Sensitivity of Blood Cell Counts in Juvenile Coho Salmon (Oncorhynchus kisutch) to Stressors Including Sublethal Concentrations of Pulpmill Effluent and Zinc

Transfer of fish from optimal to high-temperature, crowded holding conditions decreased white blood cell-thrombocyte (WBC-T) counts markedly on 12–96-h exposure. Red blood cell (RBC) counts were increased only slightly at 12 and 48 h. During 28 days acclimatization following transfer to simulated stream compartments, WBC-T counts returned to stock values within 2–4 days, following an initial decline. RBC counts were not altered appreciably.The decline in WBC-T counts of acclimatized fish to a high sublethal concentration of pulpmill effluent was greatest at 24 h. Mean RBC counts were decreased over 24–96 h, but differences were significant at 24 h only. In acclimatized fish exposed to pulpmill effluent, WBC-T counts were decreased (P < 0.001) at concentrations between 0.2 and 0.9 of the 96-h LC50. A 24-h exposure to 15% vol/vol of this effluent had considerably less effect on the WBC-T count when the effluent had been detoxified by foam fractionation.Under similar experimental conditions, exposure to sublethal levels of zinc depressed mean WBC-T counts proportional to concentration, although differences were only significant at 0.5 LC50 and greater. RBC counts were unchanged.The WBC-T response provides a reasonably rapid and sensitive method for measuring stressful levels of pulpmill effluents to salmon. Declines in WBC-T counts are attributable to reduced numbers of circulating small lymphocytes, which could result in decreased resistance of stressed fishes to disease.

To determine if changes in some hematological parameters accompanied acute exposure of juvenile coho salmon (Oncorhynchus kisutch) to dehydroabietic acid (DHAA) at three different exercise levels, clotting times, hematocrits, erythrocyte sedimentation rates, red blood cell counts, and white blood cell counts were monitored over 6-, 12-, 24-, and 48-h exposure periods to 0.75 mg/liter DHAA.White cell counts decreased significantly after 24 h exposure and there was a significant increase in clotting times after 48 h exposure to DHAA. In both cases, differences between experimentals and controls were significant only at low and intermediate exercise levels and not at a high exercise level. Hematocrits, erythrocyte sedimentation rates, and red blood cell counts were not significantly different between experimentals and controls for any of the exposure periods or exercise levels.

Channa punctatus was exposed to four different concentrations of Rutin, Taraxerol and Apigenin. Changes in some hematological parameters of Channa punctatus were assessed to determine the influence of these compounds on test fish. Fish were exposed to sublethal concentrations (80% of LC50 of 24h) of these compounds for one week. Control fish were also administered for one week. Thereafter, blood samples were obtained from the control and experimental fish. Blood was assayed for selected hematological parameters (hematocrit, hemoglobin, red blood cell count, white blood cell count total plasma protein and plasma glucose concentration). The derived hematological indices of mean corpuscular hemoglobin concentration (MCHC), mean corpuscular hemoglobin (MCH) and mean corpuscular volume (MCV) were calculated. Sublethal concentrations of these compounds caused a dose dependent decrease in hemoglobin values coupled with a decrease in hematocrit values and red blood cell counts are an obvious indication of anemia. The total white blood cell counts and the differential white blood cell counts were decreased except for the lymphocytes, where there was a slight increase. Plasma protein and glucose were also lower in exposed fish when compared with control. The hematological indices MCH, MCHC, MCV were also lowered. The result from this study reveals high mortality rate and deleterious consequences on the health of fish subjected to acute exposure of Rutin, Taraxerol and Apigenin and therefore, should not be used directly in aquaculture without having the proper knowledge.

SummaryA 74-year-old female developed pneumonia following herpes simplex encephalitis. Her white blood cell counts reached 28,400/μl, about 90% of which consisted of granulocytes. The polymorphonuclear (PMN) elastase/α1-arantitrypsin complex levels increased and reached the maximum of 5,019 ng/ml, indicating the release of a large amount of elastase derived from the granulocytes. The mechanism of PMN elastase release was most likely to be granulocyte destruction associated with phagocytosis. The cleavage of fibrinogen and fibrin by PMN elastase, independent of plasmin, was indicated by the presence of the fragments in immunoprecipitated plasma from the patient corresponding to elastase-induced FDP D and DD fragments and the absence of fragments corresponding to plasmin-induced FDP D and DD fragments on SDS-PAGE. These findings suggested that the large amount of PMN elastase released from the excessive numbers of granulocytes in this patient with herpes simplex encephalitis and pneumonia, induced the cleavage of fibrinogen and fibrin without the participation of plasmin.

Modern causal mediation theory has formalized several types of indirect and direct effects of treatment on outcomes regarding specific mediator variables. We reviewed and unified distinct approaches to estimate the “interventional” direct and indirect effects for multiple mediators and time-varying variables. This study was motivated by a clinical trial of elderly type-2 diabetic patients in which atorvastatin was widely prescribed to control patients’ cholesterol levels to reduce diabetic complications, including cardiovascular disease. Among atorvastatin’s preventive side-effects (pleiotropic effects), we focus on its anti-inflammatory action as measured by white blood cell counts. Hence, we estimate atorvastatin’s interventional indirect effects through cholesterol lowering and through anti-inflammatory action, and interventional direct effect bypassing these two actions. In our analysis, total effect (six-year cardiovascular disease risk difference) estimated by standard plug-in g-formula of −3.65% (95% confidence interval: −10.29%, 4.38%) is decomposed into indirect effect via low-density lipoprotein cholesterol (−0.90% [−1.91%, −0.07%]), via white blood cell counts (−0.03% [−0.22%, 0.11%]), and direct effect (−2.84% [−9.71%, 5.41%]) by the proposed parametric mediational g-formula. The SAS program and its evaluation via simulated datasets are provided in the Supplemental materials.
