The biopharmaceutical industry is transitioning from currently deployed batch-mode bioprocessing to a highly efficient and agile next generation bioprocessing with the adaptation of continuous bioprocessing, which reduces the capital investment and operational costs. Continuous bioprocessing, aligned with FDA’s quality-by-design (QbD) platform, is designed to develop robust processes to deliver safe and effective drugs. With the deployment of knowledge based operations, product quality can be built into the process to achieve desired critical quality attributes (CQAs) with reduced variability. To facilitate next generation continuous bio-processing, it is essential to embrace a fundamental shift-in-paradigm from “quality-by-testing” to “quality-by-design”, which requires the deployment of process analytical technologies (PAT). With the adaptation of PAT, a systematic approach of process and product understanding and timely process control are feasible. Deployment of PAT tools for real-time monitoring of CQAs and feedback control is critical for continuous bioprocessing. Given the current deficiency in PAT tools to support continuous bioprocessing, we have integrated Agilent 2D-LC with a post-flow-splitter in conjunction with the SegFlow automated sampler to the bioreactors. With this integrated system, we have established a platform for online measurements of titer and CQAs of monoclonal antibodies (mAbs) as well as amino acid concentrations of bioreactor cell culture.

The emerging new breed of IS (group decision support systems and decision simulation, multimedia, next-generation knowledge-based systems, deductive databases and DBMSs, and such like, together with the evolving networking capabilities) will have a more significant impact on the way people work than have ‘conventional’ IT products. This will have implications for what one might call the ‘cognitive style’ of the user interaction. This paper describes further a view of the organization of organizations that has already been sketched briefly elsewhere (Hutchison & Rosenberg, 1993; Hutchison, 1994), and proposes a strategy for the formal modelling of cooperative group work.

This paper presents a new concept, a ‘human-computer co-operative system’, as the next-generation knowledge-based system for application to project risk management. It first discusses the characteristics of project risks followed by the development of a common expert system for managing such risks. Then, system limitations are identified in terms of knowledge association, and a ‘human–computer co-operative system’ is proposed to overcome these limitations by explicitly incorporating human intuitive ability into the computer system. Finally, evaluations of the human–computer co-operative system are also described.

Disarmament and non-proliferation education is a key tool in curbing the spread of nuclear weapons, with a view to their elimination. This article examines the remarkable story of the International School on Disarmament and Research on Conflicts (ISODARCO) on the occasion of its fiftieth anniversary of continuous engagement in educational and training activities. ISODARCO offers a unique forum where nuclear experts from different backgrounds and approaches can meet, debate, and promote action as a transnational knowledge-based network of experts and, equally important, pass on their expertise to the ‘next generation of non-proliferation specialists’. The contribution of this small Italian NGO is indeed noteworthy, highly praised at the national and especially international level, and worth the attention of an audience broader than just non-proliferation and security experts.
