Effect of cambial dormancy state on the transport of [1-14C]indol-3-ylacetic acid in Abies balsamea shoots

Dormant attached or detached shoots of balsam fir were naturally or artificially chilled to induce different states along the rest–quiescence continuum. At the end of the chilling pretreatment, the shoots either were left intact or were debudded and treated with indol-3-ylacetic acid (IAA). The shoots were placed under controlled-environment conditions favorable for growth, and at intervals thereafter, a pulse of [1-14C]IAA was applied to the shoot apex. Measured at the end of the chilling pretreatment, [14C]IAA velocity and flux decreased with increasing duration of chilling (i.e., as rest graded into quiescence). The time required to commence cambial growth and to attain maximum rates of cambial activity and [14C]IAA transport also decreased as rest changed to quiescence. Transport in actively growing shoots exceeded that in quiescent shoots, but was similar to that in resting shoots. The [14C]IAA pulse moved basipetally as unchanged IAA, was blocked by a bark + cambium girdle, and was inhibited by abscisic acid and long-term application of exogenous IAA. The results indicate that: (1) the long-distance, cambium-located, IAA transport system demonstrated in dicotyledonous species also operates in conifers, (2) during the dormant period changes occur in [14C]IAA transport and in the cambial response to exogenous IAA, (3) the change in [14C]IAA transport is the result of change in the transporting capability of cells in the cambial zone, and (4) the change in [14C]IAA transport is not the cause of the differential response of quiescent and resting cambia to exogenous IAA.

The anatomy and histochemistry of fusiform cambial zone cells were investigated (i) in shoots producing latewood tracheids at the time of collection in July and (ii) in dormant shoots. The latter were naturally or artificially chilled to obtain different stages along the rest–quiescence continuum, then placed for 4 weeks under environmental conditions favoring growth. The transition from active growth to rest was associated with increases in RNA, protein, and total insoluble carbohydrates and in peroxidase and succinic dehydrogenase activities. The rest–quiescence transition was correlated with additional increases in RNA and protein, an increase in lipid staining, radial wall thickness and plasmalemma infolding, and a decrease in vacuolar size. The DNA level did not appear to change; however, a distinct color shift occurred in nuclei stained for total protein. During the 4-week growth period, RNA, protein, total insoluble carbohydrates, and lipids decreased, most obviously in cambia that were fully quiescent at the start of the period. Xylem production and percentage of buds flushing during the growing period increased as the initial dormancy stage changed from rest to quiescence. Little, if any, xylem production occurred in debudded shoots unless indol-3-ylacetic acid (IAA) was supplied. Endogenous and exogenous lAA induced the same anatomical and histochemical responses.

On three dates during September–December, the histochemistry and dormancy status of cambial zone cells were investigated in 6- and 19-year-old balsam fir (Abies balsamea) trees at the top of the crown (1-year-old cambium) and near the base of the bole (5- and 13-year-old cambium, respectively). The extent of dormancy on the date of collection and after 4 weeks in controlled-environment conditions favorable for growth indicated that a changeover between the dormancy stages of rest and quiescence occurred in all ages of the cambium during the experimental period. In trees of both ages, cambial dormancy began later, and the number of fusiform cells in the cambial zone was greater at the bottom of the tree than at the top. The staining intensities for total cytoplasmic RNA, protein, and insoluble carbohydrates in the fusiform cambial zone cells increased during the rest–quiescence transition at the top and bottom of the 6-year-old trees and at the top of the 19-year-old trees. However, no significant increase in RNA staining occurred at the bottom of the 19-year-old trees.

<b><i>Introduction:</i></b> Decannulation of the arteriovenous fistula (AVF) after each hemodialysis session requires a precise compression on the needle puncture site. The objective of our study was to evaluate the bleeding time (BT) needed to achieve hemostasis using WoundClot, an innovative hemostatic gauze, and to assess whether its long-term use can improve AVF preservation. <b><i>Methods:</i></b> This is a prospective single center study. Initially, the time to hemostasis after AVF decannulation was compared between WoundClot and cotton gauze in 24 prevalent hemodialysis patients. Thereafter, the patients continued to use WoundClot for 12 months and were compared to a control group consisting of 25 patients using regular cotton gauze. Follow-up data included parameters of dialysis adequacy, AVF interventions, and thrombotic events. <b><i>Results:</i></b> WoundClot use shortened significantly the time needed for hemostasis. Mean venous BT decreased by 3.99 (±4.6) min and mean arterial BT by 6.38 (±4.8) min when using WoundClot compared to cotton gauze (<i>p</i> &#x3c; 0.001). At the end of the study, dialysis adequacy expressed by spKt/V was higher in the WoundClot group compared to control (1.73 vs. 1.53, respectively, <i>p</i> = 0.047). Although patients in WoundClot group had a higher baseline BT, arterial and venous pressures did not differ between the groups after a median follow up of 10.8 months. AVF thrombosis rate was similar between the groups. <b><i>Conclusions:</i></b> WoundClot hemostatic gauze significantly reduced the time required for hemostasis after AVF decannulation and may be associated with better AVF preservation. We suggest using WoundClot for arterial BT longer than 15 min and for venous BT longer than 12.5 min.

AbstractGlobally, bird migration is occurring earlier in the year, consistent with climate-related changes in breeding resources. Although often attributed to phenotypic plasticity, there is no clear demonstration of long-term population advancement in avian migration through individual plasticity. Using direct observations of bar-tailed godwits (Limosa lapponica) departing New Zealand on a 16,000-km journey to Alaska, we show that migration advanced by six days during 2008–2020, and that within-individual advancement was sufficient to explain this population-level change. However, in individuals tracked for the entire migration (50 total tracks of 36 individuals), earlier departure did not lead to earlier arrival or breeding in Alaska, due to prolonged stopovers in Asia. Moreover, changes in breeding-site phenology varied across Alaska, but were not reflected in within-population differences in advancement of migratory departure. We demonstrate that plastic responses can drive population-level changes in timing of long-distance migration, but also that behavioral and environmental constraints en route may yet limit adaptive responses to global change.

Recently, deep learning-based techniques have shown great power in image inpainting especially dealing with squared holes. However, they fail to generate plausible results inside the missing regions for irregular and large holes as there is a lack of understanding between missing regions and existing counterparts. To overcome this limitation, we combine two non-local mechanisms including a contextual attention module (CAM) and an implicit diversified Markov random fields (ID-MRF) loss with a multi-scale architecture which uses several dense fusion blocks (DFB) based on the dense combination of dilated convolution to guide the generative network to restore discontinuous and continuous large masked areas. To prevent color discrepancies and grid-like artifacts, we apply the ID-MRF loss to improve the visual appearance by comparing similarities of long-distance feature patches. To further capture the long-term relationship of different regions in large missing regions, we introduce the CAM. Although CAM has the ability to create plausible results via reconstructing refined features, it depends on initial predicted results. Hence, we employ the DFB to obtain larger and more effective receptive fields, which benefits to predict more precise and fine-grained information for CAM. Extensive experiments on two widely-used datasets demonstrate that our proposed framework significantly outperforms the state-of-the-art approaches both in quantity and quality.

In experiments with attached and detached shoots of balsam fir, Abies balsamea L., synthetic (±)abscisic acid (ABA) (1) reduced photosynthesis and transpiration by inducing stomatal closure, (2) inhibited indoleacetic acid (IAA) - induced cambial activity in photosynthesizing and non-photosynthesizing shoots, and (3) inhibited the basipetal movement of [14C]IAA. Neither gibberellic acid nor kinetin counteracted the inhibitory effect of (±)ABA on IAA-induced cambial activity. In addition it was demonstrated that increasing the internal water stress increased the level of endogenous ABA in the phloem–cambial region of bark peelings and decreased the basipetal movement of [14C]IAA through branch sections. On the basis of these findings it is proposed that internal water stress inhibits cambial activity, partly through increasing the level of ABA; the ABA acts to decrease the provision of carbohydrates and auxin that are required for cambial growth.

AbstractOne flowering specimen of Ophrys bertolonii (s. str.) — a plant with a Circum-Adriatic distribution and hitherto unknown in other regions — was found on 7th May 2010 in the vicinity of Kunadacs (Central Hungary; N 47°00′ E 19°17′). The nearest known populations of this Mediterranean plant inhabit the Adriatic coast (ca. 450 km away) in Croatia, therefore this new occurrence represents a significant long distance dispersal event. The circumstances of the occurrence argue against introduction, but we can not decide now whether this new appearance is temporary or permanent. The permanent establishment of this strictly entomophilous plant requires the presence of its specific pollinator in the close neighbourhood of the habitat. However, no pollination was observed on the three flowers of the plant until 9th May, and one day later the stem had disappeared most likely due to grazing. Whatever the long-term fate of the plant is, this discovery represents a significant long distance (jump) dispersal event, and testifies to the dispersal ability of Ophrys species. Additionally, the appearance of a mature Adriatic plant in Central Europe fits well into the currently observed, climate change driven northward expansion of European orchids, therefore this finding most likely reflects a growing Mediterranean-like climatic influence in the region.

AbstractBacillus thuringiensis (Dipel® 36B) mixed with a sublethal concentration of acephate (Orthene®) (O, S-dimethyl acetylphosphoramidothioate), an organophosphorous insecticide, was applied at 2.35–14 l./ha to white spruce (Picea glauca) and balsam fir (Abies balsamea) trees infested with spruce budworm, Choristoneura fumiferana (Clem.). The treatment rate was 20 Billion International Units of B. thuringiensis (B.t.) activity with or without 42 g of active ingredient of acephate/ha.The ground deposit of the standard Dipel wettable powder formulation was 12% of emitted volume compared with 21–32% for the Dipel 36B flowable. The viability of B.t. spores was drastically reduced after 1 day of weathering but a high level of biological activity by the spore–crystal complex persisted for up to 20 days post-spray due probably to crystal activity.The addition of about 10% of the recommended operational rate of acephate to the B.t. suspension increased larval mortality by 34% when applied at 4.7 l./ha. Reductions in budworm populations were 97–99% in B.t. + acephate plots and 86–90% in B.t. alone plots.Plots with moderate budworm densities of up to 27 larvae/100 buds on white spruce and 36/100 on balsam fir were satisfactorily protected from excessive defoliation in the year of spray by B.t. with or without acephate. Plots with higher population densities were not satisfactorily protected based on the branch sample examination but aerial color photographs indicated good protection to the top third of the trees. Population declines were greater and defoliation and oviposition were lower in the treated plots than in the untreated checks 1 year later without further treatment. Two years later the larval population densities in all plots were low but the density was twice as high in the untreated check as in the treated plots, indicating long term suppression by the treatments. Defoliation was negligible in all plots.The treatments had no deleterious effect on spruce budworm parasitism. The data indicate that the integrated approach using Bacillus thuringiensis – chemical pesticide combinations is a viable alternative to the use of chemical pesticides alone in spruce budworm control. Large scale testing is now warranted.

This study was conducted to explore the proper time required to achieve stabilization in digestibility, serum metabolism, and rumen fermentation characteristics when different diets shift, thus providing decision-making of practical sampling frequency for basal nutritional research. For these purposes, 12 Holstein steers (body weight 467 ± 34 kg, age 14 ± 0.5 months) were equally assigned to two dietary treatments: high-density (metabolizable energy (ME) = 2.53 Mcal/kg and crude protein (CP) = 119 g/kg; both ME and CP were expressed on a dry matter basis) or low-density (ME = 2.35 Mcal/kg and CP = 105 g/kg). The samples of feces, serum, and rumen contents were collected with a 30-day interval. All data involved in this study were analyzed using the repeated measures in mixed model of SPSS. Results showed that nutrient apparent digestibility and serum metabolic parameters were stable across each monthly collection, while most rumen fermentation characteristics, namely concentrations of acetate, propionate, isobutyrate, and valerate, were affected by the interaction effects between collection period and dietary density. These findings indicate that rumen fermentation characteristics require more time to stabilize when diet shifts. It is recommended to collect ruminal digesta monthly to evaluate rumen fermentation characteristics, while unnecessary to sample monthly for digestion trials and blood tests in the long-term fattening of Holstein steers. This study may provide insights into exploring the associations between detected parameters and stabilization time, and between diet type and stabilization time when diet shifts.
