The population structure of Armillaria ostoyae and Armillaria sinapina in the central interior of British Columbia

The population structures of Armillaria ostoyae (Romagn.) Herink and Armillaria sinapina Bérubé & Dessureault were investigated at a study site near Williams Lake in the central interior of British Columbia. One hundred and twenty eight fungal isolates were examined from nine infection centers and individual genets were delineated using random amplified polymorphic DNA (RAPD) analysis and somatic incompatibility tests. Six A. ostoyae genets ranging in size from 0.70 to >15 ha were detected. The population structure of A. ostoyae was consistent with a clonal reproductive strategy, and infection centers were occupied by single A. ostoyae genets or ramets thereof. Eighteen relatively small A. sinapina genets were detected, with infection centers being occupied by multiple genets. Armillaria sinapina appears to be more pathogenic to coniferous hosts than previously reported in the region. Armillaria ostoyae appears to initiate new infections of available substrate via airborne basidiospores at a lower frequency than A. sinapina. However once established, A. ostoyae can spread quite aggressively and capture significant amounts of secondary resources, while A. sinapina is unable to do so. The results of somatic incompatibility tests used to differentiate genets corresponded with the results of RAPD analysis, with only one minor discrepancy.Key words: Armillaria, RAPD, population structure, genet, ramet, clone.

The population structure of Armillaria ostoyae (Romagn.) Herink was investigated at a study site in the interior cedar–hemlock zone of the southern interior of British Columbia. Eight 500-m long, randomly placed transects located at least 500 m apart were systematically sampled and individual genets were delineated using Random Amplified Polymorphic DNA (RAPD) analysis. Average genet intersect length was 79 m, and an estimated 88% of the area surveyed was occupied by genets <2 ha in size. The spatial distribution of genets in relation to one another was quite complex, with isolates of the same genet often non-contiguous along the transect. The location of genets could not be determined from aboveground symptom expression. Isolates collected from adjacent hosts within small groups of symptomatic hosts (infection foci) did not necessarily belong to the same genet, but single genets could occupy several infection foci. There was no relationship between geographic distance and genetic similarity of genets, suggesting random mating events for genet origin and long term maintenance of genetic identity.Key words: Armillaria, RAPD, population structure, genet, ramet, clone.

Pure cultures of Heterobasidion annosum were isolated from diseased trees in a 120-year-old Norway spruce stand. Clones of the fungus were identified by three different methods: studies of somatic incompatibility, identification of mating factors, and comparative studies of isoenzyme patterns. All of these methods yielded similar results. The study of somatic incompatibility was found to be the easiest and most reliable method for future investigations of population structures of Heterobasidion annosum. Nine clones were identified within a 60 × 60 m area. The number of trees infected by one clone varied from 1 to 13. The largest area occupied by one single clone was 30 m in diameter. Altogether, 10 different mating alleles were found, of which 2 occurred in four different clones and 2 occurred in two different clones. The vegetative spread of H. annosum between trees within the area occupied by a particular clone was confirmed by excavating root systems.

Random amplified polymorphic DNA (RAPD) analysis of DNA samples clearly distinguished between Cardantine hirsuta L. and two variants of Cardamine lilacina Hook. found in Australia. Cardarnine hirsuta, a recent immigrant, was less variable than C. lilacina, robust alpine snow-patch variant (C. lilacina-RASPV), which is an endemic plant that grows in three distinct regions in the Kosciusko alpine area and C. lilacina, intermedia variant (C. lilacina-IV), which is an endemic plant that is widespread in the same area. Comparison of RAPDs showed that, in the populations of C. lilacina-RASPV, some plants found near each other were closely related, but there was no detectable largerscale population structure. It is most likely that the present populations of C. lilacina-RASPV are relics of a single larger heterogeneous population. Cardarnine lilacina-RASPV is the only known natural host of turnip yellow mosaic tymovirus in Australia. Although in this study RAPD analysis did not indicate whether the host plant variation is influencing the variability of populations of this virus it did provide an understanding of the genetic relationships in these populations of C. lilacina.

Abstract The analysis of multiple population structures (biodemographic, genetic and socio-cultural) and their inter-relations contribute to a deeper understanding of population structure and population dynamics. Genetically, the population structure corresponds to the deviation of random mating conditioned by a limited number of ancestors, by restricted migration in the social or geographic space, or by preference for certain consanguineous unions. Through the isonymic method, surname frequency and distribution across the population can supply quantitative information on the structure of a human population, as they constitute universal socio-cultural variables. Using documentary sources to undertake the Doctrine of Belén’s (Altos de Arica, Chile) historical demography reconstruction between 1763 and 1820, this study identified an indigenous population with stable patronymics. The availability of complete marriage, baptism and death records, low rates of migration and the significant percentage of individuals registered and constantly present in this population favoured the application of the isonymic method. The aim of this work was to use given names and surnames recorded in these documentary sources to reconstruct the population structure and migration pattern of the Doctrine of Belén between 1750 and 1813 through the isonymic method. The results of the study were consistent with the ethno-historical data of this ethnic space, where social cohesion was, in multiple ways, related to the regulation of daily life in colonial Andean societies.

In this study we identified single nucleotide polymorphism (SNP) and sequence characteristic amplification region (SCAR) markers for specific identification of antler-shaped Ganoderma lucidum strains. When the partial mitochondrial SSU rDNA gene sequence of various antler- and kidney-shaped G. lucidum strains were analyzed and aligned, an SNP was found only in the antler-shaped G. lucidum strain at position 456 bp. In addition, this SNP of antler-shaped strains was digested by HinfI restriction enzyme. We further analyzed the polymorphism of various G. lucidum strains by random amplified polymorphic DNA (RAPD) analysis. In RAPD analysis, we isolated and sequenced a fragment, specific for antler-shaped G. lucidum strains. Based on this specific fragment sequence, two sets of specific primer pairs for antler-shaped G. lucidum strains were designed. PCR analysis revealed that two specific bands were observed only from antler-shaped strains. These two molecular markers will be helpful for identification of morphological characteristics of G. lucidum.

Records of uredinia and telia production on the alternate hosts of Cronartium coleosporioides in British Columbia and inoculation of Castilleja miniata with aeciospores collected from various locations showed that rust isolates from dry areas of the interior of British Columbia do not produce uredinia and may have lost the ability to do so. Collections from somewhat wetter areas produced uredinia or mixtures of uredinia and telia immediately following aeciospore inoculations, and field collections from such areas in June commonly had mixtures of uredinia and telia. Loss of the uredinial stage may be a response to climates that are often unsuitable for the spread or survival of the rust on the alternate host.
