AbstractPlant species have repeatedly evolved recognition systems between pollen and pistils that identify and reject inappropriate matings. Two of the most important systems recognize self-pollen and interspecific pollen. Outstanding questions are whether and how these two recognition systems are linked and if this association could constrain the evolution of mate choice. Our study characterizes variation in self and interspecific incompatibility in the native range of the Texas wildflower Phlox drummondii. We found quantitative variation in self-incompatibility and demonstrate that this variation is significantly correlated with variation in incompatibility with its close congener P. cuspidata. Furthermore, we find strong evidence that self and interspecific incompatibility involve common mechanisms of pollen adhesion or early pollen-tube germination. Finally, we show that P. drummondii populations that co-occur and hybridize with P. cuspidata have significantly higher interspecific incompatibility and self-incompatibility than isolated P. drummondii populations. This geographic variation suggests that the evolution of self-compatibility is constrained by selection favoring interspecific incompatibility to reduce maladaptive hybridization. To our knowledge this is the strongest evidence that a correlation between variation in self and interspecific incompatibilities could influence the evolution of pollen recognition across the range of a species.

Pollen tube growth after selfing was studied in four almond (Prunus amygdalus Batsch) families derived from crosses between self-compatible `Tuono' and self-incompatible `Ferragnès' and `Ferralise' in both directions, in order to ascertain the phenotypic expressions of the different genotypes. A differential expression of self-compatibility was observed in the seedlings of the different families. The genetic self-compatible offspring of `Ferralise' showed a lower percentage of pistils with pollen tubes at the style base and a lower number of pollen tubes at the pistil base after self-pollination than those observed in the self-compatible offspring of `Ferragnès'. This low level of self-compatibility expression observed in some `Ferralise' seedlings may be due to the inbreeding present in `Ferralise'. As a consequence, caution must be taken in almond breeding to avoid the increase of inbreeding by the utilization of related parents and to diversify the sources of self-compatibility, at present mostly limited to `Tuono.'

The ‘pollen test’ and ‘fruit set test’ following controlled crossing combinations of parents are the most commonly used methods for pollination incompatibility studies in Olea europaea L. Self-incompatibility (SI), with diagnoses based on the pollen test and pollen germination, indicating self-compatibility, is not always followed by fruit set in this species. To solve this dispute, we have reconciled all observations into a new model. Mismatches between field and laboratory data and between methods are resolved by the dual-successive-screen model (DSSM) supposing two different loci for the expression of the two SI mechanisms. Pollen/stigma is controlled by diallelic SI, or DSI, inferring two G1 and G2 compatibility/incompatibility (C/I) groups for varieties, whereas pollen tubes in ovaries are controlled by poly-allelic SI or PASI with twenty C/I groups. To explain the selfing of varieties, we have suggested that some determinants in the pollen tube and stigma are unstable and degrade (DS-D for degradation of S-determinant) after three to five days, enabling some pollen tubes to avoid being rejected, hence reaching ovules. DSI and PASI in the DSSM and DS-D mechanisms, plus the andromonoecy of the olive tree, complexify SI studies. Inferences from DSSM and DS-D mechanisms in olive orchard practice are detailed.

Pollen tube growth was studied in 10 almond [Prunus amygdalus Batsch, syn. P. dulcis (Mill.) D.A. Webb] selections of the Zaragoza breeding program, whose main objective is the development of self-compatible cultivars. Self-compatibility was evident in eight of the selections, as indicated by the fact that pollen tube growth was similar following self- and cross-pollination. In the other two selections, pollen tube growth differed following self- and cross-pollination, one showing self-incompatibility and the other an irregular progression of crossed pollen tubes. The importance of the style in sustaining pollen tube growth was evident, and pollen tube growth was influenced by style type.

AbstractPotato is the third most important staple food crop. To address challenges associated with global food security, a hybrid potato breeding system, aimed at converting potato from a tuber-propagated tetraploid crop into a seed-propagated diploid crop through crossing inbred lines, is under development. However, given that most diploid potatoes are self-incompatible, this represents a major obstacle which needs to be addressed in order to develop inbred lines. Here, we report on a self-compatible diploid potato, RH89-039-16 (RH), which can efficiently induce a mating transition from self-incompatibility to self-compatibility, when crossed to self-incompatible lines. We identify the S-locusinhibitor (Sli) gene in RH, capable of interacting with multiple allelic variants of the pistil-specific S-ribonucleases (S-RNases). Further, Sli gene functions like a general S-RNase inhibitor, to impart SC to RH and other self-incompatible potatoes. Discovery of Sli now offers a path forward for the diploid hybrid breeding program.

Self-(in)compatibility was tested in 40 new apricot cultivars from European breeding programmes. Pollen-tube growth in pistils from laboratory pollinations was analysed using the fluorescence microscopy. Cultivars were considered self-compatible if at least one pollen tube reached the ovary in the majority of pistils. Cultivars were considered self- incompatible if the growth of pollen tubes in the style stopped along with formation of characteristic swellings. Of the examined cultivars, 18 were self-compatible and 22 were self-incompatible. Fluorescence microscopy provides a relatively rapid and reliable method to determine self-incompatibility in apricot cultivars.

The reproductive biology of exotic species affects their capacity to become naturalised and invasive in non-native areas. Selfing is a common trait in many invasive plants probably because it provides reproductive assurance under low availability of pollination vectors and sexual partners. Nonetheless, the predominantly self-incompatible Australian Acacia species are among the most aggressive plants worldwide. To address whether there have been changes in selfing ability and natural reproductive success of A. longifolia during invasion, we compared one population in the invaded area (Portugal) with one population in the native range (Australia). We specifically assessed floral traits, fruit set and offspring traits for selfing and open-pollination treatments. Within each pollination treatment, no differences were found between areas, suggesting that the level of self-compatibility has not changed during invasion. However, the number of aborted seeds and seed size were significantly different between pollination treatments in Australia but not in Portugal. There were significant differences in the number of seeds per pod and in seed weight between ranges. A lower number of aborted seeds, a higher number of fully developed seeds and a greater seed size were found in the invaded area for both pollination treatments. In spite of the low selfing ability of A. longifolia in the invaded area, there was an increase in the quantity and size of the seeds produced in the new region, even for self-pollinated fruits, which might contribute to A. longifolia invasiveness.

The experimental results obtained show that the tip of the incompatible pollen tube bursts open after the outer-wall has considerably expanded in the intercellular spaces of the conducting tissue and the inner-wall has disappeared and numerous particles have accumulated in the tube cytoplasm. These particles, which measure approximately 0.2 µm in diameter and give a weak reaction to the test of Thiéry, differ in many respects from the vesicles normally present in compatible pollen tubes growing through the style; they appear to resemble, in some cases, the spheres which are discharged by the compatible pollen tubes after they have reached the embryo-sac. It is considered that these observations support the current belief that the tube wall is the site of action for the incompatibility proteins and suggest that self-incompatibility is not a passive process resulting from lack of growth stimulation but an active event which leads to the destruction of the incompatible pollen tubes. The degradation mechanism involved appears similar to the one which enables the compatible pollen tube to release its contents in the degenerated synergid and presents some analogies with the lytic process taking place in virus-infected cells. The general hypothesis is presented that the particles observed in the cytoplasm of self-incompatible pollen tubes consist of a mixture of incompatibility proteins and of basic constituents of the tube wall.
