1. The effects of an anoxic-aglycemic episode (1-3 min) on the pharmacologically isolated N-methyl-D-aspartate (NMDA)-mediated responses were examined in CA1 pyramidal hippocampal neurons in vitro. 2. An anoxic-aglycemic episode induced a long term potentiation (LTP) of the NMDA receptor-mediated field excitatory post-synoptic potentials (EPSPs). This LTP, referred to as anoxic LTP, was observed in the presence of 1) a normal Mg2+ concentration [+40.1 +/- 5% (mean +/- SE)], 2) a low Mg2+ concentration (+52.2 +/- 10%), or 3) a Mg2+ free (+49 +/- 11%), 1 h after anoxia. 3. Bath application of D-2-amino-5-phosphonovaleric acid (D-APV, 20 microM, 15-21 min) before, during, and after the anoxic-aglycemic episode, which transiently blocked the synaptic NMDA receptor mediated response, prevented the induction of anoxic LTP. 4. The intracellularly recorded NMDA receptor-mediated EPSP was also persistently potentiated by anoxia-aglycemia (+47 +/- 4%). This potentiation was not associated with changes in membrane potential or input resistance. 5. These findings provide the first evidence that an anoxic-aglycemic episode induces an LTP of NMDA receptor-mediated responses. This potentiation may participate in the cascade of events that lead to delayed neuronal death.

The effect of intracerebroventricular (icv) injection of Aβ25–35 and/or intraperitoneal (ip) application of the L-type calcium channel (VDCC) blockers verapamil or diltiazem were examined in vivo. To by-pass possible systemic actions of these agents, their effects on long-term potentiation (LTP) in the CA1 region of the in vitro hippocampal slice preparation were also examined. Application of Aβ25–35 (10 nmol in 5 μl, icv) significantly impaired LTP in vivo, as did IP injection of verapamil (1 or 10 mg/kg) or diltiazem (1 or 10 mg/kg). In the in vitro slice preparation, LTP was also depressed by prior application of Aβ25–35 (500 nmol), verapamil (20 μM), or diltiazem (50 μM). Combined application of Aβ25–35 and verapamil in either the in vivo or in vitro preparation resulted in a significant reversal of the LTP depression observed in the presence of either agent alone. However, co-application of diltiazem and Aβ25–35 failed to attenuate the depression of LTP observed in the presence of either agent alone in vivo or in vitro. Since LTP is a cellular correlate of memory and Aβ is known to be involved in Alzheimer's disease (AD), these results indicate that verapamil, a phenylalkylamine, may be useful in the treatment of cognitive deficits associated with AD.

Μέρος Ι. Αναπτυξιακές αλλαγές κατά την πρώιμη μεταγεννετική περίοδο του ανασταλτικού κυκλώματος του έσω προμετωπιαίου φλοιού σε σχέση με τον κύριο σωματοαισθητικό φλοιό.Ο προμετωπιαίος φλοιός (prefrontal cortex, PFC) ελέγχει ανώτερες γνωστικές ικανότητες που έχουν σοβαρή βλάβη στις νευροαναπτυξιακές διαταραχές. Ο PFC χαρακτηρίζεται από καθυστερημένη ωρίμανση που εκτείνεται μέχρι το τέλος της εφηβείας. Οι κυτταρικοί μηχανισμοί που ελέγχουν την πρώιμη ανάπτυξη των κυκλωμάτων του προμετωπιαίου φλοιού παραμένουν σε μεγάλο βαθμό αδιευκρίνιστοι. Η μελέτη μας αναλύει τις αναπτυξιακές κυτταρικές διεργασίες in vitro που βρίσκονται σε εξέλιξη στον έσω PFC (medial PFC, mPFC) του ποντικιού από τη δεύτερη και την τρίτη μεταγεννητική εβδομάδα ή νεογνική και προ-νεανική, αντίστοιχα, συγκριτικά με αυτές στο βαρελοειδή φλοιό (barrel cortex, BC). Τα αποτελέσματα μας δείχνουν ότι η βασική συναπτική μεταβίβαση μειώνεται με την ηλικία ως αποτέλεσμα της ταυτόχρονης μείωσης των αυθόρμητων μετασυναπτικών διεγερτικών ρευμάτων και της αύξησης των ανασταλτικών. Αν και η ολική κυτταρική πυκνότητα του mPFC μειώνεται από την δεύτερη στην τρίτη μεταγεννητική εβδομάδα, ο αριθμός των ενδονευρώνων που εκφράζουν την παρβαλβουμίνη (parvalbumin, PV) και τον υποδοχέα της σεροτονίνης (5HT3aR) αυξάνεται. Επιπλέον, τα δεδομένα μας υποδεικνύουν ότι η αυξημένη δραστικότητα του GABAA υποδοχέα οδηγεί σε αυξημένη βασική συναπτική μεταβίβαση στο νεογνικό mPFC, δείχνοντας ότι η επίδραση του GABA δεν θα μπορούσε να είναι ανασταλτική σε αυτό το χρονικό παράθυρο στον mPFC. Μέρος ΙΙ Μύες με μειωμένο αριθμό ενδονευρώνων επιδεικνύουν διαταραχές στην αυθόρμητη δραστηριότητα και στη ρυθμική δραστηριότητα των δικτύων του σωματαισθητικού /βαρελοειδούς φλοιού.Οι GABAεργικοί ενδονευρώνες είναι οι κύριοι ανασταλτικοί νευρώνες και προστατεύουν τους εγκεφαλικούς ιστούς από την υπερβολική διέγερση. Οι GABAεργικοί ενδονευρώνες του φλοιού έχουν σημαντικό ρόλο για την παραγωγή συγχρονισμένης ρυθμικής φλοιικής δραστηριότητας. Πολλές νευροψυχιατρικές διαταραχές συσχετίζονται με ανισορροπία ανάμεσα στους μηχανισμούς διέγερσης και αναστολής σε συνδυασμό με μεταβολές στη ρυθμική δραστηριότητα των νευρικών δικτύων, ειδικά σε γάμμα συχνότητες (30-80 Hz). Ερευνήσαμε τις αλλαγές της δραστηριότητας των φλοιικών δικτύων όταν απουσιάζει η αναστολή στον ενήλικο βαρελοειδή φλοιό (barrel cortex-BC) μυών. Χρησιμοποιήσαμε γενετικά τροποποιημένα ζώα ως πειραματικά μοντέλα-εργαλεία με 50% μείωση των ενδονευρώνων του ώριμου φλοιού, εξαιτίας της απαλοιφής της πρωτεΐνης Rac1 στους ενδονευρώνες που εκφράζουν το NKx2.1/Cre κατά την εμβρυική ηλικία [Rac1 conditional knockout (cKO)]. Σκοπός μας ήταν να εξετάσουμε πώς η αναπτυξιακή μείωση των φλοιικών ενδονευρώνων επηρεάζει τη βασική συναπτική μεταβίβαση, την συναπτική πλαστικότητα, την αυθόρμητη δραστηριότητα και τη ρυθμική δραστηριότητα στον ώριμο BC. Καταγραφές πεδίου διεγερτικών μετασυναπτικών δυναμικών (fEPSPs) είδαν ότα τα δίκτυα της φλοιικής στοιβάδας ΙΙ των Rac1cKO ζώων χαρακτηρίζονται από αυξημένη βασική συναπτική μεταβίβαση, μειωμένη μακροχρόνια ενδυνάμωση ( long-term potentiation, LTP) μετά από ένα έντονο ερεθισμό και ανεπηρέαστη αναλογία συζευγμένων παλμών (pair-pulse ratio, PPR). Επιπλέον, καταγραφές αυθόρμητης δραστηριότητας σε φέτες εγκεφάλου των Rac1cKO ζώων έδειξαν αυξημένη ευαισθησία και επιδεκτικότητα στην εμφάνιση αυθόρμητης δραστηριότητας. Επίσης, βρήκαμε ότι στα ζώα με μειωμένο αριθμό των φλοιικών ενδονευρώνων, η δραστηριότητα των τοπικών νευρικών δικτύων εμφανίζει μειωμένη ισχύ στις χαμηλές συχνότητες (δέλτα 0-4 Hz, θήτα 4-8 Hz, άλφα 8-12 Hz) και στις γάμμα συχνότητες (30-80 Hz) με μία πρόσθετη παρεκκλίνουσα κορυφή στις υψηλές γάμμα συχνότητες (80-150 Hz). Συνεπώς, τα αποτελέσματά μας απέδειξαν ότι η διαταραχή της GABAεργικής αναστολής αλλάζει τις συναπτικές ιδιότητες και τη συναπτική πλαστικότητα, και επιπλέον διαταράσσει τον συγχρονισμό των νευρικών δικτύων του ενήλικου BC.

In Vitro Plasticity of the Direct Feedback Pathway in the Electrosensory System of Apteronotus leptorhynchus

Wang, Daliang and Leonard Maler. In vitro plasticity of the direct feedback pathway in the electrosensory system of Apteronotus leptorhynchus. J. Neurophysiol. 78: 1882–1889, 1997. We have used field and intracellular recording from pyramidal cells in an in vitro preparation of the electrosensory lateral line lobe (ELL) of Apteronotus leptorhynchus to investigate synaptic plasticity of a direct feedback pathway: the (StF). Tetanic stimulation of the StF enhanced the StF-evoked synaptic response by 145% in field and the excitatory postsynaptic potential (EPSP) 190% in intracellular recordings. Maximal enhancement occurred at 5 s and lasted for ∼120 s. Tetanic frequencies of 100–300 Hz produced enhancement; lower or higher frequencies failed to produce statistically significant changes in EPSP amplitude. Rates of 100–200 Hz occur in vivo in the cells of origin of the StF, suggesting that this form of plasticity may be operative under natural conditions. We could not elicit either long-term potentiation or depression by any stimulation protocol of the StF; in the case of long-term potentiation, this held even when excitatory transmission was enhanced by application of bicuculline, a γ-aminobutyric acid-A antagonist. When tetanic stimulation of the StF was paired with hyperpolarization of pyramidal cells, subsequent StF-evoked EPSPs were increased by 146% (5 min posttetanus); this anti-Hebbian synaptic enhancement lasted for ∼10 min. Neither tetanic stimulation alone, hyperpolarization alone, nor tetanic stimulation paired with pyramidal cell depolarization altered StF-evoked EPSP amplitudes on this time scale. Anti-Hebbian synaptic enhancement was not blocked by the N-methyl-d-aspartate–receptor antagonist D.L-aminophosphovalerate. The in vitro demonstration of anti-Hebbian plasticity at StF synapses replicates similar in vivo results. Anti-Hebbian synaptic plasticity of the StF may be responsible in part for the ability of gymnotiform fish to reject redundant electrosensory signals.
