Development of a SCAR marker linked to the I gene in common bean

Two 24-mer SCAR primers (SW13) were developed from a previously identified 10-mer RAPD primer (OW13690) linked to the I gene, which conditions resistance to bean common mosaic virus (BCMV) in common bean. Linkage between SW13 and the I gene was tested in three F2 populations segregating for both SW13 and the I gene: N84004/Michelite (1.0 ± 0.7 cM), Seafarer/UI-114 (1.3 ± 0.8 cM), and G91201/Alpine (5.0 ± 2.2 cM). SW13 proved to be more specific and reproducible than the OW13690 RAPD marker. Using different heat-stable DNA polymerases, SW13 amplified a single 690-bp fragment linked to the I gene that more consistently permitted the identification of resistant plants. In addition, the presence of the I gene was detected using SW13 in genotypes originating from different gene pools of Phaseolus vulgaris L., indicating a broad utility of this marker for bean breeding programs. Common bean (Phaseolus vulgaris L.) is the most important edible legume in the world and is an important source of income for farmers and food for millions of families. Angular Leaf Spot (ALS), caused by the fungus Pseudocercospora griseola (Sacc.) Ferraris, is one of the most important diseases in the production of common bean with the potential to cause significant seed yield losses. An effective and environmentally friendly alternative to control this disease is the use of improved varieties that combine resistance genes of Andean and Mesoamerican origin. In this study, the response of 181 bean genotypes (coming from different breeding programs of several countries including the USA, Puerto Rico, Honduras, Ecuador, Colombia, Tanzania, Malawi, and Angola) to two angular spot isolates (races 61:11 and 63:51, prevalent in Isabela and Juana Díaz, Puerto Rico) were evaluated. Many of these genotypes contained resistant genes to different biotic and to abiotic stress. A total of 16 lines were identified to have resistance to both races. The resistant lines include the Andean breeding lines CAL 143, 277 and the Mesoamerican cultivar ‘Ouro Negro’ that possess the resistance genes Phg-5, Phg-1, Phg-3, respectively. This information can help common bean breeding programs to pyramid genes from the Andean and Mesoamerican gene pools to generate varieties with long-lasting resistance to this disease.

OAC Redstar is a determinate bush dark red kidney bean (Phaseolus vulgaris L.) cultivar with early maturity and good yield potential. It is resistant to races 1 and 15 of bean common mosaic virus as well as race 17 of anthracnose. Its seed has acceptable cooking and canning quality. In common bean (Phaseolus vulgaris L.), the expression of color in flower and seedcoat tissues requires the dominant allele of the P gene. The fully recessive p allele completely suppresses color expression in these tissues, whereas in specific genetic backgrounds, the pgri allele potentiates a grayish-white seedcoat and pale violet (nearly white) flowers with two violet dots on the banner petals. As a first step to gaining a better understanding of this important gene, we phenotypically scored an F2 population segregating for P and pgri and subsequently screened contrasting bulk DNA samples with oligonucleotide primers to uncover random amplified polymorphic DNA (RAPD) fragments. OU32300, an RAPD marker linked in coupling phase to the dominant allele, mapped 1.3 cM from P. The core 'BAT93' × 'Jalo EEP558' recombinant inbred population was scored, and the marker mapped to linkage group B7. The segregating fragment was cloned, sequenced, and shown to possess significant homology to the Ty3–gypsy class of retrotransposons. We have named the element Tpv3g. It is estimated that about 100 copies of the element are present in the common bean genome. Phylogenetic analysis placed Tpv3g in the class A group of plant retrotransposons.Key words: common bean, molecular markers, Phaseolus vulgaris L., seedcoat color, Ty3–gypsy retrotransposon.

AC Black Diamond is a high-yielding, large-seeded, shiny black dry bean (Phaseolus vulgaris L.) cultivar. It was developed from a series of crosses at the Centro Internacional de Agricultura Tropical (CIAT), Cali, Colombia, on contract to the Agriculture and Agri-Food Canada (AAFC) Research Centre, Lethbridge, with cooperation from the AAFC Research Station Morden. AC Black Diamond is well adapted to the Canadian prairies, yielding significantly more than the check cultivar, UI 906, at 122% in narrow-rows an d 106% in wide-rows. AC Black Diamond is moderately susceptible to white mold and resistant to bean common mosaic virus (BCMV). The objective of this study was to evaluate the degree of RAPD marker variability between and within commercially productive market classes representative of the Andean and Middle American gene pools of common bean (Phaseolus vulgaris L.). Six sets of near-isogenic lines were screened with oligonucleotide primers in the polymerase chain reaction-based RAPD assay. Simultaneous analyses with at least three sets of lines enabled us to score RAPD markers between the two major gene pools, races within the same gene pool, and different genotypes of the same race (within race). A “three-tiered” pattern of polymorphism was observed: between gene pools> between races> within races. The overall level of polymorphism between the Andean and Middle American gene pools was 83.4%. The overall level of polymorphism between races within the same gene pool was similar for Andean races (60.4%) and Middle American races (61.7%). The level of polymorphism between related commercial navy bean lines was 39.2% and between related commercial snap bean lines was 53.6 %. The inherent simplicity and efficiency of RAPD analyses, coupled with the number of polymorphisms detectable between related commercial genotypes, should facilitate the construction of RAPD-based genetic linkage maps in the context of populations representative of most bean breeding programs.
