The primary purpose of the present study was to determine if steroids, when conjugated by stable covalent bonds to proteins, could act as haptenic groups in inducing steroid-specific antibodies. Estrone was converted to estrone- 17-isocyanate and the latter reacted with various proteins to give estrone- 17-carbamidoprotein conjugates. Rabbits were immunized with the conjugate prepared from human serum albumin and the resulting antisera were shown by various immunochemical techniques to contain antibodies specific to the estronyl residues of this conjugate. A turbidimetric assay method for the estimation of estrone-like compounds is described.

Use of Lucifer Yellow VS as a Label in Fluorescent Immunoassays Illustrated by the Determination of Albumin in Serum

The use of a new label for fluoroimmunoassay is described. Lucifer yellow VS is a highly fluorescent vinyl sulphone dye which, under mild conditions, forms covalent bonds with amino and sulphydryl groups but is extremely stable in water. A large Stokes shift (110 nm) and an emission maximum at 540 nm give Lucifer yellow further advantages over the more commonly used labels. The use of the dye as a label has been demonstrated by developing a heterogeneous fluoroimmunoassay for human serum albumin. The fluoroimmunoassay gave comparable results to those obtained using a less specific colorimetric dye-binding assay (r = 0·97, n = 20). The advantages, limitations, and other potential uses of Lucifer yellow are discussed.

Rabbit antibody fractions of different affinities for human serum albumin were prepared by an immunosorbent technique. The fractions were used in studies on the enhancement of the precipitin reaction by polymers. Dextran increased the immune precipitation to about the same extent regardless of whether antibodies with high and low affinities were used. The effect should considerably facilitate the detection of antibodies with low precipitating ability in immunological assays. The results are discussed in terms of a steric-exclusion mechanism.

SUMMARY Thyroxine (T4) and tri-iodothyronine (T3) were coupled to human serum albumin (HSA) with carbodi-imide. By adsorption chromatography on Sephadex G-25, fractions containing purified conjugate, but not reversibly-bound T3 or T4, were obtained, and this procedure took 5 h; considerably less than the conventional dialysis technique. Highly specific high-titre antisera were produced in rabbits and guinea-pigs by injection of these fractions in Freund's adjuvant.

ZusammenfassungNach tierexperimentellen Voruntersuchungen wurde die Placentographie mit trägerfreiem 113Inm -HSA als klinische Methode eingeführt. Vor Amniocentesen und bei Verdacht auf Placenta praevia werden Placentographien geschrieben. Den Schwangeren wird eine Aktivität von 500 μCi in die Cubitalvene injiziert. Die der Aktivität entsprechende Indiummenge ist kleiner als 0,1 ng. Die fetale Strahlenbelastung liegt unter lOmrad. Bei Anwendung von 113Inm-HSA entfällt eine Blockade der mütterlichen und fetalen Schilddrüsen. Die genaue Abgrenzung einer Placenta praevia wird nicht durch eine Blasenaktivität beeinträchtigt.Es wurden bisher 19 Placentalokalisationen durchgeführt. In allen Fällen konnte der Placentasitz eindeutig festgestellt werden. Bedingt durch die lange Liegezeit beim Aufnehmen eines Szintigramms kam es in zwei Fällen zu einem Vena-Cava-Kompressions-Syndrom. Zur Verhinderung dieser klinischen Zwischenfälle werden inzwischen Placentographien mit der Anger-Kamera aufgenommen. Mit Hilfe des divergierenden Kollimators konnte der gesamte Abdominalbereich erfaßt werden. Die Aufnahmezeit konnte auf 7 — 10 Minuten verkürzt werden. Die intravenöse injizierte Aktivität betrug bei dieser Methode ebenfalls 500 μCi. Der diagnostische Aussagewert der Kamerabilder ist szintigraphischen Aufnahmen gleichwertig.
