AbstractBackgroundBovine milk provides an important source of nutrition in much of the Western world, forming components of many food products. Over many years, artificial selection has substantially improved milk production by cows. However, the genes underlying milk production quantitative trait loci (QTL) remain relatively poorly characterised. Here, we investigate a previously-reported QTL located at theCSF2RBlocus, for several milk production phenotypes, to better understand its underlying genetic and molecular causes.ResultsUsing a population of 29,350 taurine dairy cattle, we conducted association analyses for milk yield and composition traits, and identified highly significant QTL for milk yield, milk fat concentration, and milk protein concentration. Strikingly, protein concentration and milk yield appear to show co-located yet genetically distinct QTL. To attempt to understand the molecular mechanisms that might be mediating these effects, gene expression data were used to investigate eQTL for eleven genes in the broader interval. This analysis highlighted genetic impacts onCSF2RBandNCF4expression that share similar association signatures to those observed for lactation QTL, strongly implicating one or both of these genes as the cause of these effects. Using the same gene expression dataset representing 357 lactating cows, we also identified 38 novel RNA editing sites in the 3′UTR ofCSF2RBtranscripts. The extent to which two of these sites were edited also appears to be genetically co-regulated with lactation QTL, highlighting a further layer of regulatory complexity implicating theCSF2RBgene.ConclusionsThis chromosome 5 locus presents a diversity of molecular and lactation QTL, likely representing multiple overlapping effects that, at a minimum, highlight theCSF2RBgene as having a causal role in these processes.

The effectiveness of the fiber in whole cottonseed (WCS), wheat midds (WM), and rice bran was evaluated in Holstein cows in early lactation (30 to 50 days) fed mixed rations, low in neutral detergent fiber (NDF) concentration. Treatments evaluated were: (T1) control, a diet formulated to 25% NDF with 19% NDF from chopped hay (NDFH); (T2) a diet formulated to 35% NDF containing 19% NDFH with the additional 10% NDF provided by a mixture of the three by-product feeds in the concentrate; and (T3) a diet formulated to 25% NDF with 10% NDFH and the remainder provided in the concentrate from by-product feeds. All cows were fed 6% of the total DM as alfalfa hay and pangóla grass hay to meet NDFH targets. Treatment differences in DM intake (18.3 kg/d), DM intake as a percentage of body weight (3.22%), milk production (32.7 kg/d), and milk protein concentration (2.65%) were not significant (P > 0.05). Reducing the NDFH to 10% resulted in a 15% (P < 0.01) relative reduction in milk fat concentration that was not prevented by restoring the NDF level with the mixture of WCS, WM, and rice bran. Despite the fact that cows consumed more energy for milk production (NEL) from T3, the energetic efficiency of milk production (Mcal-milk/Mcal-feed) was about 15% higher (P < 0.01) when the cows consumed 19% NDFH (T1 and T2). Fiber from by-product feeds was ineffective in maintaining milk fat concentration above 3.0% and did not result in improvement in milk production or efficiency of milk production. Thus, the NDF in the mixture of by-product feeds had no ruminally effective fiber value under the conditions of this experiment.

To determine the effect of energy levels on digestible nutrient, milk production and milk quality of 7 months pregnant Bali cattle, was the purpose of this study. The study was conducted in Bali, Province of Indonesia on 12 pregnant breeding phase of pre-calving (2 months before the birth) with the parent body weight 329-340 kg/head. The treatment given is four types of Metabolizable Energy (ME) levels: 2000, 2100, 2200 and 2300/kg respectively as treatment A, B, C, and D. All ration contain 10% of crude protein. Variables measured: energy intake, digestible nutrient, milk yield, and milk quality. This research is a randomized block design. The results showed that increase energy ration until 2300 kcal ME/kg would significantly (P<0.05) increase energy intake and highest at cattle consumed ratio D is 22239.55 kcal/day. However, digestible nutrient was not affected. Milk production increased with increasing energy rations and highest (P<0.05) at cattle received treatment D is 2179.83 ml/day compared to treatment A 936.67 ml/day. Milk fat and milk lactose also highest (P<0.05) in treatment D are 8.56% and 4.76% respectively. Based on these results, it can be concluded that increase energy ration will increase energy intake, milk yield and milk fat and milk lactose of Bali cattle.

An On-Farm Trial was conducted on 24 lactating crossbred cows for assessment of feeding formaldehyde treated mustard cake (bypass protein) on milk production and economic analysis of lactating cow. Cows were divided into three groups having 8 cows each, treatments were farmers’ practice (FP); (Control): The lactating animals under this group were fedas per the feeding schedule of the farmers (5 kg. dry roughage as rice straw + 6 hrs grazing as local grass and 4 kg. commercial concentrates), T1: The lactating animals under this group were fed as per farmers practicewith 12% mustard cake of total diet was provided to the cow by replacing the same amount of commercial concentrates andT2: The lactating animals under this group were fed as per farmers practicewith 12% formaldehyde treated mustard cake of total diet was provided to the cow by replacing same amount of commercial concentrates. The average daily milk yield of lactating cows under FP, T1 and T2 was 8.58, 8.82 and 9.85 kg per cow, respectively. Differences between FP and T2 were significant. The daily increase in milk yield was 1.27 kg and 1.03 kg in cows fed T2diet over the cows fed FP and T1diet, respectively. The B: C ratios for FP, T1 and T2 groups were 2.6, 3.0 and 3.3, respectively. The feed cost reduced in T2 group by Rs. 8.64 and increased milk production by 1.27 kg in respect to FP group.

This Research Communication describes the association between genetic variation within the prolactin (PRL) gene and the milk production traits of Italian Mediterranean river buffalo (Bufala mediterranea Italiana). High resolution melting (HRM) techniques were developed for genotyping 465 buffaloes. The association of genetic polymorphism with milk production traits was performed and subsequently the effects of parity and calving season were evaluated. Single nucleotide polymorphisms (SNPs) were identified at exons 2 and 5 and at introns 1 and 2. All the SNPs were in Hardy–Weinberg equilibrium, and statistical analysis showed that the polymorphism of intron1 was significantly (P < 0·05) associated with milk yield, milk protein content and peak milk yield. The average contribution of the intron1 genotype (r2intron1) to total phenotypic variance in milk production traits was 0·09, and the TT genotype showed lower values than CC and CT genotypes. A nonsynonymous SNP was identified in exon 2, which resulted in an amino acid change from arginine to cysteine. Moreover, the polymorphism of exon 2 was associated significantly with milk fat content (P < 0·05), and the buffaloes with TT genotype showed higher total fat content than the buffaloes with CT genotype. These findings provide evidence that polymorphisms of the buffalo PRL gene are associated with milk production traits and PRL can be used as a candidate gene for marker-assisted selection in Italian Mediterranean river buffalo breeding.

This study aims to characterize the influence of short-term heat stress (HS; 4 day) in early lactating Holstein dairy cows, in terms of triggering blood metabolomics and parameters, milk yield and composition, and milk microRNA expression. Eight cows (milk yield = 30 ± 1.5 kg/day, parity = 1.09 ± 0.05) were homogeneously housed in environmentally controlled chambers, assigned into two groups with respect to the temperature humidity index (THI) at two distinct levels: approximately ~71 (low-temperature, low-humidity; LTLH) and ~86 (high-temperature, high-humidity; HTHH). Average feed intake (FI) dropped about 10 kg in the HTHH group, compared with the LTLH group (p = 0.001), whereas water intake was only numerically higher (p = 0.183) in the HTHH group than in the LTLH group. Physiological parameters, including rectal temperature (p = 0.001) and heart rate (p = 0.038), were significantly higher in the HTHH group than in the LTLH group. Plasma cortisol and haptoglobin were higher (p < 0.05) in the HTHH group, compared to the LTLH group. Milk yield, milk fat yield, 3.5% fat-corrected milk (FCM), and energy-corrected milk (ECM) were lower (p < 0.05) in the HTHH group than in the LTLH group. Higher relative expression of milk miRNA-216 was observed in the HTHH group (p < 0.05). Valine, isoleucine, methionine, phenylalanine, tyrosine, tryptophan, lactic acid, 3-phenylpropionic acid, 1,5-anhydro-D-sorbitol, myo-inositol, and urea were decreased (p < 0.05). These results suggest that early lactating cows are more vulnerable to short-term (4 day) high THI levels—that is, HTHH conditions—compared with LTLH, considering the enormous negative effects observed in measured blood metabolomics and parameters, milk yield and compositions, and milk miRNA-216 expression.

 This research aims to assess the performances of lactating Bedouin goat under arid conditions, focusing on the relationship of milk yield with body and udder morphology and growth traits. Fourteen goats and their kids were followed during early lactation (90 days post-partum). Mean daily milk yield was 0.56 kg. Milk fat, proteins, lactose, solids non-fat and minerals were respectively 34.9, 38.9, 48.8, 107 and 6.7 g L-1. Mean kids’ birth weight, adjusted body weight at 90 days of age and average daily gain were respectively 2 kg, 6.6 kg and 51.4 g day-1. Daily milk yield was correlated with goats’ body weight, withers height, rump height, udder width and udder circumference but not with linear udder score and growth performances. These results provide important data about the milk production of Bedouin goat emphasizing its correlation with body and udder morphological traits which can help to elaborate a specific breeding program for this breed.

The effect of substituting peas for soybean and canola meals as a protein source in a high-producing dairy herd was studied in 66 Holstein cows, divided into two groups based on stage of lactation, parity, level of milk production and days in milk. Two 18.5% crude protein grain concentrate diets were formulated based on the nutrient analyses of the forages available. The control grain mix contained standard protein sources, principally soybean and canola meal (SBM\CM) while the test grain mix was formulated to contain approximately 25% field peas as the major source of protein. Both grain rations were formulated to the same nutrient specifications and balanced for undegradable protein. The duration of the trial was 6 mo during which grain feeding levels were adjusted monthly based on milk yield. For cows in early lactation, 4% fat-corrected milk yield was higher (P < 0.05) for cows fed pea based concentrates (31.3 kg d−1) than for cows fed SBM\CM supplement (29.7 kg d−1). Fat-corrected milk yield was not affected by source of protein in mid- and late-lactation cows. Fat-corrected milk production was not different (P > 0.05) for cows fed SBM\CM compared with cows fed the pea supplement when cows across all stages of lactation were included in the analyses. Milk fat percent was significantly higher (P < 0.05) for early- and mid-lactation cows fed the pea supplement. The results suggest that peas can be substituted for SBM\CM as a protein source for high-producing dairy cows. SummaryTwenty-four Friesian cows were allocated to one of four diets for weeks 3–14 of lactation following 2 weeks on a common diet. The diets (kg air-dry feed/day) were 7–2 kg hay and 10·8 kg either starchy or fibrous concentrates (60S and 60F) or 3·5 kg hay and 14·0 kg either starchy or fibrous concentrates (80S and 80F). Rumen samples were taken by stomach tube in weeks 10 and 12 of lactation and the digestibility of the diets was measured with four cows per treatment during weeks 13 and 14. The digestibility of the same feeds was also measured in sheep at maintenance.The principal carbohydrate constituents of the concentrates were barley, wheat and cassava in the starchy concentrates and citrus pulp, sugar-beet pulp and wheat feed in the fibrous concentrates. The concentrates were designed to have similar concentrations of metabolizable energy (ME) and the diets were planned to provide similar intakes of digestible energy and crude protein.Milk yield and composition were very similar for treatments 60S and 60F. With the higher proportion of starchy concentrates (80S), milk yield was about 20% greater than on 60S, fat concentration fell severely but protein and lactose concentrations were unaffected. With the higher proportion of fibrous concentrates (80F), milk yield and the protein and lactose concentration were similar to values on 60F but fat concentration was lower, though not nearly so low as on 80S. Milk energy yield was reduced by the higher proportion of concentrates but was unaffected by type of carbohydrates. Live-weight changes were small.In both the sheep, consuming at maintenance, and the lactating cows consuming at about 3 times maintenance, digestibility of dry matter, organic matter and energy was higher with the higher concentrate diets but was unaffected by type of concentrate. The digestibility of fibre was greater with the fibrous concentrates but the effect of level of concentrate inclusion was inconsistent. Digestibility coefficients were consistently lower for the lactating cows than for the sheep.The proportion of acetic acid in the rumen volatile fatty acids in the cows was higher and the proportion of propionic acid was lower with the fibrous concentrates. The differences were much greater with the higher proportion of concentrates.During weeks 15–22 of lactation the cows were reallocated to concentrate treatments and given hay ad libitum. Hay intake was about 1 kg/day higher with the fibrous concentrates but the difference was not significant. Hay intake fell by about 0·6 kg/kg concentrate intake for both concentrate types. No significant differences in milk yield or composition were established, probably because of incomplete adaptation even after 8 weeks.It is concluded that at concentrate intakes of about 10 kg/day, the source of carbohydrate in the concentrates has little effect on milk production when the concentrates are of similar ME concentration. However, at higher levels of concentrate inclusion, although the diets may have similar ME concentrations, important differences in the yields of fat, protein and lactose occur due to carbohydrate source and these can be related to differences in rumen fermentation.

AbstractTwenty-eight Holstein/Friesian dairy cows were divided into four groups of seven. From weeks 4 to 15 of lactation they were given a basal diet consisting of 8 kg hay, 2 kg sugar-beet feed and 2 kg grass nuts, together with a concentrate allowance of 8 kg/day. Concentrates for group A were based on cereals and soya (control). Concentrate B contained 60 g protected fat supplement per kg; concentrate C contained 100 g lactose per kg; concentrate D contained 60 g fat supplement and 100 g lactose per kg. Milk yields were 24·6, 27·7, 25·6 and 26·5 kg/day and milk protein concentrations were 32·3, 30·7, 32·7 and 31·9 g/kg for groups A, B, C and D respectively. The effect of fat supplementation on milk yield and protein concentration was significant (P < 0·05) but the effect of lactose was not significant. Milk fat concentration was not significantly affected by treatment. It is concluded that lactose can partially alleviate the depression in milk protein concentration often observed when cows are given protected fat.

Lactation persistency and milk production are among the most economically important traits in the dairy industry. In this study, we explored the association of over 6.1 million imputed whole-genome sequence variants with lactation persistency (LP), milk yield (MILK), fat yield (FAT), fat percentage (FAT%), protein yield (PROT), and protein percentage (PROT%) in North American Holstein cattle. We identified 49, 3991, 2607, 4459, 805, and 5519 SNPs significantly associated with LP, MILK, FAT, FAT%, PROT, and PROT%, respectively. Various known associations were confirmed while several novel candidate genes were also revealed, including ARHGAP35, NPAS1, TMEM160, ZC3H4, SAE1, ZMIZ1, PPIF, LDB2, ABI3, SERPINB6, and SERPINB9 for LP; NIM1K, ZNF131, GABRG1, GABRA2, DCHS1, and SPIDR for MILK; NR6A1, OLFML2A, EXT2, POLD1, GOT1, and ETV6 for FAT; DPP6, LRRC26, and the KCN gene family for FAT%; CDC14A, RTCA, HSTN, and ODAM for PROT; and HERC3, HERC5, LALBA, CCL28, and NEURL1 for PROT%. Most of these genes are involved in relevant gene ontology (GO) terms such as fatty acid homeostasis, transporter regulator activity, response to progesterone and estradiol, response to steroid hormones, and lactation. The significant genomic regions found contribute to a better understanding of the molecular mechanisms related to LP and milk production in North American Holstein cattle.
