AbstractThe B-class of MADS-box transcription factors has been studied in many plant species, but remain functionally uncharacterized in the Rosaceae family. APETALA3 (AP3), a member of this class, controls the identity of petals and stamens in Arabidopsis thaliana. In this work, we identified two members of the AP3 lineage in the cultivated strawberry (Fragaria × ananassa): FaAP3 and FaTM6. Interestingly, FaTM6, and not FaAP3, shows an expression pattern equivalent to that of AP3 in Arabidopsis. Genome editing using Cluster Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas9 system is becoming a robust tool for targeted and stable mutagenesis of DNA. However, whether it can be efficiently used in an octoploid species such as F. × ananassa is not known. Here we report for the first time the application of CRISPR/Cas9 in F. × ananassa to characterize the function of FaTM6 in flower development. An exhaustive analysis by high-throughput sequencing of the FaTM6 locus spanning the target sites showed a high efficiency genome editing already in the T0 generation. The phenotypic characterization of the mutant lines indicates that FaTM6 plays a key role in petal and especially in anther development in strawberry. in an octoploid species such as F. × ananassa, and offer new opportunities for engineering strawberry to improve traits of interest in breeding programs.

Abstract: The objective of this work was to phenotypically characterize creole goats of the Moxotó, Azul, Canindé, Repartida, Marota, and Graúna breeds, comparing them with the Boer, Anglo Nubian, and Alpine exotic breeds, to identify the genetic groups of greater potential for carcass quality and resistance to infection caused by worm. Data on morphometric measurements were obtained for the count of worm eggs in grams of feces (WEGF), Famacha score, carcass measurements, and body weight from 308 animals, in a farm in the state of Piauí, Brazil. Repartida goats showed the highest average for wither height (61.55 cm) and rump height (62.16 cm), and Moxotó goats had the greatest ear length (13.45 cm). The Moxotó breed showed the highest means for carcass and body weight. The lowest average for body weight was observed in Azul goats (24.35 kg), and the lowest WEGF was detected in Repartida goats (200 eggs per gram of feces). The average Famacha score was 2.77. Average linkage was the method that best summarized the information on the morphometric and carcass data. The use of morphometric and carcass measurements provides satisfactory results in the phenotypic characterization of the animals. The Brazilian creole goats, which are considered resistant to worm infection and show high-quality carcass traits, may be indicated for conservation and genetic breeding programs.

AbstractJatropha curcas (jatropha) is a multipurpose plant with potential as a raw material for biofuel. In the present study, a total of 43,349 expressed sequence tags (ESTs) from J. curcas were searched for type and frequency of simple sequence repeat (SSR) markers. Five thousand one hundred and seventy-five sequences were indentified to contain 6,108 SSRs with 90.8% simple and 9.2% compound repeat motifs. One hundred and sixty-three EST-SSRs were developed and used to evaluate the transferability and genetic relatedness among 4 accessions of J. curcas from China, Mexico, Thailand and Vietnam; 5 accessions of congeneric species, viz. J. gossypiifolia, dwarf J. integerrima, normal J. integerrima, J. multifida, J. podagrica; and Ricinus communis. The polymorphic markers showed 75.56–85.19% transferability among four species of Jatropha and 26.67% transferability across genera in Ricinus communis. Investigation of genetic relatedness showed that J. curcas and J. integerrima are closely related. EST-SSRs used in this study demonstrate a high efficiency of cross species/genera amplification and are useful for identifying genetic diversity of jatropha and its close taxa and to choose the desired related species for wide crossing to improve new varieties of jatropha. The markers can also be further exploited for genetic resource management and genetic improvement of related species/genera through marker-assisted breeding programs.

High temperatures can negatively affect lettuce production by promoting early bolting, which leads to increased levels of latex accumulation in the leaves, causing them to become bitter. The lack of adaptation of this culture to such conditions made it an object of study for plant breeding programs, resulting in well succeeded studies. However, little is known about the genes that regulate lettuce flowering. A better understanding of the complex genic interactions involved in the process of lettuce floral initiation is of great importance, since it can enable the development of late-bolting cultivars through plant genetic transformation. MADS-box transcriptional factors are key flowering regulators and have been extensively studied during the flowering process in several species. Thus, this study aimed to identify and characterize the Lettuce MADS-box gene family through the use of bioinformatics tools. The computational analysis consisted in gene prediction, alignment, and phylogenetic analysis. 91 sequences of putative MADS-box genes were identified and characterized by a phylogenetic study of 20 MADS-box genes. Future studies comprising mutants for these genes in plant model species and in lettuce will enable a better understanding of the functions performed by these genes during lettuce flowering, as well as, a better comprehension of this process.

Across the three domains of life, B-family DNA polymerases play a variety of roles in both DNA repair and DNA replication processes. We examine the phenotypic consequences of loss of the putative repair polymerases PolB2 and/or PolB3 in the crenarchaeon Sulfolobus islandicus. We detect a modest growth advantage when cells lacking the polymerase are grown in unperturbed conditions. Further, we observe a striking insensitivity of the mutant lines to acute treatment with the oxidizing agent, hydrogen peroxide. In addition, cells lacking PolB3 show enhanced sensitivity to the DNA damaging agent 4-NQO. Our data therefore suggest that these non-essential DNA polymerases may influence DNA repair pathway choice in these hyperthermophilic aerobes.

Bronchoscopes have been linked to outbreaks of nosocomial infections. The phenotypic and genomic profiles of bronchoscope-associated Klebsiella aerogenes isolates are largely unknown. In this work, a total of 358 isolates and 13 isolates were recovered from samples after clinical procedures and samples after decontamination procedures, respectively, over the five months. Antimicrobial susceptibility testing found seven K. aerogenes isolates exhibiting a low-level resistance to antimicrobial agents. Among seven K. aerogenes isolates, we found five sequence types (STs) clustered into three main clades. Collectively, this study described for the first time the phenotypic and genomic characteristics of bronchoscope-associated K. aerogenes.
