Responsiveness of the interrenal tissue of yellow perch (Perca flavescens) from contaminated sites to an ACTH challenge test in vivo

The effects of chronic toxic stress on the hypothalamo-pituitary-interrenal (HPI) axis were investigated in yellow perch (Perca flavescens) captured at a reference site (Lake Memphremagog) and two sites contaminated with heavy metals and organic contaminants (Ile Perrot and Iles de la Paix, Lake St. Louis) in spring, summer, and fall. Cortisol secretion of the fish was stimulated in situ by an acute capture stress or by an i.p. injection of 4 IU/100 g body mass of porcine corticotropin (ACTH1-39). The response to both these challenges was lower in perch from the highly contaminated site than in perch from the reference site in the spring but not in summer. In fall, fish from the highly contaminated site had, as in spring, a lower response to ACTH than fish from the reference site. The reduced ability of perch to respond to capture stress or to ACTH indicates that the interrenal tissue in fish from contaminated sites is functionally impaired. Cortisol-impaired fish also had abnormal carbohydrate metabolism. The reduced ability of wild fish from contaminated sites to respond to a standardized ACTH challenge may be used as an early indicator of contamination-induced chronic stress.

The characteristic elevation of plasma cortisol levels in response to an acute stress of capture was impaired in both male and female yellow perch (Perca flavescens) from lakes contaminated by heavy metals. The impairment of the cortisol stress response was observed in fish 4 + years and older whereas the capacity to elevate plasma cortisol levels of fish younger than 4 + was not significantly different at contaminated and reference sites. The responsiveness to ACTH of the interrenal tissue of 4 + yellow perch was evalutated in vitro to determine whether the impairment of the cortisol stress response is caused by a dysfunction of the interrenal tissue or if the dysfunction is located elsewhere in the hypothalamo-pituitary-interrenal axis controlling the secretion of cortisol. The amount of cortisol secreted by the interrenal tissue of yellow perch from a contaminated site in response to a 10-min stimulation with 10-7 M ACTH was significantly lower compared with fish from the reference site. These results indicate that the impairment of the cortisol stress response observed in fish from polluted sites is caused, at least in part, by a dysfunction of the interrenal tissue.

A study was conducted to ascertain the health of winter flounder (Pleuronectes americanus) sampled near a naval facility contaminated with polychlorinated biphenyls (PCBs), using length-mass relationship, hematological values, histopathology, and parasitism as bioindicators of stress. Samples were collected by SCUBA divers about 2 km (Broad Cove) and 5 km (Fox Harbour) down-current from the dockyard and a reference site (Fox Island) 5 km offshore in Placentia Bay, Newfoundland. Length, mass, and condition factor were significantly greater in both sexes sampled at the reference site than at the two contaminated sites, whereas macroscopic anomalies, including fin necrosis, infestation with ectoparasitic metacercariae of the digenean Cryptocotyle lingua, and histopathological lesions in the liver, kidney, spleen, and gills, were more frequent at the latter locations. A greater variety of prey were consumed by fish taken at Fox Island than at Fox Harbour, while food was absent in several samples from Broad Cove. Three endoparasites, Ceratomyxa acadiensis, Echinorhynchus gadi, and Steringophorus furciger, were less prevalent and abundant in flounder sampled at Broad Cove than at the other two sites. These results suggest a connection between PCB contamination and impaired fish health. In addition, anomalies were more prevalent in samples collected nearest to the contaminated site than in those collected farther away.

Zinc is an essential micronutrient for freshwater fish but can be toxic to them at elevated concentrations. Therefore, the regulation of zinc uptake is important in maintaining homeostasis when fish are chronically exposed to elevated zinc in nature. This study examined the kinetics of in vivo branchial and in vitro intestinal zinc uptake in wild yellow perch (Perca flavescens) from metal-contaminated and reference lakes in northern Ontario. The results showed that the branchial zinc uptake involves high-affinity transport sites, whereas the intestinal zinc uptake involves low-affinity transport sites. Interestingly, significant alterations in the branchial zinc uptake (reduced affinity, increased maximum transport rate) but no apparent changes in the intestinal zinc uptake characteristics were observed in the metal-impacted yellow perch population relative to the reference population. Subsequently, no differences in zinc concentrations of gill, liver, and whole body were recorded between reference and metal-impacted yellow perch populations. Overall, our study indicated that the gill, not the gut, likely plays a critical role in maintaining the zinc homeostasis in wild fish under chronic exposure.

We have conducted studies with juvenile yellow perch (Perca flavescens) over a period of 20 weeks to address the question of the interaction between water- and lipid-soluble antioxidant vitamins. Fish (2·25±0·14 g) were divided into twelve groups, and triplicate groups were fed one of four casein-based, semi-purified diets formulated to contain low or high vitamin E levels of either 5 or 160 mg/kg without or with vitamin C supplementation (250 mg/kg). Diets were designated as −C−E, −C+E, +C−E, or +C+E, respectively. The fish fed the +C+E diet showed significantly higher weight gain, feed intake, and feed efficiency than the groups fed vitamin C-deficient diets. Total ascorbate concentrations of liver were significantly higher in fish fed vitamin C-supplemented diets than in fish fed the vitamin C-deficient diet after 16 and 20 weeks. The liver α-tocopherol concentrations were increased by supplemental vitamin C in vitamin E-deficient dietary groups which indicates a sparing or regenerating effect of vitamin C on vitamin E. Fish fed vitamin C-deficient diets (−C−E and −C+E) exhibited severe deficiency symptoms, such as scoliosis, lens cataracts, anorexia, and haemorrhages. The cumulative mortality was significantly higher in the −C−E groups. The thiobarbituric acid-reactive substances value was significantly higher in blood plasma of fish fed a diet unsupplemented with both vitamins. The findings in the present study with yellow perch support the hypothesis that vitamin C regenerates and/or spares vitamin E in vivo.

Parasites were examined in yellow perch, Perca flavescens (Mitchill, 1814), from four localities ranging in degree of pollution in the St. Lawrence River, Quebec, Canada, to examine the effects of the most prevalent parasite species on expression of biomarkers of oxidative stress. Various biomarkers appeared to be affected by the infection levels of Apophallus brevis Ransom, 1920 and genus Diplostomum von Nordmann, 1832. For certain biomarkers, interactions between infection level and pollution type were detected for A. brevis, Diplostomum spp., and genus Ichthyocotylurus Odening, 1969. Activity of glutathione reductase in gill tissue decreased with increasing numbers of A. brevis, but only at the two most polluted localities. Catalase activity in kidney increased with numbers of Diplostomum spp. at the polluted localities, but not at the two least contaminated sites. Results suggest that parasites may affect expression of biomarkers of pollution and that pathogenicity of parasites may be enhanced under polluted conditions. Exposure to contaminants appears to reduce tolerance, but not resistance, to parasites in yellow perch in this system. This type of immunosuppression may be widespread in polluted ecosystems.

Luminescent Vibrio harveyi is common in sea and estuarine waters. It produces several virulence factors and negatively affects larval penaeid shrimp in hatcheries, resulting in severe economic losses to shrimp aquaculture. Although V. harveyi is an important pathogen of shrimp, its pathogenicity mechanisms have yet to be completely elucidated. In the present study, isolates of V. harveyi were isolated and characterized from diseased Penaeus monodon postlarvae from hatcheries in Kerala, India, from September to December 2016. All 23 tested isolates were positive for lipase, phospholipase, caseinase, gelatinase and chitinase activity, and 3 of the isolates (MFB32, MFB71 and MFB68) showed potential for significant biofilm formation. Based on the presence of virulence genes, the isolates of V. harveyi were grouped into 6 genotypes, predominated by vhpA+ flaB+ ser+ vhh1- luxR+ vopD- vcrD+ vscN-. One isolate from each genotype was randomly selected for in vivo virulence experiments, and the LD50 ranged from 1.7 ± 0.5 × 103 to 4.1 ± 0.1 × 105 CFU ml-1. The expression of genes during the infection in postlarvae was high in 2 of the isolates (MFB12 and MFB32), consistent with the result of the challenge test. However, in MFB19, even though all genes tested were present, their expression level was very low and likely contributed to its lack of virulence. Because of the significant variation in gene expression, the presence of virulence genes alone cannot be used as a marker for pathogenicity of V. harveyi.

The study aims to isolate, characterize, and examine probiotic bacteria's inhibitory ability against Vibrio harveyi bacteria, both in-vitro and in vivo. Methods used in the study consist of 1) An Isolation of Candidate Probiotic Bacteria, 2) An Antagonistic Test of Candidate Probiotic Bacteria in vitro, 3) An Identification of Bacteria, 4) A Pathogenicity Test of Candidate Probiotic Bacteria, 5) An Antagonistic Test of Candidate Probiotic Bacteria against V. harveyi in vivo. According to the isolation of candidate probiotic bacteria, there are 18 isolated candidate probiotic. After being tested for its inhibitory ability in vitro, there are 8 isolates with zone of inhibition as follows: isolate MM 7 from intestine (22 mm), isolate MM 6 from intestine (12 mm), isolate MM 10 from sea water (10 mm), isolate MM 5 from intestine (9 mm), isolate MM 4 from intestine (8 mm), isolate MM 3 from intestine (7 mm), isolate MM 2.2 from intestine (7 mm), isolate MM 2.1 from intestine (7 mm). Eight genera of the candidate probiotic bacteria is derived from Portunid crab, they are Staphylococcus, Streptococcus, bacillus, vibrio, Alcaligenes, Lactobacillus, micrococcus. Before proceeding the V. harveyi bacterial challenge test in vivo, three potential isolates consisting of MM6, MM7 and MM10 as the probiotic bacteria are pathogenicity-tested against V. harveyi. The survival rate of Portunid crab on pathogenicity test using MM6, MM7 and MM10 generates 91.11-100%, while the control generates 100% survival rate. Variance analysis result through post-hoc Tukey's Honest Significant Difference (HSD) test at 95% confidence interval indicates that isolate MM7 and MM10 are significantly able to increase hatchling Portunid crab's survival rate.

A 51 year-old male was admitted to our hospital with chief complaints of fever, dry cough and dyspnea. Chest X -ray films and his history of taking Chinese medicine for liver dysfunction were suggestive of drug-induced pneumonitis. Lymphocyte stimulation test (LST) to causative Chinese medical drugs of Sho-saiko-to and Dai-saiko-to was negative with peripheral blood lymphocytes (PBL), but was positive with Iymphocytes from bronchoalveolar lavage fluid (BALF). In vivo challenge test for Sho-saiko-to was positive. The LST with BALF-lymphocytes proved to be very useful in making a diagnosis of drug-induced pneumonitis.
