Curves are presented showing the growth in liquid egg of Salmonella bareilly, S. manhattan, S. typhi-murium, S. oranienburg, S. typhi, Escherichia coli, Aerobacter aerogenes, Staphylococcus aureus, Streptococcus fecalis and S. pyogenes, and of Salmonella bareilly in reconstituted egg. Streptococcus pyogenes does not grow in egg and dies off rapidly at temperatures above 60° F. (15.6 °C.). The other organisms generally grow well in liquid egg at temperatures above 60° F. (15.6 °C.). Liquid and reconstituted egg should therefore be maintained well below this temperature to prevent the multiplication of Salmonella and other possible pathogens.

Experiments were made to find out whether the common communion cup is likely to serve as a vehicle for the transmission of infection.A silver chalice and sacramental wine containing 14·5% of alcohol were used. Observations with volunteers showed that the number of organisms deposited on the rim of the chalice varied from person to person, but was usually quite small—less than 100.Rotation of the cup was of no benefit except to those partaking during the first round, since the saliva deposited on the rim by each person in turn remained to contaminate the cup during the second round, and the combined effect of the alcohol and the silver of the chalice was not rapid enough to destroy the contaminating organisms before rotation of the cup was completed. On the other hand the use of a linen cloth or purificator led to a diminution of about 90% in the bacterial count of the cup.Organisms in saliva deposited on the interior of the dry chalice suffered some diminution in numbers within 8 min., presumably as the result of the disinfectant action of the silver, but the effect was too small to be of significance. When suspended in wine and deposited on the internal surface of the chalice Escherichia coli suffered a substantial reduction within 3 min., Streptococcus pyogenes was destroyed completely; but Staphylococcus aureus was affected to a much less extent.Various experiments designed to measure the disinfectant action of wine, and of silver and wine together, showed that the augmenting effect of silver on the disinfectant action of the alcohol was quite small. Strep. pyogenes proved to be far more sensitive to alcohol than Esch. coli, Staph. aureus and Serratia marcescens. Under the conditions of the experiment these last three organisms were not destroyed for 10–12 min., whereas Strep. pyogenes perished within 1½ min.The results of our work are in general agreement with those of previous workers, and show that the organisms deposited on the rim of the communion cup are not destroyed within the short time—5 sec. as an average—elapsing between the partaking of the sacrament by each successive communicant. It must therefore be admitted that the common communion cup may serve as a means of transmitting infection. Reasons are given, however, for believing that the risk of transmission is very small, and probably much smaller than that of contracting infection by other methods in any gathering of people.Such risk as there is could be greatly diminished by the use of a purificator for wiping the cup between each communicant, and could be abolished completely by substituting individual cups or by the practice of intinction.

Persistent foam formation occurring during blending of jelly powder (JP), whole egg powder (WEP) and skim milk power (SMP) was demonstrated. Reduction of foaming was uniformly best achieved by using the Colworth Stomacher 400 (STOM) for mixing or blending under a vacuum (VAC). Two hundred, 1000 and 6000 ppm of Dow Corning Antifoam Agent (DAF) was necessary to reduce foaming of JP, SMP and WEP, respectively. Escherichia coli was recovered in similar numbers from JP, SMP and WEP by normal blending (N), STOM, VAC or by the application of 200 ppm of DAF. Significantly lower recovery of Staphylococcus aureus from JP resulted when N or VAC was employed and from SMP when STOM was used. Streptococcus faecalis showed significantly lower recoveries from JP and WEP when STOM or VAC and DAF at 200 ppm were used.

ABSTRACT The C30 carotene synthase CrtM from Staphylococcus aureus and the C40 carotene synthase CrtB from Erwinia uredovora were swapped into their respective foreign C40 and C30 biosynthetic pathways (heterologously expressed in Escherichia coli) and evaluated for function. Each displayed negligible ability to synthesize the natural carotenoid product of the other. After one round of mutagenesis and screening, we isolated 116 variants of CrtM able to synthesize C40 carotenoids. In contrast, we failed to find a single variant of CrtB with detectable C30 activity. Subsequent analysis revealed that the best CrtM mutants performed comparably to CrtB in an in vivo C40 pathway. These mutants showed significant variation in performance in their original C30 pathway, indicating the emergence of enzymes with broadened substrate specificity as well as those with shifted specificity. We discovered that Phe 26 alone determines the specificity of CrtM. The plasticity of CrtM with respect to its substrate and product range highlights the potential for creating further new carotenoid backbone structures.

Studies were made to compare the growth and death of Staphylococcus aureus, Streptococcus faecalis and Escherichia coli in skim milk concentrated by ultrafiltration to that in unconcentrated skim milk. Skim milk was volume concentrated to 2× in laboratory-scale stirred UF cells. Behavior of the organisms was analyzed in four inoculated milk samples: 2× retentate, 1× water-diluted retentate, milk equivalent (retentate plus permeate) and unconcentrated skim milk. Growth of each organism and of total aerobes did not vary in the four milk samples at either 7 or 13°C. For S. faecalis and E. coli, D-values for samples heated to 62.7°C did not significantly differ in the four milk samples (p>0.01). The D-value of S. aureus in water-diluted retentate was slightly but significantly lower than those in the other three milk samples (p<0.01), possibly due to the lowered lactose level in this sample.

Nosocomial infection is a rising problem in developing countries. Microorganisms Isolated from three private and public hospital environmental surfaces in Akure Metropolis, Ondo State, Nigeria were investigated in this study. Bacterial and fungal organisms were isolated and compared among the three hospitals. The study revealed that bacteria were the most predominant microorganisms found in the hospital environmental surfaces than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria but Aspergillus funmigatus was the predominant fungi. The result showed that the microbial loads of the public hospitals were higher than that of the private hospitals. The bacteria load of the male ward was found to be higher than that of the female ward while the fungal loads of each of the hospital environmental surfaces of female were higher than that of the male. The study revealed that bacteria were the most predominant microorganisms found in the hospital environment than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria. All the hospital environmental surfaces were contaminated with one or more microorganisms in the course of the research.

The phagocytic capacity of macrophages from C3H/H3J mice was assessed against lipopolysaccharide-producing (Escherichia coli) and -nonproducing (Staphylococcus aureus) bacteria. Despite their gene-coded unresponsiveness to lipopolysaccharide endotoxin and lymphokines and their defective tumoricidal activity, proteose peptone-induced C3H/HeJ macrophages did not display a defective phagocytic capacity, but rather displayed an enhanced phagocytosis of both bacterial strains compared with macrophages from closely related C3H/HeN mice. Unstimulated peritoneal resident C3H/HeJ macrophages, on the other hand, displayed a normal phagocytic activity toward E. coli and enhanced phagocytosis toward S. aureus.

The hygiene status of most abattoirs and retail outlets in Nigeria is very poor, and it contributes to the unacceptable level of bacteria load in beef, and poses a health risk to consumers. This study examined the bacteriological analysis of beef production chain in Birnin Kebbi metropolis. A total of 100 samples of meat, water and surface swab in Main Market, GRA, Badariya, Bayan Kara and Rafin Atiku, Birnin Kebbi Central abattoir and retail meat shops were collected aseptically, processed and analyzed. Meat from retail outlets were mostly contaminated by Staphylococcus aureus (24.4%), followed by Escherichia coli (17.3%), Klebsiella pneumonia (9.8%), Salmonella typhimurium (9.1%), Entrobater aerogenes (8.5%), Streptococcus pyogenes (8.1%), Proteus vulgaris (7.8%), Micrococcus luteus (7.8%) and Pseudomonas aeruginosa (7.2%). In the abattoirs contact surfaces, contamination occurred mostly by Staphylococcus aureus (23.4%), Salmonella typhimurium (18.2%), Escherichia coli (16.9%), Entrobater aerogenes (9.1%), Streptococcus pyogenes (7.8%), Klebsiella pneumonia, Proteus vulgaris, and Pseudomonas aeruginosa (6.5% each) and Micrococcus luteus (5.2%). The findings revealed a high bacterial load in both abattoir and retail meat outlets. In order to safeguard the health of the public against the risks of food borne infections, there is a need to educate and advocate good sanitation and meat handling practices in the abattoir and beef retail outlets.
