AbstractThe sterile insect technique (SIT) is widely used in integrated pest management programs for the control of the Mediterranean fruit fly (medfly), Ceratitis capitata. The genetic interactions between the released individuals from the genetic sexing strains (GSS), used for SIT applications worldwide, and wild individuals have not been studied. Under the hypothesis that a number of Vienna GSS individuals released to the field might not be completely sterile and may produce viable offspring, we have analyzed medfly Spanish field populations to evaluate the presence of Vienna strain genetic markers. To this goal, we have used contrasted nuclear and mitochondrial genetic markers, and two novel sets of nuclear polymorphisms with the potential to be markers to discriminate between Vienna and wild individuals. Nuclear Vienna markers located on the 5th chromosome of Vienna males have been found in 2.2% (19 from 875) of the Spanish wild medfly females captured at the area where SIT is applied. In addition, a female-inherited mitochondrial Vienna marker has been found in two from the 19 females showing nuclear Vienna markers. The detection of several of these markers in single individuals represents evidence of the introgression of Vienna strain into natural populations. However, alternative explanations as their presence at low frequency in wild populations in the studied areas cannot be fully discarded. The undesired release of non-fully sterile irradiated GSS individuals into the field and their interactions with wild flies, and the potential environmental implications should be taken into account in the application of the SIT.

The Sterile Insect Technique (SIT) is an effective pest control method developed worldwide against many species of Fruit Flies. It involves the release of male insects sterilized, at pupae instar, by exposure to ionizing radiation.  The Area-wide control of medfly (Ceratitis capitata), with SIT based on sterile males release combined to other control techniques, was implemented since 2017 as a pilot area in Moulouya Perimeter in North-eastern Morocco. The main objective of this study is to evaluate the effectiveness of this tool in controlling Medfly and estimate sterile male population densities from data collected from trap catches after the release of a pre-determined number of sterile males in five Citrus orchards (0, 500, 1000 and 3000 sterile males per hectare) and the calculation of FTD (fly/trap/day) of sterile and wild males. The result showed that the percentage of recaptured males and FTD Sterile indices were related to release density by power function regression, while the sterile to wild ratio and release density were linear regression-adjusted. The finding confirmed the effectiveness of release in reducing the fly population in the TIS area by reducing the rate of citrus infestation in field and export shipments.

Μελετήθηκε η επίδραση του ενδοκυτταρικού βακτηρίου Wolbachia pipientis σε βασικές παραμέτρους της ζωής της μύγας της Μεσογείου, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), καταγράφηκε η σύσταση της βακτηριακής κοινότητας του πεπτικού της συστήματος και μελετήθηκε η προβιοτική δράση του απομονωθέντος βακτηριακού είδους Enterobacter sp. στη φυλή Vienna 8 GSS του εντόμου, η οποία εκτρέφεται μαζικά για την κάλυψη των αναγκών των προγραμμάτων εξαπόλυσης στειρωμένων εντόμων (Sterile Insect Technique). Στην πρώτη ενότητα πειραμάτων που πραγματοποιήθηκαν στο εργαστήριο Εντομολογίας και Γεωργικής Ζωολογίας του Πανεπιστημίου Θεσσαλίας μελετήθηκαν σε εργαστηριακές συνθήκες και με έντομα αποικιών που εκτρέφονταν σε μικρούς πληθυσμούς, βασικές βιολογικές και συμπεριφορικές προσαρμογές του εντόμου συνεπεία της βακτηριακής μόλυνσης με Wolbachia. Τα αποτελέσματα έδειξαν ότι η μόλυνση με Wolbachia: α) δεν επηρέασε την πρόοδο της αναπαραγωγικής ωρίμανσης των θηλυκών ούτε το συνολικό αριθμό ώριμων αυγών στις ωοθήκες τους, τόσο εκείνων που τράφηκαν στο στάδιο του ενηλίκου με υδρολυμένη μαγιά και ζάχαρη όσο και εκείνων που τράφηκαν μόνο με ζάχαρη, β) μείωσε τη συχνότητα του σεξουαλικού καλέσματος των αρσενικών της μύγας της Μεσογείου. Η αρνητική επίδραση της μόλυνσης με Wolbachia ήταν εντονότερη στα αρσενικά που είχαν πρόσβαση σε πρωτεΐνη (υδρολυμένη μαγιά) σε σχέση με τα αρσενικά που τρέφονταν αποκλειστικά με ζάχαρη. Επίσης, η εκδήλωση του σεξουαλικού καλέσματος ως ένδειξη της ταχύτητας αναπαραγωγικής ωρίμανσης των αρσενικών έδειξε ότι η βακτηριακή μόλυνση καθυστέρησε τη σεξουαλική ωρίμανση των αρσενικών τόσο σε εκείνα που τράφηκαν με υδρολυμένη μαγιά και ζάχαρη όσο και εκείνων που τράφηκαν μόνο με ζάχαρη, γ) μείωσε δραστικά την ανταγωνιστικότητα σύζευξης των αρσενικών όταν μελετήθηκε σε μικρά ατομικά κλουβιά (όγκου 400 cm3), και τέλος δ) δεν επηρέασε σημαντικά την αποθήκευση σπέρματος στις σπερματοθήκες των θηλυκών. Επίσης, στο πλαίσιο της πρώτης ενότητας πειραμάτων μελετήθηκε η επίδραση της μόλυνσης με Wolbachia των αρσενικών και των θηλυκών ατόμων της μύγας της Μεσογείου στη δεκτικότητα σύζευξης και επανασύζευξης, καθώς και η επίδραση του ιστορικού των συζεύξεων στα δημογραφικά χαρακτηριστικά (ωοπαραγωγή, εκκόλαψη των αυγών, διάρκεια ζωής) μολυσμένων και μη-μολυσμένων με Wolbachia θηλυκών της μύγας της Μεσογείου. Εν συντομία, τα αποτελέσματα έδειξαν ότι: α) η μόλυνση είτε των θηλυκών είτε των αρσενικών τροποποίησε τα ποσοστά σύζευξης και επανασύζευξης, β) τα μολυσμένα με Wolbachia θηλυκά απόθεσαν περισσότερα αυγά σε σύγκριση με τα μη-μολυσμένα θηλυκά. Η μολυσματική κατάσταση των αρσενικών δεν επηρέασε την ωοπαραγωγή. Ο αριθμός των συζεύξεων (μία ή δύο) των θηλυκών δεν επηρέασε την ωοπαραγωγή τους, γ) η μόλυνση των θηλυκών με Wolbachia μείωσε δραματικά τα ποσοστά εκκόλαψης των προνυμφών. Επιπλέον, το σπέρμα των μολυσμένων αρσενικών έδειξε πως ανταγωνίζεται επιτυχώς το σπέρμα των μη-μολυσμένων αρσενικών για τη γονιμοποίηση των αυγών των μη-μολυσμένων θηλυκών, δ) τα μολυσμένα με Wolbachia επανασυζευγμένα θηλυκά έζησαν περισσότερο από τα μη-μολυσμένα επανασυζευγμένα θηλυκά, παρότι η διάρκεια ζωής δεν διέφερε μεταξύ των μολυσμένων και μη-μολυσμένων θηλυκών που συζεύχθηκαν μόνο μία φορά. Επιπλέον, η επανασύζευξη των θηλυκών επέφερε κόστος στη διάρκεια ζωής μόνο των μη-μολυσμένων θηλυκών. Αντίθετα, η επανασύζευξη των μολυσμένων θηλυκών αύξησε τη διάρκεια ζωής τους σε σύγκριση με τα μολυσμένα θηλυκά που συζεύχθηκαν μόνο μία φορά. Η μολυσματική κατάσταση των αρσενικών δεν επηρέασε τη επιβίωση των θηλυκών σεξουαλικών συντρόφων τους. Η δεύτερη πειραματική ενότητα επικεντρώθηκε στην αξιολόγηση της προοπτικής συνεισφοράς των συμβιωτικών οργανισμών (Wolbachia, βακτήρια του πεπτικού συστήματος) στη μαζική παραγωγή εντόμων υψηλής βιολογικής ποιότητας για εφαρμογή σε προγράμματα καταπολέμησης εντόμων. Τα πειράματα πραγματοποιήθηκαν στους χώρους του εργαστηρίου FAO/IAEA Insect Pest Control Laboratory (IPCL), στο Seibersdorf της Αυστρίας. Χρησιμοποιήθηκαν έντομα αποικιών της μύγας της Μεσογείου που εκτρέφονταν σε συνθήκες μαζικής εκτροφής μικρής κλίμακας και εξετάσθηκε η επίδραση του Wolbachia σε βασικές βιολογικές παραμέτρους της αρμοστικότητας των εντόμων καθώς και η προβιοτική δράση του Enterobacter sp.. Τα αποτελέσματα έδειξαν ότι το βακτήριο Wolbachia επηρέασε σημαντικά αρκετές παραμέτρους της αρμοστικότητας (fitness) της μύγας της Μεσογείου σε συνθήκες μαζικής εκτροφής μικρής κλίμακας. Η μείωση των ποσοστών εκκόλαψης των προνυμφών και συνεπώς ο μειωμένος αριθμός ανακτηθέντων ενηλίκων, αποτέλεσε το πιο αξιοσημείωτο των ευρημάτων. Επιπλέον, διαπιστώθηκε μείωση στη διάρκειας ανάπτυξης των ανηλίκων σταδίων του πληθυσμού Vienna 8 GSS ως συνέπεια της μόλυνσης. Η φυλή wCer2 του βακτηρίου δεν επηρέασε ούτε τη διάρκεια ζωής ούτε την ωοπαραγωγή των θηλυκών της μύγας της Μεσογείου, παρότι η φυλή wCer4 φαίνεται να επάγει μείωση τόσο της διάρκειας ζωής όσο και της ωοπαραγωγής των μολυσμένων θηλυκών. Τα αποτελέσματά μας έδειξαν επίσης, ότι η ανταγωνιστικότητα σύζευξης των αρσενικών σε κλωβούς υπαίθρου, η ικανότητα πτήσης των ενηλίκων και η ικανότητα επιβίωσης υπό συνθήκες έλλειψης νερού και τροφής επηρεάζονται σημαντικά από τη φυλή του εντόμου καθώς και από τη φυλή του βακτηρίου (αλληλεπίδραση των δύο γενοτύπων). Τα αποτελέσματα της μελέτης των βακτηρίων του πεπτικού συστήματος συνοψίζονται ως εξής: α) από το πεπτικό σύστημα προνυμφών και ενηλίκων εντόμων (αρσενικών και θηλυκών) απομονώθηκαν τα γένη των βακτηρίων Providencia, Enterobacter, και Acinetobacter, β) η προσθήκη Enterobacter sp., ιδιαίτερα με τη μορφή ζωντανών βακτηρίων στην τροφή των προνυμφών, οδήγησε σε αυξημένο αριθμό νυμφών και ενηλίκων καθώς και σε ταχύτερη ανήλικη ανάπτυξη, κυρίως των αρσενικών, γ) η προσθήκη βακτηρίων στην προνυμφική τροφή δεν επηρέασε το βάρος των νυμφών, την αναλογία φύλου, την ανταγωνιστικότητα σύζευξης των αρσενικών, την ικανότητα πτήσης των ενηλίκων και την ικανότητα επιβίωσης υπό συνθήκες έλλειψης νερού και τροφής. Τα δεδομένα των παραπάνω πειραμάτων συμβάλλουν στην πληρέστερη κατανόηση των βιολογικών μηχανισμών δράσης του συμβιωτικού βακτηρίου Wolbachia και των αλλαγών που επιφέρει η μόλυνση σε βασικές παραμέτρους της βιολογίας και συμπεριφοράς της μύγας της Μεσογείου. Επιπλέον, υπό το πρίσμα των ιδιαίτερων συνθηκών που συνεπάγεται η μαζική εκτροφή των εντόμων, αξιολογήθηκε η επίδραση της βακτηριακής μόλυνσης με Wolbachia και της χορήγησης Enterobacter sp. μέσω της τροφής των προνυμφών σε βασικές βιολογικές παραμέτρους, οι οποίες είναι ενδεικτικές της «ποιότητας» των παραγόμενων εντόμων. Συζητείται η προοπτική συνεισφοράς των συμβιωτικών οργανισμών (Wolbachia, βακτήρια του πεπτικού συστήματος) στην καταπολέμηση της μύγας της Μεσογείου μέσω των προγραμμάτων εξαπόλυσης στειρωμένων εντόμων (Sterile Insect Technique, Incompatible Insect Technique) και προτείνονται δυνατότητες περαιτέρω έρευνας.

The Impact

Ceratitis capitata (medfly) is one of the most devastating crop pests worldwide. The Sterile Insect Technique (SIT) is a control method that is based on the mass rearing of males, their sterilization, and release in the field. However, the effectiveness of the technique depends on the quality of the released males and their fitness. We previously isolated and selected a probiotic bacteria (Enterobacter sp.), from wild-caught medflies, according to criteria that improved biological quality traits of reared medfly males.We firstly evaluated the impact of the irradiation on the expression of different immune and stress genes in the medfly sterile males. Expression was measured at differents time points ranging from 0 to 168 h after irradiation to capture the response of genes with distinct temporal expression patterns. Then, we supplemented the larval diet with previously isolated Enterobacter sp.strain, live and autoclaved at various concentrations to see whether the probiotic treatments affect, through their protective role, the gene expression level, and quality traits. The irradiation had significant effect on the genes attacin, cecropin, PGPR-LC, hsp23, and hsp70 level expression. The expression of attacin and PGPR-LC was up-regulated while that of cecropin was down-regulated. Hsp genes showed decreased levels between 0 and 18 h to peak at 72 h. However, the supplementation of the probiotic strain, either live or autoclaved, was statistically significant only for attacingene. However, significant interaction time x probiotic was noticed for attacin, cecropin, hsp23 and hsp70. The probiotic treatments also improved the quality control parameters like pupal weight. From this work we can conclude that a consortium of parabiotics (autoclaved probiotics) treatment will be recommended in insectaries considering both the beneficial effects on mass reared insects and its general safety for insectary workers and for environment.

Abstract The genetic sexing strain (GSS) of the Mediterranean fruit fly (Ceratitis capitata (Wiedemann)) Vienna 8D53− is based on a male-linked translocation system and uses two selectable markers for male-only production, the white pupae (wp) and the temperature sensitivity lethal (tsl) genes. In this GSS, males emerge from brown pupae and are resistant to high temperatures while females emerge from white pupae, are sensitive to high temperatures. However, double homozygous females (wp tsl/wp tsl) exhibit a slower development rate compared to heterozygous males (wp+tsl+/wp tsl) during the larval stage, which was attributed to the pleiotropic effects of the tsl gene. We present the first evidence that this slower development is due to a different gene, here namely slow development (sd), which is closely linked to the tsl gene. Taking advantage of recombination phenomena between the two loci, we report the isolation of a novel temperature sensitivity lethal strain using the wp mutation as a morphological marker, which showed faster development (wp tsl FD) during the larval stage and increased in its temperature sensitivity compared with the normal tsl strain. Moreover, the introgression of this novel wp tsl FD combined trait into the Vienna 8D53− GSS, resulted in a novel Vienna 8D53− FD GSS, where females showed differences in the thermal sensibility, larval development speed, and productivity profiles. The modification of these traits and their impact on the mass rearing of the GSS for sterile insect technique applications are discussed.
