AbstractAutism Spectrum Disorder results in deficit in social interaction, non-verbal communication and social reciprocity. Cognitive tasks pertaining to emotion processing are often preferred to distinguish the ASD children from the typically developing ones. We analysed the role of face and emotion processing in ASD and explored the feasibility of using EEG as a neural marker for detecting ASD. Subjects performed a visual perceptual task with face and nonface stimuli. Successful ASD detection was possible as early as 50 ms. post stimulus onset. Alpha and Beta oscillations seem to best identify autistic individuals. Multivariate pattern analysis and source localization studies points to the role of early visual processing and attention rather than emotion and face processing in detecting autism.

Neuroimaging and EEG studies have shown that passive observation of the full body and of specific body parts is associated with 1) activity of an occipito-temporal region named the extrastriate body area (EBA), 2) amplitude modulations of a specific posterior event-related potential (ERP) component (N1/N190), and 3) a theta-band (4–7 Hz) synchronization recorded from occipito-temporal electrodes compatible with the location of EBA. To characterize the functional role of the occipito-temporal theta-band increase during the processing of body-part stimuli, we recorded EEG from healthy participants while they were engaged in an identification task (match-to-sample) of images of hands and nonbody control images (leaves). In addition to confirming that occipito-temporal electrodes show a larger N1 for hand images compared with control stimuli, cluster-based analysis revealed an occipito-temporal cluster showing an increased theta power when hands are presented (compared with leaves) and show that this theta increase is higher for identified hands compared with nonidentified ones while not being significantly different between not identified nonhand stimuli. Finally, single trial multivariate pattern analysis revealed that time-frequency modulation in the theta band is a better marker for classifying the identification of hand images than the ERP modulation. The present results support the notion that theta activity over the occipito-temporal cortex is an informative marker of hand visual processing and may reflect the activity of a network coding for stimulus identity. NEW & NOTEWORTHY Hands provide crucial information regarding the identity of others, which is a key information for social processes. We recorded EEG activity of healthy participants during the visual identification of hand images. The combination of univariate and multivariate pattern analysis in time- and time-frequency domain highlights the functional role of theta (4–7 Hz) activity over visual areas during hand identification and emphasizes the robustness of this neuromarker in occipito-temporal visual processing dynamics.

Purpose Play is a critical aspect of children's development, and researchers have long argued that symbolic deficits in play may be diagnostic of developmental disabilities. This study examined whether deficits in play emerge as a function of developmental disabilities and whether our perceptions of play are colored by differences in language and behavioral presentations. Method Ninety-three children participated in this study (typically developing [TD]; n = 23, developmental language disorders [DLD]; n = 24, attention-deficit/hyperactivity disorder [ADHD]; n = 26, and autism spectrum disorder [ASD]; n = 20). Children were videotaped engaging in free-play. Children's symbolic play (imagination, organization, elaboration, and comfort) was scored under conditions of both audible language and no audible language to assess diagnostic group differences in play and whether audible language impacted raters' perception of play. Results Significant differences in play were evident across diagnostic groups. The presence of language did not alter play ratings for the TD group, but differences were found among the other diagnostic groups. When language was audible, children with DLD and ASD (but not ADHD) were scored poorly on play compared to their TD peers. When language was not audible, children with DLD were perceived to play better than when language was audible. Conversely, children with ADHD showed organizational deficits when language was not available to support their play. Finally, children with ASD demonstrated poor play performance regardless of whether language was audible or not. Conclusions Language affects our understanding of play skills in some young children. Parents, researchers, and clinicians must be careful not to underestimate or overestimate play based on language presentation. Differential skills in language have the potential to unduly influence our perceptions of play for children with developmental disabilities.

ABSTRACTBackground: Gut microflora influences neural development through complex mechanisms. Feeding practices, especially breastfeeding influence gut microbiome and thereby play a pivotal role in immune and neural development. Current understandings of the role of healthy distal gut microflora in the development of immune and neural systems provide insights into immunological mechanisms as one of the possible etiologies in autism spectrum disorder (ASD). Studies have shown that optimal breastfeeding is associated with lower odds of being at-risk for ASD and children with ASD are suboptimally breastfed. Methods: The feeding practices of children with ASD (n = 30) was compared to their typically developing siblings as matched controls (n = 30). Information regarding feeding practices was collected from mothers through a semi-structured questionnaire. Results: About 43.3% of children with ASD received exclusive breastfeeding, whereas 76.7% of their typically developing siblings were exclusively breastfed. Exclusive breastfeeding was associated with lower odds for ASD (odds ratio [OR] = 0.166; 95% confidence interval [CI] = 0.025–0.65), while early introduction of top feeds was associated with higher odds (OR = 6; 95% CI = 1.33–55.19). Difficulties in breastfeeding were attributed to child-related factors in 13.2% of the children. Conclusion: Children with ASD are suboptimally breastfed compared to their typically developing siblings. Exclusive breastfeeding may confer protection in vulnerable children. Further studies on larger prospective sample are required to establish the association.

AbstractValue-based decision-making is ubiquitous in every-day life, and critically depends on the contingency between choices and their outcomes. Only if outcomes are contingent on our choices can we make meaningful value-based decisions. Here, we investigate the effect of outcome contingency on the neural coding of rewards and tasks. Participants performed a reversal-learning paradigm in which reward outcomes were contingent on trial-by-trial choices, and performed a ‘free choice’ paradigm in which rewards were random and not contingent on choices. We hypothesized that contingent outcomes enhance the neural coding of rewards and tasks, which was tested using multivariate pattern analysis of fMRI data. Reward outcomes were encoded in a large network including the striatum, dmPFC and parietal cortex, and these representations were indeed amplified for contingent rewards. Tasks were encoded in the dmPFC at the time of decision-making, and in parietal cortex in a subsequent maintenance phase. We found no evidence for contingency-dependent modulations of task signals, demonstrating highly similar coding across contingency conditions. Our findings suggest selective effects of contingency on reward coding only, and further highlight the role of dmPFC and parietal cortex in value-based decision-making, as these were the only regions strongly involved in both reward and task coding.

Viewpoint-invariant face recognition is thought to be subserved by a distributed network of occipitotemporal face-selective areas that, except for the human anterior temporal lobe, have been shown to also contain face-orientation information. This review begins by highlighting the importance of bilateral symmetry for viewpoint-invariant recognition and face-orientation perception. Then, monkey electrophysiological evidence is surveyed describing key tuning properties of face-selective neurons—including neurons bimodally tuned to mirror-symmetric face-views—followed by studies combining functional magnetic resonance imaging (fMRI) and multivariate pattern analyses to probe the representation of face-orientation and identity information in humans. Altogether, neuroimaging studies suggest that face-identity is gradually disentangled from face-orientation information along the ventral visual processing stream. The evidence seems to diverge, however, regarding the prevalent form of tuning of neural populations in human face-selective areas. In this context, caveats possibly leading to erroneous inferences regarding mirror-symmetric coding are exposed, including the need to distinguish angular from Euclidean distances when interpreting multivariate pattern analyses. On this basis, this review argues that evidence from the fusiform face area is best explained by a view-sensitive code reflecting head angular disparity, consistent with a role of this area in face-orientation perception. Finally, the importance is stressed of explicit models relating neural properties to large-scale signals.

To make adaptive decisions in a social context, humans must identify relevant agents in the environment, infer their underlying strategies and motivations, and predict their upcoming actions. We used functional magnetic resonance imaging, in conjunction with combinatorial multivariate pattern analysis, to predict human participants’ subsequent decisions in an incentive-compatible poker game. We found that signals from the temporal-parietal junction provided unique information about the nature of the upcoming decision, and that information was specific to decisions against agents who were both social and relevant for future behavior.

The fusiform face area (FFA) is considered to be a highly specialized brain module because of its central importance for face perception. However, many researchers claim that the FFA is a general visual expertise module that distinguishes between individual examples within a single category. Here, I circumvent the shortcomings of some previous studies on the FFA controversy by using chess stimuli, which do not visually resemble faces, together with more sensitive methods of analysis such as multivariate pattern analysis. I also extend the previous research by presenting chess positions, complex scenes with multiple objects, and their interrelations to chess experts and novices as well as isolated chess objects. The first experiment demonstrates that chess expertise modulated the FFA activation when chess positions were presented. In contrast, single chess objects did not produce different activation patterns among experts and novices even when the multivariate pattern analysis was used. The second experiment focused on the single chess objects and featured an explicit task of identifying the chess objects but failed to demonstrate expertise effects in the FFA. The experiments provide support for the general expertise view of the FFA function but also extend the scope of our understanding about the function of the FFA. The FFA does not merely distinguish between different exemplars within the same category of stimuli. More likely, it parses complex multiobject stimuli that contain numerous functional and spatial relations.

AbstractFunctional brain connectivity is altered in children and adults with autism spectrum disorder (ASD). Mapping pre-symptomatic functional disruptions in ASD could identify infants based on neural risk, providing a crucial opportunity to mediate outcomes before behavioral symptoms emerge.Here we quantify functional connectivity using scalable EEG measures of oscillatory phase coherence (6-12Hz). Infants at high and low familial risk for ASD (N=65) underwent an EEG recording at 3 months of age and were assessed for ASD symptoms at 18 months using the Autism Diagnostic Observation Schedule-Toddler Module. Multivariate pattern analysis was used to examine early functional patterns that are associated with later ASD symptoms.Support vector regression (SVR) algorithms accurately predicted observed ASD symptoms at 18 months from EEG data at 3 months (r=0.76, p=0.02). Specifically, lower frontal connectivity and higher right temporo-parietal connectivity predicted higher ASD symptoms. The SVR model did not predict non-verbal cognitive abilities at 18 months (r=0.15, p=0.36), suggesting specificity of these brain alterations to ASD.These data suggest that frontal and temporo-parietal dysconnectivity play important roles in the early pathophysiology of ASD. Early functional differences in ASD can be captured using EEG during infancy and may inform much-needed advancements in the early detection of ASD.

This study examined the natural social interest of 193 (95 boys, 98 girls) typically developing preschoolers aged 41 to 77 months ( M = 61.71 years, SD = 8.48 years) toward their included peers with autism spectrum disorder (ASD; N = 16, M = 66.8, SD = 8.80). A hierarchical linear model examined the role of endogenous (within-child) and exogenous (environmental) variables in explaining typically developing children’s natural social behaviors. Results indicated the significant contribution of three variables: (a) typically developing children’s prosocial behaviors, (b) teachers’ attitudes about their training regarding inclusion, and (c) areas of inclusion support needed by the specific child with ASD. Discussion highlighted possible interactions among these variables within the social–ecological theoretical framework, for explaining peer relations of typically developing children with peers with ASD in regular preschools.

Phelan–McDermid syndrome (PMS) is one of the most common genetic forms of autism spectrum disorder (ASD). While sensory reactivity symptoms are widely reported in idiopathic ASD (iASD), few studies have examined sensory symptoms in PMS. The current study delineates the sensory reactivity phenotype and examines genotype–phenotype interactions in a large sample of children with PMS. Sensory reactivity was measured in a group of 52 children with PMS, 132 children with iASD, and 54 typically developing (TD) children using the Sensory Assessment for Neurodevelopmental Disorders (SAND). The SAND is a clinician-administered observation and corresponding caregiver interview that captures sensory symptoms based on the DSM-5 criteria for ASD. Children with PMS demonstrated significantly greater hyporeactivity symptoms and fewer hyperreactivity and seeking symptoms compared to children with iASD and TD controls. There were no differences between those with Class I deletions or sequence variants and those with larger Class II deletions, suggesting that haploinsufficiency of SHANK3 is the main driver of the sensory phenotype seen in PMS. The syndrome-specific sensory phenotype identified in this study is distinct from other monogenic forms of ASD and offers insight into the potential role of SHANK3 deficiency in sensory reactivity. Understanding sensory reactivity abnormalities in PMS, in the context of known glutamatergic dysregulation, may inform future clinical trials in the syndrome.
