Morphology and morphogenesis of synnemata of Ceratocystis ulmi were observed under a scanning electron microscope and a transmission electron microscope. Synnematal development in C. ulmi can be divided into four stages: (1) synnema initial formation, (2) stalk development, (3) conidiophore formation, and (4) conidium production. Observations on each stage of development are described and illustrated.

Gastritis from returning bile is a common disease, but the reason for the disease is not clear. As the pathologic ultrastructure research progresses, it has drawn attention to the ultrastructural change of cells in gastric mucosa by clinical workers. We observed gastric mucosa tissues of 15 patients suffering from gastritis with a transmission electron microscope (TEM) and a scanning electron microscope (SEM). It is the first report in China that fungus exists in the lamina propria of gastric mucosa tissue. The result is as follows.The gastric mucosa tissues of 15 patients suffering from gastritis were acquired by stomachoscopy. Both TEM and SEM specimens were prepared by the usual methods. Under the TEM, the epithelial surface became higher and larger. Mitochondria of the cells were swollen and cristae were disrupted. There were vacuoles in the cells. The nucleus showed disorder, heterochromatin became darker, and nucleolae could be observed.

Transmission electron microscope(TEM) and scanning electron microscope(SEM) were widely used in experimental tumor studies. They are useful for evaluation of cellular transformation in vitro, classification of histological types of tumors and treating effect of tumors. We have obtained some results as follows:1. Studies on the malignant transformation of mammalian cells in vitro. Syrian golden hamster embryo cells(SGHEC) were transformed in vitro by ThO2 and/or ore dust. In a few days after dust added into medium, some dust crystals were phagocytized. Two weeks later, malignant transformation took place. These cells were of different size, nuclear pleomorphism, numerous ribosomes, increasing of microvilli on cell surface with various length and thickness, and blebs and ruffles(Figs. 1,2). Myelomonocytic leukemic transformation of mouse embryo cells(MEC) was induced in vitro by 3H-TdR. Transformed cells were become round from fusiform. The number of mitochondria and endoplasmic reticulum was reduced, ribosomes and nucleoli increased, shape of nuclei irregular, microvilli increased, and blebs and ruffles appeared(Fig. 3).

Pollen grains of 20 taxa from two genera of the Liliaceae were examined and compared by LM (light microscope), SEM (scanning electron microscope) and pollens of four taxa were also examined with TEM (transmission electron microscope). Pollen grains shed as monads. They are monosulcate and ellipsoidal. Fritillaria crassifolia subsp. crassifolia Freyn & Smt. sometimes sheds the pollen as dyads. Exine is semitectate and the tectum is perforate. Columellae are simplicolumellate. Ectexine is thicker than endexine. Exine sculpture (ornamentation) is reticulate, reticulate-rugulate, rugulate and retipilate in Asparagus pollens and reticulate, suprareticulate, rugulate-reticulate and striate-reticulate in Fritillaria pollens. Το ψευδοαποφολιδωτικό σύνδρομο (pseudoexfoliation syndrome, PEX), το οποίο ενοχοποιείται για αυξημένες διεγχειρητικές επιπλοκές κατά την φακοθρυψία, απαντάται αρκετά συχνά στην Ελλάδα. Κλινικά παρατηρήθηκε ότι ο υδροδιαχωρισμός πραγματοποιείται πιο εύκολα στους ψευδοαποφολιδωτικούς ασθενείς. Δύο κύριοι στόχοι τέθηκαν στην παρούσα έρευνα: α) η υπερμικροσκοπική μελέτη του προσθίου περιφακίου με τη βοήθεια των ηλεκτρονικών μικροσκοπίων διελεύσεως (transmission electron microscope, ΤΕΜ) και σαρώσεως (scanning electron microscope, SEM) και β) η κλινική σύγκριση της ευκολίας του υδροδιαχωρισμού μεταξύ ασθενών με και χωρίς ψευδοαποφολιδωτικό σύνδρομο. Ασθενείς-Μέθοδος: Συνολικά στη μελέτη συμμετείχαν 149 ασθενείς με γεροντικό καταρράκτη, άνω των 60 ετών, εκ των οποίων οι 60 εμφάνιζαν ψευδοαποφολιδωτικό σύνδρομο. Όλοι οι ασθενείς υποβλήθηκαν σε ανεπίπλεκτη φακοθρυψία από τον ίδιο χειρουργό. Τα πρόσθια περιφάκια που ελήφθησαν με συνεχή κυκλοτερή καψουλόρρηξη από τους 34 ασθενείς εξετάστηκαν με ΤΕΜ και τα 14 από αυτά επιπλέον με SEM. Στην κλινική μελέτη, συμμετείχαν οι υπόλοιποι 115 ασθενείς. Για τον υδροδιαχωρισμό σε αυτούς τους ασθενείς χρησιμοποιήθηκαν 3 σύριγγες ινσουλίνης γεμάτες με 0.5 ml BSS (balanced salt solution) που εγχυόταν στην 3η , 6η και 9η ώρα και αξιολογούνταν η επίτευξη ή όχι της πλήρους κινητοποίησης του φακού. Αποτελέσματα: Στους ασθενείς με ψευδοαποφολιδωτικό σύνδρομο, ανεξαρτήτως του βιομικροσκοπικού τύπου του καταρράκτη, τόσο με το ΤΕΜ όσο και με το SEM, εντοπίστηκαν εκσεσημασμένες υποεπιθηλιακές αλλοιώσεις. Η επιφάνεια του επιθηλίου προς τις φακικές ίνες εμφανιζόταν συνολικά πιο ανώμαλη και το ύψος των επιθηλιακών κυττάρων παρουσίαζε μεγάλο εύρος. Παράλληλα απεικονίστηκε εξωκυττάριο, άμορφο, κρυσταλλικό ή μικροκοκκώδες υλικό και μεμβρανώδεις ή ωοειδείς δομές. Επιπλέον, πέρα από το τυπικό ινιδώδες υλικό πάνω από τη βασική μεμβράνη του προσθίου περιφακίου προς τον πρόσθιο θάλαμο, επιμήκη ινίδια εντοπίζονταν με σαφήνεια και εντός της βασικής μεμβράνης. Στο επιθήλιο όλων των ασθενών παρατηρήθηκαν εκφυλιστικού τύπου αλλοιώσεις, όπως διάχυτο ενδοκυττάριο και εξωκυττάριο οίδημα, κενοτόπια, ψευδοπολυστιβάδωση, λύση των κυττάρων και διαταραχή της σύνδεσής τους τόσο μεταξύ τους όσο και με τη βασική μεμβράνη. Οι αλλοιώσεις αυτές ήταν πιο έντονες και πιο συχνές στους ασθενείς με ψευδοαποφολιδωτικό σύνδρομο. Τέλος, καταγράφηκαν αποπτωτικά κύτταρα και στις δύο ομάδες. Τα παρακείμενα επιθηλιακά κύτταρα απεικονίζονταν να αναλαμβάνουν τον ρόλο των μακροφάγων και αποπτωτικά σωμάτια εντοπίζονταν εντός φαγοσωμάτων. Στην κλινική μελέτη, ο υδροδιαχωρισμός πραγματοποιούταν επιτυχώς με την μέθοδο που περιγράψαμε στο 92,8% των ασθενών με ΡΕΧ, έναντι του 64,8% των ασθενών μαρτύρων, αποδεικνύοντας ότι επιτελείται ευκολότερα στο ψευδοαποφολιδωτικό σύνδρομο (Chi-square test, p=0.001). Συμπέρασμα: Στο ψευδοαποφολιδωτικό σύνδρομο καταγράφηκαν για πρώτη φορά υποεπιθηλιακές αλλοιώσεις, στην περιοχή διασύνδεσης του επιθηλίου με τις υποκείμενες φακικές ίνες. Προτείνουμε ότι οι εν λόγω αλλοιώσεις προκύπτουν από την εκφύλιση τόσο των επιθηλιακών κυττάρων όσο και των φακικών ινών, συμβάλλοντας στην χαλαρή σύνδεση του επιθηλίου με τον φλοιό. Αυτό δικαιολογεί τη διαφορετική κλινική συμπεριφορά των ιστών και τον ευκολότερο υδροδιαχωρισμό στους ψευδοαποφολιδωτικούς ασθενείς, όπως διαπιστώθηκε και στην κλινική μελέτη. Επιπρόσθετα, για πρώτη φορά καταγράφηκαν ινιδώδεις εναποθέσεις ψευδοαποφολιδωτικού υλικού και εντός της βασικής μεμβράνης του προσθίου περιφακίου ασθενών με ΡΕΧ, προσφέροντας νέα στοιχεία για την ιστοπαθογενετική προσέγγιση του ψευδοαποφολιδωτικού συνδρόμου, τα οποία θα μπορούσαν να βοηθήσουν στην αποσαφήνιση των μηχανισμών προέλευσης του φακικού ψευδοαποφολιδωτικού υλικού. Κυρίως στους ψευδοαποφολιδωτικούς ασθενείς, καταγράφηκαν επιπλέον σοβαρές επιθηλιακές αλλοιώσεις οι οποίες είναι συγκρίσιμες με αλλοιώσεις που έχουν περιγραφεί σε σοβαρές παθολογίες και νόσους, όλες εκ των οποίων είχαν αποκλειστεί από την παρούσα μελέτη. Πρόκειται, τέλος, για την πρώτη μελέτη ηλεκτρονικού μικροσκοπίου διελεύσεως, την πιο ειδική μέθοδο ανίχνευσης της απόπτωσης, η οποία κατέγραψε ανθρώπινα αποπτωτικά κύτταρα σε ασθενείς με καταρράκτη.

In a previous study of impurity gettering in Si it was found that Au-containing precipitates were formed at the surfaces of wafers which had been equilibrated with Au at 1200°C and then subjected to B diffusion at 1000°C. The work reported in summary here relates to the characterization of the precipitates and to the construction of a model for their growth. This investigation involved the use of both the transmission electron microscope (TEM) and the scanning electron microscope (SEM). Full details will be reported elsewhere.

In this discussion the words “high resolution imaging” of a solid sample in the scanning electron microscope (SEM) mean that details can be resolved that are considerably smaller than the penetration depth of the incident electron beam (EB) into the specimen. “Atomic resolution” in either the transmission electron microscope (TEM) or scanning transmission electron microscope (STEM) means that columns of atoms are resolved.Image contrasts in the backscattered electron (BSE) image are strongly affected by the specimen tilt and by the position and energy sensitivity of the BSE detector. The expression “BSE image” generally implies that the specimen is normal to the beam and the detector is above it. This shows compositional variations in the specimen with a spatial resolution limited by the spreading of the EB during the initial stages of penetration. This is similar in basic principle to the Z-Contrast method in the STEM that shows atomic resolution from a thinned single crystal mounted in the magnetic field of the focusing lens.

The Shanwang Fauna has been one of the well-known Miocene Konservat-Lagerstätten for a long time, but the high-fidelity preservation of the fossils has been often ignored. This paper illustrates the exceptional preservation displayed by some fossil insects from the Shanwang Formation. The microstructures of the preserved exoskeletons were analyzed using a scanning electron microscope and a transmission electron microscope. The epicuticle, procuticle, and some fine details such as cuticular scales could be recognized from the structural remains. These details could enable the identification of valued taxonomic and phylogenetic features and contribute to understanding the diagenetic environment.
