The purpose of this research was to identify the relationship of characteristic and knowledge of farmers to the successful of artificial insemination (AI) in Aceh Besar Districts. The number of respondents is 93 people from twenty three districts in Aceh Besar, selected purposively as the sample of this research. The instruments used in this research are questionnaire and direct observation in the field, while the observed variables are: farmer characteristics, farmer knowledge and artificial insemination result. The results of this research revealed that the characteristics and knowledge of the farmers were good and showed a real relationship to the successfulness of AI in Aceh Besar Districts. The average service per conception (S / C) in Aceh Besar Districts during 2017 was 1.32 while the conception rate (CR) was 86.36%. The results were in accordance with the standard of general guidance of Upsus Siwab Year 2017.

1. The relationship of semen age to conception rate was studied at two artificial-insemination centres and a subcentre of one of these. Data relating to 50,213 first inseminations were included in the analysis.2. It is shown that external conditions can affect the rate of decrease of conception rate with semen age.3. Records giving c.r.'s for each day of semen age may reveal centre differences that cannot be detected in records giving mean c.r.'s only.4. Information may be lost if data are grouped by months rather than recorded by collections.5. The rate of decrease should not be used for comparing centres until the various factors affecting it (e.g. breed differences) are more fully understood. Further investigations should not be limited to one centre.

Melhorar o desempenho reprodutivo é importante para aumentar a lucratividade das propriedades. Assim, nós avaliamos a contagem de folículos antrais (CFA), o escore de condição corporal (ECC), o peso vivo e o comportamento animal sobre à taxa de concepção de vacas Brahman submetidas à inseminação artificial em tempo-fixo (IATF). Vacas (n=122) receberam um protocolo convencional de IATF de três manejos (D0, 8 e 10) a base de estrógeno e progesterona, além do monitoramento da expressão de estro antes da inseminação. O ECC e o comportamento animal foram avaliados em todos os dias do protocolo, o peso foi mensurado no D0 e no diagnóstico de gestação (D40) e a CFA no D0. Para análise, estabeleceu-se os grupos de CFA (baixa, intermediária e alta), do comportamento (calma, inquieta e agitada), do peso (ganhando, mantendo e perdendo) e do ECC (ganhando, mantendo e perdendo). A taxa de concepção geral foi de 50%, e não esteve associada (p>0,05) a classificação da CFA (baixa/52,6%, intermediária/50,9% e alta/45,4%), do peso (ganhando/43,0%, mantendo/54,7% e perdendo/47,1%), do ECC (ganhando/44,0%, mantendo/54,3% e perdendo/37,5%) e do comportamento (calma/43,0%, inquieta/54,7% e agitada/47,1%). No entanto, a baixa CFA apresentou maior (p=0,05) proporção de animais com alta intensidade de estro (94,7%) e foi observado maior escore de reatividade (p=0,001) no primeiro dia de manejo da IATF em relação aos outros dias. Vacas com baixa CFA resultaram em maior proporção de expressão de estro e o primeiro dia de manejo da IATF determinou uma maior reatividade animal em relação aos outros dias do protocolo.   Palavras-chave: Condição Corporal. Inseminação Artificial. Performance Reprodutiva. Peso Vivo.    Abstract Improving reproductive performance is important to increase farm profitability. The  relationship of antral follicle count (AFC), body condition score (BCS), weight, and animal behavior on the conception rate in cows submitted to timed artificial insemination (TAI). Cows (n=122) received a conventional TAI protocol of three managements (D0, 8, and 10) based on estrogen and progesterone, in addition to monitoring estrus expression before insemination. The BCS and behavior score were assessed every day of the TAI protocol. Weight was measured at D0 and in the pregnancy diagnosis (D40), and AFC was determined at D0. For analysis, the groups were established according to AFC (low, intermediate, and high), behavior animal (calm, restless, and agitated), weight variation (gaining, maintaining, and losing), and BCS variation (gaining, maintaining, and losing). The overall conception rate was 50%, and it was not associated with (P>0.05) the AFC classification (low/52.6%, intermediate/50.9%, and high/45.4%), weight variations (gaining/43.0%, maintaining/54.7%, and losing/47.1%), BCS variations (gaining/44.0%, maintaining/54.3%, and losing/37.5%) and animal behavior (calm/43.0%, restless/54.7%, and agitated/47.1%). However, the low AFC showed a higher (P=0.05) proportion of animals with high intensity of estrus expression (94.7%). In addition, a higher score for animal reactivity (P=0.001) was noted on the first day of the management of TAI protocol than to the other days of the hormonal protocol. The low AFC resulted in a higher proportion of cows with high estrus expression and the management practices for TAI determined a greater animals reactivity at the beginning of the breeding program.   Keywords: Body Condition. The aim of the study was to replace the first GnRH in the Ovsynch protocol by selecting cows bearing corpus luteum as well as follicles in a defined stage of development at PGF2α administration. Additionally, various terms of GnRH administration after PGF2α were tested. Seventy five non-pregnant cows bearing corpus luteum were divided into groups according to the phase of follicular development on D 0 (day of PGF2α administration)-growth (GR, follicles 3.0-7.9 mm in diameter), early dominance (ED, dominant follicle 8.0-14.9 mm) and late dominance (LD, dominant follicle 15.0-23.0 mm). In addition, the cows were divided into groups according to the terms of GnRH administration (24, 48 or 72 h after PGF2α). In this way, groups GR 48 (n = 5), GR 72 (n = 6), ED 24 (n = 10), ED 48 (n = 12), ED 72 (n = 12), LD 24 (n = 10), LD 48 (n = 10) and LD 72 (n = 10) were established. Growth of ovulatory follicle, term of ovulation, insemination and conception rates as well as relation of the size of preovulatory follicle (day of ovulation) to the size of following corpus luteum (day 14) were evaluated. The highest level of synchronization of ovulation (100% on D 3) as well as conception rate (50%) was achieved in group ED 48. This protocol gives an opportunity of timing artificial insemination to 18-24 hours after GnRH administration, as ovulation occurs with a high probability within 24 - 48 hours after GnRH administration.

This study aimed to characterize estrus sign/s which best predict the time of ovulation and determine ovarian responses to different  synchronization protocols and conception rates to fixed-time artificial insemination in Boran*Holstein crossbred heifers. In the first part of the study, twenty-seven heifers were observed for estrus signs (from induced and natural) and the relationship of various estrus signs with ovulation time was evaluated by using ultrasonography. In the second part, 91 Boran*Holstein crossbred heifers were randomly grouped into three groups. Group one heifers (n=28) received 100μg gonadotropin-releasing hormone (GnRH) on day zero (D0), 500μg  prostaglandin F2α (PGF2α) on day 7 and 100μg of GnRH on day 9. Group 2 heifers (n=32) were treated as group one but additionally  received progesterone as controlled internal drug release (CIDR). Group three heifers (n=31) were treated as those in group 2 but without injection with GnRH on day 0. In all the 3 groups’ insemination was made at 19h of the second GnRH. The results showed that irrespective of estrus source type (induced or natural), score for standing to be mounted, mounting other heifers, and non-receptive mount by other heifers showed a strong positive correlation with ovulation time (r=0.67, P<0.05). Standing estrus duration, and time elapsed from standing estrus to ovulation were shorter (P<0.05) in induced estrus. The conception rate was 39.3% in synchronized ovulation in the absence of CIDR. The conception rate in timed insemination was 56.3% when ovulation was synchronized by combining CIDR, PGF2α, and GnRH. In conclusion injection of GnRH on day zero together with treatment with CIDR improved ovulation rate, the number of new corpus luteum, and conception rate. Heifers that ovulated within the first 24h after timed insemination had a higher (P<0.05) conception rate than heifers ovulated after 24h of timed insemination. Cycling heifers at day zero had a greater (P<0.05) conception rate than non-cycling.

Effect of one-day delaying progesterone administration at the beginning of 5-day Cosynch protocol was investigated in Holstein heifers for the first artificial insemination (AI). Heifers received a synchronized ovulation and timed AI (TAI) with CIDR inserted on day (d) 0 (CIDR-5; n = 206) or d 1 (CIDR-4; n = 192). In both group, GnRH was administered on d 0 followed by a single PGF2α injection and CIDR removal five days later from GnRH. On d 8, TAI and GnRH administration were concurrently conducted. Heifers detected in estrus up to 24 h prior to TAI were inseminated without GnRH administration. Rates of ovulation, accessory CL formation and new dominant follicle development following initial GnRH injection did not differ between groups. P/AI did not differ between CIDR-4 (44.3%, [85/192]) and CIDR-5 (51.9%, [107/206]) groups, respectively. Pregnancy per AI (P/AI) was significantly (p < 0.01) declined as heifers’ age (12–13, 14, 15, 16 and17–21 months) proceeded in CIDR-4 group (55.6%, 52.1%, 37.9%, 35.7%, 32.4%) compared to those in CIDR-5 group (60.0%, 50.0%, 53.9%, 51.5%, 46.2%) respectively. In conclusion, there is no benefit for delaying CIDR administration in 5-day Cosynch protocol in dairy heifers. However, higher P/AI in CIDR-5 group in older heifers can be considered for reproductive management.
