Selection of Optimum Process, Solvent and Drying Method for Extraction of Antioxidants

Satu uji kaji telah dijalankan untuk pemilihan proses optimum, pelarut, dan teknik pengeringan bagi pengekstrakan antioksidan daripada buah Morinda citrifolia. Pengekstrakan telah dijalankan dengan soxhlet, ultrasonik dan tekanan tinggi hidrostatik yang diubahsuai pada 100 bar dan 50°C dengan 15 gram serbuk Morinda dalam 200 ml pelarut. Pelarut yang telah digunakan ialah air, etanol, dan etil asetat. Hasil penurasan kemudiannya dikeringkan melalui vakum dan pengering semburan. Aktiviti antioksidan atau peratusan ekstrak perencatan ditentukan dengan radikal Di-Phenyl picryl hydrazyl. Hasil ekstrak juga telah ditentukan dalam kajian ini. Pengoptimuman telah dilakukan dengan menggunakan kaedah gerak balas permukaan. Hasil kajian ini menunjukkan bahawa, aktiviti antioksidan atau peratusan perencatan DPPH radikel bebas yang paling tinggi dipamerkan daripada ekstrak yang diperolehi dari proses HHPE dengan etil asetat sebagai pelarut yang dikeringkan melalui vakum. Daripada kajian ini juga, hasil ekstrak yang tertinggi diperolehi melalui proses HHPE pada 100 bar dan 50°C, di mana etil asetat digunakan sebagai pelarut dengan teknik pengeringan vakum. Di samping itu, didapati juga bahawa keadaan pengekstrakan dan pelarut secara langsungnya memberi kesan kepada pengekstrakan antioksidan daripada buah Morinda citrifolia jika dibandingkan dengan teknik pengeringan. Kata kunci: Aktiviti antioksidan, radikel DPPH, pengekstrakan tekanan tinggi, Morinda citrifolia, pengekstrakan ultrasonik, pengekstrakan soxhlet An experimental study was performed for optimum process, solvent and drying method for the extraction of antioxidants from Morinda citrifolia fruit. Extractions were performed by soxhlet, ultrasonic and modified version of high hydrostatic pressure extraction at 100 bar and 50°C with 15 grams of powdered morinda in 200 ml of solvent. Water, ethanol and ethyl acetate were used as solvents. The filtrate was dried by vacuum and spray dryers. The antioxidant activity or inhibiton percentage of the extracts was determined by Di-Phenyl picryl hydrazyl radical scavenging method. The yield was also determined. Optimization was performed using response surface methodology. It was found that the highest antioxidant activity or inhibition percentage of DPPH free radicals was exhibited by the extract obtained from HHPE process with ethyl acetate as solvent and vacuum dried. It was also found that the highest yield was obtained from HHPE process at 100 bar and 50°C with ethanol as solvent and vacuum dried. It was noted that the nature of the process and the solvent used significantly contributed to the extraction of antioxidants from Morinda citrifolia fruit when compared with the drying methods. Turmeric (Curcuma longa L.) is known for its health benefits. Several previous studies revealed that curcumin, the main active compound in turmeric, has antioxidant capacity. It has been previously demonstrated that puffing, the physical processing using high heat and pressure, of turmeric increases the antioxidant and anti-inflammatory activities by increasing phenolic compounds in the extract. The current study sought to determine if high hydrostatic pressure extraction (HHPE), a non-thermal extraction at over 100 MPa, aids in the chemical changes and antioxidant functioning of turmeric. 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) analyses were conducted and assessed the content of total phenol compounds in the extract. The chemical changes of curcuminoids were also determined by high performance liquid chromatography (HPLC). Among the three variables of ethanol concentration, pressure level, and treatment time, ethanol concentration was the most influential factor for the HHPE of turmeric. HHPE at 400 MPa for 20 min with 70% EtOH was the optimal extraction condition for the highest antioxidant activity. Compositional analysis revealed that 2-methoxy-4-vinylphenol was produced by puffing. Vanillic acid and ferulic acid content increased with increasing HHPE time. Synergistic effect was not observed on antioxidant activity when the turmeric was sequentially processed using puffing and HHPE.

Noni blossoms have a long history of medicinal uses in tropical areas. This study was conducted to investigate the major phytochemical components, toxicological properties, and antioxidant activity of noni blossoms. An HPLC-PDA method was developed and validated for the identification and quantification of major components. The major phytochemicals were iridoid glycosides, deacetylasperulosidic acid and asperulosidic acid, and flavonoids, quercetin-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside and kaempferol-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside, each present at 3.764, 3.576, 1.513, and 3.096 mg/g, respectively. The aqueous extract of noni blossoms, at 500 μg/mL, exhibited greater antioxidant activity in the 2,2-diphenylpicrylhydrazyl radical scavenging assay than green tea (88.11 ± 0.01% versus 76.60 ± 0.05%). A primary DNA damage test in E. coli PQ37 (SOS-chromotest) and a twenty-four hour brine shrimp toxicity test did not reveal any genotoxic or cytotoxic activity. These results provide a useful reference for the identification of noni blossoms as well as preliminary evaluation of safety and efficacy. Further evaluation of the potential applications of noni blossoms is warranted.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

We investigated the effects of cooking (steaming and microwaving) and processing (freeze-drying and hot-air-drying) methods on the antioxidant activity of broccoli florets. 2,2-diphenyl-1-picrylhydrazyl (DPPH•), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS•), and alkyl• free radical scavenging assays were employed to assess anti-oxidant potentials. The cytoprotective effect against oxidative damage induced by H2O2 was studied using hepatocellular carcinoma (HepG2) cells. Anti-proliferative effects were assessed in MCF-7 and MDA-MB-231 breast cancer cells. L-sulforaphane in broccoli extracts was quantified using high-performance liquid chromatography (HPLC). Steam and microwave treatments caused increases in total polyphenol content (TPC), whereas the total flavonoid content (TFC) decreased following steam treatment. A slight increase in TFC was observed in the microwaved samples. Extracts of all broccoli samples showed almost identical radical scavenging and cytoprotective effects. HPLC demonstrated that steamed (3 min)-freeze-dried (F-S3) and microwaved (2 min)-freeze-dried (F-M2) samples exhibited elevated levels of L-sulforaphane. In addition, the F-S3 and F-M2 extracts displayed strong anti-proliferative effects in MCF-7 cells, which correlated with L-sulforaphane content. As we observed no significant decrease in the antioxidant activity of broccoli florets, the cooking and processing methods and conditions studied here are recommended for broccoli.

Antioxidant activities of acetone and ethyl acetate extracts from Metaplexis japonica Makino, one of famous medicine plants in the eastnorth region of China, named luomo in Chinese, were examined by a DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical-scavenging assay and a β-carotene-linoleic acid test. In DPPH, the antioxidant activity of the acetone extracts, ethyl acetate extracts and derivative were IC50 were 313.21, 266.92 and 118.78μg/mL, respectively. In the β-carotene-linoleic acid test, IC50 were 285.09, 351.57 and 123.89μg/mL. It was concluded that Metaplexis japonica Makino and its derivatives might be a potential natural source of antioxidants .

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

Objective: This study was designed to determine the Total phenol, flavonoid content, antioxidant and free radical scavenging properties of different solvent extracts of Piper umbellatum.  Methods: Different solvent extracts evaluated with DPPH radical scavenging activity and Reducing power activity.Results: The presence of phenol and flavonoid showed highly in the methanol extracts than ethyl acetate and acetone extracts. All the extracts have various level of antioxidant activity. Methanol solvent extract have good extraction and show significant antioxidant activity. The effect of reducing power of methanol extract revealed good antioxidant activity compare with other tested extracts.Conclusion: On the basis of the above results we concluded that methanol extract of Piper umbellatum whole plant extracts shows significant antioxidant activity than ethyl acetate and acetone extracts.Keywords: Piper umbellatum, Antioxidant, DPPH, FRAP assay, Polar, Non polar solvents

<p> </p><p>Red dragon fruit (<em>H. Polyrhizus</em>) is one of the the plants that has a great potential as natural antioxidant. This study tested the activity of radical scavenging of 2-2' diphenyl -1- pikril hidrazil (DPPH) in the methanol extract, as well as in the soluble and insoluble fractions of ethyl acetate of red dragon fruit peel. This research is carried out through various stages, such as: extraction and fractionation to obtain both insoluble fraction and soluble fractions of ethyl acetate. Antioxidant activity test is conducted by the method of thin layer chromatography and spectrophotometry.<strong> </strong>Antioxidant activity test, IC<sub>50 </sub>values of methanol extract, ethyl acetate soluble fraction, and insoluble fraction of ethyl acetate had been obtained consecutively as much as 241.19 µg /mL, 8.34  µg/mL, 46.84 µg/mL. The soluble fraction of ethyl acetate had greater antioxidant activity compared to the methanol extract and the insoluble fractions of ethyl acetate.</p>

Jussara (Euterpe edulis) fruit is a strong candidate for exportation due to its high content of anthocyanins. However, its rapid perishability impairs its potential for further economic exploration, highlighting the relevance of producing ready-to-drink juices by applying innovative processing, such as high hydrostatic pressure (HHP). The effect of HHP (200, 350, and 500 MPa for 5, 7.5, and 10 min) on anthocyanins content and antioxidant activity (AA) by FRAP and TEAC assays, and the most effective HHP condition on overall sensory acceptance and stability of jussara juice, were investigated. While mild pressurization (200 MPa for 5 min) retained anthocyanins and AA, 82% of anthocyanins content and 46% of TEAC values were lost at the most extreme pressurization condition (500 MPa for 10 min). The addition of 12.5% sucrose was the ideal for jussara juice consumer acceptance. No significant difference was observed for overall sensory acceptance scores of unprocessed (6.7) and HHP-processed juices (6.8), both juices being well-accepted. However, pressurization was ineffective in promoting the retention of anthocyanins and AA in jussara juice stored at refrigeration temperature for 60 days, probably due to enzymatic browning.
