A new blood steroid, 11-ketotestosterone, has been isolated from postspawned male sockeye salmon. The steroid was identified by chromatography, infrared spectra, chemical transformation, and synthesis. This steroid is present at a concentration of 12 μg/100 ml of plasma and probably was reported as corticosterone by other investigators. Evidence is presented in support of the occurrence of corticosterone at a very low concentration in salmon plasma. The concentrations of cortisol, cortisone, corticosterone, and total Porter–Silber positive steroids were determined in large plasma samples taken from postspawned fish of both sexes.

17 Hydroxyprogesterone

Cortisone, cortisol, and 17-hydroxyprogesterone were isolated from the plasma of prespawning Fraser River sockeye salmon. The female plasma contained double the concentration of cortisone and cortisol found in male plasma, 41 μg and 26 μg/100 ml respectively as compared with 22 μg and 11 μg/100 ml.Corticosterone is probably present at a very low concentration and neither 11-desoxycorticosterone nor aldosterone was detected in the plasma from approximately 100 fish. Cortisone, cortisol, and total 17-hydroxysteroids were quantitatively determined in plasma samples obtained from two pure races of sockeye captured at various stages of sexual maturity. The combined concentration of cortisone and cortisol which, at the mouth of the river was three times that of the normal human, increased to as high as 17 times this level following spawning. The results are discussed in relation to the death of the fish.

Glucuronoside conjugation was indicated by the use of a specific glucuronidase inhibitor, glucosaccharo-1, 4-lactone, and the inability of the enzyme to hydrolyze testosterone sulphate.

Testosterone was isolated from both the "free" and conjugated steroid fractions obtained from the plasma of spawned female sockeye salmon. The structure of the steroid was confirmed by several criteria including a sulphuric acid chromogen and infrared spectra. Evidence is presented for the presence of testosterone in the conjugated steroid fraction prepared from spawned male sockeye. Several unsuccessful attempts were made to detect dehydroepiandrosterone and androsterone.

SUMMARY A new tetrazolium reducing compound, present as a conjugate which could be hydrolysed by β-glucuronidase, has been isolated from the urine of normal men and women and oophorectomized-adrenalectomized women, all of whom had received 100 mg./day 6α-methyl-17α-acetoxyprogesterone (MAP) either subcutaneously or orally. Detailed studies of its characteristics, including degradation reactions, ultraviolet and infrared spectra, combined with studies on known steroids, suggest that the new compound is 6β,17α,21-trihydroxy-6α-methyl-Δ4-pregnene-3,20-dione (21-acetate). It is suggested that MAP in vivo undergoes hydroxylation at C-21 in the absence of ovarian and adrenal tissue which up to the present have been considered essential for this metabolic process.

LC-SPE/cryo NMR and MS methodologies have been developed and employed for a rapid structure determination of 4″-tetrahydrofurfuryl macrozone reaction mixture components. Macrozones, novel conjugates of azithromycin, and thiosemicarbazones have shown very good in vitro antibacterial activities against susceptible and some resistant bacterial strains and are promising agents for further development. The post-column multiple trapping of the chromatographically separated reaction mixture components on the SPE cartridges increased the sensitivity and together with cryogenically cooled NMR probe made it possible to identify and structurally characterize main 4″-tetrahydrofurfuryl macrozone reaction mixture compounds including those present at very low concentration level. This approach has several advantages over a classical off-line procedure, efficiency and low solvent consumption being the two most important ones. All identified components were process-related. It has been demonstrated that two different kinds of compounds with respect to structure were identified, i.e., macrolide-related and thiosemicarbazone-related ones. This methodology can serve as a platform for reliable and effective macrolides reaction components structure profiling, serving as both isolation and identification tools.
