Endophytic Phomopsis species: host range and implications for diversity estimates

Foliar endophyte assemblages of teak trees growing in dry deciduous and moist deciduous forests of Nilgiri Biosphere Reserve were compared. A species of Phomopsis dominated the endophyte assemblages of teak, irrespective of the location of the host trees. Internal transcribed spacer sequence analysis of 11 different Phomopsis isolates (ten from teak and one from Cassia fistula) showed that they fall into two groups, which are separated by a relatively long branch that is strongly supported. The results showed that this fungus is not host restricted and that it continues to survive as a saprotroph in teak leaf, possibly by exploiting senescent leaves as well as the litter. Although the endophyte assemblage of a teak tree growing about 500 km from the forests was also dominated by a Phomopsis sp., it separated into a different group based on internal transcribed spacer sequence analysis. Our results with an endophytic Phomopsis sp. reinforce the earlier conclusions reached by others for pathogenic Phomopsis sp., i.e., that this fungus is not host specific, and the species concept of Phomopsis needs to be redefined.Key words: fungal endophytes, Tectona grandis, fungal diversity, Phomopsis, host restriction.

Infundibulicybe rufa sp. nov., is described from Jiuzhaigou Biosphere Reserve, southwestern China. It is characterized by the combination of the following characters: umbilicate to slightly infundibuliform, reddish brown pileus; decurrent, cream lamellae; cylindrical stipe concolorous with the pileus surface. Molecular phylogenetic analyses using the nuclear ribosomal internal transcribed spacer (ITS) region indicates that I. rufa is closely related to I. mediterranea and I. bresadolana. A description, line drawings, phylogenetic placement and comparison with allied taxa for the new taxon are presented.

Radiocarbon analysis in annual rings of a teak tree (Tectona grandis) is reported in comparison with previously published results. Samples (disks) were collected from Hoshangabad (22°30′N, 78°E), Madhya Pradesh, in central India. The previously published sample was collected from Thane (19°12′N, 73°E), Maharashtra, near the west coast of India (Chakraborty et al. 1994). Two short Δ14C time series were reconstructed with these tree samples to capture the bomb peak of atmospheric 14C and the spatial variability in this record. These time series represent the periods 1954–1977 and 1959–1980 for Hoshangabad and Thane, respectively. The 14C peaks in these places appear around 1964–1965. The Hoshangabad tree records a peak Δ14C value of 708 ± 8%, which conforms to the peak value of Northern Hemisphere Zone 3 as described in Hua and Barbetti (2004). But the peak Δ14C at Thane is somewhat less (630 ± 8%) probably due to the dilution by fossil fuel CO2 free of 14C emanating from the neighboring industrial areas. This depletion of peak values has been used to estimate the local emission of fossil fuel CO2, which is approximately 2.3% of the background atmospheric CO2 concentration.

Molecular identification of eighteenTermitomycesspecies collected from two states, Ondo and Ekiti in Nigeria was carried out using the internal transcribed spacer (ITS) region. The amplicons obtained from rDNA ofTermitomycesspecies were compared with existing sequences in the NCBI GenBank. The results of the ITS sequence analysis discriminated between all theTermitomycesspecies (obtained from Ondo and Ekiti States) andTermitomycessp. sequences obtained from NCBI GenBank. The degree of similarity of T1 to T18 to gene ofTermitomycessp. obtained from NCBI ranges between 82 and 99 percent.Termitomycesspecies from Garbon with ascension number AF321374 was the closest relative of T1 to T18 except T12 that has T. eurhizus and T. striatus as the closet relative. Phylogenetic tree generated with ITS sequences obtained from NCBI GenBank data revealed that T1 to T18 are more related toTermitomycesspecies indigenous to African countries such as Senegal, Congo, and Gabon.

Lantana camara is a dominant invasive shrub in many protected areas of India including the Nilgiri Biosphere Reserve (NBR).  We conducted a study to assess the regeneration potential of endemic native (shola) trees under different levels of Lantana infestation in the upper plateau of NBR.  A total of 61 plots in a total area of 0.73ha were sampled, out of which 0.57ha was in Lantana dominated sites and 0.16ha in undisturbed shola forests.  The plots were classified as per the level of Lantana infestation (intensive, moderate, and low infestation). We found that regeneration of shola trees, including endemics decreased with increasing intensity of Lantana invasion.  No regeneration occurred in the intensively infested plots whereas regeneration was high in undisturbed shola forests.

Enterocytozoon bieneusi is a common intracellular parasite that infects a wide range of hosts, including humans and companion animals, raising concerns of zoonotic transmission. However, there is limited epidemiological information on the prevalence and genotypes of E. bieneusi in sheltered dogs and cats in Sichuan province, southwestern China. A total of 880 fecal samples were collected from shelters in different cities of Sichuan province, including 724 samples from dogs, and 156 samples from cats. Enterocytozoon bieneusi was determined by sequence analysis of the ribosomal internal transcribed spacer (ITS). Overall, the prevalence of E. bieneusi was 18% (158/880), and the parasite was detected in 18.8% (136/724) and 14.1% (22/156) of the dogs and cats examined, respectively. Sequence analysis revealed the presence of five genotypes in dogs, including three known genotypes CD9 (n = 92), PtEb IX (n = 41), and Type IV (n = 1), and two novel genotypes SCD-1 (n = 1) and SCD-2 (n = 1). Similarly, four genotypes were identified in cats, including CD9 (n = 11), Type IV (n = 6), D (n = 4), and PtEb IX (n = 1). Genotypes D and Type IV have previously been identified in humans and are reported in sheltered dogs and cats in the present study, indicating that these animals could be as potential sources of human microsporidiosis infections.
