AbstractThis study aimed to investigate the effects of active dry yeast (ADY) on rumen microbial composition and slaughter performance of beef cattle. Thirty-two finishing beef cattle (simmental crossbred cattle ♂ × cattle-yaks ♀), with an average body weight of 110 ± 12.85 kg, were randomly assigned to one of four treatments: the low plane of nutrition group (Control), low plane of nutrition group + ADY 2 g/head/d (ADY2), low plane of nutrition group + ADY 4 g/head/d (ADY4) and high plane of nutrition group (HPN). ADY supplementation increased average daily gain (P<0.001), and the carcass weight of ADY4 group had no significant difference with HPN group (P>0.05). The serum glutamic-pyruvic transaminase activity in control and ADY4 group was higher than HPN group (P=0.001). The neutral detergent fiber (P=0.022) and acid detergent fiber (P=0.043) digestibility in HPN group was greater than control, but no difference was obtained among ADY2, ADY4 and HPN group (P>0.05). The rumen ammonium nitrogen content in control was greater than ADY2 and ADY4 group (P=0.003), and no difference was obtained ADY2, ADY4 and HPN group (P>0.05). The propionic acid content in the rumen in ADY2, ADY4, and HPN group were greater than control group (P<0.001). The simpson (P=0.014) and shannon (P=0.045) indexes in control and HPN group were greater than ADY4 group. At the phylum level, the relative abundance of Firmicutes in the HPN group was higher than ADY4 group (P=0.015). At the genus level, HPN and ADY4 were clustered together, and the relative abundance of Ruminococcaceae UCG-002 in ADY4 group was higher than control and HPN group (P=0.004). In conclusion, supplementation ADY 4 g/head/d shift the rumen microbial composition of beef cattle fed low plane of nutrition to a more similar level with cattle fed with HPN diet, produced comparable carcass weight with HPN diet.

Abstract BackgroundChalazion is a chronic inflammatory granuloma of the meibomian gland formed on the basis of the obstruction of the meibomian gland drainage duct and the retention of secretions. It is one of the most common clinically eye diseases in children. Chronic inflammation of the meibomian glands is responsible for this disease, and the gut flora is thought to be involved in the inflammatory process. In this study, we investigated the relationship between intestinal microbial composition and children's chalazion.MethodsFecal samples were collected from 21 children with chalazion and 26 healthy children. DNA was extracted from fecal stool samples and 16S rRNA sequences in the gut flora were detected by using second second-generation sequencing technology. The results were used to compare the composition of the microbiome between patients and healthy controls.ResultsAccording to Alpha Diversity and Beta Diversity analysis, we found that there was no significant difference in bacteria diversity and relative abundance between the two groups. We compared the flora of the control group and the diseased group through Lefse analysis, and screened out 11 different species. Based on the absolute abundance of species, 43 different species were selected. Anosim analysis and metastats analysis were used to compare the flora of the control group and the diseased group again. At the species level, we found that gut_metagenome and human_gut_metagenome are the common differences in species levels obtained from the above analysis. Finally, corrplot correlation analysis was performed, suggesting that gut_metagenome has a great correlation with the number, ulceration, and recurrence of chalazion in children.ConclusionsThere was no significant difference in the diversity index and relative abundance of flora in children with chalazion compared with healthy children, but there were significant differences in some bacterial species. The gut_metagenome strains identified in this study were significantly related to the growth, ulceration, and relapse of children with chalazion. It is suggested that gut_metagenome may be a microbiological indicator which is independent of clinicopathologic factors but associated with chalazion disease .* These authors have contributed equally to this work.

Interaction between the dietary fiber and the gut microbes can regulate host bile acid metabolism. This study sought to explore the effects of guar gum combined with pregelatinized waxy maize starch (GCW) in a gestation diet on reproductive performance, gut microbiota composition, and bile acid homeostasis of sows. A total of 61 large white sows were randomly grouped into the control (n = 33) and 2% GCW (n = 28) groups during gestation. GCW diet increased birth-weight of piglets, and decreased the percentage of intrauterine growth restriction (IUGR) piglets. In addition, dietary GCW reduced gut microbial diversity and modulated gut microbial composition in sows on day 109 of gestation. The relative abundance of bile salt hydrolase (BSH) gene-encoding bacteria, Lactobacillus and Bacteroides decreased after GCW administration, whereas no significant difference was observed in the fecal level of total glycine-conjugated and taurine-conjugated bile acids between the two groups. Dietary GCW increased the relative abundance of Ruminococcaceae (one of few taxa comprising 7α-dehydroxylating bacteria), which was associated with elevated fecal deoxycholic acid (DCA) in the GCW group. GCW administration lowered the concentrations of plasma total bile acid (TBA) and 7α-hydroxy-4-cholesten-3-one (C4) (reflecting lower hepatic bile acid synthesis) at day 90 and day 109 of gestation compared with the control diet. Furthermore, the levels of plasma glycoursodeoxycholic acid (GUDCA), tauroursodeoxycholic acid (TUDCA) and glycohyocholic acid (GHCA) were lower in the GCW group compared with the control group. Spearman correlation analysis showed alterations in the composition of the gut microbiota by GCW treatment was associated with improved bile acid homeostasis and reproductive performance of sows. In conclusion, GCW-induced improves bile acid homeostasis during gestation which may enhance reproductive performance of sows.

The objective of this study was to evaluate the effect of Hibiscus tiliaceus flower extracts supplementation in concentrate on performance, nutrient digestibility and blood metabolites of Bali cattle. Sixty of male Bali cattle averaging 225 ± 09.15 kg initial body weight were used in this study. They were divided into two groups. The first group (as control) was cattle fed ammoniated rice straw and concentrate (R1) and the second group was fed as the same as the control group, but concentrate was supplemented with H. tiliaceus flower extract at 200 ppm/kg of dry matter (R2). Variables measured were dry matter digestibility (DMD), organic matter digestibility (OMD), neutral detergent fiber digestibility (NDFD), and acid detergent fiber (ADFD) using total collection method. Other variables were blood glucose and urea, average daily gain (ADG), feed conversion (FC) dan feed efficiency (FE). T-test showed that DMD, OMD, ADFD, and NDFD in the second group (R2) were higher (P<0.05) than control (R1). However, there was no significant difference (P>0.05) between performance (ADG, FC, FE) and blood parameters of urea and glucose between the groups fed diets supplemented with H. tiliaceus flower extracts and control group. There was an increase in ADG and FE on R2 (0.63 ± 0.23 kg and FE 5.54 ± 1.89) compared to R1 (0.54 ± 0.32 kg and 0.54 ± 0.32). The study concluded that supplementation of Hibiscus tiliaceus flower extracts as feed additive in the concentrate diet can increase nutrient digestibility and tend to improve the performance of Bali cattle.

Abstract Background Dietary fibers are widely considered to be beneficial to health as they produce nutrients through gut microbial fermentation while facilitating weight management and boosting gut health. To date, the gene expression profiles of the carbohydrate active enzymes (CAZymes) that respond to different types of fibers (raw potato starch, RPS; inulin, INU; pectin, PEC) in the gut microbes of pigs are not well understood. Therefore, we investigated the functional response of colonic microbiota to different dietary fibers in pigs through metatranscriptomic analysis. Results The results showed that the microbial composition and CAZyme structure of the three experimental groups changed significantly compared with the control group (CON). Based on a comparative analysis with the control diet, RPS increased the abundance of Parabacteroides, Ruminococcus, Faecalibacterium and Alloprevotella but decreased Sutterella; INU increased the relative abundance of Fusobacterium and Rhodococcus but decreased Bacillus; and PEC increased the relative abundance of the Streptococcus and Bacteroidetes groups but decreased Clostridium, Clostridioides, Intestinibacter, Gemmiger, Muribaculum and Vibrio. The gene expression of CAZymes GH8, GH14, GH24, GH38, GT14, GT31, GT77 and GT91 downregulated but that of GH77, GH97, GT3, GT10 and GT27 upregulated in the RPS diet group; the gene expression of AA4, AA7, GH14, GH15, GH24, GH26, GH27, GH38, GH101, GT26, GT27 and GT38 downregulated in the INU group; and the gene expression of PL4, AA1, GT32, GH18, GH37, GH101 and GH112 downregulated but that of CE14, AA3, AA12, GH5, GH102 and GH103 upregulated in the PEC group. Compared with the RPS and INU groups, the composition of colonic microbiota in the PEC group exhibited more diverse changes with the variation of CAZymes and Streptococcus as the main contributor to CBM61, which greatly promoted the digestion of pectin. Conclusion The results of this exploratory study provided a comprehensive overview of the effects of different fibers on nutrient digestibility, gut microbiota and CAZymes in pig colon, which will furnish new insights into the impacts of the use of dietary fibers on animal and human health.

ABSTRACT The objective of this work was to evaluate the effect of increasing levels of guava agro-industrial waste (GAW) on the ingestive behavior of Santa Inês lambs. Forty non-castrated sheep of the Santa Inês breed were used, at an initial weight of 21.33 ± 2.62 kg, and at the age of 120 days, distributed in a completely randomized design, with five treatments and fed with increasing GAW levels (0.0, 7.5, 15.0, 22.5 and 30.0%). The average daily gain (ADG) showed orthogonal contrast (P <0.05), in which the animals that ingested the control diet had lower performances. The animals with GAW added to the diet obtained more significant weight gains reflected by a better feed conversion. Among the variables, idleness, feeding, and total chewing time had a significant effect (P<0.05); animals spent more time idle at the 30.0% inclusion level and less time in the control group. However, the means of the 7.5, 15.0, and 22.5% GAW inclusion levels were statistically similar to the control group and the 30.0% level. There was a significant effect (P<0.05) on feed efficiency, with the lowest values being presented for the control group and the highest values at the 30% inclusion level of GAW. The 30.0% GAW feed for lambs in confinement reduces feed time, and total chewing prolongs idleness and increases feed efficiency without compromising dry matter intake, neutral detergent fiber intake, and ADG.

Abstract Background: Studies have reported a high prevalence of periodontal disease in individuals with intellectual disability (ID). The aim of this study was to compare the relative abundance of periodontal pathogens (red, orange, yellow, purple, and green complexes) between individuals with ID and healthy controls.Methods: Of the 31 subjects enrolled in this study, 16 with severe ID were selected from the outpatient clinic of the Special Care Dentistry of Hiroshima University Hospital, and 14 healthy subjects were selected from the outpatient clinic of another department at the same hospital. Dental plaque was sampled after oral examination. Decayed, missing, and filled permanent teeth (DMFT) counts were obtained and periodontal measurements were taken using the papillary-marginal-attached (PMA) index, plaque index (PI), gingival index (GI), and probing depth (PD). Gene sequencing (16S rRNA) was performed for each sample using next-generation high-throughput sequencing methods. The relative abundance of the periodontal pathogens and the clinical parameters were compared. An unpaired t-test was used to compare the oral health status in both groups and analysis of covariance was performed to compare the relative abundance of each pathogen.Results: No statistically significant difference in DMFT was observed between the two groups. However, significant differences in the median PMA index, PI, and GI were noted between the groups (P < 0.0001). In addition, the mean PD in the ID group was significantly higher than that in the control group (P < 0.0001). The relative abundances of Tannerella spp. and Treponema spp. were significantly higher in the ID group when compared to the control group at the genus level (P = 0.0383 and 0.0432, respectively); alternatively, the relative abundance of Porphyromonas spp. was significantly lower in the ID group (P < 0.0001).Conclusions: The PMA index, PI, GI, and PD were significantly lower in the ID group than in the control group. On the other hand, no significant difference in DMFT was observed between the two groups. Furthermore, our findings indicate that Tannerella forsythia might be more closely associated with periodontal disease than Porphyromonas gingivalis in individuals with ID.

Abstract Heat stress continues to be a challenge for feedlot producers, forcing physiological adaptations in beef cattle that can have a negative impact on gain and carcass quality. Feedlots may want to incorporate systems that aid in mitigating heat stress, including sprinkler systems, which are commonly found on dairies. The objective of this study was to determine the impact of sprinkler systems on the body temperature of growing feedlot steers applied at three different temperature thresholds. Thirty-two Angus-cross beef steers (298 ± 15 SD days of age) were randomly assigned to 8 pens. Treatments were assigned to pens according to location to avoid drainage issues, with one replicate located on the eastern side of the feedlot and the other replicate located on the western side. Treatments including no sprinklers (C), high temperature threshold sprinklers (HT), moderate temperature threshold sprinklers (MT) or low temperature threshold sprinklers (LT). Sprinklers (flow rate: 5.11 L/min) were activated at 33°C (HT), 28°C (MT), and 23°C (LT) for five minutes at corresponding temperatures every thirty minutes for 57 days. Rectal temperature of steers in their pens was measured in the morning (0800 h), afternoon (1500 h), and evening (1900 h) three days each week (Monday, Wednesday, Friday) for a total of 19 experimental days. Weather measurements, including ambient temperature, windspeed, humidity and solar radiation were recorded at each measurement time along with the maximum values for each day. The change in body temperature (ΔBT) between the morning and afternoon was affected by ambient temperature (P &lt; 0.01); MT (P = 0.02) and LT (P = 0.02) – different than C. There was no significant difference between sprinkler treatments and the control group (P &gt; 0.05). Day affected (ΔBT) between the morning and afternoon (P &lt; 0.05) and the afternoon and evening (P &lt; 0.05). When averaged by day, the control group was significantly higher than MT (P = 0.04) between the morning and afternoon. The control group was also significantly higher than LT between the morning and afternoon (P = 0.03) and the afternoon and evening (P &lt; 0.01). The change in steer body temperature between afternoon and morning was affected by ambient temperature, and averaged across days, lowering the temperature threshold for sprinkling decreased in the afternoon and evening body temperature increase in steers.

A study was conducted to evaluate the effectiveness of Synovex® H as an agent for prenatal androgenization in beef cattle. Thirty Charolais-cross heifers, exposed to a fertile bull during a 55-d spring breeding season, were randomly assigned to a control group or implanted with three times the normal dose of Synovex® H between days 20 and 75 of gestation. Serum concentrations of testosterone, estradiol, and progesterone, calving rate, and growth performance were determined in the pregnant heifers. Testosterone levels were greater (P < 0.001) in treated heifers than in control heifers. Synovex® H treatment had no affect on estradiol levels. Progesterone levels were lowered (P < 0.05) in treated heifers. Synovex® H treatment decreased (P < 0.05) calving rate and birth weight of calves born to treated heifers. Calving ease scores were not affected by Synovex® H treatment. Weight was not affected; however, average daily gain was improved 7% by Synovex® H treatment (0.88 vs. 0.82 ± 0.04 kg d−1, treated and control heifers, respectively). These results are interpreted to suggest that Synovex® H is not an appropriate androgenizing agent for cattle due to adverse effects on progesterone concentrations during pregnancy and on calving rate. <p>This study aimed to evaluate the effect of replacing soybean meal with soybean in multiple supplements on nutritional parameters, microbial efficiency and productive and reproductive performance of heifers grazing in <em>Urochloa decumbens </em>during the drought period. Were used 39 crossbred heifers of initial age and initial weight of 21 months and 309.5±7 kg, respectively. The experimental design was completely randomized with four treatments with eight replicates, and a control treatment with seven replications. Two treatments had soybean meal as the protein source and two treatments had soybean as the protein source, containing 25% and 40% crude protein. The amount of supplement offered was 1.0 kg/animal/day. The animals of the control group received only mineral salt <em>ad libitum</em>. The supplemented animals had higher average daily gain (ADG) than control animals (P&lt;0.10), and there was no difference in ADG among the supplements (P&gt;0.10). There was an effect of supplementation (P &lt;0.10) on intake of dry matter (DM), organic matter (OM), crude protein (CP), ether extract (EE), non-fiber carbohydrates (NFC), total digestible nutrients (TDN) and neutral detergent fiber corrected for ash and protein (NDF). There were no differences (P&gt;0.10) on intake of OM and DM grazing between the supplemented and non-supplemented. Supplementation improved DM digestibility and all constituents of the diet (P&lt;0.10). It was found that the provision of multiple supplements optimizes the performance of heifers grazing during the dry season, and that the substitution of soybean meal by soybean did not improve productive performance of animals.</p>

In order to investigate the effect of gentle handling on the behavior, performance, and meat quality of pigs from weaning to slaughter, 144 6-week-old weaned hybrid Min piglets (a native breed) were selected and divided into a handling group (HG: 9 pens × 8 pigs) and a control group (CG: 9 pens × 8 pigs). After 6 weeks of handling, we observed and then evaluated the pigs’ responses to a handler with behavioral scores. Moreover, we measured heart rate and production performance. Three pigs were randomly selected from each of the 18 pens and divided into a handling group (HG: n = 27) and a control group (CG: n = 27), and the HG pigs were given gentle handling until slaughter. Subsequently, we evaluated meat quality and the production performance of six pigs from each group. The results show that AA test (approaching–avoidance test) scores in HG pigs, the number of contacts with the handler and absence of contact with the handler, were significantly higher than in the CG pigs (p < 0.01). The occurrences of avoidance and looking at the handler were lower in the HG than in the CG group (p < 0.01); however, heart rate was not found to be significantly different between the two groups (p = 0.63). No significant difference was found in the average daily gain, average daily feed intake, and feed conversion ratio between the two groups during the two periods (p > 0.05). The b* value was determined 45 min after slaughter, and it was significantly lower in the HG than that in the CG group (p = 0.002). Furthermore, 2 h after slaughter, the L value of the HG group was significantly higher than that of the CG group (p = 0.047), but no difference was observed in carcass quality or other meat quality indicators between the two groups (p > 0.05). The results indicate that gentle handling could reduce pigs’ anxiety and increase their willingness to approach the handler, increasing the intimacy of the pigs and handlers. However, long-term gentle handling had little effect on pig performance, carcass quality, and meat quality.
