Engineering Meniscus Form and Function via Multi-Layer Cell-Seeded Nanofibrous Scaffolds With Circumferentially Aligned Fibers

The menisci are crescent-shaped fibrocartilaginous tissues which function to transmit and distribute complex loading patterns between the femur and tibia of the knee joint. Meniscus function in tension arises from highly aligned collagen fibers which run in a circumferential manner between insertion sites on the tibial plateau (1,2). However, the meniscus is often injured, and partial removal of the meniscus represents the most commonly performed orthopaedic surgery, despite the fact that its removal increases the likelihood of osteoarthritis in the long-term (3). Tissue engineered scaffolds have emerged as a promising alternative to replace portions of the damaged meniscus (4). Toward replacement, we have developed aligned nanofibrous scaffolds that can recapitulate the mechanical anisotropy of the meniscus (5,6). More recently, we have developed an approach to replicate the circumferential macroscopic orientation of fibers using a novel electrospinning method (7). However, these organized scaffolds are relatively thin (<1 mm), and so multilayer scaffolds will be required to replicate the anatomic size of the native tissue. Moreover, cellular interactions will be crucial to the long-term function of these scaffolds. Thus, the objective of this study was to evaluate the morphological characteristics and mechanical properties of single layer and multilayer circumferentially aligned (CircAl) scaffolds seeded with mesenchymal stem cells (MSCs) and to compare them to scaffolds featuring linearly aligned (LinAL) fibers. We hypothesized that with increasing time in culture, matrix formed between layers would provide mechanical reinforcement, particularly in CircAl scaffolds where fiber rotation would be more likely to occur.

Neomycin was instilled daily, uni-or bilaterally, into the middle ear of guinea pigs for three months. The cochleae were examined, by light and electron microscopy, six months after the end of treatment. The organs of Corti of the treated ears were completely destroyed, and in the most advanced lesions, were substituted by a single layer of very thin flat cells. In the spiral ganglion only some glial cells and a few neurons could be observed. All surviving neurons were myelinated, and their ultrastructure was greatly altered, with disappeared, losing it amorphous and filamentous components. The spiral limbus and the stria vascularis were atrophic and were also covered, in the final stages, by flat elongated cells. In view of its morphological characteristics, this epithelium may arise from the displacement of the interdental cells and perhaps from the cochlear surface of Reissner's membrane.

The use of sewage sludge in agriculture and recovery of degraded areas has been shown as a promising alternative for its final destination. Studies on micronutrient levels after sludge application are necessary to avoid soil contamination at toxic levels. The objective of this work was to verify the micronutrient contents in the soil profile and pH, up to one-meter-deep, nine years after the application of sewage sludge and planting of native species of the Atlantic Forest. The experiment was implemented in a degraded Quartzeneic Neosol and conducted in randomized blocks with four replicates and eight treatments, consisting of six doses of sewage sludge (0, 2.5, 5, 10, 15 and 20 Mg ha-1, with supplementation of potassium due to low concentration in the residue), besides the control treatment, mineral fertilization and only potassium supplementation. After nine years, the contents of all micronutrients evaluated presented a significant response to the application of the treatments, and the application of sewage sludge provided an increase in their contents. Soil pH remained stable at sites receiving mineral fertilization and potassium supplementation. Only manganese and zinc showed mobility in the soil profile. The application of sewage sludge in degraded soil increases the micronutrient content and decreases its movement in the soil profile, and the application of the maximum dose of the residue does not provide toxic levels of these elements in the soil in the long term.

ABSTRACT The long-term effects of interferon treatment on cell lines that maintain human papillomavirus type 31 (HPV-31) episomes have been examined. High doses and prolonged interferon treatment resulted in growth arrest of HPV-positive cells, with a high percentage of cells undergoing apoptosis. These effects were not seen with interferon treatment of either normal human keratinocytes or cells derived from HPV-negative squamous carcinomas, which exhibited only slight decreases in their rates of growth. Within 2 weeks of the initiation of treatment, a population of HPV-31-positive cells that were resistant to interferon appeared consistently and reproducibly. The resistant cells had growth and morphological characteristics similar to those of untreated cells. Long-term interferon treatment of HPV-positive cells also resulted in a reduction in HPV episome levels but did not significantly decrease the number of integrated copies of HPV. Cells that maintained HPV genomes lacking E5 were sensitive to interferon, while cells expressing only the E6/E7 genes were resistant. In contrast, cells that expressed E2 from a tetracycline-inducible promoter were found to be significantly more sensitive to interferon treatment than parental cells. This suggests that at least a portion of the sensitivity to interferon could be mediated through the E2 protein. These studies indicate that cells maintaining HPV episomes are highly sensitive to interferon treatment but that resistant populations arise quickly.

ABSTRACT: Cultivars of the genus Nopalea are known in Brazil for being tolerant to cochineal carmine attacks, thus making the cultivation of this genus a promising alternative for mitigating the negative effects of this insect on the production of biomass. With the objectives of characterizing morphologically spineless forage cactus varieties and identify morphological characteristics that may be the focus in spineless forage cactus breeding programs, an experiment was conducted in a completely randomized block design with 11 treatments and four replications. The variety Alagoas showed the highest values of weight, area and volume of cladodes. The varieties Negro Michoacan F7 and V7, Tamazunchale V12 showed the highest values of the cladode area index, the total volume of cladodes and total fresh mass production. The varieties Negro Michoacan V7 and F7 presented the highest water use efficiency and dry mass yield. Cladode volume showed the highest correlation coefficients with the fresh weight of cladodes. Aiming the release of varieties for biomass production, varieties Negro Michoacan F7, V7 and Tamazunchale V12 may substitute the Miúda variety. The number and cladode area index may be used as criteria for selection of superior varieties in breeding programs.

Due to their distinctive physicochemical properties, platinum nanoparticles (PtNPs) have emerged as a material of interest for a number of biomedical therapeutics. However, in some instances NP exposure has been correlated to health and safety concerns, including cytotoxicity, activation of cellular stress, and modification to normal cell functionality. As PtNPs have induced differential cellular responses in vitro, the goal of this study was to further characterize the behavior and toxicological potential of PtNPs within a HepG2 liver model. This study identified that a high PtNP dosage induced HepG2 cytotoxicity. However, lower, subtoxic PtNP concentrations were able to elicit multiple stress responses, secretion of proinflammatory cytokines, and modulation of insulin-like growth factor-1 dependent signal transduction. Taken together, this work suggests that PtNPs would not be overtly toxic for acute exposures, but sustained cellular interactions might produce long term health consequences.

Inland waters are warming at highly variable rates that often differ from regional air temperature trends. This variable warming is partially attributable to an individual lake’s geographical and morphological characteristics. In very large lakes, significant intralake variability in long-term warming trends has also been observed. In light of this intralake and interlake heterogeneity of lake surface water temperature (LSWT) and LSWT trends, we revisit the 1.1 km Advanced Very High Resolution Radiometer (AVHRR) record for the Laurentian Great Lakes. In this work, we have assembled a long-term (1986–2016) and high-spatial-resolution (0.018°) daily LSWT dataset using AVHRR record. Subtracting an empirically-determined mean diurnal cycle mitigates the effects of varying observation times. Adjustments in the georegistration of the images are made to reduce the impact of AVHRR navigational errors on the earlier platforms. Both the original daily composites, and a gap-filled product using locally weighted interpolation methods will be made available to support fine-scale physical and environmental research in the region.

Medium lacking serum but supplemented with milk will support the growth of sparse cells in culture. Milk obtained within 8 h after the birth of a calf (day 1 colostrum) is the most effective in supporting proliferation. In mixed cultures of early-passage bovine embryonic kidney (BEK) or early-passage calf kidney (CK) cells, both epithelial cells and fibroblasts grow in Dulbecco's modified eagle's medium (DMEM) supplemented with serum. However, only cells that appear to be epithelial-like grow in DMEM supplemented with colostrum. Sparse cultures of early-passage human and rat fibroblasts that grow readily in DMEM supplemented with serum do not grow in DMEM supplemented with colostrum. Canine kidney epithelial cells (MDCK), when plated sparsely, grow exponentially in DMEM supplemented with day 1 bovine colostrum. The generation time is 26 h, the same growth rate as in DMEM supplemented with calf serum. The MDCK cells can be subcultured and regrown to confluence repeatedly in colostrum-supplemented DMEM. Growth in DMEM supplemented with colostrum does not alter the morphological characteristics of the MDCK cells, which are polygonal, contain microvilli at the apical surface, and are connected by tight junctions and desmosomes. MDCK cells do not proliferate in DMEM supplemented with milk obtained 1 wk after the birth of a calf.

AbstractPurposeThe lack of long-term patency of synthetic vascular grafts currently available on the market has directed research towards improving the performance of small diameter grafts. Improved radial compliance matching and tissue ingrowth into the graft scaffold are amongst the main goals for an ideal vascular graft.MethodsBiostable polyurethane scaffolds were manufactured by electrospinning and implanted in subcutaneous and circulatory positions in the rat for 7, 14 and 28 days. Scaffold morphology, tissue ingrowth, and mechanical properties of the scaffolds were assessed before implantation and after retrieval.ResultsTissue ingrowth after 24 days was 96.5 ± 2.3% in the subcutaneous implants and 77.8 ± 5.4% in the circulatory implants. Over the 24 days implantation, the elastic modulus at 12% strain decreased by 59% in direction of the fibre alignment whereas it increased by 1379% transverse to the fibre alignment of the highly aligned scaffold of the subcutaneous implants. The lesser aligned scaffold of the circulatory graft implants exhibited an increase of the elastic modulus at 12% strain by 77% in circumferential direction.ConclusionBased on the observations, it is proposed that the mechanism underlying the softening of the highly aligned scaffold in the predominant fibre direction is associated with scaffold compaction and local displacement of fibres by the newly formed tissue. The stiffening of the scaffold, observed transverse to highly aligned fibres and for more a random fibre distribution, represents the actual mechanical contribution of the tissue that developed in the scaffold.

Epidemiological studies on air pollution in Mexico often use the environmental concentrations of pollutants as measured by monitors closest to the home of participants as exposure proxies, yet this approach does not account for the space gradients of pollutants and ignores intra-city human mobility. This study aimed to develop high-resolution spatial and temporal models for predicting long-term exposure to PM2.5 and NO2 in ~16,500 participants from the Mexican Teachers’ Cohort study. We geocoded the home and work addresses of participants, and used secondary source information on geographical and meteorological variables as well as other pollutants to fit two generalized additive models capable of predicting monthly PM2.5 and NO2 concentrations during the 2004-2019 period. Both models were evaluated through 10-fold cross-validation, and showed high predictive accuracy with out-of-sample data and no overfitting (CV-RMSE=0.102 for PM2.5 and CV-RMSE=4.497 for NO2). Participants were exposed to a monthly average of 24.38 (6.78) mg/m3 of PM2.5 and 28.21 (8.00) ppb of NO2 during the study period. These models offer a promising alternative for estimating PM2.5 and NO2 exposure with high spatio-temporal resolution for epidemiological studies in the Mexico City Metropolitan Area.

Purpose The purpose of this paper is to verify the effects of extrusion temperature on orthotropic behaviour of the mechanical properties of parts obtained by fused filament fabrication (FFF) under quasi-static tensile loads. Design/methodology/approach Tensile tests were performed on single layer specimens fabricated in polylactic acid (PLA) and acrylonitrile butadiene styrene (ABS) to evaluate the mechanical properties at different extrusion temperatures and raster orientations (0°, 45° and 90°). Furthermore, a detailed study of morphological characteristics of the single layer samples cross-section and of the bonding quality among adjacent deposited filaments was performed by scanning electron microscopy to correlate the morphology of materials with mechanical behaviour. Findings The results show that the orthotropic behaviour of FFF-printed parts tends to reduce, while the mechanical properties improved with increase in extrusion temperature. Furthermore, the increase in extrusion temperature led to an improvement in inter-raster bonding quality and in the compactness and homogeneity of the parts. Originality/value The relation between the extrusion temperature, orthotropic behaviour and morphological surface characteristics of the single layer specimen obtained by FFF has not been previously reported.
