Genetic factors determining plasticity in flax genotrophs

Changes in the amount of nuclear DNA can be induced in 'Stormont Cirrus', a plastic variety (PL) of flax, by different environments, but not in the linseed variety 'Royal' (R) which is not plastic. PL is thought to contain nuclear and cytoplasmic factors, not possessed by R, which are necessary for the plastic characters to appear. Further crosses have shown that the PL nuclear factor plays a direct part in the determination of plasticity and that the cytoplasmic factor is maintained in the presence of both PL and R nuclei. Insofar as the genetic analysis has been taken, the distribution of genetic factors determining plasticity (+) or nonplasticity (−) in PL and R are as follows: nuclear factor (PL, +; R, −); cytoplasmic factor (PL, +; R, −); nuclear factor for synthesizing de nouveau cytoplasmic factor (PL, −; R, −); nuclear factor for maintaining the cytoplasmic factor (PL, +; R, +); site at which DNA changes occur (PL, +; R, +). It is suggested that the nuclear factor is activated by the cytoplasmic factor, and must come directly from a cytoplasm containing the cytoplasmic factor to be operative.Key words: flax, genotype plasticity, plasticity, genotrophs.

Heritable changes can be induced in the amount of nuclear DNA of the Stormont Cirrus variety (PL) of flax when grown in the specific environments of nitrogen and phosphorus, but such changes did not occur in variety Royal (R). Changes occurred in the amount of nuclear DNA of R when the PL nuclear and cytoplasmic factors were introduced by crossing. In the experiments reported here, tests were made on the plasticity of the F1's of their reciprocal crosses to PL and R to understand why L and S genotrophs are stable. The results suggest that there may be a feedback from nucleus to cytoplasm in L0, which inactivates the cytoplasmic factor. Possibly a paramutation-like mechanism adjusts the different amount of DNA, or heterochromatin, between the homologous chromosomes in heterozygotes. L3 and S3 DNA has reverted to the same amount as in PL. This reversion in DNA appears to be accompanied by an increase in stability rather than in any gain in plasticity. The nuclear DNA difference between large (L) and small (S) genotrophs, induced in the flax variety 'Stormont Cirrus' (PL), was investigated using Feulgen photometry. L and S genotrophs were crossed, respectively, to the PL genotype and the parental, and F1 generations were grown in greenhouse (T1) and field (T2) temperatures. Results demonstrate that (i) the DNA values show increased reversion in the crosses, compared with the parents at T1 and less reversion in the crosses, than in the parents, at T2; (ii) more reversion occurs in the PL maternal crosses than in the PL paternal crosses; and (iii) the PL genotroph possess a nuclear and a cytoplasmic factor which plays a direct part in determining nuclear DNA changes in the chromosomes of the stable genotype.Key words: rDNA, DNA amounts, genotrophs, temperature effects.

This chapter reviews traditional and new zooplankton sampling techniques, sample preservation, and sample analysis, and provides the sources where in-depth discussion of these topics is addressed. The net systems that have been developed over the past 100+ years, many of which are still in use today, can be categorized into eight groups: non-opening/closing nets, simple opening/closing nets, high-speed samplers, neuston samplers, planktobenthos plankton nets, closing cod-end samplers, multiple net systems, and moored plankton collection systems. Methods of sample preservation include preservation for sample enumeration and taxonomic morphological analysis, and preservation of samples for genetic analysis. Methods of analysis of zooplankton samples include determination of biomass, taxonomic composition, and size by traditional methods; and genetic analysis of zooplankton samples.

Nuclear factor-κB (NF-κB) is a ubiquitously expressed cellular factor regulated by the cytoplasmic factor inhibitor protein κBα (IκBα). Activation of NF-κB by cytokines, including tumor necrosis factor-α (TNF-α), requires the phosphorylation and degradation of IκBα. An anti-apoptotic role for NF-κB has recently been suggested. In the present study, we ascertained whether death-promoting signals and apoptosis mediated by TNF-α are suppressed by NF-κB in postnatal ventricular myocytes. Stimulation of myocytes with TNF-α resulted in a 12.1-fold increase ( P < 0.01) in NF-κB-dependent gene transcription and DNA binding compared with controls. This was accompanied by a corresponding increase in the NF-κB target protein A20 as determined by Western blot analysis. Vital staining revealed that TNF-α was not cytotoxic to myocytes and did not provoke apoptosis. Adenovirus-mediated delivery of a nonphosphorylatable form of IκBα to inactivate NF-κB prevented TNF-α-stimulated NF-κB-dependent gene transcription and nuclear NF-κB DNA binding. Importantly, myocytes stimulated with TNF-α and defective for NF-κB activation resulted in a 2.2-fold increase ( P < 0.001) in apoptosis. To our knowledge, the data provide the first indication that a functional NF-κB signaling pathway is crucial for suppressing death-promoting signals mediated by TNF-α in ventricular myocytes.

Pistacia lentiscus L. is a dioecious shrub or small tree with high drought resistance, native in the entire Mediterranean basin. The variety ‘Chia’ (mastic tree) is cultivated for the production of mastic gum (a resin produced after wounding of the trunks and thick branches) in the Island of Chios (Greece), but it also has a significant ornamental value. In the present study, ten male and ten female genotypes of P. lentiscus var. Chia from the natural habitats of Chios were selected and examined with respect to the rooting efficiency of their shoot cuttings. The germination ability of the seeds was also tested. The 20 plants were morphologically described, mainly with respect to traits related to their ornamental value. Furthermore, leaves were collected from the 20 genotypes, from which genomic DNA (gDNA) was isolated, followed by amplification of gDNA fragments using the polymerase chain reaction (PCR) and inter simple sequence repeat (ISSR) primers. This was done aiming both at the determination of their genetic distance and the establishment of possible correlations between the amplified bands and certain morphological traits. The results of the study showed that there were differences among the genotypes for both methods of propagation. Regarding the rooting of the shoot cuttings, the best genotype was 8M (cv. ‘Psilophyllos’), achieving a rooting rate of 62.5% in winter with the application of indolebutyric acid (potassium salt of IBA), while regarding the seed germination capacity the genotype 2F exhibited the highest germination rate (57%). Genetic analysis using ISSRs separated the plants into four groups, one group consisting of male genotypes, one of the female genotypes, one consisting of members from both genders and a fourth containing a single male genotype. The genetic analysis of the male genotypes only produced a dendrogram showing the cultivars clustering in three different groups. Regardless of the genetic analysis, it seems that there were correlations between the ISSR markers and the leaf traits and also the gender and the asexual propagation. These correlations can assist future breeding programs of P. lentiscus var. Chia.

Ixodes scapularis Say, 1821 (the black-legged tick) is becoming established in Canada. The northwards expansion of I. scapularis leads to contact between I. scapularis and Ixodes cookei Packard, 1869, a well-established tick species in Eastern Canada. Examination of I. cookei and I. scapularis collected from New Brunswick revealed ticks with ambiguous morphologies, with either a mixture or intermediate traits typical of I. scapularis and I. cookei, including in characteristics typically used as species identifiers. Genetic analysis to determine if these ticks represent hybrids revealed that four had I. cookei derived mitochondrial DNA but I. scapularis nuclear DNA. In one case, the nuclear sequence showed evidence of heterozygosity for I. scapularis and I. cookei sequences, whereas in the others, the nuclear DNA appeared to be entirely derived from I. scapularis. These data strongly suggest genetic hybridization between these two species. Ixodes cookei and hybrid ticks were readily collected from humans and companion animals and specimens infected with Borrelia burgdorferi Johnson et al., 1984, the causative agent of Lyme disease, were identified. These findings raise the issue of genetic introgression of I. scapularis genes into I. cookei and warrant reassessment of the capacity of I. cookei and I. cookei × I. scapularis hybrids to vector Borrelia infection.

According to an analysis of published data, only 20% of patients with the new coronavirus infection develop severe life-threatening complications. Currently, there are no known biomarkers, the determination of which before the onset of the disease would allow assessing the likelihood of its severe course. The purpose of this literature review was to analyze possible genetic factors characterizing the immune response to the new coronavirus infection that could be associated with the expression of angiotension-converting enzyme 2 (ACE-2) and related proteins as predictors of severe Corona virus disease 2019 (COVID-19). We analyzed original articles published in Medline, PubMed and Scopus databases from December 2019 to November 2020. For searching articles, we used the following keywords: New coronavirus infection, Severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2), COVID-19, severe course, complications, thrombosis, cytokine storm, ACE-2, biomarkers. In total, 3714 publications were selected using the keywords, of which 8 were in congruence with all the criteria. The literature analysis of the association of immunogenic characteristics and the expression of ACE-2 and related proteins with the development of severe COVID-19 revealed following genetic factors: HLA-B*46:01 genotype, CXCR6 gene hypoexpression, CCR9 gene expression, TLR7, rs150892504 mutations in the ERAP2 gene, overexpression of wild-type ACE-2, TMPRSS2 and its different polymorphisms. Genes, associated with the severe course, are more common among men. According to the analysis data, it can be assumed that there are population differences. However, the diagnostic significance of the markers described must be confirmed with additional clinical studies.

In crosses between the two taxa Medicago striata Bast, and M. littoralis Rohde a high mortality of gametes and seedlings, and sterility of some plants were noted which were not related to gross chromosomal rearrangements. Although the F1, F2 and F3 generations from reciprocal crosses differed in chlorophyll deficiencies (indicating a cytoplasmic influence) a genic cause became evident from segregations for chlorophyll characters in the F2 and F3. Transference of the cytoplasmic factor by the pollen is indicative.Segregation for pod coiling direction indicated that the character was determined by one or two genetic factors of which the clockwise coiling direction is recessive. The spininess appeared to be determined by one genetic factor, of which the spineless allele is recessive.On the basis of genetic differences (especially on the built-in repulsion systems for normal chlorophyll development of opposite species) the two taxa should be considered two different species.
