Extracts from primary leaves of Lee wheat were prepared at various days following inoculation with races of leaf rust and tested for ribonuclease (RNase) activity. As early as 24 hours after inoculation there was a marked increase in the specific activity of the enzyme in extracts of rusted host tissues. A further increase in activity was observed during later stages of infection, with the susceptible and resistant reacting tissue differing only in the degree of their response. Extracts from noninoculated control leaves exhibited a constant RNase activity throughout the period of observation. The germination medium and extracts from germinating uredospores contained comparatively little RNase activity. No direct evidence was obtained either for the possible release of the enzyme from particulate cellular fractions of the host tissue as a result of infection or for the removal of an RNase inhibitor in the host tissue responding to infection.

Maximum germination of flodman thistle [Cirsium flodmanii(Rydb.) Arthur ♯3CIRFL] occurred at alternating temperatures of 10 to 20 and 20 to 30 C. Seeds germinated (20%) in 1.7 M NaCl. Light was important in seed germination; however, its absence could be overcome by adding 8.6 × 10-5M GA3(gibberellic acid) to the germination medium. The greatest seedling emergence occurred when seeds were planted on the soil surface and was lower when seeds were buried at any depth in the soil. Osmotic potentials at −0.5 MPa and greater reduced germination, but 3% of the seeds germinated at −1.3 MPa. Flodman thistle seedlings tolerated average plant water potentials of −0.9 and −1.1 MPa, but they died when the water potential was decreased to −1.5 MPa. Flodman thistle seedlings 43 days old and with four true leaves resprouted after topgrowth removal. Regrowth was evident on 75% of the clipped plants 13 days after initial topgrowth removal. Flodman thistle rosettes spread more rapidly and rosette survival was enhanced more in a weed-free nursery than in a weedy pasture.

Torpedo-stage somatic embryos were selected from actively growing cultures and trasferred to embryo maintenance medium [MS with (per liter) 412.5 mg NH4NO3, 475 mg KNO3, 1 g myo-inositol, 90 g sucrose, 2 g activated charcoal, and 7 g TC agar] supplemented with either 0%, 2.5%, 5%, 7.5%, or 10% polyethylene glycol (PEG) for 4, 8, or 12 weeks. Embryos placed on treatment media were transferred directly to grape somatic embryo germination medium [MS with (per liter) 1 g myo-inositol, 30 g sucrose, 1 M benzyladenine, and 7 g TC agar] once their PEG treatment was terminated. The number of embryos that germinated was recorded 4 weeks after transfer to somatic embryo germination medium. The number of germinated embryos that differentiated into plants was recorded at 8 weeks. There was no difference in germination rates and embryo differentiation among embryos incubated on medium with or without PEG for 4 weeks. A difference in embryo growth rate was observed after 8 weeks on medium with PEG. Embryo grew fastest on media containing 5% or 7.5% PEG. In addition, embryos grown on medium with 5% or 7.5% PEG were morphologically similar to zygotic embryos.

Physic nut (Jatropha curcas Linn.) has the potential as a source of sustainable biofuels. Somatic embryo proliferation of J. curcas may cause somaclonal variations. This research aimed to investigate somaclonal variations of J. curcas somatic embryo derived-plantlet using ISSR markers. Somatic embryos of J. curcas at the globular phase were cultured on liquid MS medium supplemented with 0, 0.5, 1.0, 1.5, and 2.0 mg L-1 of 2,4-D. Parameter observed were embryos weight, embryos volume, colour, and size of embryos. After proliferation, the embryos were cultured on a germination medium until the cotyledonary phase. The results showed that proliferation of J. curcas somatic embryos was optimal, with the highest weight and volume,  at MS medium added with 1 mg L-1 2,4-D.

Abstract The content of IAA in stem rust-infected susceptible wheat leaves shows a highly pronounced maximum 5-6 days after inoculation, shortly prior to the onset of sporulation. This auxin increase can not only be caused by a reduced degradation of IAA. Considerable amounts of IAA are also found in urediospores and germlings; the IAA is in part released by them into the germination medium. IAA applied exogenously to wheat leaves is channelled into two different degradation path­ways: (a) into the peroxidase-catalysed decarboxylation which leads to indole-3-methanol and subsequent products as well as into (b) a non-decarboxylative path which leads to a number of oxindolic compounds. Furthermore, IAA conjugates such as IAAglc and IAAsp are formed. The formation of the products is characteristically dependent upon the concentration of the IAA applied. In rust fungus-infected wheat leaves, all IAA metabolites occur which are known in healthy leaves. The mode of their formation after “feeding” of radioactively-labelled IAA leads to the conclusion that the main part of the IAA in the infected leaves is present in a pool which does not permit a rapid exchange with the IAA taken up. The results lead to the hypothesis that IAA is present, to a major extent, in the structures of the fungus and is probably also produced by it.

AbstractPollen germination is crucial for the process of plant development which strongly depends on the presence of carbohydrates as a primary source of energy. In this study, we analyzed the differential effects of four sugars with varying concentrations on the pollen germination of Vinca rosea Linn. and Cucumis melo var. utilissimus Duth. & Fuller. using Brewbaker and Kwack’s medium as germination medium and hanging drop method after an incubation period of 1h. Sucrose and glucose supported the pollen germination but galactose and fructose did not support and even considerably inhibited the pollen germination of Vinca rosea. Whereas in pollen germination of Cucumis melo var. utilissimus, all the four sugars supported the pollen germination. The study suggests that 15% sucrose, for Vinca rosea, and 12% galactose, for Cucumis melo var. utilissimus, supports in achieving the highest pollen germination percentage when added to the pollen germination medium.

A laboratory experiment was conducted on germination papers to study the effect of fluoride (F) at 0 (T1), 50 (T2), 100 (T3), 200 (T4), 250 (T5) and 300 (T6) ppm on germination and morphological parameters in wheat (Triticum aestivum L.) variety, HUW-234 at 2, 4 and 6 days after initiation of germination process. Fluoride toxicity caused reduction in germination per cent, germination index, coefficient of velocity of germination and germination energy (%) while mean germination time increased with fluoride concentration. Root and shoot lengths and dry matters decreased with increased concentrations of fluoride. Ratio of root: shoot weight increased with increased concentration of fluoride. Elongation of seminal roots was adversely affected by increased fluoride level. Increased fluoride level in the germination medium decreased RGR of seedlings progressively. Present study revealed that enhanced fluoride concentration in germination medium caused reduction in germination and germination related parameters.
