Factors affecting the management of petunia and tomato late blight, caused by Phytophthora infestans, under greenhouse conditions were investigated. Late blight-infected petunias (Petunia × hybrida) and tomatoes (Lycopersicon esculentum) each produced sporangia that were dispersed throughout the greenhouse via air currents. Infected petunias produced and released fewer sporangia than infected tomatoes, but infected petunias released sporangia two times longer. Surface-directed irrigation reduced disease incidence compared with overhead irrigation that wetted the foliage. The fungicides dimethomorph-mancozeb, fosetyl-Al, azoxystrobin, and dipo-tassium phosphonate/phosphate suppressed late blight development, as did the plant defense activator acibenzolar-S-methyl. All products were applied twice at 7-day intervals. The other plant defense activator (harpin protein) and the bioantagonists (Trichoderma harzianum, Glio-cladium virens, and Bacillus subtilis) were ineffective at the rates tested.

A single lime application of 5.4 t/ha per 20 cm increment of soil to a depth of 60 cm controlled bacterial wilt (Psendomonas solanacearum E. Smith) up to 2 years in tomato (Lycopersicon esculentum Mill.) growing in a naturally infested soil. `Sunny' with 64% disease incidence, was the most susceptible, `Capitan' (20%) was intermediate, and `Caraibo' (5%) and `Hawaii 7997' (2%) were the least susceptible. Incorporation of lime 40 to 60 cm deep reduced bacterial wilt incidence in 'Capitan' by 75%, and 60% in `Sunny'. The number of bacteria in the soil was not affected by lime treatment. Soil pH and total Caincreased linearly with time and with depth of lime incorporation. Bacterial wilt control in `Sunny' and `Capitan' was associated with an increase in pH. Liming to 40 or 60 cm generally increased leaf Ca and decreased Mn, Zn, and Cu. `Hawaii 7997' contained significantly higher concentrations of Ca, Mg, and K, and lower Cu compared to 'Sunny'. `Hawaii 7997' produced significantly higher yields than the other cultivars. Deep lime incorporation increased fruit weight of `Sunny' by 184% and 'Capitan' by 79% as compared with 0 lime.

Integrated commercial blue-green algae extracts and bioagents treatments against vegetables root rot incidence when used as soil drench under greenhouse and plastic house conditions were evaluated. All applied treatments reduced significantly root rot incidence at both pre- and postemergence growth stages of cucumber, cantaloupe, tomato, and pepper plants compared with untreated check control. In pot experiment, the obtained results showed that treatments ofTrichoderma harzianumorBacillus subtiliseither alone or combined with commercial algae extracts were significantly superior for reducing root rot disease for two tested vegetable plants compared with the other tested treatments as well as control. It is also observed that rising concentrations of either algae products, Oligo-X or Weed-Max, were reflected in more disease reduction. Promising treatments for controlling root rot disease incidence were applied under plastic houses conditions. As for field trails carried out under plastic houses conditions at different locations, the obtained results revealed that the applied combined treatments significantly reduced root rot incidence compared with fungicide and check control treatments. At all locations it was observed that Weed-Max (2 g/L) +Bacillus subtilissignificantly reduced disease incidence of grown vegetables compared with Oligo-X (2 mL/L) +Trichoderma harzianumtreatments. An obvious yield increase in all treatments was significantly higher than in the control. Also, the harvested yield in applied combined treatments at all locations was significantly higher than that in the fungicide and control treatments.

Pseudomonas syringae pv. apii, causal agent of bacterial leaf spot (BLS) of celery, was first identified in California in 1989. By 1991, BLS was apparent in all celery-growing areas of the state. Greenhouse-produced transplants were affected most severely, and disease incidence approached 100% in some greenhouses. In this study, sources of inoculum and factors contributing to disease development were investigated in three Salinas Valley greenhouse operations during the 1991, 1992, and 1993 celery transplant seasons (January to August). Epiphytic P. syringae pv. apii was not detected on celery transplants until April or May of each year. Increased epiphytic populations preceded BLS outbreaks, and high-pressure, overhead irrigation favored bacterial infiltration and disease development. In seed-wash assays, P. syringae pv. apii was recovered from 5 of 24 commercial celery seed lots. In field tests, epiphytic P. syringae pv. apii was found on umbels of inoculated celery plants, and seeds from these plants were heavily contaminated with P. syringae pv. apii. Contaminated seed produced seedlings with large epiphytic P. syringae pv. apii populations. Hot-water treatment (50°C for 25 min) eliminated >99.9% of seed contamination. Based on these results, disease management techniques are proposed.

During polymer flooding, certain amount of polymer would be lost. Polymer retention causes sweep volume expanding on one side, it also causes polymer loss on the other. Therefore, it is a very important topic to study the influencing factors of polymer retention. There are many factors affecting polymer retention process. This paper mainly studied the influence from dynamic factors such as polymer solution concentration, injection rate, injection time, injected pv number. This paper investigated the influence of these factors on polymer retention process, and optimized these factors to minimize polymer loss in reservoir.

A standard sand – cornmeal – nutrient salt medium, for use in the production of artificial inoculum of Helminthosporium sativum, is described. This inoculum induces uniform plant disease development when thoroughly incorporated with the planting soil. The results presented clearly demonstrate the necessity of using a series of infestation levels in studies of factors affecting disease development in artificially infested soil. It is shown that considerable reliance can be placed on treatment comparisons within an experiment but that comparisons between experiments are much less accurate. The role of a toxin (or toxins) in disease development in barley seedlings has been demonstrated. The toxic activity was found to be distinct from that frequently encountered on addition of organic matter to soil. Results obtained suggest that toxin adsorption by the soil may play an important role in reducing disease incidence and severity.

ABSTRACT The tet(L) gene of Bacillus subtilis confers low-level tetracycline (Tc) resistance. Previous work examining the >20-fold-inducible expression of tet(L) by Tc demonstrated a 12-fold translational induction. Here we show that the other component of tet(L) induction is at the level of mRNA stabilization. Addition of a subinhibitory concentration of Tc results in a two- to threefold increase in tet(L) mRNA stability. Using a plasmid-borne derivative of tet(L) with a large in-frame deletion of the coding sequence, the mechanism of Tc-induced stability was explored by measuring the decay of tet(L) mRNAs carrying specific mutations in the leader region. The results of these experiments, as well as experiments with a B. subtilis strain that is resistant to Tc due to a mutation in the ribosomal S10 protein, suggest different mechanisms for the effects of Tc on translation and on mRNA stability. The key role of the 5" end in determining mRNA stability was confirmed in these experiments. Surprisingly, the stability of several other B. subtilis mRNAs was also induced by Tc, which indicates that addition of Tc may result in a general stabilization of mRNA.

ABSTRACT Daptomycin is the first of a new class of cyclic lipopeptide antibiotics used against multidrug-resistant, gram-positive pathogens. The proposed mechanism of action involves disruption of the functional integrity of the bacterial membrane in a Ca2+-dependent manner. We have used transcriptional profiling to demonstrate that treatment of Bacillus subtilis with daptomycin strongly induces the lia operon including the autoregulatory LiaRS two-component system (homologous to Staphylococcus aureus VraSR). The lia operon protects against daptomycin, and deletion of liaH, encoding a phage-shock protein A (PspA)-like protein, leads to threefold increased susceptibility. Since daptomycin interacts with the membrane, we tested mutants with altered membrane composition for effects on susceptibility. Deletion mutations of mprF (lacking lysyl-phosphatidylglycerol) or des (lipid desaturase) increased daptomycin susceptibility, whereas overexpression of MprF decreased susceptibility. Conversely, depletion of the cell for the anionic lipid phosphatidylglycerol led to increased resistance. Fluorescently labeled daptomycin localized to the septa and in a helical pattern around the cell envelope and was delocalized upon the depletion of phosphatidylglycerol. Together, these results indicate that the daptomycin-Ca2+ complex interacts preferentially with regions enriched in anionic phospholipids and leads to membrane stresses that can be ameliorated by PspA family proteins.

AIM: To assess the changes in anterior chamber parameters and examine the factors associated with changes in the intraocular pressure (IOP) in individuals who have undergone phacoemulsification surgery. METHODS: It is a longitudinal analysis of secondary clinical data collected from 105 non-glaucomatous eyes (82 patients) undergoing a cataract surgery. We studied the association between anterior chamber parameters, grade of cataract, demographics, and changes in the IOP over a period of three weeks. We also evaluated the association between the pressure-depth (PD) ratio and changes in the IOP during this time. RESULTS: The mean age (SD) of the 82 patients was 60.1±7.8y. The mean±standard deviation (SD) IOP was 15.06±3.36 mm Hg pre-operatively; it increased to 15.75±4.21 mm Hg on day one (P=0.20). In the multifactorial models, the mean IOP was -1.715 (95%CI: -2.795, -0.636) mm Hg on day 21±5 compared with the pre-operative values. The anterior chamber depth (ACD), axial length, age, sex, and grade of cataract were not significantly associated with changes in the IOP. Each unit increase in the PD ratio was associated with an increase in the mean IOP by 1.289 mm Hg (95%CI: 0.906, 1.671). After adjusting for pre-operative PD ratio, none of the other variables (ACD, axial length, temporal angle) were significantly associated with changes in mean IOP. CONCLUSION: The PD ratio was the single most important factor associated with the changes in post-operative IOP over three weeks post-surgery.

ABSTRACT The Bacillus subtilis competence transcription factor ComK is required for establishment of competence for genetic transformation. In an attempt to study the ComK factor further, we explored the genes regulated by ComK using the DNA microarray technique. In addition to the genes known to be dependent on ComK for expression, we found many genes or operons whose ComK dependence was not known previously. Among these genes, we confirmed the ComK dependence of 16 genes by using lacZ fusions, and three genes were partially dependent on ComK. Transformation efficiency was significantly reduced in an smf disruption mutant, although disruption of the other ComK-dependent genes did not result in significant decreases in transformation efficiency. Nucleotide sequences similar to that of the ComK box were found for most of the newly discovered genes regulated by ComK.
