THE CYTOGENETICS OF A DITELOTETRASOMIC LINE IN BARLEY

A ditelotetrasomic line (14+2−1S) carrying two telocentric chromosomes in addition to the normal complement was isolated in the selfed progeny of a monotelotrisomic (14+1S) of barley. The plants were shorter and later maturing than the disomic, and highly sterile. In diakinesis and metaphase I cells at meiosis, the extra telos showed a high frequency of pairing with each other and with their normal homologues, resulting in 6″ + (t″ + 1″) and 7″ + t″ as the most frequent configurations. Regular meiotic behavior at subsequent stages resulted in a 42.2% frequency of pollen transmission of the extra telos. Selfed progenies gave rise to 7.1% diploids, 45.7% monotelotrisomics and 47.2% ditelotetrasomics.

In two hybrids between Triticum timopheevi Zhuk. and T. durum Desf., which have irregular meioses, metaphase I was subdivided into four developmental stages, early, medium, late, and very late. This subdivision was based on the presence in the anthers of other stages that occurred together with metaphase I. It was then discovered that in metaphase I cell populations there was a progressive increase of univalents from the early and medium stages to the very late stage. This phenomenon can be explained on the assumption that metaphase I is of shorter duration in the less irregular cells which pass into anaphase I earlier than the more irregular cells. As a consequence of this developmental phenomenon at metaphase I, the anaphase I and telophase I cells in the late anthers contained fewer lagging chromosomes than the anaphase I and telophase I cells in the very late anthers. Despite these numerical differences, the degrees of lagging were remarkably similar in both stages; approximately 70% of these univalents lagged at late and very late stages in both hybrids.During metaphase I many univalents of the irregular cells moved towards the equatorial plate, became oriented, and lagged at anaphase I and telophase I. It was found that the univalents of the least irregular cells accumulated somewhat faster at the plates than those of the more irregular cells.Considering the relationships between all of the available data, the hypothesis is advanced that when a certain number of univalents have accumulated at the equatorial plate a state of equilibrium is established and anaphase I is initiated. On the basis of this hypothesis an explanation of the trends observed at metaphase I is given.

Somaclonal variation occurs among regenerants from tissue culture of many plant species. Our objective was to determine whether cytogenetic variation contributes to somaclonal variation in cotton (Gossyptum hirsutum L.,2n = 4x = 52). Of 117 somaclones of cotton regenerated from 18-month-old callus cultures of 'SJ-2' and 'SJ-5' cultivars, 35 were analyzed for meiotic abnormalities. The population of somaclones was extremely varied in phenotype, most plants being strikingly aberrant in phenotype. Fertility was generally poor: 84% failed to set bolls and only 5% set 10 or more bolls in a field environment. Only one of the somaclones (3%) formed 26 bivalents at metaphase I. Fourteen were nonsynaptic to partially synaptic at metaphase I. Synaptic abnormalities impaired fertility and precluded thorough metaphase analysis. Chromosome numbers obtained for 32 plants ranged from 49 to 53, and only 1 plant was hyperaneuploid. No plant was polyploid. Chromosomal abnormalities in plants with normal metaphase pairing included univalents, unequal bivalents, rod bivalents, trivalents, open quadrivalents, and centric fragments. Seventeen hypoaneuploid plants formed a V-shaped trivalent at metaphase I, constituting a high frequency of tertiary monosomy. The high frequencies of aneuploidy and tertiary monosomy indicate that cytogenetic anomalies are a major source of somaclonal variation in cotton. It is hypothesized that (i) primary cytogenetic events during cotton cell culture give rise to breakage – fusion – bridge (BFB) cycles, (ii) BFB cycles accrue during culture, (iii) BFB cycles cause loss of chromatin, and (iv) BFB cycles are resolved by the formation of stable tertiary chromosomes with mono-centric activity. The hypothesis accounts mechanistically for the coincidence of chromatin deficiencies and chromatin exchange involved implicitly in tertiary monosomy, as well as for the relatively high frequency of tertiary monosomy among somaclones.Key words: aneuploid, monosomic, synaptic, sterility, Gossypium.

The multivalents that appeared in the decaploid strain of Agropyron elongatum (2n = 10x = 70), a relative of wheat, ranged from trivalent to decavalent. Few univalents occurred. The metaphase I chromosome association in 12 cells where all configurations could clearly be identified averaged 0.42 ring X + 0.17 chain X + 0.42 ring VIII + 0.17 branched VIII + 0.25 chain VIII + 0.17 chain VII + 1.17 ring VI + 0.33 branched VI + 0.5 chain VI + 1.67 ring IV + 0.42 branched IV + 0.58 chain IV + 0.08 branched III + 0.17 chain III + 12.58 ring II + 3.75 open II + 0.25 I. The occurrence of decavalents, up to two in one cell, and of a cell with five multivalents, each of which involved more than five chromosomes, and many multivalents of ring shape indicated that the strain is autodecaploid.The chromosome associations of each cell can be interpreted as seven groups of 10 homologous chromosomes. The high frequency of bivalents indicated a tendency toward reduced multivalent formation, for which an explanation is suggested.Key words: Agropyron elongatum, meiotic configuration, decaploid, multivalent.

A ditelosomic line was crossed with 12 identified monosomics. The metaphase I pairing was studied in the 40 + t – chromosome progenies. The telocentric chromosome was found to be homologous to the three previously identified monosomics namely, −7, −10, and −20 suggesting that the same chromosome is missing in all three lines. None of the chromosomes in monosomic condition used in the crosses except ST-7 and ST-17 from A. byzantina were involved in the reciprocal translocation present between Sun II and Garry and Rodney.The gene for normal vs. abaxial curling of the leaves was located on the short arm of chromosome 20. The genes for diploidisation and normal vs. kinky neck were located on 20L.

Edible chia seeds, purchased from the local markets in Guatemala, Mexico, and southern California, were used for species identification, chromosome counts, karyotype construction, and meiotic analyses. Plants raised from those seeds had ovate leaves, dense racemose inflorescences, pale-blue flowers, and were identified from herbarium specimens as Salvia hispanica L. Mitotic analyses of root tips from 50 plants showed 2n = 12—the lowest chromosome number in the genus. Chromosomes were small, ranging from 2 to 3.5 pm. One pair of chromosomes was metacentric (with the long arm: short arm ratio, r = 1.5), four pairs were submetacentric (r = 2.6 to 3.6), and one pair was telocentric (r = 12). Meiosis was regular and six bivalents were observed at metaphase I. Ring and rod bivalents averaged 1.53 ± 1.05 and 4.47 ± 1.05, respectively;

Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired disorder associated with absence of expression of phosphatidylinositol (PI)- linked membrane proteins from circulating hematopoietic cells of multiple lineages. Recent work demonstrated that decay accelerating factor, one such PI-linked protein, bears the Cromer-related blood group antigens. This study demonstrated that other high incidence antigens, including Cartwright (Yta/Ytb), Holley-Gregory (Hy/Gya), John Milton Hagen (JMH), and Dombrock (Doa/Dob), are absent from the complement-sensitive (PNH III) erythrocytes of patients with PNH. The relatively normal, complement-insensitive erythrocytes from the same patients express these antigens normally. Therefore, these antigens most likely reside on PI-linked proteins absent from PNH III, but not PNH I, erythrocytes.

Complete reconstructions of all the bivalents were made from electron micrographs of serial sections through six pollen mother cells at metaphase I of meiosis in Triticum aestivum (hexaploid bread wheat). At least two of these metaphases contained interlocked pairs of bivalents. In one, two ring bivalents were interlocked, while in another a rod bivalent ran through the centre of a ring bivalent. Two other groups of bivalents were too closely appressed to allow separation into individual bivalents and may have contained interlocks. Meiosis in other anthers of the same plants examined by light microscopy was considered normal. The frequency of interlocking found was much higher than reported from light-microscope spreads. Not all interlocks in metaphase I cells need adversely affect meiosis, but knowledge of their regularity and form may facilitate understanding the processes of chromosome pairing.

Meiosis was studied in two diploid (2n = 2x = 24) siblings of Solanum phureja Juz. et Buk. and in 11 disomic and 2 trisomic descendants. The diploid siblings carry the same heterozygous interchange and either one or two inversions. The frequency of quadrivalents at diakinesis/metaphase I in these clones was 0.56 and 0.62 per pollen mother cell. In two plants from the first inbred generation (I1) this frequency was about the same but in some other I1 plants and a full sib the frequency was substantially lower, varying from 0.00 to 0.16. Most quadrivalents, 78–83%, were rings. A variety of quadrivalent configurations at diakinesis and metaphase I was observed, giving rise to balanced and unbalanced gametes. The absence of ring quadrivalents in trisomic descendants of one of the siblings implied that tertiary trisomics or primaries being homozygous for the interchange were present in the I1 generation. Regular chromosome distribution (12–12) at anaphase I occurred in 46.5 and 73.2% of the pollen mother cells studied in the two original clones. Irregularities, such as 11–13 distribution, lagging chromosomes, and a bridge and fragment, were detected on average in 2.7, 3.3, and 32.5%, respectively, of the anaphase I cells analysed. In hybrids from crosses between 6 primary trisomics as females with the interchange heterozygote, the involvement in the interchange of chromosomes 3 and 12 was clearly demonstrated.Key words: Solanum phureja, interchange heterozygote, chromosome identification, interchange trisomic, meiosis.

The somatic number, meiotic behavior, morphology and ecology of five forms of Paspalum dilatatum Poir are reported. The sexual type yellow anther (2n = 40) showed essentially regular meiotic behavior. The common form (2n = 50) presented 20 II and 10 I in about 85% of the cells studied at diakinesis and metaphase I. Pauciciliatum (2n = 40) and Uruguaiana (2n = 60) showed a wide range of chromosome configurations at these stages; finally, Torres presented uniformly 60 univalents. These results are compared with morphologic and ecologic data and some suggestions about the possible mode of origin of these forms are presented.

Ten telocentric chromosomes of diploid Agropyron elongatum (Host.) P.B. (2n = 14) were added to the chromosome complement of Triticum aestivum L. emend. Thell. The ditelosomic additions were crossed with Triticum speltoides (Tausch) Gren. ex Richter, and in the tetraploid hybrids the pairing frequencies of the telosomes were determined, expressed as percent of PMC's in which a telosome paired at metaphase I. All Agropyron telosomes paired with Triticum chromosomes. The pairing frequencies ranged from 4.4% to 41.2% of the PMC's, it is concluded that none of the ten Agropyron chromosome arms has a homologous partner among the four Triticum genomes involved. The pairing frequencies did not correlate with the lengths of the telosomes. Pairing of the Agropyron telosomes in these tetraploid hybrids approximated the chromosome pairing that occurred in a diploid hybrid T. tauschii (Coss.) Schmal. (the donor of the D genome of T. aestivum) × A. elongatum.
