THE EFFECT OF UNSATURATION OF DIETARY FAT AND OF ANTIOXIDANTS ON THE DEVELOPMENT OF LIVER DAMAGE

The addition of unsaturated fatty acids, even in small amounts, to diets low in vitamin E, selenium, and the sulphur-containing amino acids greatly accelerated the development of acute liver necrosis in rats. The production of this lesion was shown also to be affected in a similar manner by the nature of the dietary fat in the ration consumed immediately prior to the removal of protective substances such as the sulphur-containing amino acids; unsaturated fatty acids or lipid again acted as predisposing factors.Gamma-tocopherol incorporated in the diet at a level of 0.02% and injected at a level of 1.5 mg/rat/day was equally as effective as alpha-tocopherol in preventing liver necrosis. The protective effect of gamma-tocopherol against the development of liver necrosis lends support to the thesis that alpha-tocopherol protects against liver damage by virtue of its antioxidant activity.

Vitamin E is the general term for all tocopherols and tocotrienols, of which alpha-tocopherol is the natural and biologically most active form. Although gamma-tocopherol makes a significant contribution to the vitamin E CONTENT in foods, it is less effective in animal and human tissues, where alpha-tocopherol is the most effective chain-breaking lipid-soluble antioxidant. The antioxidant function of vitamin E is critical for the prevention of oxidation of tissue PUFA. Animal experiments have shown that increasing the degree of dietary fatty acid unsaturation increases the peroxidizability of the lipids and reduces the time required to develop symptoms of vitamin E deficiency. From these experiments, relative amounts of vitamin E required to protect the various fatty acids from being peroxidized, could be estimated. Since systematic studies on the vitamin E requirement in relation to PUFA consumption have not been performed in man, recommendations for vitamin E intake are based on animal experiments and human food intake data. An intake of 0.6 mg alpha-tocopherol equivalents per gram linoleic acid is generally seen as adequate for human adults. The minimum vitamin E requirement at consumption of fatty acids with a higher degree of unsaturation can be calculated by a formula, which takes into account the peroxidizability of unsaturated fatty acids and is based on the results of animal experiments. There are, however, no clear data on the vitamin E requirement of humans consuming the more unsaturated fatty acids as for instance EPA (20:5, n-3) and DHA (22:6, n-3). Studies investigating the effects of EPA and DHA supplementation have shown an increase in lipid peroxidation, although amounts of vitamin E were present that are considered adequate in relation to the calculated oxidative potential of these fatty acids. Furthermore, a calculation of the vitamin E requirement, using recent nutritional intake data, shows that a reduction in total fat intake with a concomitant increase in PUFA consumption, including EPA and DHA, will result in an increased amount of vitamin E required. In addition, the methods used in previous studies investigating vitamin E requirement and PUFA consumption (for instance erythrocyte hemolysis), and the techniques used to assess lipid peroxidation (e.g. MDA analysis), may be unsuitable to establish a quantitative relation between vitamin E intake and consumption of highly unsaturated fatty acids. Therefore, further studies are required to establish the vitamin E requirement when the intake of longer-chain, more-unsaturated fatty acids is increased. For this purpose it is necessary to use functional techniques based on the measurement of lipid peroxidation in vivo. Until these data are available, the widely used ratio of at least 0.6 mg alpha-TE/g PUFA is suggested. Higher levels may be necessary, however, for fats that are rich in fatty acids containing more than two double bonds.

Feeding of pig rations containing high amounts of unsaturated fatty acids either in fresh or oxidized condition was demonstrated to possess strongly ulcerogenic properties. The addition of amino acids or selenium, or the increase of dietary protein did not change the number of the lesions. Vitamin E seemed to produce a certain protective effect, but this appeared to be partial and slight. The mode of action of vitamin E in preventing gastric lesions is discussed.

Abstract One of the commodities produced from the ocean that have a bioactive compounds is a sea slug (Discodoris sp.). That is very interesting to study mainly deals with the nature of both chemical and biochemical characteristics and their use for the field of food and health. The aims of this research was to determine the nutrient content of sea slug, determaine chemical content and determine antioxidant activity. The experiment was conducted with several stages: sample preparation, analysis of nutrient content, extraction bioactive compound, analysis of chemical content and antioxidant activity.  Sea slug from the island of Madura Pamekasan potential as a source of protein, fat, and minerals. Sea slug has esensial amino acids amount to 5.57% was dominated by leucin of 1.42%. Nonesensial amino acids amount to 6.54% wich was dominated by glutamic acid 2.19%.  Saturated fatty acids amounted to 27.53% of sea slug was dominated by palmitic (C16: 0) which was 13.36%. Unsaturated fatty acids amounted to 34.66%, which was dominated by the essential fatty acids linolenic (C18: 3, n-3) 20.91%. The highest yield of the crude extract was ethanol and contained alkaloid compounds, steroids, phenols, carbohydrates and reducing sugar compound. The meat of sea slug with ethanol solvent has an IC best 50 antioxidant activity at 441, 12 ppm.

A histologic examination of spontaneous cases of sawdust livers in cattle indicated that the focal liver necrosis was an expression of vitamin E-selenium deficiency. The condition was reproduced in Hereford steers by feeding a diet rich in polyunsaturated fatty acids and poor in protein, vitamin E. and selenium. Lesions also occurred in the kidney, heart, skeletal muscled and pylorus. Addition of dictary protein or injection of selenium partially prevented the condition. Cellular anoxia with formation of hyalinc bodies in the liver and kidney was considered to be the common denominator of the degenerative changes. Due to the relatively mild tissue changes, plasma GOT and OCT determinations were found to be of no diagnostic value.

n the current paper, we aimed to analyze the way in which packing (different polyethylene types) influence the quality of pasteurized melange during storage, packing being realized in units of 1 kg (Tetra Pak) (batch Lexp-1) and units of 5 kg (Bag in box) (batch Lexp-2). Products were stored during a period of 28 days at a temperature of +4�C, qualitative determinations being realized in first day (day 0), at 7 days, at 14 days, 21 days and in day 28 of storage. Were effectuated a sensorial examination and chemical analysis were was established the content in dry matter (%), water (%), proteins (%), content in essential amino acids (isoleucine, methionine, tryptophan, phenylalanine) and non-essential amino acids (alanine, histidine, glycine, serine) (mg/100g) as well as the content in lipids (%) establishing their profile by identification of some saturated fatty acids (16:0 mg/100g and 18:0 mg/100g) and unsaturated fatty acids (16:1 mg/100g and 18:1 mg/100g). After sensorial examinations, the first modifications were observed at the checking effectuated in day 21 for batch Lexp-2, the obtained score being of 18 points, and at checking effectuated in day 28 was given a score of 18 points for melange belonging to batch Lexp-1 and only 14 points for melange from batch Lexp-2. Differences were recorded also in case of chemical composition of products, so for protein content at batch Lexp-1 in first checking day was obtained a mean of 12.730�0.24% and at batch Lexp-2 12.614�0.22%. Differences between those two batches were insignificant (p[0.05). In case of fat content, at the end of storage period was obtained a mean of 11.256�0.06% for batch Lexp-1 and 11.244�0.11% for batch Lexp-2, differences being insignificant (p [ 0.05). Regarding the profile of amino acids and fatty acids, the mean values obtained during whole storage period oscillated from one stage to another, but the differences between those two batches were insignificant (p [ 0.05). Pasteurized egg melange suffers certain sensorial modifications during storage, especially on consistency and colouring, modifications which are accentuated mainly by storage conditions. Type of polyethylene utilized for this product hadn�t influenced the nutritive qualities of product.

Camelina sativa L is a cruciferous oilseed plant. This plant is cultivated as an oilseed crop mainly in Europe and in North America and over the past years the cultivation has arranged in our country. The analyzed oil is obtained from the seeds of Camelina sativa L, growing in Bornuur, Tuv province. The goal of this study was to determine the physical and chemical characteristics and fatty acids composition of Camelina sativa L seed oil cultivated in Mongolia. According to our analysis total lipid was determined 38.52 %, moisture 4.80 % and total mineral elements 4.02 %, respectively. Mineral elements in Camelina sativa L seeds contain calcium (0.56 %), phosphorous (1.22 %), potassium (1.39 %), magnesium (0.53 %) in dominated amounts; iron, zinc, manganese and copper in trace amounts. Eight nonessential amino acids in seeds of this plant with total amount of 75.9 % were identified; phenylalanine was detected in highest amount among the all identified amino acids, while lysine, tryptophan and arginine are followed. The following characteristics in Camelina sativa seeds oil were determined. The refractive index was 1.4774 at 20°C, the peroxide value of fresh oil was 0.03 meq H2O2 /kg, saponification value 185.8 mg KOH/g, iodine value 143.33 g J2 and acidic value 6.27 mg KOH /g. Carotenoid was determined as 16.77 mg %, by spectrometry in Camelina sativa seeds oil. The analysis of fatty acids composition showed that there are 12.5 % saturated and 87.5 % unsaturated fatty acids. In particular, oleic acid (C18:1) 14.0 %, linoleic acid (C18:2) 9.0 %, α-linolenic acid (C18:3) 10.5 % and gondoic acid (C20:1) 32.8 %, were composed the major part of unsaturated fatty acids.
