Genetic control of bud phenology in pole-size trees and seedlings of coastal Douglas-fir

The extent to which bud phenology is genetically controlled and related to growth traits was examined in seedlings and pole-size trees of coastal Douglas-fir (Pseudotsugamenziesii var. menziesii (Mirb.) Franco). Data on bud burst, bud set, and stem growth were collected from pole-size trees of 60 open-pollinated families growing in four plantations, and from seedlings of 45 of these same families growing in three trials. In both age-classes, bud burst was under moderate to strong genetic control (h2 ≥ 0.44) and family breeding values were stable across test environments, indicating that this trait could be readily altered in breeding programs. Bud set was inherited strongly in pole-size trees (h2 = 0.81) but weakly in seedlings (h2 < 0.30). Both bud burst and bud set were positively correlated with growth in seedlings and pole-size trees. Thus, selection for greater growth at either age-class is expected to delay bud burst and bud set. We also evaluated the accuracy of two alternatives for assessing bud burst phenology in pole-size trees compared with the traditional method. We show that bud-burst date on lateral branches can be used to accurately rank both individuals and families for bud-burst date on less accessible leader shoots. In addition, we found that families can be ranked for mean bud-burst date by the proportion of trees per family that have flushed on a given scoring day. This method is only effective, however, when between 25 and 75% of all trees in the test have flushed at the time of scoring.

Genetic control of cold hardiness in two-year-old seedlings was compared with that in 7-year-old saplings of 40 open-pollinated families in each of two breeding populations (Coast and Cascade) of coastal Douglas-fir (Pseudotsuga menziesii var. menziesii (Mirb.) Franco) from western Oregon. In addition, the efficacy of bud phenology traits as predictors of cold hardiness at the two stages was explored. Fall and spring cold hardiness were assessed using artificial freeze testing. Similar genetic control of cold hardiness in seedlings and saplings is suggested by strong type-B genetic correlations (rB) between the two ages for fall and spring cold injury traits (rB[Formula: see text] 0.78) and by similar trends in individual tree heritability estimates (hi2), e.g., hi2was greater in spring (h–i2= 0.73) than in fall (h–i2= 0.36) and greater in the Coast population (h–i2= 0.69) than in the Cascade population (h–i2= 0.40) at both ages. Strong responses to direct selection are expected for spring cold hardiness at both ages and for fall cold hardiness in seedlings, even under mild selection intensities. Similar heritabilities in seedlings and saplings, and strong genetic correlations between ages for cold-hardiness traits, ensure that selection at one age will produce similar gains at the other age. Type-A genetic correlations (rA) between fall and spring cold hardiness were near zero in the Cascade population (rA= 0.08 and -0.14 at ages 2 and 7, respectively) but were moderate and negative in the Coast population (rA= -0.54 and -0.36, respectively). Bud-burst timing appears to be a suitable surrogate to artificial freeze testing for assessing spring cold hardiness in both seedlings and saplings, as is bud set timing for assessing fall cold hardiness in seedlings, but bud set timing is a poor predictor of fall cold hardiness in saplings.

Genetic variation in fall cold damage in coastal Douglas-fir ( Pseudotsuga menziesii (Mirb.) Franco var. menziesii ) was measured by exposing excised branches of seedlings from 666 source locations grown in a common garden to freezing temperatures in a programmable freezer. Considerable variation was found among populations in fall cold hardiness of stems, needles, and buds compared with bud burst, bud set, and biomass growth after 2 years. Variation in fall cold hardiness was strongly correlated (r = 0.67) with cold-season temperatures of the source environment. Large population differences corresponding with environmental gradients are evidence that natural selection has been important in determining genetic variation in fall cold hardiness, much more so than in traits of bud burst (a surrogate for spring cold hardiness), bud set, and growth. Seed movement guidelines and breeding zones may be more restrictive when considering genetic variation in fall cold hardiness compared with growth, phenology, or spring cold hardiness. A regional stratification system based on ecoregions with latitudinal and elevational divisions, and roughly corresponding with breeding zones used in Oregon and Washington, appeared to be adequate for minimizing population differences within regions for growth and phenology, but perhaps not fall cold hardiness. Although cold hardiness varied among populations, within-population and within-region variation is sufficiently large that responses to natural or artificial selection may be readily achieved.

The consequences for growth and phenology of early selection for height or its growth components were evaluated in 160 open-pollinated families of Douglas-fir (Pseudotsugamenziesii (Mirb.) Franco) from southwestern Oregon. Seedlings from two inland and two coastal populations (40 families each) were grown for two growing seasons in a common garden. Predicted response to selection suggests that risk of low juvenile–mature correlation and maladaptation with early selection would be less in the inland than in the coastal region. A phenological event that influences a common growth pattern seems to account for the difference in response. Early bud set in the 1st year was genetically correlated with larger overwintering buds in seedlings from both inland and coastal regions. These larger buds yielded a large increment of predetermined growth in the 2nd year, followed by little or no free growth and early bud set. Seedlings with late bud set in the 1st year had the converse pattern. Inland seedlings set buds much earlier on the average than did coastal seedlings; hence seedlings from the two regions had different growth patterns. Risks that can attend early selection for height generally would be decreased in both regions by selecting for predetermined growth, but several qualifications are discussed.

The relative density profile from pith to bark was examined in young Douglas-fir (Pseudotsugamenziesii (Mirb.) Franco) stems at 1.3, 0.7, and 0.4 m above the ground. The trees represented 22 half-sib families growing at two progeny test locations. The objective was to evaluate the reliability of wood samples taken from below breast height (1.3 m) for selection for relative density in young coastal Douglas-fir progeny tests. The relative density profile appeared to be different for the first 6 or 7 years from the pith at the different sampling heights, but the difference decreased with cambial age. Genetic correlation estimates were sufficiently high to indicate identical genetic control at the three sampling heights, but heritability estimates were higher at 1.3 and 0.7 m than at 0.4 m. Sampling at 0.7 m is as effective as at 1.3 m. This allows reliable selection 1 to 2 years earlier than by sampling at 1.3 m.

Growth, phenology, and cold hardiness of seedlings from 74 populations of Douglas-fir (Pseudotsugamenziesii var. glauca (Beissn.) Franco) from central Idaho were compared in four contrasting environments. Analyses of 3-year-old seedlings revealed population differentiation for eight variables: bud burst, bud set, multiple flushing, height, deviation from regression of 3-year height on 2-year height, spring frost damage, fall frost damage, and winter injury. These analyses, as well as high intercorrelations among population means, suggested that adaptations result from a balance between selection for a high growth potential in mild environments and selection for cold hardiness in severe environments. Consequently, genetic variation among populations was closely related to the elevation, geography, and climate of the seed source.

AbstractTo increase quality and amount of wood production in Turkish red pine (Pinus brutiaTen.), genetic control of wood specific gravity (WSG), tracheid length and growth traits was investigated in Ceyhan1A progeny trial by evaluating 168 families originated from six clonal Turkish red pine seed orchards. Wood samples were taken by destructive sampling during the rouging of this trial at the age of seven. Differences among the 168 families for mean WSG was large (ranged from 0.35 to 0.62), as indicated by high individual (0.42±0.07) and family mean (0.55±0.03) heritabilities. Family differences and high heritabilities were also observed for all growth traits and tracheid length. Genetic correlations between WSG and growth traits were insignificant (near zero), while low and insignificant negative phenotypic correlations among the same traits were also observed. Predicted genetic gain for single trait selection at age of seven was low for WSG (0.37%), but substantial for stem volume (8.4%) in phenotypic seed orchards. However, the first generation clonal seed orchards consisting of the best 30 clones yielded higher genetic gains (5.2% for WSG and 35% for stem volume). These preliminary results suggest that selection for wood characteristics and growth traits in Turkish pine could be practiced at early ages for short rotation (about 30 years) in industrial plantations.

Program pemuliaan ikan mas varietas Punten dilakukan dengan seleksi individu terhadap karakter bobot ikan. Pembentukan populasi dasar untuk kegiatan seleksi dilakukan dengan memijahkan secara massal induk ikan mas yang terdiri atas 20 induk betina dan 21 induk jantan yang dikoleksi dari daerah Punten, Kepanjen (delapan betina dan enam jantan), Kediri (tujuh betina dan 12 jantan), Sragen (27 betina dan 10 jantan), dan Blitar (15 betina dan 11 jantan). Larva umur 10 hari dipelihara selama empat bulan. Selanjutnya dilakukan penjarangan sebesar 50% dan benih dipelihara selama 14 bulan untuk dilakukan seleksi dengan panduan hasil sampling 250 ekor individu setiap populasi. Seleksi terhadap calon induk dilakukan saat umur 18 bulan pada populasi jantan dan betina secara terpisah dengan memilih berdasarkan 10% bobot ikan yang terbaik. Calon induk yang terseleksi kemudian dipelihara hingga matang gonad, kemudian dipilih sebanyak 150 pasang dan dipijahkan secara massal. Didapatkan respons positif dari hasil seleksi berdasarkan bobot ikan, yaitu 49,89 g atau 3,66% (populasi ikan jantan) dan 168,47 g atau 11,43% (populasi ikan betina). Nilai heritabilitas untuk bobot ikan adalah 0,238 (jantan) dan 0,505 (betina).Punten carp breeding programs were carried out by individual selection for body weight trait. The base population for selection activities were conducted by mass breeding of parent consisted of 20 female and 21 male collected from area Punten, eight female and six male (Kepanjen), seven female and 12 male (Kediri), 27 female and 10 male (Sragen), 15 female and 11 male (Blitar). Larvae 10 days old reared for four moths. Then after spacing out 50% of total harvest, the offspring reared for 14 months for selection activity based on the sampling of 250 individual each population. Selection of broodstock candidates performed since 18 months age on male and female populations separately by selecting based on 10% of fish with best body weight. Candidates selected broodstocks were then maintained until mature. In oder to produce the next generation 150 pairs were sets and held for mass spawning. The results revealed that selection response were positive, 49.89 g (3.66%) for male and 168.47 (11.43%) for female. Heritability for body weight is 0.238 (male) and 0.505 (female).

Nuclear receptor subfamily 6, group A, member 1 (NR6A1), as an important member of the nuclear receptor family, plays an important role in regulating growth, metabolism, and differentiation of embryonic stem cells. For this reason, the NR6A1 gene is considered to be a promising candidate for economic traits and was found to be associated with body size traits in many livestock. However, no studies have been conducted on NR6A1 in donkeys so far. Thus, in this research, we focused on donkeys and identified a 13 bp deletion in intron-1 of the NR6A1 gene among 408 individuals from Guanzhong and Dezhou donkeys using polyacrylamide gel electrophoresis. Three genotypes were identified, namely II, ID, and DD. The association analysis indicated that the body lengths and body heights5f genotype II individuals were significantly different to those of genotype ID in Dezhou donkeys. Conclusively, the 13 bp deletion was associated with growth traits in both Guanzhong donkeys and Dezhou donkeys, indicating that the NR6A1 gene could be a possible candidate gene in marker-assisted selection for donkey breeding programs.

SUMMARYA total of 245 test cross progenies obtained by crossing cytoplasmic male sterile (CMS) lines and restorer (RHA) lines were evaluated in replicated trials in 1995 and 1997. Experiments were carried out in two locations (Osimo, East Central Italy and Budrio, Northern Italy) in 1995, whereas in 1997 trials were conducted only in Osimo. General combining ability (GCA) and specific combining ability (SCA) of the parents was evaluated to identify genotypes suitable to be used as testers for breeding programs and to verify the performance of hybrids. The results obtained in 1995 were used to make test crosses in 1996 to be evaluated in 1997. The GCA and SCA of RHA and CMS lines were nearly always significant for all the studied traits when lines were randomly taken in 1995. When selected testers were used on the basis of the results of the first year, the SCA of new RHA lines and GCA of new CMS lines were not significant for achene yield, but they were for the other traits.The best GCA estimates for RHA were often higher than those of CMS, indicating that selection for RHA could be more useful than for CMS. As expected, GCA was always lower than SCA. CMS and RHA, with high variance among their test crosses for yield and many other traits, could be used as testers. The lack of a tester with high variance in all traits requires more than one tester in evaluating lines. The GCA of a line can change in function of the germplasm with which it is combined.

The juvenile period represents a serious impediment in olive breeding programs. Seedlings with long juvenile period are of very low interest for the breeder because their evaluation considerably delays the first stages of the breeding process. For this reason, the influence of seedling vigour (measured as plant height or stem diameter) on the characteristics at the adult stage was studied to establish useful negative preselection criteria on the basis of that relationship. Olive progenies from crosses and open pollinations of 12 different parents carried out in 1998 and 1999 were evaluated in the greenhouse and, afterwards, during the first 3 years of bearing in the open field. The results obtained indicate that early evaluation and selection for juvenile period can be performed at the seedling stage in olive progenies on the basis of vigour measurements. Selection for short juvenile period was valid irrespective of parentage and, therefore, could be efficient in a general context. No relationship between juvenile period and yield or fruit traits was found so that this preselection criterion would have no adverse effects on these characters.
