Cecropin A (CA) fungicidal properties were explored. Nongerminated and germinated Aspergillus spp. and Fusarium spp. conidia were treated with CA. CA achieved complete lethality at < or = 25 microM (99 micrograms/ml) for germinating, but not nongerminating, conidia of Aspergillus spp. CA achieved total lethality for nongerminated and germinating conidia of Fusarium spp at 1.5 microM (6 micrograms/ml). MIC and minimal lethal concentration assays in buffered RPMI medium gave similar results.

ABSTRACT We have previously reported poor correlation between the in vitro fungicidal activity of LY303366 and MIC results in RPMI medium based upon the manufacturer’s suggested susceptibility endpoint, lack of visual growth. Additionally, we have noted a significant trailing effect with LY303366 when MICs are determined in RPMI medium. These observations have led us to evaluate an alternative susceptibility endpoint for LY303366, an 80% reduction in growth compared with control (similar to that utilized for azoles). Two isolates each of Candida albicans,Candida glabrata, and Candida tropicalis were selected for testing. MICs were determined for LY303366 in RPMI 1640 medium buffered with morpholinepropanesulfonic acid. MICs were determined with suggested (MIC100) and experimental (MIC80) endpoints. The minimal fungicidal concentration (MFC) of LY303366 for each isolate was also determined. Time-kill curves were determined in RPMI medium with each isolate at concentrations of LY303366 ranging from 0.125 to 16× MIC80to assess the correlation between MIC80 and fungicidal activity. Lastly, fungi exposed to LY303366 were examined via scanning electron microscope (SEM) for evidence of drug-induced ultrastructure change. MIC80s for test isolates ranged from 0.015 to 0.12 μg/ml and were consistently three to five wells less than MIC100s. Good correlation was observed between fungicidal activity, as assessed by kill curves, and the MIC80. SEM data revealed significant ultrastructure changes induced by LY303366 even at sub-MIC80s. Based on our results demonstrating better correlation between MIC80 and fungicidal activity, i.e., time-kill curves and MFCs, we suggest that 80% reduction in visible growth be utilized as the endpoint for susceptibility determinations with LY303366 in RPMI medium.

The fungicidal properties of the synthetic peptide D4E1 were studied with nongerminated and germinating conidia of Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Fusarium moniliforme, and Fusarium oxysporum. The minimal lethal concentrations (MLC) needed to kill 100% of germinating conidia ofA. fumigatus, A. flavus, and A. niger were 12.5, 12.5, and 25 µM, respectively. The MLC value for nongerminated and germinating conidia of both Fusarium spp. was 3.0 µM. Except for A. fumigatus, D4E1 was inactive against the nongerminated conidia of the Aspergillus spp. Physicochemical studies showed D4E1 complexed with ergosterol, a sterol present in conidial walls. Cholesterol, present in nongerminated conidia of F. moniliforme, had a greater affinity for D4E1 than did ergosterol. D4E1 was more resistant to fungal and plant protease degradation than the natural peptide, cecropin A. These in vitro results suggest D4E1 is a candidate for transgenic expression in plants to enhance host resistance to fungal infection.Key words: peptides, fungicidal, Aspergillus spp., Fusarium spp., sterols.

The aim of the study was to determine the effectiveness of selected seven commercial essential oils (EsO) (grapefruit, lemongrass, tea tree (TTO), thyme, verbena, cajeput, and Litsea cubeba) on isolates of common Central European parasitic fungal species of Fusarium obtained from infected wheat kernels, and to evaluate the oils as potential natural fungicides. The study was conducted in 2 stages. At each stage, the fungicidal activity of EsO (with concentrations of 0.025; 0.05; 0.125; 0.25; 0.50; 1.0, and 2.0%) against Fusarium spp. was evaluated using the disc plate method and zones of growth inhibition were measured. At the first stage, the fungistatic activity of EsO was evaluated against four species of Fusarium from the Polish population (F. avenaceum FAPL, F. culmorum FCPL, F. graminearum FGPL and F. oxysporum FOPL). The correlation coefficient between the mycelial growth rate index (T) and the fungistatic activity (FA) was calculated. At the second stage, on the basis of the mycelium growth rate index, the effectiveness of the EsO in limiting the development of Fusarium isolates from the German population (F. culmorum FC1D, F. culmorum FC2D, F. graminearum FG1D, F. graminearum FG2D and F. poae FP0D) was assessed. The first and second stage results presented as a growth rate index were then used to indicate essential oils (as potential natural fungicides) effectively limiting the development of various common Central European parasitic species Fusarium spp. Finally, the sensitivity of four Fusarium isolates from the Polish population and five Fusarium isolates from the German population was compared. The data were compiled in STATISTICA 13.0 (StatSoft, Inc, CA, USA) at the significance level of 0.05. Fusarium isolates from the German population were generally more sensitive than those from the Polish population. The sensitivity of individual Fusarium species varied. Their vulnerability, regardless of the isolate origin, in order from the most to the least sensitive, is as follows: F. culmorum, F. graminearum, F. poae, F. avenaceum and F. oxysporum. The strongest fungicidal activity, similar to Funaben T, showed thyme oil (regardless of the concentration). Performance of citral oils (lemongrass and Litsea cubeba) was similar but at a concentration above 0.025%.

Fusariumes un género fúngico amplio y diverso de diferentes complejos deespecies, causante de una gran variedad de enfermedades en plantas, productor dediversas toxinas y representa un importante patógeno oportunista en humanos. Laidentificación de las especies de Fusarium ha sido por mucho tiempo una tareacompleja y controversial. Esto es debido principalmente a la aplicación de diferentessistemas taxonómicos y la inherente variabilidad morfológica de algunas de estasespecies. Estas características requieren de la revisión por parte de un expertomicólogo, con el fin de lograr un acertado y confiable diagnóstico, el cual es crucialen el manejo de enfermedades o infecciones y estudios de diversidad genética. EnColombia, se ha reportado un incremento anual del 317 % de casos de infeccionescausadas por Fusarium, entre 1995 y 2003, sin embargo en centros especializados anivel nacional en micología médica, no se lleva a cabo un diagnóstico a nivel deespecie. El objetivo de este estudio fue el de establecer la identidad de aislamientosclínicos de Fusarium, mediante el uso de un marcador molecular. Para lograr esteobjetivo se llevó a cabo la identificación de los 59 aislamientos mediante consulta enla base de datos Fusarium-ID con base en secuencias codificantes del factor deelongación de la traducción EF-1a. Los resultados obtenidos permitieron observar laagrupación de los 59 aislamientos en tres complejos de especies: Fusariumoxysporum(FOSC), Fusarium solani (FSSC) y Fusarium incarnanatum-equiseti(FIESC). Basado en los resultados, se observa que el uso de las secuenciascodificantes para el factor de elongación de traducción permiten una confiableclasificación de los aislamientos de origen clínico y permite ratificar la utilidad queposee este marcador molecular en los distintos complejos de Fusarium.

El comercio del café es uno de los renglones económicos más importantes a nivel mundial, sin embargo, también es susceptible a contaminaciones desde su cosecha hasta su transformación. Para la realización de este estudio se recolectaron muestras de café cereza procedentes de diferentes municipios productores de Norte de Santander y posteriormente en el laboratorio se aislaron y caracterizaron fenotípicamente los siguientes agentes fúngicos del grano de café: Aspergillus  spp., Penicillium spp., Fusarium spp., Cladosporium  spp., Mucor spp. y  Rhizopus spp. con ayuda de claves taxonómicas. Los hongos que presentaron una alta incidencia fueron Aspergillus spp. y Fusarium spp. Entre las tres variables fisicoquímicas analizadas en el café cereza (pH, actividad de agua, % de humedad) se puede afirmar con certeza que la actividad de agua y el porcentaje de humedad influyen directamente en el número y tipo de aislamientos fúngicos obtenidos.
