Time course of anatomical changes to stem vascular tissues of alfalfa, Medicago sativa, from probing injury by the potato leafhopper, Empoasca fabae

We used a videomicrography technique to apply standard-duration pulses of leafhopper probing damage to alfalfa stems, or manually punctured stems with an implement resembling leafhopper stylets, to compare damage induction and chronicle the plant's anatomical responses over time. Plants were examined at 0.5, 1, 2, 4, and 8 days after probing. Leafhopper probing occurred primarily in phloem tissue: stylets damaged and deposited sheath saliva primarily in phloem. Between days 1 and 4, collapsed phloem cells and sheath saliva deposits were dissolved and removed as surviving cells adjacent to stylet pathways underwent enlargement and division. By day 8, phloem tissue assumed a near-normal appearance when viewed in cross section, although it is not known whether phloem functionality was restored. In contrast, xylem tissue suffered reduction in size, quantity, and total cross-sectional area of mature tracheary elements. These effects to vascular tissue may be responsible for all subsequent symptoms of hopperburn in alfalfa. Damage to phloem and xylem in mechanically punctured stems was slight and was similar for all time points. Studies (1—4) have shown that it is possible to distinguish different stages of phloem tissue differentiation in the developing roots of Lemna minor by examination in the transmission, scanning, and optical microscopes. A disorganized meristem, immediately behind the root-cap, gives rise to the vascular tissue, which consists of single central xylem element surrounded by a ring of phloem parenchyma cells. This ring of cells is first seen at the 4-5 cell stage, but increases to as many as 11 cells by repeated radial anticlinal divisions. At some point, usually at or shortly after the 8 cell stage, two phloem parenchyma cells located opposite each other on the ring of cells, undergo an unsynchronized, periclinal division to give rise to the sieve element and companion cell. Because of the limited number of cells involved, this developmental sequence offers a relatively simple system in which some of the factors underlying cell division and differentiation may be investigated, including the distribution of diffusible low atomic weight elements within individual cells of the phloem tissue.

The severity and duration of ischemia-reperfusion injury is hypothesized to play an important role in the ability of the heart subsequently to recover contractility. Permeabilized trabeculae were prepared from a rat model of ischemia-reperfusion injury to examine the impact on force generation. Compared with the control perfused condition, the maximum force (Fmax) per cross-sectional area and the rate of tension redevelopment of Ca2+-activated trabeculae fell by 71% and 44%, respectively, during ischemia despite the availability of a high concentration of ATP. The reduction in Fmax with ischemia was accompanied by a decline in fiber stiffness, implying a drop in the absolute number of attached cross bridges. However, the declines during ischemia were largely recovered after reperfusion, leading to the hypothesis that intrinsic, reversible posttranslational modifications to proteins of the contractile filaments occur during ischemia-reperfusion injury. Examination of thin-filament proteins from ischemic or ischemia-reperfused hearts did not reveal proteolysis of troponin I or T. However, actin was found to be glutathionylated with ischemia. Light-scattering experiments demonstrated that glutathionylated G-actin did not polymerize as efficiently as native G-actin. Although tropomyosin accelerated the time course of native and glutathionylated G-actin polymerization, the polymerization of glutathionylated G-actin still lagged native G-actin at all concentrations of tropomyosin tested. Furthermore, cosedimentation experiments demonstrated that tropomyosin bound glutathionylated F-actin with significantly reduced cooperativity. Therefore, glutathionylated actin may be a novel contributor to the diverse set of posttranslational modifications that define the function of the contractile filaments during ischemia-reperfusion injury.

✓ Currently, diagnosis and management of disorders involving nerves are generally undertaken without images of the nerves themselves. The authors evaluated whether direct nerve images obtained using the new technique of magnetic resonance (MR) neurography could be used to make clinically important diagnostic distinctions that cannot be readily accomplished using existing methods. The authors obtained T2-weighted fast spin—echo fat-suppressed (chemical shift selection or inversion recovery) and T1-weighted images with planes parallel or transverse to the long axis of nerves using standard or phased-array coils in healthy volunteers and referred patients in 242 sessions. Longitudinal and cross-sectional fascicular images readily distinguished perineural from intraneural masses, thus predicting both resectability and requirement for intraoperative electrophysiological monitoring. Fascicle pattern and longitudinal anatomy firmly identified nerves and thus improved the safety of image-guided procedures. In severe trauma, MR neurography identified nerve discontinuity at the fascicular level preoperatively, thus verifying the need for surgical repair. Direct images readily demonstrated increased diameter in injured nerves and showed the linear extent and time course of image hyperintensity associated with nerve injury. These findings confirm and precisely localize focal nerve compressions, thus avoiding some exploratory surgery and allowing for smaller targeted exposures when surgery is indicated. Direct nerve imaging can demonstrate nerve continuity, distinguish intraneural from perineural masses, and localize nerve compressions prior to surgical exploration. Magnetic resonance neurography can add clinically useful diagnostic information in many situations in which physical examinations, electrodiagnostic tests, and existing image techniques are inconclusive.

AbstractLignin is a coproduct of biofuel and paper industries, which exhibits binding qualities when mixed with water. Lignin is an ideal candidate for a sustainable stabilization of unpaved roads. To this end, an experimental program was devised and carried out to quantify effects of lignin on compaction and early age shear strength behaviors of sand. Samples were prepared by mixing a particular type of coproduct called calcium lignosulfonate (CaL) with sand and water. Based on the extensive analyses of six series of strength tests, it was found that a normalized cohesion increased with an increasing normalized areas ratio. Normalizations were carried out by dividing the cohesion and area ratio by gravimetric CaL content whereby the area ratio was obtained by dividing the portion of the cross-sectional area occupied with lignosulfonate-water (CaL-W) paste by the total cross-sectional area. While the increase in the normalized cohesion eventually leveled out, the cohesion peaked at 6% of CaL. Thus, sand-CaL-water (S-CaL-W) mixes sustained larger shear stresses than dry sand for a range of normal stresses below the limiting normal stress. Consequently, the early age behavior indicates that adding CaL-W to sand is clearly beneficial in the near-surface applications in dry sand.

ABSTRACTWe collected peat sediments (sediment core ADK13083002) from Haven Lake on the north side of Adak Island (central Aleutian Islands, Alaska) to determine whether the vegetation has changed. We confirmed the presence of six tephra layers, including Forty Years (0.3 cal ka BP), T2, YBO (3.3 cal ka BP), Intermediate (6.4 cal ka BP), Main (9.5 cal ka BP), and T6. We identified four major pollen assemblage zones (HL-1 to HL-4, in descending order) in the cored sediment. HL-1 was dominated by Ranunculaceae and Empetrum pollen; H-2 was dominated by Poaceae, Ranunculaceae, and Empetrum pollen; HL-3 was dominated by Poaceae and Empetrum pollen; and HL-4 was dominated by Poaceae, Cyperaceae, Lycopodiaceae, and Empetrum pollen. Small charcoal particles, likely transported from a distance, were found at low frequencies until 6.4 cal ka BP. The total cross-sectional area of charcoal particles increased to 1500 μm2 or more by 6.4 cal ka BP, implying that the large charcoal particles originated from nearby Aleut settlements, which were established around the same time.

Background: Cognitive impairment affects half of the multiple sclerosis (MS) patient population and is an important contributor to patients’ daily activities. Most cognitive impairment studies in MS are, however, cross-sectional or/and focused on the early disease stages. Objective: We aim to assess the time course of decline of different cognitive domains. Methods: We collected neuropsychological data on 514 MS patients to construct Kaplan-Meier survival curves of the tests included in the Neuropsychological Screening Battery for MS (NSBMS) and the Symbol Digit Modalities Test (SDMT). Cox-proportional hazard models were constructed to examine the influence of MS onset type, age at onset, gender, depression and level of education on the time course, expressed as age or disease. Results: Survival curves of tests focusing on information processing speed (IPS) declined significantly faster than tests with less specific demands of IPS. Median age for pathological decline was 56.2 years (95% CI: 54.4–58.2) on the SDMT and 63.9 years (95% CI: 60–66.9) on the CLTR, a memory task. Conclusion: In conclusion, IPS is the cognitive domain not only most widely affected by MS but it is also the first cognitive deficit to emerge in MS.

AbstractDNA methylation age acceleration (DNAmAA, derived from an epigenetic clock) and relative leukocyte telomere length (rLTL) are widely accepted biomarkers of aging. Nevertheless, it is still unclear which aspects of aging they represent best. Here we evaluated longitudinal associations between baseline rLTL and DNAmAA (estimated with 7-CpG clock) and functional assessments covering different domains of aging. Additionally, we made use of cross-sectional data on these assessments and examined their association with DNAmAA estimated by five different DNAm age measures.Two-wave longitudinal data was available for 1,083 participants of the Berlin Aging Study II (BASE-II) who were re-examined on average 7.4 years after baseline as part of the GendAge study. Functional outcomes were assessed with Fried’s frailty score, Tinetti mobility test, falls in the past 12 months (yes/no), Finger-floor distance, Mini Mental State Examination (MMSE), Center for Epidemiologic Studies Depression Scale (CES-D), Activities of Daily Living (ADL), Instrumented ADL (IADL) and Mini Nutritional Assessment (MNA).Overall, we found no evidence for an association between the molecular biomarkers measured at baseline, rLTL and DNAmAA (7-CpG clock), and functional assessments assessed at follow-up. Similarly, a cross-sectional analyses of follow-up data did also not show evidence for associations of the various DNAmAA measures (7-CpG clock, Horvath’s clock, Hannum’s clock PhenoAge, and GrimAge) with functional assessments.In conclusion, neither rLTL nor 7-CpG DNAmAA were able to predict impairment in the analyzed assessments over a ∼7 year time-course. Similarly, DNAmAA as estimated by five epigenetic clocks was not a good cross-sectional marker of health deterioration either.

The α7β1-integrin is a heterodimeric transmembrane protein that adheres to laminin in the extracellular matrix, representing a critical link that maintains structure in skeletal muscle. In addition to preventing exercise-induced skeletal muscle injury, the α7-integrin has been proposed to act as an intrinsic mechanosensor, initiating cellular growth in response to mechanical strain. The purpose of this study was to determine the extent to which the α7-integrin regulates muscle hypertrophy following eccentric exercise. Wild-type (WT) and α7-integrin transgenic (α7Tg) mice completed a single bout of downhill running exercise (−20°, 17 m/min, 60 min), and gastrocnemius-soleus complexes were collected 1, 2, 4, and 7 days (D) postexercise (PE). Maximal isometric force was maintained and macrophage accumulation was suppressed in α7Tg muscle 1D PE. Mean fiber cross-sectional area was unaltered in WT mice but increased 40% in α7Tg mice 7D PE. In addition, a rapid and striking fivefold increase in embryonic myosin heavy chain-positive fibers appeared in α7Tg mice 2D PE. Although Pax7-positive satellite cells were increased in α7Tg muscle 1D PE, the number of nuclei per myofiber was not altered 7D PE. Phosphorylation of mammalian target of rapamycin (mTOR) was significantly elevated in α7Tg 1D PE. This study provides the first demonstration that the presence of the α7β1-integrin in skeletal muscle increases fiber hypertrophy and new fiber synthesis in the early time course following a single bout of eccentric exercise. Further studies are necessary to elucidate the precise mechanism by which the α7-integrin can enhance muscle hypertrophy following exercise.

Abstract We studied the time-course of stem response for six months following complete girdling in branches of Aquilaria sinensis to determine the potential role of interxylary phloem (IP) in this response. It was found that the vascular cambium, as well as its derivative secondary xylem and phloem, regenerated fully through redifferentiation of IP. We confirmed that vascular cambium regenerated within one month after girdling based on observation of new vessels, IP, and secondary phloem fibers. The time-course study showed that IPs made connections with each other, merged, and became larger through the proliferation of IPs parenchyma cells and the cleaving of secondary xylem in a narrow zone 400 to 1000 μm deep inside the girdled edge. This led to the formation of a complete circular sheath of vascular cambium, followed by the regeneration of vascular tissue. It is worth noting that the secondary xylem is regenerated always following the formation of a thick belt of wound xylem.

The time course of angiogenesis during hypertrophy of the rat plantaris muscle was studied by using a unilateral, synergistic ablation model. Animals ( n = 6/group) were euthanized 2, 5, 7, 15, 21, and 30 days postmyectomy. Sections from both the hypertrophied and contralateral muscles were simultaneously stained for capillaries and muscle fiber type. Mean fiber cross-sectional area (FA) and various indexes of capillarity were determined by using a video analysis system. The capillary supply to individual fibers, assessed as the FA supplied per capillary contact, remained unchanged until day 21 (compared with day 2) and exhibited a significant increase at day 30. Analysis of the time course of capillary development on the basis of the number of capillary contacts per fiber, and of hypertrophy on the basis of FA, yielded half-lives of 10.1 and 11.2 days, respectively. It was concluded that angiogenesis during muscle overload is tightly coupled to the changes in FA, which could suggest that the two processes are initiated and/or driven by some common factor(s).
