderate in Synthetic line. In conclusion, the females from Synthetic line have a higher total newborn than those from Local population, as a consequence of a higher number of released oocytes and embryos that successfully reach implantation. However, a higher uterine crowding in Synthetic line seems to limit survival of foetuses that reach term of gestation, while ovulation rate is the principal limiting factor of total newborn in Local population.</p>

A total of 32 prepubertal gilts of Yorkshire and Landrace breeding were selected at 138 d and fed ad libitum a 16.2% crude protein diet formulated to provide 13.1 MJ DE kg−1. From selection until the end of second estrus, the gilts were exposed to a boar for 30 min d−1 to facilitate the detection of pubertal and second estrous periods. From 14 d after puberty, the gilts received daily injections of either porcine growth hormone at 90 μg kg−1 body weight (GH; n = 20) or vehicle (CT; n = 12) until 24 h after the onset of second estrus and were then killed 9 d later to determine ovulation rate. Gilts not displaying a second estrus by 24 day after puberty were considered anestrus and the injection regime was halted. Anestrous gilts were killed 30–32 d after puberty and their ovaries examined for the presence of corpora albicantia and the absence of corpora lutea. Blood samples were obtained from all gilts at 14, 17, and 20 d after puberty. There was no treatment effect on the duration of the estrous cycle (20.8 vs. 21.3 d for GH and CT, respectively), but while all CT gilts cycled normally, only 55% of the GH gilts had a second estrus (P < 0.01). In those gilts having a second estrus, the daily injection of growth hormone increased ovulation rate (14.3 vs. 12.4 for GH and CT respectively; P < 0.03). Serum type 1 insulin-like growth factor (IGF-1) concentrations were higher (P < 0.001) in GH than in CT gilts, but there was no difference between cycling and anestrous GH gilts. We suggest that the effect of growth hormone on ovulation rate was mediated by increased secretion of IGF-1. The etiology of the high incidence of anestrus is, however, not known.
