Abstract BackgroundStress-induced dopaminergic (DAergic) neuronal death in the midbrain region is the primary cause of Parkinson’s disease (PD). Approximately 2% of the global population aged above 65 years is affected with PD. Various factors are responsible for the death of DAergic neurons, among which mitochondrial dysfunction, oxidative stress, misfolded protein aggregation and neuroinflammation are the primary factors. From the discovery of L-dopa, multiple drugs were discovered to improve lifestyle of PD patients, but they failed due to their multiple side effects. Tinospora cordifolia (Tc), a medicinal herb has been used in traditional medicines to treat neurodegenerative diseases. In our previous study, the neuroprotective role of Tc against MPTP-intoxicated Parkinsonian mice was reported. Here, we further explore the neuroprotective molecular mechanisms of Tc in Rotenone (ROT) intoxicated mouse model through proteomics approach.MethodsMice were pretreated with Tc extract by oral administration, followed by ROT-intoxication (2mg/kg body wt. for 35 days, subcutaneous). Rotarod, catalepsy, footprint and pole tests were carried out at 35th day to observe the neuroprotective effects of Tc on motor impairment caused by ROT in PD mice. Protein from nigrostriatal region of the mid brain was isolated, and label free quantification (LFQ) was carried out to identify differentially expressed protein (DEPs) in control vs. PD and PD vs. treatment group. Bioinformatics analysis of DEPs was carried out to explore the molecular pathway, cellular location, molecular function of proteins.ResultsIn this study, we report 800 DEPs in control vs. PD and 133 in PD vs. Treatment group. In silico tools clearly demonstrate significant enrichment of biochemical and molecular pathways with DEPs which are known to be important for PD progression, including, mitochondrial gene expression, hypothetical network for drug addiction, PD pathways, TGF-β signaling, Alzheimer’s disease, Odorant GPCRs and chemokine signaling pathway.ConclusionThis study provides a novel insight for the disease progression in PD mouse. More importantly, it demonstrates that Tc exerts the therapeutic effects through the regulation of multiple pathways to protect DAergic neurons.

Nigrospora oryzae was isolated as an endophytic fungus from the leaves of Coccinia grandis, a popular medicinal plant used to control diabetes. Fermentation of the fungus in potato dextrose broth and chromatographic purification of the ethyl acetate extracts of the broth and mycelium yielded two phenazine secondary metabolites, which were identified as phenazine-1-carboxylic acid (1) and phenazine-1-carboxamide (2) by comparing their spectral data with those reported in the literature. Compound 2, isolated in high yield (1 g/4 L medium), showed strong antifungal activity against the plant pathogen Cladosporium cladosporioides. This is the first report of the isolation of N. oryzae as an endophytic fungus of C. grandis. These phenazines have never been isolated from any fungal source. Antifungal activity of 2 against C. cladosporioides is reported for the first time.

AbstractThe fungal extract as well as the 3 biosynthetic compounds, (S)-(+)-2-cis-4-trans-abscisic acid (1), , 7'-hydroxy-abscisic acid (2) and 4-des-hydroxyl altersolanol A (3) from the endophytic fungus, Nigrospora oryzae, isolated from Combretum dolichopetalum leaf were investigated for their antidiabetic potential.The antidiabetic activity was determined in alloxan-induced diabetic mice by monitoring their fasting blood sugar for 9 h.The extract and the compounds (1–3) significantly (p<0.001) reduced the fasting blood sugar of the diabetic mice.The present study has shown that the biosynthetic products of the endophytic fungus, N. oryzae, exhibited strong antidiabetic activity. It has further shown that endophytic fungi could be an alternative source of novel compounds for management of diabetes.

Two new compounds, named as (2 R,3 S)-pinobanksin-3-cinnamate (1), and 15α-hydroxy-(22 E,24 R)-ergosta-3,5,8(14),22-tetraen-7-one (2), were isolated from the endophytic fungus Penicillium sp. FJ-1 of Acanthus ilicifolius Linn. Their structures were elucidated on the basis of spectroscopic analysis. Additionally, compound 1 exhibited potent neuroprotective effects on corticosterone-damaged PC12 cells, and compound 2 showed potent cytotoxicity on glioma cell lines.

The endophytic microbiota is considered to be one of the consistent and noble souce of potential and unique natural amalgams. These natural amalgams carry diverse pharmaceutical significance which the reason for their importance among research fields. The diversity of plants carries much more diversity of the endophytes as their mutual parts where both are benefited from each other. The current work deals with the isolation of the endophytic fungus from Tinospora cordifolia, for which the leaves were used after the surface sterilization, followed by the production of secondary metabolite by the endophytic isolates through submerged fermentation technique. The produced metabolite was extracted by liquid-liquid extraction technique, which was further used for evaluating its antifungal potential against Candida albicans and the obtained results show their considerable potential. The GC-MS profiling of secondary metabolite was conducted to determine the presence of some bioactive compounds in them, and as a result, some potential compounds detected are Levoglucosenone, Silanediol, Nonane, D-Allose, 5-Hydroxymethylfurfural. Since these compounds are biologically important in various aspects which gives the diversified application to the secondary metabolites. The study concludes the potential of secondary metabolites from endophytic fungus of Tinospora cordifolia and further investigation can be approached on determining the same from other plants, and also evaluating another bioactive potential of secondary metabolites.
