Notes on the biology of some native orchids of Thunder Bay, their endophytes and symbionts

Field studies were made of eight terrestrial orchid species of the Thunder Bay region during their active growing period. In four of them roots were examined for infection, then isolation and identification of endophytes attempted.Seeds were collected from all species and, along with those of two alien ones, were grown on various media in axenic culture. Their germination and development were studied and described.Seeds were also tested in dixenic cultures with locally isolated endophytes and four fungi known to be good symbionts with other orchids. The nature of infection was finally assessed and discussed.In the field the roots of mature plants of the orchid species differed in their degree of infection and reaction to their endophytes. Only two species of Rhizoctonia were isolated. These are described.In axenic culture, germination of some species was affected by the organic (and inorganic) components of the media, in others it was not. For growth they all responded differently to such components, but most were intolerant of casamino acids, yeast extract, and potato extract. Generally there seemed to be a direct correlation between percentage of germination and amounts of subsequent growth, but a less defined one with the number of cells per seed.In dixenic cultures the orchids responded differently also to the fungi. Their reaction to infection had no relation to their ability to germinate on poor media or to germinate and grow on richer ones. Only one good symbiotic association was established. It was between Goodyera oblongifolia from British Columbia and Rs10, a rice pathogen from Malaysia.The results and their implications are briefly discussed.

This paper continues the publication of new data obtained during field studies in remote and poorly studied regions of Laos in 2015–2018. It provides descriptions of two new species (Bulbophyllum brachyscapum Aver. and Sunipia saccata Aver.), emended descriptions of two previously published but insufficiently known species (Cleisostoma verrucosum Aver., Oberonia vesiculifera Aver.) and reports of eight species newly recorded in the flora of Laos (Biermannia calcarata Aver., Bulbophyllum farreri (W. W. Sm.) Seidenf., Didymoplexiella trichechus (J. J. Sm.) Garay, Eria eriopsidobulbon C. S. P. Parish et Rchb. fil., Habenaria ciliolaris Kraenzl., Liparis caudata Aver. et K. S. Nguyen, Odontochilus umbrosus (Aver.) Ormerod, and Porpax ustulata (C. S. P. Parish et Rchb. fil.) Rolfe). Appropriate citations of regional literature, colour illustrations, data on types and distribution, descriptions, etymology, as well as notes on taxonomy and related species are provided for the studied species.

SummaryAlthough lipid signaling has been shown to serve crucial roles in mammals and plants, little is known about this process in filamentous fungi. Here we analyse the contribution of phospholipase D (PLD) and its product phosphatidic acid (PA) in hyphal morphogenesis and growth of Epichloë festucae and Neurospora crassa, and in the establishment of a symbiotic interaction between E. festucae and Lolium perenne. Growth of E. festucae and N. crassa PLD deletion strains in axenic culture, and for E. festucae in association with L. perenne, were analysed by light-, confocal- and electron microscopy. Changes in PA distribution were analysed in E. festucae using a PA biosensor and the impact of these changes on endocytic recycling and superoxide production investigated. We found that E. festucae PldB and the N. crassa ortholog, PLA-7, are required for polarized growth, cell fusion and ascospore development, whereas PldA/PLA-8 are dispensable for these functions. Exogenous addition of PA rescues the cell-fusion phenotype in E. festucae. PldB is also crucial for E. festucae to establish a symbiotic association with L. perenne. This study identifies a new component of the cell-cell communication and cell fusion signaling network that controls hyphal morphogenesis and growth in filamentous fungi.

Based on data from the Department of Culture and Tourism of Bandung, Bandung has a legendary street food culinary that is highly favored by culinary enthusiasts who visit this city. Street food culinary is snacks that have been around for a long time with authentic flavors and stories behind, some of popular street food culinary are bandros, combro, colenak, ketan bakar, cireng ​​and others. The rapid development of culinary potential in this city has caused many new street foods that enriches culinary diversity in Bandung so that culinary enthusiasts need an information media contains of information about culinary in this city, especially authentic street food culinary which is starting to be hard to find. Through qualitative methods and data collection techniques by means of field studies such as observation, interviews and questionnaires, it is necessary to design an application-based information media. The final results of this research is user interface design for the media that informs Bandung street food culinary. Inspired by the word kabita which comes from Sundanese means tempted to taste food, was chosen as the name of the application that informs culinary street food in the city of Bandung that aims to facilitate culinary enthusiasts to get that information

The endocrine disrupting chemical nonylphenol (NP) is a technical product which consists of a complex mixture of nonylphenols with different alkyl side-chain isomers. Since the bio-degradation of each NP isomer may lead to its own range of metabolites, the isolation and identification of transformation products is very difficult. In order to overcome this difficulty, the nonylphenol isomer 4(3′,5′-dimethyl-3′-heptyl)-phenol (p353NP) was synthesized, and its degradation by an axenic culture of Sphingomonas TTNP3 was investigated with [ring-U-14C]-labelled and non-labelled p353NP including a time-course study. Radioactive mass balancing resulted in different polar soluble fractions, in insoluble radioactivity associated with biomass, and volatile radioactivity in the form of the mineralization product 14CO2. In the extracellular media, the presence of nonanol corresponding to the nonyl chain of the NP isomer was confirmed and its concentration was determined during the course of fermentation. No other radioactive compounds were detected beside the parent isomer. Radioactive metabolites were only found in the intracellular fraction of S. TTNP3.

Penelitian ini mengadaptasi penelitian dan pengembangan pendidikan model Borg & Gall dengan menyederhanakan menjadi 2 tahapan sebagai berikut (1) tahap pra-pengembangan, tahapan ini meliputi (a) kajian literatur dan penelitian relevan dan (b) studi lapangan, (2) tahap pengembangan, tahapan ini meliputi (a) penyusunan draf, (b) validasi ahli, (c) uji coba model skala kecil, (d) uji coba model skala besar, dan (e) produk akhir. Uji coba skala kecil dilakukan terhadap 30 anak kelas atas SDN Aditirto Pejagoan Kebumen, Provinsi Jawa Tengah. Uji coba skala besar dilakukan terhadap 130 anak kelas atas dari SD Negeri 1 Karangpoh, 26 siswa SD Negeri 1 Logede, 22 siswa SD Negeri 4 Pejagoan, 24 siswa SD Negeri 1 Kebulusan, dan 30 siswa SD Negeri 3 Kebulusan Kecamatan Pejagoan, Kabupaten Kebumen, Provinsi Jawa Tengah. Instrumen yang digunakan untuk mengumpulkan data adalah wawancara, kuisioner penilaian karakter permainan tradisional, kuisioner observasi pengembangan permainan tradisional, kuisioner observasi keefektifan model permainan tradisional, kuisioner penilian observasi ahli media, dan kuisioner evaluasi siswa. Hasil penelitian berupa pengembangan model permainan tradisional terdiri dari 3 jenis permainan yaitu: (1) permainan goteng, (2) lari papan/segi empat, dan (3) kasti tangan. Berdasarkan penilaian para ahli materi dan guru penjasorkes dapat disimpulkan bahwa pengembangan model permainan tradisional yang disusun sangat baik dan efektif, sehingga model permainan layak digunakan untuk pembelajaran pendidikan jasmani pada siswa kelas atas. Developing Models of Traditional Games to Build Characters of Elementary School Students of Upper Grades AbstractThe study was conducted through two stages by adapting the research and development of educational models Borg & Gall as follows (1) pre-development stage, this stage include (a) review of relevant literature and research, and (b) field studies, (2) the development stage, this stage include; (a) drafting, (b) validation expert, (c) testing of small-scale models, (d) testing large-scale models, and (e) of the final product.The small-scale tryout was conducted by involving 30 students of upper grades of state elementary schools (SES) of Aditirto, Pejagoan sub-district, Kebumen regency, Central Java porovinsi. The large-scale tryout was conducted by involving 130 students of upper grades of SES 1 of Karangpoh, 26 students of SES 1 of Logede, 22 students of SES 4 of Pejagoan, 24 students of SES 1 of Kebulusan, and 30 students of SES 3 of Kebulusan Pejagoan sub-district, Kebumen regency, Central Java porovinsi. The data collecting instruments were an interview guideline, a questionnaire to assess characters of traditional games, a questionnaire to assess the traditional game development, a questionnaire for the assessment by the media expert, and a questionnaire for the evaluation by students. The results of the study of the development of models of traditional games were three types of games, i.e: (1) goteng game, (2) board/rectangle running, and (3) hand kasti. Based on the assessment by the materials experts and the teachers of physical, sports, and health education, it could be concluded that the developed modals of traditional games were very good and effective so that they were appropriate to be used in the learning physical education for the students of upper grades.

The research has been done about isolation and identification on degradator bacterium of petroleum waste which symbions with sponge from Melawai Beach, Balikpapan. The research objectives; knowing sponge species, symbioning bacterium isolate and its ability of petroleum waste degradation. The research method were analysis of sponge Morphology analysis, isolation of symbions bacterium, identification, and activity testing at petroleum waste media which is coming from wells result of processing of PT. Chevron. The research result obtained the morphology analysis concluded that the sponges is Callyspongia sp. Bacterium symbion isolation obtained that two isolates which were given each symbols isolate A with the rounded shape and crem color whereas isolate B with yellow color and jagged shape. Coloring analysis of isolate obtained that isolate A is negative gram bacterium type and isolate B is positive gram bacterium type. Activity isolate testing at petroleum waste media, isolate A did not grow significantly at the media which marked with the existence of layer transparent on the media, isolate B growth significantly which marked with existence of solidity on the media, and the combination of isolate A+B also showed growth which marked with the existence of layer transparent and solidity on media. The conclusions research, sponge species is Callyspongia sp which contained two isolates bacterium namely negative gram bacterium and positive gram bacterium and both of the isolates can grow at media of petroleum waste.

Orchid seeds are very small, dust like in appearance, fusiform in shape, lacking endosperm and have undifferentiated embryo covered by transparent seed coat. Mycorrhizal association is required for seed germination of naturally growing orchids. In this symbiotic association, orchid species are dependent on mycorrhizal association for supply of mineral nutrients. In in-vitro condition, such demand of minerals may be compensated by external supply of sugar and mineral nutrients that are required for seed germination of orchid. Several orchids are responded by this asymbiotic seed culture and have commercial importance. Therefore, formulation of efficient in-vitro protocol is important for commercially important orchid species as well as endangered orchid species for conservation. This review paper is focused on various aspects of asymbiotic seed germination of orchids and the role of organic additives in successful seed germination.

Local wisdom is known as a cultural asset that develops in an area, all policies and customs that play a role in Aceh will become a culture that is respected and appreciated in Aceh. In its development, local wisdom is very influential in protecting people's thoughts, including the seeds of radicalism. The existence of radicalism in Aceh was most evident during the conflict between the Republic of Indonesia and the separatist movement in Aceh. After the peace period of the MOU Helsingki took place, radicalism in Aceh did not occur openly. Only ripples of thought trying to incite from within the disapproval of the current government. You could say, the turmoil arose because of the influence of political color, where the current population of Aceh is quantitatively more inclined to the failed presidential candidate. The problem raised in this study is how to revitalize the values ​​of Aceh's local wisdom which are threatened with fading due to the globalization of foreign cultures. This research design uses qualitative with reference sources based on literature and field studies. The analysis technique is descriptive. The findings of this study are that local wisdom is ideal in countering the existence of radicalism in Aceh through the media meuseumeuraya, tengku authorities, implementing the values ​​of ‘hadih madja’, sub-district da'i programs, and so on. In conclusion, strengthening local wisdom in Aceh is very urgent because the character of the Acehnese people respects traditional values ​​and authority more than the government authority.  Abstrak: Kearifan lokal dikenal sebagai aset budaya yang berkembang di suatu daerah, semua kebijakan dan kebiasaan yang memainkan peran keacehan akan menjadi suatu kultur yang dihormati dan dihargai di Aceh. Pada perkembangannya kearifan lokal sangat berpengaruh dalam memproteksi pemikiran masyarakat termasuk bibit-bibit radikalisme. Eksistensi radikalisme di Aceh paling kentara terjadi pada masa konflik antara NKRI dan gerakan sparatis di Aceh. Setelah masa damai MOU Helsingki terjadi, radikalisme di Aceh tidak terjadi secara terbuka. Hanya riak-riak pemikiran yang mencoba menghasut dari dalam tentang ketidaksetujuan terhadap pemerintahan saat ini. Bisa dibilang, gejolak itu muncul karena pengaruh warna politik yang mana saat ini penduduk Aceh secara kuantitas lebih condong pada calon Presiden yang gagal terpilih. Adapun masalah yang diangkat dalam penelitian ini adalah bagaimana merevitalisasikan nilai-nilai kearifan lokal Aceh yang terancam pudar akibat serangan globalisasi budaya luar. Design penelitian ini menggunakan kualitatif dengan sumber referensi berbasis kajian pustaka dan lapangan. Teknik analisis berupa deskriptif. Temuan dari penelitian ini adalah kearifan lokal sangat ideal dalam menangkal eksistensi radikalisme di Aceh melalui media meuseumeuraya, otoritas tengku,implementasi nilai hadih madja, program da’i kecamatan dan lain sebagainya.  Kesimpulannya, penguatan kearifan lokal di Aceh sangat urgen karena karakter masyarakat Aceh lebih menghormati nilai dan otoritas adat dibanding otoritas pemerintah. Date Palm (Phoenix dactylifera) is a very important and commonly growing tree found in Saudi Arabia. The fruit is nutritionally important and adds to the economy of the country. Isolation and identification of the phytopathogenic mycoflora is an important tool in the diagnosis of the pathogenic genera which are damaging to the palm under study. This is the first attempt to isolate and identify the phytopathogenic fungal flora found on the phylloplane and other plant parts of date palm from Ad Darb region of Jizan province during the year 2018. The media employed for the study was Potato Dextrose Agar on which 0.5 cm direct pieces of the diseased parts (phylloplane, fruit, rachis and trunk) of the date palm were inoculated. The isolated fungi were analysed qualitatively and quantitatively and the results interpreted in the form of table, bar diagram and photomicrographs. The mycoflora is represented by Aspergillus, Botrytis, Ceratocystis, Fusarium, Graphiola, Penicillium, Rhizopus ,Saprolegnia and Syncephalastrum. One hundred and fifty four fungal isolates were obtained from the nine represented genera. Aspergillus was the predominant genera with seventy four isolates. This study will help in the protection and conservation of this very important plant in the province. The study can also help the plant pathologist in disease forecasting and effective monitoring of the different diseases of the date palm thereby helping in the management and minimization of economic loss.

This study aimed to isolate and identify the bacteria Salmonella sp in spleen of male layer chicken that died in farms Sibreh, Aceh Besar. A sample of the research is the spleen of male layer chickens. Isolation of Salmonella sp performed by inoculating spleen that were pierced using a swab on the media Selenite Cystine Broth (SCB) then planted on selective media Salmonella Shigella Agar (SSA) and colony morphology was observed, continued with observation of Gram stain under microscopic. Furthermore, colonies of suspected Salmonella sp planted in the media MR-VP (Oxoid), Indol (Difco), Simmons's Citrate Agar (Oxoid), Triple Sugar Iron Agar (TSIA), Sulfite Indol Motiliti (SIM), and media sugars (sucrose, manitol, glucose, lactose , maltose). The results of this study were analyzed descriptively. The research shows a results from a total of 33 samples, we found that 15 samples are positive that can be isolated and identified as Salmonella sp. Therefore it can be concluded that Salmonella sp is possible the death cause of male layer chicken in farms Sibreh, Aceh Besar
