DIFFERENCES IN NITROGEN CONTENT IN EXTRACTS OF CHILLED AND NON-CHILLED TISSUES

In an attempt to elucidate the increase of growth promotion of tissue cultures by extracts obtained from tissues chilled before extraction, determinations of nitrogen content were made on (a) fresh, and chilled chick embryos, (b) extracts prepared from fresh and chilled chick embryos, (c) fresh and chilled human placenta, (d) extracts prepared from fresh and chilled human placenta.There was no difference in dry weight, nitrogen content, phosphorus content, and nitrogen: phosphorus ratio between chilled and fresh chick embryos and chilled and fresh human placenta. There was, however, a significant difference in the nitrogen content of the extracts prepared from fresh and from chilled tissues, It is suggested that chilling of tissues for 7 days at + 4 °C. prior to extraction may render the cells more easily destructible during extraction, and thus supply a better yield of nutritive substances.

The respiration rates and tissue contents of nitrogen, phosphorus, and titratable acidity were determined for apple fruit from trees receiving differential fertilizer treatments U, N, P, NP, and NPK (U, no fertilizer; N, 2 lb ammonium sulphate; P, 4 1b superphosphate; and K, 1 Ib potassium sulphate, per tree per year). The results obtained (expressed on a dry weight basis) are summarized as follows: Respiration rate: N > NP = NPK = U > P. Total nitrogen content: NPK > NP = N > U = P. Ethanol-insoluble nitrogen content: NPK = NP = N > U = P. Ethanol-soluble nitrogen content: NPK > NP = N > U = P. Phosphorus content : P > NP = NPK > U > N. Titratable acidity content: NPK > N > NP = U > P. The incidence of internal breakdown in stored fruit from these treatments has positively correlated with respiration rate per cell and negatively correlated with phosphorus content. Phosphorus content was negatively correlated with cell volume and respiration rate. Mechanisms by which phosphorus content might regulate the incidence of internal breakdown are discussed.

Aims: This study was done to evaluate the effects of T. harzianum AAUT14 and B.subtilis AAUB95 on chocolate spot (B. fabae) and growth promotion of faba bean. Study Design: A completely randomized block design was utilized. Place and Duration of Study: The study was conducted at Kulumsa Agricultural Research Center, 8º2'N and 39º10’E, Kulumsa, June-November, 2018. Methodology: Two trails (Trial-1 Ashebeka and Trial-2 Hachalu) were employed. We included T1-Control (B.f only); T2-T. harzianum AAUT14+ B.f; T3-B. subtilis AAUB95+B.f; T4-T. harzianum AAUT14+B. subtilis AAUB95+B.f; T5- MORE 720 WP+B.f; T6- ORZEB+B.f as treatments of the study. The disease development was assessed together with yield and related parameters. Results: In trail 1, a reduction that varied from 31-61% for disease incidence and 13-33% of severity over T1 upon 70 days after sowing. Upon 90 days, the disease incidence and severity was reduced to 20-50% and 36-51%, respectively. Trichoderma harzianum AAUT14+B.subtilis AAUB95 (T4) reduced the disease incidence and severity showing no significance difference (P=.05) with the chemical fungicide, MORE 720 WP (T5) upon 70 and 90 days of sowing. In trial 2, the microbial inoculants reduced the disease incidence and severity to 28-63% and 17-30% upon 70 days. Likewise, the disease incidence and severity was reduced to 23-51% and 37-54% upon 90 days. In addition, the AUDPC ranged from 1586.1-2250.0%DSU in trial 1 and 1382.0-2454.5%DSU in trial 2. Moreover, leaf area of 68.95cm2 was displayed by T4 in trial 1 and 54.14cm2 in trial 2. In addition, T 4 indicated, 62% and 49% increment of hundred seed dry weight and grain yield estimate in trial 1, and 56% and 55%, increase in trial 2 compared to the uninoculated control. The percentage of healthy pods was 90% and 88.87% in trial 1 and 2, respectively, in the treatment that received T4 and followed by T2 that showed 70.40 and 78.86% in trial 1 and 2, respectively. T4 resulted 4391.45kg/ha and 4378.12kg/ha, that followed by T2 with 3764.58kg/ha and 3654.17kg/ha of yield estimate in trial 1 and 2, respectively. 27-42% and 26-41% of harvest index was exhibited in trial 1 and trial 2, respectively. Furthermore, the seed nitrogen content increased from 33-70% in trial 1 and 29-62% in trial 2. The seed nitrogen content showed 33-70% and 29-62% increment in trial 1 and 2, respectively. Even though the fungicides (T5 and T6), protected the faba bean plants from chocolate spot, there was <10% of seed nitrogen and crudeprotein content increment over the untreated control plants in both varieties. Conclusion: The mixture of T. harzianum AAUT14 and B. subtilis AAUB95 or in some cases T. harzianum AAUT14 performed best on controlling chocolate spot, growth promotion and yield increment of faba bean.

The aim of this research is to evaluate the effect of distiller dried grain (DDG) of rice husk in quail feed which effectively reduces important chemical elements in feces. The results showed that administration of DDG influenced the absorption of nutrients carbon, nitrogen, phosphorus and calsium in the quail intestine. Substitution of 10% DDG proportion in quail formulation not significantly (P>0.05) effect on carbon content in feces. Carbon content  infeces of quail at proportion of 20% DDG were significantly (P<0.01) higher than 10%. Nitrogen content in feces of quail at proportion of 20% DDG significant (P<0.05) higher than  both 10 and 0%.Futhermore nitrogen content in quail feces at 10% DDG significant (P<0.05) higher than 0%. Phosphorus content in feces quails at proportion of 10 and 20% DDG significantly (P<0.05) lower than 0%. Calcium content in quail feces at 10% and 0% not significant (P>0.05) but both were significantly (P<0.05) lower than 20%. The results of this study can be concluded that the substitution of 10% DDG of rice husk in quail feed formulations can increase absorption of phosphorus but decreases the absorption of carbon, nitrogen and calcium   Keyword: chemical elements, distillater dried grain, feces, quail

A block of Jonathan apple trees were treated for 4 years as individual tree plots with nil, nitrogen, nitrogen + phosphorus, and nitrogen + potassium applied as sprays during two periods, either in spring before bud differentiation or after bud differentiation in mid January. The treatments had no effect on the levels of phosphorus or potassium in the fruit. Spring applications had no significant effect on the nitrogen content, but summer applications a marked one, though there was no evidence of a cumulative build-up or any effect on cell number per fruit. These increases in nitrogen content were accompanied by small increases in breakdown level, but only when an inherent seasonal susceptibility existed. Only a diminishing proportion of the additional nitrogen was incorporated into the protein fraction, and the preclimacteric respiration rate was not affected significantly by the increase in protein. Except for percentage dry matter, there was a wide variation in the values between trees of the variables, which permitted a multiple regression analysis. The regression of breakdown and cell volume was positive and highly significant in all years. Except for the year of very low incidence, there was a significant positive relationship between breakdown and nitrogen level, and a consistent negative relationship between breakdown and phosphorus level which was significant in the years of high breakdown incidence. No relationship could be shown between breakdown and potassium. Similar results were obtained by using a percentage fresh weight or percentage dry weight basis for the nitrogen, phosphorus, and potassium contents, or by using actual values or a log (x + 1) transformation for the variables, though there were some advantages in using the latter. The results suggest an important physiological interaction between breakdown and cell volume, nitrogen, and phosphorus.

Plants of L. temulentum were induced to flower by exposure to 1 long day. Shoot apices harvested from vegetative plants prior to the long�day exposure (day I) and others harvested on days IV and VI were analysed for length, fresh weight, total and residual (after extraction for solubles and lipids) dry weight, total and residual nitrogen content, residual phosphorus, and RNA and DNA phosphorus content. Cell number per apex was also established for two harvests.

A factorial experiment was conducted with a completely randomized design to evaluate the effects of planting date and density on calendula herbs and peppermint. It had 3 replicates and was done in Khosroshahr research farm, Tabriz in 2006. Under studied factors were: 3 planting dates (10 May, 25 May and 10 June) in 4 densities (25, 35, 45, 55) of the plant in square meters. The results of variance a nalysis showed that there was 1% probability significant difference between the effects of planting date and bush density on the leave number, bush height and the bush dry weight. But the mutual effect of the plant date in mentioned traits density was insignificant. Regarding the traits mean comparison, the total maximum dry weight was about the 55 bush density in mm. Also, the bush high density in mm causes the bush growth and its mass reduction. When there is the density grain, the flower number will increase due to bush grain in surface unit. Overall, we can conclude that 10 June planting and 45 bush density in mm is the most suitable items and results in favored production with high essence for these crops.

SummaryThis study aims to develop a biocontrol agent against Fusarium oxysporum f.sp. radicis-lycopersici (FORL) in tomato. For this, a set of 23 bacterial endophytic isolates has been screened for their ability to inhibit in vitro the growth of FORL using the dual plate assay. Three isolates with the most sound antagonistic activity to FORL have been qualitatively screened for siderophore production, phosphates solubilization and indolic acetic acid (IAA) synthesis as growth promotion traits. Antagonistic values of the three candidates against FORL were respectively: 51.51 % (EB4B), 51.18 % (EB22K) and 41.40 % (EB2A). Based on 16S rRNA gene sequence analysis, the isolates EB4B and EB22K were closely related to Enterobacter ludwigii EN-119, while the strain EB2A has been assigned to Leclercia adecarboxylata NBRC 102595. The promotion of tomato growth has been assessed in vitro using the strains EB2A, EB4B and EB22K in presence of the phytopathogen FORL. The treatments with the selected isolates increased significantly the root length and dry weight. Best results were observed in isolate EB4B in terms of growth promotion in the absence of FORL, improving 326.60 % of the root length and 142.70 % of plant dry weight if compared with untreated controls. In the presence of FORL, the strain EB4B improved both root length (180.81 %) and plant dry weight (202.15 %). These results encourage further characterization of the observed beneficial effect of Enterobacter sp. EB4B for a possible use as biofertilizer and biocontrol agent against FORL.

Nurse crops are often recommended in prairie restoration planting. This work investigated several alternative nurse crops to determine their utility in prairie planting. Nurse crops were composed of increasing densities (900, 1800, or 2700 seeds/m2) of partridge pea, spring oats, spring barley, Canada wild rye, or equal mixtures of partridge pea and one of the grasses. The experimental design was a randomized complete-block set in two sites with three blocks per site and 48 treatments per block. Each 3 × 3-m plot contained 1 m2 planted in Dec. 1995 or Mar. 1996 with an equal mix of seven prairie species. The nurse crops were sown over each nine square meter area in April 1996. Plots lacking nurse crops served as controls. Evaluated data consisted of weed pressure rankings and weed and prairie plant dry weight. Nurse crop treatments had a significant effect on weed pressure in both sites. Barley (1800 and 2700 seeds/m2) as well as partridge pea + barley (2700 seeds/m2) were most effective at reducing weed pressure. When weed and prairie plant biomass values were compared, a significant difference was observed for site quality and planting season. Prairie plant establishment was significantly greater in the poorly drained, less-fertile site and spring-sown plots in both sites had significantly higher prairie biomass values. Overall, after two seasons, there was no advantage in using nurse crops over the control. Among nurse crop treatments, oats were most effective in reducing weed competition and enhancing prairie plant growth.

Salicylic acid (SA) is a common plant-produced signal molecule that is responsible for inducing tolerance to a number of biotic and abiotic stresses. An experiment was, therefore, conducted to test whether acetyl salicylic acid (ASA) application at various concentrations through seed immersion or foliar spray would protect muskmelon [Cucumis melo L. (Reticulatus Group)] seedlings subjected to chilling stress. Twenty-one-day-old plants pre-treated with ASA (0, 0.1, 0.25, 0.50 or 1.0 mM) were subjected to chilling stress for 72 h at 3 ± 0.5°C. ASA, applied either through seed immersion or foliar spray, was effective within the range of 0.1 to 1 mM in inducing tolerance to chilling stress in muskmelon seedlings; however, there was no significant difference between application methods. ASA significantly and curvilinearly affected all seedling growth and stress indicator variables tested except shoot dry weight. The best protection was obtained from seedlings pre-treated with 0.5 mM ASA. The highest ASA concentration used was slightly less effective in providing chilling stress protection. Even though both methods provided similar means of protection, due to its simplicity and practicality, immersion of muskmelon seeds prior to sowing in 0.5 mM ASA would be a more desirable method to induce tolerance to chilling stress.
