Pod necrosis and dieback of terminal portions of stems, commonly called bud blight, are observed in soybean fields and associated with Tobacco streak virus (TSV), an understudied virus of soybean. Furthermore, many TSV-infected plants are asymptomatic. The objectives of this study were to characterize the distribution and seasonal progress of TSV-infected plants in both natural and controlled epidemics, and the agronomic impact of TSV on soybean in plots with controlled introduction of inoculum. Incidence of TSV-positive samples ranged from 17 to 56% in a general survey. In the presence of natural sources of inoculum, the incidence of TSV-infected plants ranged from 10 to 95% depending on cultivar and location, and peaked at growth stage R2, but detection dropped dramatically at R5. During 2001, significant yield loss and incidence of mottled seed were associated with TSV, but results were confounded by a high incidence of SMV. In 2002, SMV was controlled by cultivar selection, and a 25% reduction in grain yield was attributable to TSV. The incidence of mottled seed and green stem syndrome were low in the presence of TSV. Reductions in plant density and final plant height contributed to reduced yields. However, no significant differences were found in seed number per plant and 100-seed weight. Data indicate that plant mortality was the main mechanism by which TSV caused yield loss in induced epidemics.

Tobacco streak virus (TSV) has an extensive plant host range, but until recently has not been a common problem in North American soybean. TSV is associated with bud blight and yield loss due to reduced plant height and density, and delayed seed development and plant maturity. TSV has been reported in recent years in Illinois, Iowa, Kansas, and Wisconsin, as well as Ontario, Canada. Presence of the virus was confirmed by ELISA. In an Iowa field with high incidence, regression analysis indicated a significant positive relationship between incidence and seed moisture. TSV is seed and pollen transmitted, as well as being spread by thrips. Increasingly variable weather creating favorable conditions for thrips may be facilitating the rise of TSV. The combination of increased incidence of TSV with associated potential for yield loss and few known management methods brings attention to the need for further research. The effect of a range of sowing rates on scald infection and yield in barley cv. Stirling was studied to investigate if increased plant density will lead to more ears/m2 and therefore compensate for yield losses due to scald. Increased sowing rate (50 v. 100 kg/ha) did not compensate for yield loss due to scald. However, significant yield increases (3.0 v. 4.0 t/ha) with increased sowing rates (25 v. 125 kg/ha) occurred when scald was controlled. Such yield in creases were accompanied by increases in the number of ears/m2 and number of grains/ha, but decreases in number of grains/ear and 100-grain weight. The severity of scald infection was not affected by sowing rates.

More than 300 red raspberry cultivars and selections were screened for raspberry (Rubus idaeus L.) bushy dwarf virus (RBDV), tobacco streak virus (TSV), and tomato ringspot virus (TomRSV) using enzyme-linked immunosorbent assay in three naturally infected breeding program selection plots at Corvallis, Ore. All genotypes tested negative for TSV and TomRSV. The RBDV incidence in primocane-fruiting cultivars and selections was 67%; in floricane-fruiting genotypes, it was 34%. The pattern of RBDV infection in the field showed no discernible trend. The high incidence may have been due to use of infected parents, propagation of infected genotypes, and pollen transmission. `Willamette', considered to be immune to the common strain of RBDV, along with 14 clones that had been in the field 10 years or longer, tested negative. The high incidence of RBDV in the breeding plots may provide an opportunity to identify resistant parents for breeding programs. An early seedling screening method for RBDV susceptibility is desirable to eliminate highly susceptible genotypes from the program and maintain a lower incidence of RBDV within the breeding plots.

Background: Peanut stem necrosis disease (PSND) caused by Tobacco streak virus (TSV) is a major constraint for groundnut production in Andhra Pradesh (A.P.). However, studies on prevalence and spread of the disease confined to only few districts of A.P. with this background current study focused on incidence and spread of the disease in entire state of A.P. Further an isolate of TSV occurring in A.P. characterized on the basis of genetic features by comparing with other TSV isolates originated from different hosts and locations from world.Methods: Roving survey was conducted during kharif 2017-18 in groundnut growing districts of Andhra Pradesh (A.P.) for peanut stem necrosis disease incidence. Groundnut plants showing PSND symptoms were collected and tested with direct antigen coating enzyme linked immunosorbent assay (DAC-ELISA). Groundnut samples found positive by ELISA once again tested by reverse transcription polymerase chain reaction (RT-PCR). The representative TSV-GN-INDVP groundnut isolate from Prakasham district was maintained on cowpea seedlings by standard sap inoculation method in glasshouse for further molecular characterization. The Phylogenetic tree for coat protein (CP) gene was constructed using aligned sequences with 1000 bootstrap replicates following neighbor-joining phylogeny.Result: Thirty-eight (52.7%) of seventy-two groundnut samples collected from different locations in A.P were given positive reaction to TSV by DAC-ELISA. For the first time, PSND incidence observed in coastal districts (Krishna, Guntur, Sri Pottisriramulu Nellore, Prakasham) of A.P. Maximum PSND incidence recorded from Bathalapalli (22.2%) and the minimum incidence in Mulakalacheruvu (4.1%). The coat protein (CP) gene of TSV-GN-INDVP groundnut isolate was amplified by RT-PCR and it shared maximum per cent nucleotide identity (97.51-98.62%) with TSV isolates from groundnut and other different crops reported in India. All Indian isolates cluster together irrespective of crop and location based on the phylogenetic analysis.

Experiments were conducted in open field to assess the effect of seeding season and density on the yield, the chemical composition and the accumulation of total tannins in grains of two chickpea (<em>Cicer arietinum</em> L.) cultivars (<em>Pascià</em> and <em>Sultano</em>). Environmental conditions and genetic factors considerably affected grain yield, nutrient and total tannins content of chickpea seeds, giving a considerable range in its qualitative characteristics. Results confirmed cultivar selection as a central factor when a late autumn-early winter sowing is performed. In effect, a more marked resistance to Ascochyta blight (AB) of <em>Sultano</em>, allowed better agronomic performances when favourable-to-AB climatic conditions occur. Winter sowing appeared to be the best choice in the Mediterranean environment when cultivating to maximise the grain yield (+19%). Spring sowing improved crude protein (+10%) and crude fibre (+8%) content, whereas it did not significantly affect the accumulation of anti-nutrients compounds such as total tannins. The most appropriate seeding rate was 70 seeds m–2, considering that plant density had relatively little effect on the parameters studied.

ABSTRACTMolecular Marker-based Selection on F2 Progeny for Development of Promising Rice Lines Resistant to Brown PlanthopperBrown planthopper (BPH) is the major insect pest of rice and accounts for significant yield loss. This experiment was aimed to develop BC1F1 and F3 brown planthopper resistant rice lines. Selection on the basis of SSR markers can be done by using two polymorphic SSR markers, i.e., RM586 dan RM8213, which screened from eight SSR markers for BPH resistant. Sixty-three F2 genotypes from IP-1 (derived from IR-64 x PTB-33) population and twenty F2 genotypes from PP-11 (derived from Pandan Wangi x PTB-33) population were selected and will be used for further research by selfed and backcrossed to recipient parents. Chi-squares test for segregation of DNA bands in F2 generation showed that RM8213 fitted with 1:2:1 Mendelian ratio for controlling photosynthetic rates and trichomes length in IP-1 population. This information could be used in programs to develop a durable brown planthopper resistant rice cultivar.Keywords: BPH, F2 population, Moleculer marker, SSRABSTRAKWereng coklat merupakan salah satu hama utama pada tanaman padi yang mampu menurunkan produksi padi secara nyata. Penelitian ini bertujuan untuk memperoleh galur-galur padi F2 yang memiliki marka-marka yang berasosisasi dengan ketahanannya terhadap wereng coklat. Seleksi pada galur padi F2 hasil persilangan telah dilakukan melalui teknik marka molekuler Simple Sequence Repeat (SSR) menggunakan dua marka SSR yang menunjukkan polimorphisme yaitu RM586 dan RM8213 dari delapan marka yang diskrining. Sebanyak 63 genotip dari populasi IP-1 (hasil persilangan IR-64 x PTB-33) dan 20 genotip dari populasi PP-11 (hasil persilangan Pandan Wangi x PTB-33) untuk disilangkan sendiri maupun disilang balik dengan tetua recipient. Selain itu, hasil analisis Chi-Kuadrat untuk segregasi pita DNA menunjukkan bahwa primer RM8213 memiliki rasio 1:2:1 (dominasi tidak sempurna) dalam mengontrol karakter laju fotosintesis dan panjang trikoma terhadap wereng coklat pada populasi IP-1. Informasi yang diperoleh dari penelitian ini nantinya dapat digunakan untuk program perakitan kultivar padi tahan wereng coklat yang durable.Kata Kunci: Marka molekuler, Populasi F2, SSR, Wereng coklat
