Study ofTrichoderma viridemetabolism under conditions of the restriction of oxidative processes

Trichoderma viride was capable of growth and conidiation in the presence of high concentrations of the uncoupler 3,3′,4′,5-tetrachlorosalicylanilide (up to 100 µmol·L–1) and of the respiration inhibitor mucidin (up to 100 µmol·L–1) in both submerged and surface cultivation. When vegetative mycelia were cultivated on the solid Czapek-Dox medium with yeast autolysate under an anaerobic and CO2-containing atmosphere, the growth was observed only rarely but the microorganism survived as long as 3 months under these conditions. Major products of metabolism of both aerobic and anaerobic submerged mycelia were identified by means of1H-NMR measurements. Major products excreted to the medium under aerobic conditions were succinic and citric acids. Major metabolites present in the submerged mycelia were γ-aminobutyric (and glutamic) acid and alanine. Under anaerobic conditions, citric acid was not excreted into the medium but ethanol appeared. Its production could not be increased upon increasing the sugar concentration. The appearance of secondary metabolites was found to be modified by oxygen availability during the mycelial growth. Results suggest that the vegetative form of T. viride is capable of fermentative metabolism characterized by the production of ethanol and succinate and that the excretion of carboxylic acids is developmentally regulated and modified by oxygen availability.Key words: Trichoderma, mycelia, anaerobiosis, citrate, succinate, ethanol, GABA.

ABSTRACT The frequency of transconjugants were compared for the incompatibility (Inc) P-1 and P-7 plasmids pBP136 and pCAR1 under aerobic and anaerobic conditions. Filter mating assays were performed with one donor strain and one recipient strain using different donors of Pseudomonas and recipient strains, including Pseudomonas, Pantoea, and Buttiauxella. Under anaerobic condition, frequencies of transconjugants for both plasmids were 101-103-fold lower than those under aerobic condition regardless of whether aerobically or anaerobically grown donors and recipients were used. To compare the transconjugant ranges under aerobic and anaerobic conditions, conjugation was performed between the donor of pBP136 and recipient bacteria extracted from environmental samples. Several transconjugants were uniquely obtained from each aerobic or anaerobic condition. Our findings indicate that a plasmid can differently spread among bacteria depending on the oxygen concentrations of the environment.

Aeration by mechanical agitation of 15% wheat mash fermented by Aerobacillus polymyxa inhibited the formation of 2,3-butanediol and particularly of ethanol. Aeration of similar mashes by passage of finely dispersed air or oxygen at the rate of 333 ml. per minute per litre of mash increased the rate of formation and yield of 2,3-butanediol but inhibited ethanol formation. However, the over-all time required for the completion of fermentation was not shortened from the usual 72 to 96 hr. required for unaerated mashes. There was no evidence of a shift from fermentative to oxidative dissimilation. Under aerobic conditions, the final butanediol–ethanol ratio was approximately 3:1. Anaerobic conditions, as produced by the passage of nitrogen or hydrogen through the mash, increased the rate of formation of both butanediol and ethanol and shortened the fermentation time to about 48 hr. Under these conditions, the butanediol–ethanol ratio was reduced to about 1.3:1.0. Carbon dioxide gave a butanediol–ethanol ratio resembling that of anaerobic fermentation but did not reduce fermentation time.

Three bioagents (Trichoderma viride, T. harzianum and Gliocladium virens) and five biopesticides (Achook, Neemgold, Wannis, Spictaf and Neemazal) were evaluated under in vitro and in vivo conditions against Colletotrichum lindemuthianum. All the three antagonistic fungi caused significant inhibition of mycelial growth, maximum being with T. viride (69.21%) followed by T. harzianum (64.20%). Among the biopesticides tested at four concentrations, Wanis applied @ 1000 ?l/ml caused maximum inhibition of 82.12 per cent followed by Spictaf (52.85%). T. viride and Wanis @ 1000 ?l/ml were most effective in reducing the seed borne infection. Integration of bioagents with Bavistin showed that disease can be effectively managed with seed dressing either with Bavistin or biopesticide followed by foliar treatment with fungicide or biopesticide.
